    2013-05-01

Labor-intensive multistep biological tasks, such as the construction and cloning of DNA molecules, are prime candidates for laboratory automation. Flexible and biology-friendly operation of robotic equipment is key to its successful integration in biological laboratories, and the efforts required to operate a robot must be much smaller than the alternative manual lab work. To achieve these goals, a simple high-level biology-friendly robot programming language is needed. We have developed and experimentally validated such a language: Programming a Robot (PaR-PaR). The syntax and compiler for the language are based on computer science principles and a deep understanding of biological workflows. PaR-PaR allows researchers to use liquid-handling robots effectively, enabling experiments that would not have been considered previously. After minimal training, a biologist can independently write complicated protocols for a robot within an hour. Adoption of PaR-PaR as a standard cross-platform language would enable hand-written or software-generated robotic protocols to be shared across laboratories.

  11. Cummins MD & HD Accessory Hybridization CRADA -Annual Report FY15

    SciTech Connect (OSTI)

    Deter, Dean D.

    2015-10-01

There are many areas of MD and HD vehicles that can be improved by new technologies and optimized control strategies. Component optimization and idle reduction need to be addressed, this is best done by a two part approach that includes selecting the best component technology, and/or architecture, and optimized controls that are vehicle focused. While this is a common focus in the light duty industry it has been gaining momentum in the MD and HD market as the market gets more competitive and the regulations become more stringent. When looking into systems optimization and idle reduction technologies, affected vehicle systems must first be considered, and if possible included in the new architecture to get the most benefit out of these new capabilities. Typically, when looking into idle reduction or component optimization for MD/HD, the vehicle s accessories become a prime candidate for electrification or hybridization. While this has already been studied on light duty vehicles (especially on hybrids and electric vehicles) it has not made any head way or market penetration in most MD and HD applications. If hybrids and electric MD and HD vehicles begin to break into the market this would be a necessary step into the ability to make those vehicles successful by allowing for independent, optimized operation separate from the engine.

CoMD-Em is a software implementation suite of the CoMD [4] proxy app using different emerging programming models. It is intended to analyze the features and capabilities of novel programming models that could help ensure code and performance portability and scalability across heterogeneous platforms while improving programmer productivity. Another goal is to provide the authors and venders with some meaningful feedback regarding the capabilities and limitations of their models. The actual application is a classical molecularmore » dynamics (MD) simulation using either the Lennard-Jones method (LJ) or the embedded atom method (EAM) for primary particle interaction. The code can be extended to support alternate interaction models. The code is expected ro run on a wide class of heterogeneous hardware configurations like shard/distributed/hybrid memory, GPU's and any other platform supported by the underlying programming model.« less

  1. Technical Insights for Saltstone PA Maintenance

    SciTech Connect (OSTI)

    Flach, G.; Sarkar, S.; Mahadevan, S.; Kosson, D.

    2011-07-20

The Cementitious Barriers Partnership (CBP) is a collaborative program sponsored by the US DOE Office of Waste Processing. The objective of the CBP is to develop a set of computational tools to improve understanding and prediction of the long-term structural, hydraulic, and chemical performance of cementitious barriers and waste forms used in nuclear applications. CBP tools are expected to better characterize and reduce the uncertainties of current methodologies for assessing cementitious barrier performance and increase the consistency and transparency of the assessment process, as the five-year program progresses. In September 2009, entering its second year of funded effort, the CBP sought opportunities to provide near-term tangible support to DOE Performance Assessments (PAs). The Savannah River Saltstone Disposal Facility (SDF) was selected for the initial PA support effort because (1) cementitious waste forms and barriers play a prominent role in the performance of the facility, (2) certain important long-term behaviors of cementitious materials composing the facility are uncertain, (3) review of the SDF PA by external stakeholders is ongoing, and (4) the DOE contractor responsible for the SDF PA is open to receiving technical assistance from the CBP. A review of the current (SRR Closure & Waste Disposal Authority 2009) and prior Saltstone PAs (e.g., Cook et al. 2005) suggested five potential opportunities for improving predictions. The candidate topics considered were (1) concrete degradation from external sulfate attack, (2) impact of atmospheric exposure to concrete and grout before closure, such as accelerated slag and Tc-99 oxidation, (3) mechanistic prediction of geochemical conditions, (4) concrete degradation from rebar corrosion due to carbonation, and (5) early age cracking from drying and/or thermal shrinkage. The candidate topics were down-selected considering the feasibility of addressing each issue within approximately six months, and

