Diatoms are eukaryotic microalgae that contain genes from various sources, including bacteria and the secondary endosymbiotic host. Due to this unique combination of genes, diatoms are taxonomically and functionally distinct from other algae and vascular plants and confer novel metabolic capabilities. Based on the genome annotation, we performed a genome-scale metabolic network reconstruction for the marine diatom Phaeodactylum tricornutum. How diatoms respond to environmental perturbations such as elevated carbon concentrations in the atmosphere is currently poorly understood. Physiological and genetic control mechanisms for plant adaptation to high temperature and elevated CO2

Acclimations of the stomatal response to CO2 were characterized. Stomata from the model plant used, Vicia faba, are very sensitive to ambient CO2 when grown in growth chambers as compared to stomata from green house grown leaves. The different CO2 sensitivities of growth chamber and green house grown guard cells was confirmed by reciprocal transfer experiments. Stomata acclimated to their new environment and acquired the CO2 sensitivity typical of that environment. A mechanism for CO2 sensing was also characterized. The ecological prominence of diatoms in the ocean environment largely results from their superior competitive ability for dissolved nitrate (NO₃^-). To investigate the cellular and genetic basis of diatom NO₃^- assimilation, in this paper we generated a knockout in the nitrate reductase gene (NR-KO) of the model pennate diatom Phaeodactylum tricornutum. In NR-KO cells, N-assimilation was abolished although NO₃^- transport remained intact. Unassimilated NO₃^- accumulated in NR-KO cells, resulting in swelling and associated changes in biochemical composition and physiology.
