    In 2007, the U.S. Department of Energy joined the California Energy Commission in funding a project to begin to examine the technical, economic and institutional (policy and regulatory) aspects of energy-efficient community development. That research project was known as the Chula Vista Research Project for the host California community that co-sponsored the initiative. The researches proved that the strategic integration of the selected and economically viable buildings energy efficiency (EE) measures, photovoltaics (PV), distributed generation (DG), and district cooling can produce significant reductions in aggregate energy consumption, peak demand and emissions, compared to the developer/builder's proposed baseline approach. However, the central power plant emission reductions achieved through use of the EE-DG option would increase local air emissions. The electric and natural gas utility infrastructure impacts associated with the use of the EE and EE-PV options were deemed relatively insignificant while use of the EE-DG option would result in a significant reduction of necessary electric distribution facilities to serve a large-scale development project. The results of the Chula Vista project are detailed in three separate documents: (1) Energy-Efficient Community Development in California; Chula Vista Research Project report contains a detailed description of the research effort and findings. This includes the methodologies, and tools used and the analysis of the efficiency, economic and emissions impacts of alternative energy technology and community design options for two development sites. Research topics covered included: (a) Energy supply, demand, and control technologies and related strategies for structures; (b) Application of locally available renewable energy resources including solar thermal and PV technology and on-site power generation with heat recovery; (c) Integration of local energy resources into district energy systems and existing energy

    The purpose of this report is to present this Consultant`s findings regarding the nature and extent of the mineral estate of the United States at National Petroleum Reserve No. 2 (NPR-2), Buena Vista Hills Field, Kern County, California. Determination of the mineral estate is a necessary prerequisite to this Consultant`s calculation of estimated future cash flows attributable to said estate, which calculations are presented in the accompanying report entitled ``Phase II Final Report, Study of Alternatives for Future Operations of the Naval Petroleum and Oil Shale Reserves, NPR-2, California.`` This Report contains a discussion of the leases in effect at NPR-2 and subsequent contracts affecting such leases. This Report also summarizes discrepancies found between the current royalty calculation procedures utilized at NPR-2 and those procedures required under applicable agreements and regulations. Recommendations for maximizing the government`s income stream at NPR-2 are discussed in the concluding section of this Report.

    Aligning large vertebrate genomes that are structurally complex poses a variety of problems not encountered on smaller scales. Such genomes are rich in repetitive elements and contain multiple segmental duplications, which increases the difficulty of identifying true orthologous SNA segments in alignments. The sizes of the sequences make many alignment algorithms designed for comparing single proteins extremely inefficient when processing large genomic intervals. We integrated both local and global alignment tools and developed a suitemore » of programs for automatically aligning large vertebrate genomes and identifying conserved non-coding regions in the alignments. Our method uses the BLAT local alignment program to find anchors on the base genome to identify regions of possible homology for a query sequence. These regions are postprocessed to find the best candidates which are then globally aligned using the AVID global alignment program. In the last step conserved non-coding segments are identified using VISTA. Our methods are fast and the resulting alignments exhibit a high degree of sensitivity, covering more than 90% of known coding exons in the human genome. The GenomeVISTA software is a suite of Perl programs that is built on a MySQL database platform. The scheduler gets control data from the database, builds a queve of jobs, and dispatches them to a PC cluster for execution. The main program, running on each node of the cluster, processes individual sequences. A Perl library acts as an interface between the database and the above programs. The use of a separate library allows the programs to function independently of the database schema. The library also improves on the standard Perl MySQL database interfere package by providing auto-reconnect functionality and improved error handling.« less

    Comparison of DNA sequences from different species is a fundamental method for identifying functional elements in genomes. Here we describe the VISTA family of tools created to assist biologists in carrying out this task. Our first VISTA server at http://www-gsd.lbl.gov/VISTA/ was launched in the summer of 2000 and was designed to align long genomic sequences and visualize these alignments with associated functional annotations. Currently the VISTA site includes multiple comparative genomics tools and provides users with rich capabilities to browse pre-computed whole-genome alignments of large vertebrate genomes and other groups of organisms with VISTA Browser, submit their own sequences of interest to several VISTA servers for various types of comparative analysis, and obtain detailed comparative analysis results for a set of cardiovascular genes. We illustrate capabilities of the VISTA site by the analysis of a 180 kilobase (kb) interval on human chromosome 5 that encodes for the kinesin family member3A (KIF3A) protein.