  18. RADIATION PRESSURE DETECTION AND DENSITY ESTIMATE FOR 2011 MD

We present our astrometric observations of the small near-Earth object 2011 MD (H ? 28.0), obtained after its very close fly-by to Earth in 2011 June. Our set of observations extends the observational arc to 73days, and, together with the published astrometry obtained around the Earth fly-by, allows a direct detection of the effect of radiation pressure on the object, with a confidence of 5?. The detection can be used to put constraints on the density of the object, pointing to either an unexpectedly low value of ?=(640330)kg m{sup ?3} (68% confidence interval) if we assume a typical probability distribution for the unknown albedo, or to an unusually high reflectivity of its surface. This result may have important implications both in terms of impact hazard from small objects and in light of a possible retrieval of this target.

Recent studies show that compounding polyamide 6 (PA 6) with a PA 6 polyether block copolymers made by reaction injection molding (RIM) or continuous anionic polymerization in a reactive extrusion process (REX) result in blends with high impact strength and high stiffness compared to conventional rubber blends. In this paper, different high impact PA 6 blends were prepared using a twin screw extruder. The different impact modifiers were an ethylene propylene copolymer, a PA PA 6 polyether block copolymer made by reaction injection molding and one made by reactive extrusion. To ensure good particle matrix bonding, the ethylene propylene copolymer was grafted with maleic anhydride (EPR-g-MA). Due to the molecular structure of the two block copolymers, a coupling agent was not necessary. The block copolymers are semi-crystalline and partially cross-linked in contrast to commonly used amorphous rubbers which are usually uncured. The combination of different analysis methods like atomic force microscopy (AFM), transmission electron microscopy (TEM) and scanning electron microscopy (SEM) gave a detailed view in the structure of the blends. Due to the partial cross-linking, the particles of the block copolymers in the blends are not spherical like the ones of ethylene propylene copolymer. The differences in molecular structure, miscibility and grafting of the impact modifiers result in different mechanical properties and different blend morphologies.

The behaviour of sheared colloidal suspensions with full hydrodynamic interactions (HIs) is numerically studied. To this end, we use the hybrid stochastic rotation dynamics-molecular dynamics (SRD-MD) method. The shear viscosity of colloidal suspensions is computed for different volume fractions, both for dilute and concentrated cases. We verify that HIs help in the collisions and the streaming of colloidal particles, thereby increasing the overall shear viscosity of the suspension. Our results show a good agreement with known experimental, theoretical, and numerical studies. This work demonstrates the ability of SRD-MD to successfully simulate transport coefficients that require correct modelling of HIs.

  2. Kinetic energy deficit in the symmetric fission of /sup 259/Md. [Light particle emission in /sup 256/Fm fission

    SciTech Connect (OSTI)

    Hulet, E.K.; Wild, J.F.; Lougheed, R.W.; Baisden, P.A.; Dougan, R.J.; Mustafa, M.G.

    1980-10-01

The fragment energies of about 725 coincidence events have now been observed in the spontaneous fission (SF) decay of 105-min /sup 259/Md since its discovery in 1977. The fission of /sup 259/Md is characterized by a symmetric mass distribution, similar to those of /sup 258/Fm and /sup 259/Fm, but with a broad total kinetic energy (anti TKE) distribution which peaks at about 195 MeV, in contrast to those of /sup 258/Fm and /sup 259/Fm, for which the anti TKE is about 240 MeV. This kinetic energy deficit, approx. 40 MeV, has been postulated to be due to the emission of hydrogen-like particles by /sup 259/Md at the scission point in a large fraction of the fissions, leaving the residual fissioning nucleus with 100 protons. The residual nucleus would then be able to divide into two ultrastable tin-like fission fragments, but with less kinetic energy than that observed in the SF of /sup 258/Fm and /sup 259/Fm, because of binding-energy losses and a reduction in the Coulomb repulsion of the major fragments. To test this hypothesis, counter-telescope experiments aimed at detecting and identifying these light particles were performed. In 439 SF events 3 + 3 protons of the appropriate energy were observed, too few to account for the kinetic energy deficit in the fission of /sup 259/Md. There seems to be no explanation for this problem within the framework of current fission theory. These results are discussed along with preliminary measurements of light-particle emission in the SF of /sup 256/Fm. 5 figures.

