High-performance liquid chromatography with electro-chemical detection (HPLCEC) has become well accepted as a sensitive method for determining a wide variety of organic and inorganic compounds. In these laboratories, the authors have been interested in developing analytical methods for trace metal determinations of metals such as Cu, Ni, Cr(III), Cr(VI), Mn, Fe, Co, Hg, and Pb by the HPLCES method in industrial effluents. However, the instrumentation developed in these earlier studies has not been fully automated as required for long-term monitoring. This paper describes the design of a new completely automated HPLEC system for on-line monitoring.

Here we integrate a high-pressure electroosmotic pump (EOP), a nanoflow gradient generator, and a capillary column into a miniaturized liquid chromatographic system that can be directly coupled with a mass spectrometer for proteomic analysis. We have recently developed a low-cost high-pressure EOP capable of generating pressure of tens of thousands psi, ideal for uses in miniaturized HPLC. The pump worked smoothly when it was used for isocratic elutions. When it was used for gradient elutions, generating reproducible gradient profiles was challenging; because the pump rate fluctuated when the pump was used to pump high-content organic solvents. Determination of catechol-O-methyltransferase activity in brain tissue by high-performance liquid chromatography with on-line radiochemical detection

A sensitive assay for catechol-O-methyltransferase (COMT) activity by high-performance liquid chromatography with on-line radiochemical detection was described. The method was based on the measurement of /sup 3/H- labeled 3-O- and 4-O-methylated products of the substrate, 3,4- dihydroxybenzoic acid, using S-adenosyl-L-(methyl-/sup 3/H)methionine as the methyl donor, or the measurement of /sup 14/C-labeled 3-O- and 4-O-methylated products of the substrate, (7-/sup 14/C)dopamine. The reaction products were determined from the incubation mixture after removal of protein by injecting an aliquot into the liquid chromatograph. High-Pressure Open-Channel On-Chip Electroosmotic Pump for Nanoflow High Performance Liquid Chromatography

Here, we construct an open-channel on-chip electroosmotic pump capable of generating pressures up to ~170 bar and flow rates up to ~500 nL/min, adequate for high performance liquid chromatographic (HPLC) separations. A great feature of this pump is that a number of its basic pump units can be connected in series to enhance its pumping power; the output pressure is directly proportional to the number of pump units connected.