    VISTA is a comprehensive suite of programs and databases developed by and hosted at the Genomics Division of Lawrence Berkeley National Laboratory. They provide information and tools designed to facilitate comparative analysis of genomic sequences. Users have two ways to interact with the suite of applications at the VISTA portal. They can submit their own sequences and alignments for analysis (VISTA servers) or examine pre-computed whole-genome alignments of different species. A key menu option is the Enhancer Browser and Database at http://enhancer.lbl.gov/. The VISTA Enhancer Browser is a central resource for experimentally validated human noncoding fragments with gene enhancer activity as assessed in transgenic mice. Most of these noncoding elements were selected for testing based on their extreme conservation with other vertebrates. The results of this enhancer screen are provided through this publicly available website. The browser also features relevant results by external contributors and a large collection of additional genome-wide conserved noncoding elements which are candidate enhancer sequences. The LBL developers invite external groups to submit computational predictions of developmental enhancers. As of 10/19/2009 the database contains information on 1109 in vivo tested elements - 508 elements with enhancer activity.

    SuperCDMS (Super Cryogenic Dark Matter Search) is a leading direct dark mat-ter search experiment which uses solid state detectors (Ge crystals) at milliKelvintemperatures to look for nuclear recoils caused by dark matter interactions in the de-tector. `Weakly Interacting Massive Particles' (WIMPs) are the most favoured darkmatter candidate particles. SuperCDMS, like many other direct dark matter searchexperiments, primarily looks for WIMPs. The measurement of both the ionizationand the lattice vibration (phonon) signals from an interaction in the detector allow itto discriminate against electron recoils which are the main source of background forWIMP detection.SuperCDMS currently operates about 9 kg of Ge detectors at the Soudan under-ground lab in northern Minnesota. In its next phase, SuperCDMS SNOLAB plansto use 100-200 kg of target mass (Ge) which would allow it to probe more of theinteresting and and as of yet unexplored parameter space for WIMPs predicted bytheoretical models. The SuperCDMS Queen's Test Facility is a detector test facilitywhich is intended to serve as detector testing and detector research and developmentpurposes for the SuperCDMS experiment.A modifed detector called the HiZIP (Half-iZIP), which is reduced in complex-ity in comparison to the currently used iZIP (interleaved Z-sensitive Ionization and Phonon mediated) detectors, is studied in this thesis. The HiZIP detector designalso serves to discriminate against background from multiple scatter events occurringclose to the surfaces in a single detector. Studies carried out to compare the surfaceevent leakage in the HiZIP detector using limited information from iZIP data takenat SuperCDMS test facility at UC Berkley produce a highly conservative upper limitof 5 out of 10,000 events at 90% condence level.     A series of integral effect tests (IETs) was performed by the Korea Atomic Energy Research Inst. (KAERI) using the VISTA integral test loop (VISTA-ITL) as a small-scale IET program. Among them this paper presents major results acquired from the safety-related IETs with the VISTA-ITL facility for the SMART design. Three small-break loss-of-coolant accident (SBLOCA) tests of safety injection system (SIS) line break, shutdown cooling system (SCS) line break and pressurizer safety valve (PSV) line break were successfully performed and the transient characteristics of a complete loss of flowrate (CLOF) was simulated properly with the VISTA-ITL facility.     VISTA was conceived through a detailed systems analysis as a viable, realistic, and defensible spacecraft concept based on advanced ICF technology but existing or near-term technology for other systems. It is a conical self-contained single-stage piloted spacecraft in which a magnetic thrust chamber directs the plasma emissions from inertial confinement fusion (ICF) targets into a rearward exhaust. VISTA's propulsion system is therefore unique because it is based on (1) a rather mature technology (ICF), which is known to work with sufficient driver input; (2) direct heating of all expellant by the fusion process, thus providing high mass flow rates without significant degradation of jet efficiency; and (3) a magnetic thrust chamber, which avoids the plasma thermalization and resultant degradation of specific impulse that are unavoidable with the use of mechanical thrust chambers. VISTA therefore has inherently high power/mass ratios and high specific impulses. With advanced ICF technology, ultra-fast roundtrips (RTs) to objects within the solar system are possible (e.g., {ge}145 days RT to Mars, {ge}7 years RT to Pluto). Such short-duration missions are imperative to minimize the human physiological deteriorations arising from zero gravity and the cosmic-radiation. In addition, VISTA offers on-board artificial gravity and propellant-based shielding from cosmic rays, thus reducing the physiological deteriorations to insignificant levels. In this paper, we give an overview of the various vehicle systems for this concept, estimate the general missions performance capabilities for interplanetary missions, and describe in detail the performance for the baseline mission of a piloted roundtrip to Mars with a 100-ton payload. Items requiring further research include a reduction of the wet mass from its baseline value of 6,000 metric tons, and the development of fast ignition or its equivalent to provide target gains in excess of several hundred.     We have developed Phylo-VISTA (Shah et al., 2003), an interactive software tool for analyzing multiple alignments by visualizing a similarity measure for DNA sequences of multiple species. The complexity of visual presentation is effectively organized using a framework based upon inter-species phylogenetic relationships. The phylogenetic organization supports rapid, user-guided inter-species comparison. To aid in navigation through large sequence datasets, Phylo-VISTA provides a user with the ability to select and view data at varying resolutions. The combination of multi-resolution data visualization and analysis, combined with the phylogenetic framework for inter-species comparison, produces a highly flexible and powerful tool for visual data analysis of multiple sequence alignments.