This review article describes our neutron scattering experiments made in the past four years for the understanding of the single-particle (hydrogen atom) dynamics of a protein and its hydration water and the strong coupling between them. We found that the key to this strong coupling is the existence of a fragile-to-strong dynamic crossover (FSC) phenomenon occurring at around T L = 225±5 K in the hydration water. On lowering of the temperature toward FSC, the structure of hydration water makes a transition from predominantly the high density form (HDL), a more fluid state, to predominantly the low density formmore » (LDL), a less fluid state, derived from the existence of a liquid–liquid critical point at an elevated pressure. We show experimentally that this sudden switch in the mobility of hydration water on Lysozyme, B-DNA and RNA triggers the dynamic transition, at a temperature T D = 220 K, for these biopolymers. In the glassy state, below T D , the biopolymers lose their vital conformational flexibility resulting in a substantial diminishing of their biological functions. We also performed molecular dynamics (MD) simulations on a realistic model of hydrated lysozyme powder, which confirms the existence of the FSC and the hydration level dependence of the FSC temperature. Furthermore, we show a striking feature in the short time relaxation ( β -relaxation) of protein dynamics, which is the logarithmic decay spanning 3 decades (from ps to ns). The long time α -relaxation shows instead a diffusive behavior, which supports the liquid-like motions of protein constituents. We then discuss our recent high-resolution X-ray inelastic scattering studies of globular proteins, Lysozyme and Bovine Serum Albumin. We were able to measure the dispersion relations of collective, intra-protein phonon-like excitations in these proteins for the first time. We found that the phonon energies show a marked softening and at the same time their population increases

Graphene oxide (GO) was prepared by oxidation of graphite flakes by a mixture of H{sub 2}SO{sub 4}/H{sub 3}PO{sub 4} and KMnO{sub 4} based on Marcano's method. Two different masterbatches containing GO (33.3%) and polyamide-6 (PA6) (66.7%) were prepared both via solvent casting in formic acid and by melt mixing in a mini-extruder (Haake). The two masterbatches were then used to prepare PA6-based nanocomposites with a content of 2% in GO. For comparison, a nanocomposite by direct mixing of PA6 and GO (2%) and PA6/graphite nanocomposites were prepared, too. The oxidation of graphite into GO was assessed by X-ray diffraction (XRD), Micro-Raman spectroscopy, scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS) analyses. All these techniques demonstrated the effectiveness of the graphite modification, since the results put into evidence that, after the acid treatment, interlayer distance, oxygen content and defects increased. SEM micrographs carried out on the nanocomposites, showed GO layers totally surrounded by polyamide-6, this feature is likely due to the strong interaction between the hydrophilic moieties located both on GO and on PA6. On the contrary, no interactions were observed when graphite was used as filler. Mechanical characterization, carried out by tensile and dynamic-mechanical tests, marked an improvement of the mechanical properties observed. Photoluminescence and EPR measurements were carried out onto nanoparticles and nanocomposites to study the nature of the interactions and to assess the possibility to use this class of materials as semiconductors or optical sensors.

