CYTOCHEMICAL REACTION FOR DELTA(5)-3 BETA-HYDROXYSTERIOD DEHYDROGENASE WAS STUDIED IN MONOLAYERS OF TESTICULAR CELLS GROWN IN VITRO. CELLS IN CULTURES ESTABLISHED FROM TESTICULAR TISSUE OF IMMATURE RATS WERE ESSENTIALLY NEGATIVE FOR DELTA(5)-3 BETA-HYDROXYSTERIOD DEHYDROGENASE. CELLS IN CULTURES ESTABLISHED FROM TESTICULAR TISSUE OF SIMILAR ANIMALS PRETREATED WITH HUMAN CHORIONIC GONADOTROPIN (HCG) WERE STRONGLY POSITIVE FOR DELTA(5)-3 BETA-HYDROXYSTEROID DEHYDROGENASE. ADDITION OF HCG TO THE CULTURE MEDIUM FAILED TO INDUCE A POSITIVE REACTION FOR THIS ENZYME. THESE STUDIES SHOW THAT TESTICULAR CELLS WHICH MAY BE ACTIVE IN STEROID BIOSYNTHESIS CAN BE GROWN IN VITRO.