    Identifying transcriptional regulatory elements represents a significant challenge in annotating the genomes of higher vertebrates. We have developed a computational tool, rVISTA, for high-throughput discovery of cis-regulatory elements that combines transcription factor binding site prediction and the analysis of inter-species sequence conservation. Here, we illustrate the ability of rVISTA to identify true transcription factor binding sites through the analysis of AP-1 and NFAT binding sites in the 1 Mb well-annotated cytokine gene cluster1 (Hs5q31; Mm11). The exploitation of orthologous human-mouse data set resulted in the elimination of 95 percent of the 38,000 binding sites predicted upon analysis of the human sequence alone, while it identified 87 percent of the experimentally verified binding sites in this region.

  1. VISTA -- A Vehicle for Interplanetary Space Transport Application Powered by Inertial Confinement Fusion

    Inertial Confinement Fusion (ICF) is an ideal technology to power self-contained single-stage piloted (manned) spacecraft within the solar system because of its inherently high power/mass ratios and high specific impulses (i.e., high exhaust velocities). These technological advantages are retained when ICF is utilized with a magnetic thrust chamber, which avoids the plasma thermalization and resultant degradation of specific impulse that are unavoidable with the use of mechanical thrust chambers. We started with Rod Hyde's 1983 description of an ICF-powered engine concept using a magnetic thrust chamber, and conducted a more detailed systems study to develop a viable, realistic, and defensible spacecraft concept based on ICF technology projected to be available in the first half of the 21st century. The results include an entirely new conical spacecraft conceptual design utilizing near-existing radiator technology. We describe the various vehicle systems for this new concept, estimate the missions performance capabilities for general missions to the planets within the solar system, and describe in detail the performance for the baseline mission of a piloted roundtrip to Mars with a 100-ton payload. For this mission, we show that roundtrips totaling {ge}145 days are possible with advanced DT fusion technology and a total (wet) spacecraft mass of about 6000 metric tons. Such short-duration missions are advantageous to minimize the known cosmic-radiation hazards to astronauts, and are even more important to minimize the physiological deteriorations arising from zero gravity. These ICF-powered missions are considerably faster than those available using chemical or nuclear-electric-propulsion technologies with minimum-mass vehicle configurations.   2. VISTA Enhancer Browser--A Database of Tissue-Specific HumanEnhancers

    Despite the known existence of distant-acting cis-regulatoryelements in the human genome, only a small fraction of these elements hasbeen identified and experimentally characterized in vivo. This paucity ofenhancer collections with defined activities has thus hinderedcomputational approaches for the genome-wide prediction of enhancers andtheir functions. To fill this void, we utilize comparative genomeanalysis to identify candidate enhancer elements in the human genomecoupled with the experimental determination of their in vivo enhanceractivity in transgenic mice (1). These data are available through theVISTA Enhancer Browser (http://enhancer.lbl.gov). This growing databasecurrently contains over 250 experimentally tested DNA fragments, of whichmore than 100 have been validated as tissue-specific enhancers. For eachpositive enhancer, we provide digital images of whole-mount embryostaining at embryonic day 11.5 and an anatomical description of thereporter gene expression pattern. Users can retrieve elements near singlegenes of interest, search for enhancers that target reporter geneexpression to a particular tissue, or download entire collections ofenhancers with a defined tissue specificity or conservation depth. Theseexperimentally validated training sets are expected to provide a basisfor a wide range of downstream computational and functional studies ofenhancer function.