For over 40 years the US Department of Energy (DOE) and its predecessors have made Californium-252 ({sup 252}Cf) available for a wide range of industries including medical, nuclear fuels, mining, military and national security. The Radiochemical Engineering Development Center (REDC) located within the Oak Ridge National Laboratory (ORNL) processes irradiated production targets from the High Flux Isotope Reactor (HFIR). Operations in Building 7930, Cell G provide over 70% of the world's demand for {sup 252}Cf. Building 7930 was constructed and equipped in the mid-1960s. Current operations for {sup 252}Cf processing in Building 7930, Cell G require use of through-the-wall manipulators and the PaR Remote Handling System. Maintenance and repairs for the manipulators is readily accomplished by removal of the manipulator and relocation to a repair shop where hands-on work can be performed in glove boxes. Contamination inside cell G does not currently allow manned entry and no provisions were created for a maintenance area inside the cell. There has been no maintenance of the PaR system or upgrades, leaving operations vulnerable should the system have a catastrophic failure. The Cell G PaR system is currently being operated in a run to failure mode. As the manipulator is now 40+ years old there is significant risk in this method of operation. In 2006 an assessment was completed that resulted in recommendations for replacing the manipulator operator control and power centers which are used to control and power the PaR manipulator in Cell G. In mid-2008 the chain for the bridge drive failed and subsequent examinations indicated several damaged links (see Figure 1). To continue operations the PaR manipulator arm is being used to push and pull the bridge as a workaround. A retrieval tool was fabricated, tested and staged inside Cell G that will allow positioning of the bridge and manipulator arm for removal from the cell should the PaR system completely fail. Every action in biology is performed by dynamic proteins that convert between multiple states in order to engage their functions. Often binding to various ligands is essential for the rates of desired transitions to be enhanced. The goal of computational biology is to study these transitions and discover the different states to fully understand the protein's normal and diseased function, design drugs to target/bias specific states, and understand all of the interactions in between. We have developed a new methodology that is capable of calculating the absolute free energy of proteins while taking into account all the interactions with the solvent molecules. The efficiency of the new scheme is an order of magnitude greater than any existing technique. This method is now implemented in the massively parallel popular MD program package NAMD. This now makes it possible to calculate the relative stability of different conformational states of biological macromolecules as well as their binding free energies to various ligands.

The structure of the X(3872) meson is unknown. Different competing models of the cc exotic state X(3872) exist, including the possibilities that this state is either a mesonic molecule with dominating D0D*0 + c.c. composition, a ccqq tetraquark, or a cc-gluon hybrid state. It is expected that the X(3872) state is rather strongly coupled to the pp channel and, therefore, can be produced in pp and pA collisions at PANDA. We propose to test the hypothetical molecular structure of X(3872) by studying the D or D* source stripping reactions on a nuclear residue.

  20. The structure and IR signatures of the arginine-glutamate salt bridge. Insights from the classical MD simulations

    2015-06-07

Salt bridges and ionic interactions play an important role in protein stability, protein-protein interactions, and protein folding. Here, we provide the classical MD simulations of the structure and IR signatures of the arginine (Arg)glutamate (Glu) salt bridge. The Arg-Glu model is based on the infinite polyalanine antiparallel two-stranded ?-sheet structure. The 1 ?s NPT simulations show that it preferably exists as a salt bridge (a contact ion pair). Bidentate (the end-on and side-on structures) and monodentate (the backside structure) configurations are localized [Donald et al., Proteins 79, 898915 (2011)]. These structures are stabilized by the short {sup +}NH?O{sup ?} bonds. Their relative stability depends on a force field used in the MD simulations. The side-on structure is the most stable in terms of the OPLS-AA force field. If AMBER ff99SB-ILDN is used, the backside structure is the most stable. Compared with experimental data, simulations using the OPLS all-atom (OPLS-AA) force field describe the stability of the salt bridge structures quite realistically. It decreases in the following order: side-on > end-on > backside. The most stable side-on structure lives several nanoseconds. The less stable backside structure exists a few tenth of a nanosecond. Several short-living species (solvent shared, completely separately solvated ionic groups ion pairs, etc.) are also localized. Their lifetime is a few tens of picoseconds or less. Conformational flexibility of amino acids forming the salt bridge is investigated. The spectral signature of the Arg-Glu salt bridge is the IR-intensive band around 2200 cm{sup ?1}. It is caused by the asymmetric stretching vibrations of the {sup +}NH?O{sup ?} fragment. Result of the present paper suggests that infrared spectroscopy in the 20002800 frequency region may be a rapid and quantitative method for the study of salt bridges in peptides and ionic interactions between proteins. This region is usually not considered in

