Estimating Michaelis-Menten or Langmuir isotherm constants by weighted nonlinear least squares

The authors derive a nonlinear least squares fitting method for determining either Michealis-Menten equation of Langmuir adsorption isotherm constants from experimental data for microbial growth rate. Data points can be weighted unequally if their relative precisions are known. Comparison of this method with the commonly used linearization methods, using simulated data sets containing normally distributed random errors, showed that this method yields more accurate and precise estimates of the constants than any of the linearizations. A short BASIC program to facilitate computation is presented.

In the three-compartment model of transfer of native glucose and (18F)fluorodeoxyglucose (FDG) into brain, both transport across the blood-brain barrier and phosphorylation by hexokinase can be described by the Michaelis-Menten equation. This permits the use of fixed transport (tau = K*1/K1) and phosphorylation (psi = k*3/k3) ratios and a common partition volume (Ve = K1/k2) for tracer and glucose. By substituting transfer constants of FDG for those of glucose, using tau and psi, the lumped constant was determined directly by positron tomography. Non-cyclic Geometric Phase In Stochastic Processes: Corrections To Michaelis-menten Kinetics And Applications To A Cell Growth Model

We generalize the concept of the geometric phase in stochastic kinetics to a noncyclic evolution. Its application is demonstrated on kinetics of the Michaelis-Menten reaction. It is shown that the noncyclic geometric phase is responsible for the correction to the Michaelis-Menten law when parameters, such as a substrate concentration, are changing with time. We also discuss a model, where this correction qualitatively changes the outcome of reaction kinetics.