    The Buena Vista Hills field is located about 25 miles southwest of Bakersfield, in Kern County, California, about two miles north of the city of Taft, and five miles south of the Elk Hills field. The Antelope Shale zone was discovered at the Buena Vista Hills field in 1952, and has since been under primary production. Little research was done to improve the completion techniques during the development phase in the 1950s, so most of the wells are completed with about 1000 ft of slotted liner. The proposed pilot consists of four existing producers on 20 acre spacing with a new 10 acre infill well drilled as the pilot CO{sub 2} injector. Most of the reservoir characterization of the first phase of the project will be performed using data collected in the pilot pattern wells. This is the first annual report of the project. It covers the period February 12, 1996 to February 11, 1997. During this period the Chevron Murvale 653Z-26B well was drilled in Section 26-T31S/R23E in the Buena Vista Hills field, Kern County, California. The Monterey Formation equivalent Brown and Antelope Shales were continuously cored, the zone was logged with several different kinds of wireline logs, and the well was cased to a total depth of 4907 ft. Core recovery was 99.5%. Core analyses that have been performed include Dean Stark porosity, permeability and fluid saturations, field wettability, anelastic strain recovery, spectral core gamma, profile permeametry, and photographic imaging. Wireline log analysis includes mineral-based error minimization (ELAN), NMR T2 processing, and dipole shear wave anisotropy. A shear wave vertical seismic profile was acquired after casing was set and processing is nearly complete.

    The Buena Vista Hills field is located about 25 miles southwest of Bakersfield, in Kern County, California, about two miles north of the city of Taft, and five miles south of the Elk Hills field. The Antelope Shale zone was discovered at the Buena Vista Hills field in 1952, and has since been under primary production. Little research was done to improve the completion techniques during the development phase in the 1950s, so most of the wells are completed with about 1000 ft of slotted liner. The proposed pilot consists of four existing producers on 20 acre spacing with a new 10 acre infill well drilled as the pilot CO{sub 2} injector. Most of the reservoir characterization of the first phase of the project will be performed using data collected in the pilot pattern wells. This is the first annual report of the project. It covers the period February 12, 1996 to February 11, 1997. During this period the Chevron Murvale 653Z-26B well was drilled in Section 26-T31S/R23E in the Buena Vista Hills field, Kern County, California. The Monterey Formation equivalent Brown and Antelope Shales were continuously cored, the zone was logged with several different kinds of wireline logs, and the well was cased to a total depth of 4907 ft. Core recovery was 99.5%. Core analyses that have been performed include Dean Stark porosity, permeability and fluid saturations, field wettability, anelastic strain recovery, spectral core gamma, profile permeametry, and photographic imaging. Wireline log analysis includes mineral-based error minimization (ELAN), NMR T2 processing, and dipole shear wave anisotropy. A shear wave vertical seismic profile was acquired after casing was set and processing is nearly complete.

    Energy consumption in California fell in 1991 for the first time in five years. The State`s economy was especially hard hit by a continuing national recession. The construction industry for the second year experienced a dramatic downturn. Energy use in the industrial sector showed a modest increase, but consumption in other end-use categories declined. The decrease in energy used in transportation can be traced to a substantial fall in the sales of both highway diesel fuels and vessel bunkering fuels at California ports, the latter reflecting a mid-year increase in taxes. Gasoline sales by contrast increased as did the number of miles traveled and the number of automobiles in the State. Production in California`s oil and gas fields was at 1990 levels thus arresting a steady decline in output. Due to enlarged steam flooding operations, production at several fields reached record levels. Also countering the decline in many of California fields was new production from the Port Arguello offshore field. California natural gas production, despite a modest 1991 increase, will not fill the use within the State. Petroleum comprised more than half of the State`s energy supply principally for transportation. Natural gas use showed a small increase. Oil products play virtually no role in electrical production. The largest single source of electricity to the State is imports from the Pacific Northwest and from coal-fired plants in the Southwest. Combined contributions to transmitted electricity from renewable and alternate sources declined as hydropower was constrained by a prolonged drought and as geothermal power from the largest and oldest field at The Geysers fell. Windpower grew slightly; however solar power remained at 1990 levels and made no substantial contribution to total power generation.