In this study the MMI ConfidentDesign multiscale approach (consisting in a non-linear multiscale simulation based on DIGIMAT registered including the injection modeling of the filled polymer and a multiscale mechanical model using the fiber orientation tensor resulting from the injection) has been combined with an orthotropic damageable elastic simulation. The anisotropic properties (including rupture criterion) are estimated and a multiscale simulation including the heterogeneous material properties issued from injection process is done. The impact of fiber ratios is then investigated. The structural simulation predicts stresses localized close to the punch, as well in injected PA66 than in composite part. Greater the fiber volume ratio, greater the modulus and more brittle the composite.

A more reactive liner material is needed for use as liner and cruciform material in tritium producing burnable absorber rods (TPBAR) in commercial light water nuclear reactors (CLWR). The function of these components is to convert any water that is released from the Li-6 enriched lithium aluminate breeder material to oxide and hydrogen that can be gettered, thus minimizing the permeation of tritium into the reactor coolant. Fourteen zirconium alloys were exposed to 2.5 kPa water vapor in a helium stream at 300 C over a period of up to 35 days. Experimental alloys with aluminum, yttrium, vanadium, titanium, and scandium, some of which also included ternaries with nickel, were included along with a high nitrogen impurity alloy and the commercial alloy Zircaloy-2. They displayed a reactivity range of almost 500, with Zircaloy-2 being the least reactive.

We present classical molecular dynamics (MD) with Modified Embedded Atom Model (MEAM) simulations to investigate the role of primary radiation defects and radiogenic helium as factors affecting the phase stability of PuGa alloys in cooling–heating cycles at ambient pressure. The models of PuGa alloys equilibrated at ambient conditions were subjected to cooling–heating cycles in which they were initially cooled down to 100 K and then heated up to 500 K at ambient pressure. The rate of temperature change in the cycles was 10 K/ns. The simulations showed that the initial FCC phase of PuGa alloys undergo polymorphous transition in coolingmore » to a lower symmetry α'-phase. All the alloys undergo direct and reverse polymorphous transitions in the cooling–heating cycles. The alloys containing vacancies shift in both transitions to lower temperatures relative to the defect-free alloys. The radiogenic helium has much less effect on the phase stability compared to that of primary radiation defects (in spite of the fact that helium concentration is twice of that for the primary radiation defects). Lastly, this computational result agrees with experimental data on unconventional stabilization mechanism of PuGa alloys.« less

Here, age dating of nuclear material can provide insight into source and suspected use in nuclear forensic investigations. We report here a method for the determination of the date of most recent chemical purification for uranium materials using the 235U-231Pa chronometer. Protactinium is separated from uranium and neptunium matrices using anion exchange resin, followed by sorption of Pa to an SiO2 medium. The concentration of 231Pa is measured by isotope dilution mass spectrometry using 233Pa spikes prepared from an aliquot of 237Np and calibrated in-house using the rock standard Table Mountain Latite and the uranium isotopic standard U100. Combined uncertainties of age dates using this method are 1.5 to 3.5 %, an improvement over alpha spectrometry measurement methods. Model ages of five uranium standard reference materials are presented; all standards have concordant 235U-231Pa and 234U-230Th model ages.

    2013-12-15

Purpose: Catheter-directed thrombolysis (CDT) is a highly effective approach in the treatment of deep venous thrombosis (DVT). There are no data on the primary use of CDT with argatroban and tissue plasminogen activator (tPA) in patients without heparin-induced thrombocytopenia (HIT). The aim of this study was to evaluate the efficacy and safety of the combined administration of argatroban and tPA during CDT for massive DVT in patients without HIT. Methods: Thirty-three patients with massive symptomatic iliac and femoropopliteal DVT underwent CDT with tPA and argatroban within 28 {+-} 6 h of presentation. The dose of tPA was 0.75-1 mg/h through the infusion port and that of argatroban at 0.3-1 {mu}g/kg/min through the side port of the sheath. The patients were evaluated for the efficacy and safety of CDT and recurrent symptomatic venous thromboembolism (VTE) at a mean follow-up of 22 months. Results: There was no bleeding or iatrogenic pulmonary embolism with the CDT regimen we used. Grade III lysis (complete resolution of thrombus on venography) was achieved in 30 patients (91 %). In 3 patients with additional inferior vena cava filter thrombosis, further thrombectomy of the filter was required. No patient developed recurrent VTE. Conclusion: Concomitant administration of argatroban and tPA is a highly safe and effective regimen for CDT for massive DVT.

