Despite the importance of kinases' catalytic activity regulation in cell signaling, detailed mechanisms underlying their activity regulation are poorly understood. Herein, using insulin-like growth factor 1 receptor kinase (IGF-1RK) as a model, the mechanisms of kinase regulation by its activation loop (A-loop) phosphorylation were investigated through molecular dynamics (MD) and alchemical free energy simulations. Characterization of the TPR-MET oncogene p65 and the MET protooncogene p140 protein-tyrosine kinases

The proteins encoded by the human TPR-MET oncogene (p65/sup tpr-met/) and the human MET protooncogene (p140/sup met/) have been identified. The p65/sup tpr-met/ and p140/sup met/, as well as a truncated TPR-MET product expression in Escherichia coli, p50/sup met/, are autophosphorylated in vitro on tyrosine residues. Using the immunocomplex kinase assay, p140/sup met/ activity was detected in various human tumor epithelial cell lines. The Mst–Lats kinase cascade is central to the Hippo tumor-suppressive pathway that controls organ size and tissue homeostasis. The adaptor protein Mob1 promotes Lats activation by Mst, but the mechanism remains unknown. Here, we show that human Mob1 binds to autophosphorylated docking motifs in active Mst2. This binding enables Mob1 phosphorylation by Mst2. Phosphorylated Mob1 undergoes conformational activation and binds to Lats1. We determine the crystal structures of phospho-Mst2–Mob1 and phospho-Mob1–Lats1 complexes, revealing the structural basis of both phosphorylation-dependent binding events.