    Air Products and Chemicals, Inc. has completed a comprehensive, multiyear project to demonstrate a hydrogen infrastructure in California. The specific primary objective of the project was to demonstrate a model of a real-world retail hydrogen infrastructure and acquire sufficient data within the project to assess the feasibility of achieving the nation's hydrogen infrastructure goals. The project helped to advance hydrogen station technology, including the vehicle-to-station fueling interface, through consumer experiences and feedback. By encompassing a variety of fuel cell vehicles, customer profiles and fueling experiences, this project was able to obtain a complete portrait of real market needs. The project also opened its stations to other qualified vehicle providers at the appropriate time to promote widespread use and gain even broader public understanding of a hydrogen infrastructure. The project engaged major energy companies to provide a fueling experience similar to traditional gasoline station sites to foster public acceptance of hydrogen. Work over the course of the project was focused in multiple areas. With respect to the equipment needed, technical design specifications (including both safety and operational considerations) were written, reviewed, and finalized. After finalizing individual equipment designs, complete station designs were started including process flow diagrams and systems safety reviews. Material quotes were obtained, and in some cases, depending on the project status and the lead time, equipment was placed on order and fabrication began. Consideration was given for expected vehicle usage and station capacity, standard features needed, and the ability to upgrade the station at a later date. In parallel with work on the equipment, discussions were started with various vehicle manufacturers to identify vehicle demand (short- and long-term needs). Discussions included identifying potential areas most suited for hydrogen fueling stations

    Building energy benchmarking is the comparison of whole-building energy use relative to a set of similar buildings. It provides a useful starting point for individual energy audits and for targeting buildings for energy-saving measures in multiple-site audits. Benchmarking is of interest and practical use to a number of groups. Energy service companies and performance contractors communicate energy savings potential with ''typical'' and ''best-practice'' benchmarks while control companies and utilities can provide direct tracking of energy use and combine data from multiple buildings. Benchmarking is also useful in the design stage of a new building or retrofit to determine if a design is relatively efficient. Energy managers and building owners have an ongoing interest in comparing energy performance to others. Large corporations, schools, and government agencies with numerous facilities also use benchmarking methods to compare their buildings to each other. The primary goal of Task 2.1.1 Web-based Benchmarking was the development of a web-based benchmarking tool, dubbed Cal-Arch, for benchmarking energy use in California commercial buildings. While there were several other benchmarking tools available to California consumers prior to the development of Cal-Arch, there were none that were based solely on California data. Most available benchmarking information, including the Energy Star performance rating, were developed using DOE's Commercial Building Energy Consumption Survey (CBECS), which does not provide state-level data. Each database and tool has advantages as well as limitations, such as the number of buildings and the coverage by type, climate regions and end uses. There is considerable commercial interest in benchmarking because it provides an inexpensive method of screening buildings for tune-ups and retrofits. However, private companies who collect and manage consumption data are concerned that the identities of building owners might be revealed and

    Energy consumption in the state of California decreased about 3% in 1993 reflecting continuation of the recession that was manifest in a moribund construction industry and a high state unemployment that ran counter to national recovery trends. Residential/commercial use decreased slightly reflecting a mild winter in the populous southern portion of the state, a decrease that was offset to some extent by an increase in the state population. Industrial consumption of purchased energy declined substantially as did production of self-generated electricity for in-house use. Consumption in the transportation sector decreased slightly. The amount of power transmitted by the utilities was at 1992 levels; however a smaller proportion was produced by the utilities themselves. Generation of electricity by nonutilities, primarily cogenerators and small power producers, was the largest of any state in the US. The growth in the number of private power producers combined with increased amounts of electricity sold to the public utilities set the stage for the sweeping proposals before the California Public Utility Commission to permit direct sales from the nonutilities to retail customers. California production of both oil and natural gas declined; however, to meet demand only the imports of natural gas increased. A break in the decade-long drought during the 1992--1993 season resulted in a substantial increase in the amount of hydroelectricity generated during the year. Geothermal energy`s contribution increased substantially because of the development of new resources by small power producers. Decline in steam production continued at The Geysers, the state`s largest field, principally owned and managed by a public utility. Increases in windpower constituted 1--1/2% of the total electric supply--up slightly from 1992. Several solar photo voltaic demonstration plants were in operation, but their contribution remained small.

    California oil producers face a loss of production this year because of constraints on pipeline and tanker transportation to Los Angeles area refineries. The potential bottleneck is occurring at a time when Outer Continental Shelf production is near capacity from Chevron Corp.'s Point Arguello project at the same time production is increasing from Exxon Corp.'s nearby Santa Ynex Unit (SYU) expansion. Both megaprojects must compete for pipeline space with onshore crude producers, notably in California's San Joaquin Valley (SJV). Recent development limiting transportation options include: An indefinite shutdown of Four Corners Pipe Line Co.'s 50,000 b/d Line No. 1, damaged by the Jan. 17 earthquake; Loss of a tanker permit by Chevron and partners for offshore Point Arguello production; Permanent shutdown of Exxon's offshore storage and treatment (OST) facility, which since 1981 has used tankers to transport about 20,000 b/d of SYU production from the Santa Barbara Channel to Los Angeles. The OST, the first commercial floating production system in the US -- placed in the Santa Barbara Channel in 1981 after a decade of precedent setting legal and political battles -- was shut down Apr. 4.     California energy consumption increased in 1994 in keeping with a recovery from the previous mild recession years. Although unemployment remained above the national average, other indicators pointed to improved economic health. Increased energy use was registered principally in the residential/commercial and transportation end-use sectors. A cooler-than-usual winter and spring was reflected in increased consumption of natural gas, the principal space-heating fuel in the state. Because of low water levels behind state dams, utilities turned to natural gas for electrical generation and to increased imports from out-of- state sources to meet demand. Other factors, such as smaller output from geothermal, biomass, and cogenerators, contributed to the need for the large increase in electrical supply from these two sources. Nonetheless, petroleum dominated the supply side of the energy equation of the state in which transportation requirements comprise more than one-third of total energy demand. About half of the oil consumed derived from California production. Onshore production has been in slow decline; however, in 1994 the decrease was compensated for by increases from federal offshore fields. Until 1994 production had been limited by regulatory restrictions relating to the movement of the crude oil to onshore refineries. State natural gas production remained at 1993 levels. The increased demand was met by larger imports from Canada through the recent expansion of Pacific Transmission Company`s 804 mile pipeline. Deregulation of the state`s utilities moved ahead in 1994 when the California Public Utilities Commission issued its proposal on how to restructure the industry. Public hearings were conducted in which the chief issues were recovery of the utilities` capital investments, conflicts with the Public Utilities Policies Act, management of power transactions between new suppliers and former utility customers, and preservation of energy conservation programs

The flight was part of the Airborne Greenhouse Gas Emissions Survey, a collaboration between Berkeley Lab, the National Oceanic and Atmospheric Administration, and the University of California, and UC Davis to pinpoint the sources of greenhouse gases in central California.     Over the past several years, four of the largest investor-owned California utilities have completed more than 50 evaluation studies designed to measure the energy and demand impacts of their demand-side management (DSM) programs. These four are: Pacific Gas and Electric (PG and E), Southern California Edison (SCE), Southern California Gas (SoCalGas), and San diego Gas and Electric (SDG and E). These studies covered residential, commercial, industrial, and agricultural DSM programs and provided a wealth of information on program impacts. The objective of this report is to summarize the results of these DSM evaluation studies in order to describe what DSM has achieved in California, to assess how well these achievements were forecast, and to compare the effectiveness of different types of DSM programs. This report documents the sizable investment made by the California utilities in their 1990--92 DSM programs. Between 1990 and 1992, the four utilities spent $772 million on energy-efficiency/conservation programs. This report also summarizes the realization rates estimated by the 50+ evaluation studies. Realization rates are defined as ex-post net savings estimates divided by ex-ante net savings estimates. Realization rates are summarized for 158 programs and program segments.