    1993-06-01

This report presents analytical analyses of the insensitive munitions container PA103, with modified design features for a static internal pressure of 500 psi and for a dynamic impact resulting from a 7-ft free fall onto a rigid surface. The modified design features addressed by the analyses were the inclusion of a score pattern on the container cylindrical body and a plastic plate (fuse) sandwiched between metal flanges on the container end. The objectives of both the pressure and impact analyses were to determine if the induced stresses at the score patterns in the cylindrical body of the container were sufficient to induce failure. Analytical responses of the container to the imposed loads were obtained with finite element analysis methodology. The computer codes ABAQUS and VEC/DYNA3D were used to obtain the results. Results of the pressure analysis indicate that failure of the container body would be expected to occur at the score pattern for a static internal pressure of 500 psi. Also, results from three impact orientations for a 7-ft drop indicate that membrane stresses in the vicinity of the score pattern are above critical crack growth stress magnitudes, especially at low ([minus]60[degrees]F) temperatures.

  16. Reduction of VOC emissions from metal dip coating applications -- Canam Steel Corporation Point of Rocks, MD case study

    SciTech Connect (OSTI)

    Monfet, J.P.

    1997-12-31

The reduction of VOC emissions from metal dip coating applications is not an environmental constraint, it is an economic opportunity. This case study shows how the industry can reap economic benefits from VOC reductions while improving air quality. The Canam Steel Corporation plant located in Point of Rocks, MD operates dip tanks for primer application on fabricated steel joists and joist girders. This process is presently subject to a regulation that limits the paint VOC content to 3.5 pounds per gallon of coating less water. As a result of the high paint viscosity associated with that regulation, the paint thickness of the dipped steel is thicker than the customers` specifications. Most of the VOC emissions can therefore be associated with the excess of paint applied to the products rather than to the required thickness of the coating. The higher paint usage rate has more than environmental consequences, it increases the cost of the applied coating. The project is to reduce the paint usage by controlling the viscosity of the coating in the tank. Experimental results as well as actual mass balance calculations show that using a higher VOC content paint would reduce the overall VOC emissions. The author explained the project to the Maryland Department of the Environment (MDE) Air and Radiation Management Administration. First, the MDE agreed to develop a new RACT determination for fabricated steel dipping operations. The new regulation would limit the amount of VOC than can be emitted to dip coat a ton of fabricated steel. Second, the MDE agreed to allow experimentation of the higher VOC content paint as a pilot project for the new regulation. This paper demonstrates the need for a RACT determination specific to fabricated steel dipping operations.

The microstructure and surface properties of wollastonite (W) and MW powders were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy-dispersive X-ray spectrometry (EDS) and X-ray diffraction (XRD), respectively. The microstructure of W, MW and silane modified MW (SMW) powders were characterized by Fourier translation infrared spectroscopy (FTIR). The mechanical properties, heat distortion temperature (HDT) and oxygen index (OI) of PA6 composites having different fillers were discussed. It was shown that the surface of wollastonite was coated with a layer of 33 nm thickness of Mg(OH){sub 2} grains and the distribution of which was uniform. The number of the hydroxyl groups on the surface of wollastonite powder increased after coated with Mg(OH){sub 2}. Filling 30% of SMW powder in PA6 the mechanical properties, HDT and OI were notably enhanced.

  18. Intercomparison of the seasonal cycle in 200 hPa kinetic energy in AMIP GCM simulations

    SciTech Connect (OSTI)

    Boyle, J.S.

    1996-10-01

The 200 hPa kinetic energy is represented by means of the spherical harmonic components for the Atmospheric Model Intercomparison Project (AMIP) simulations, the National Center for Environmental Prediction/National Center for Atmospheric Research (NCEP/NCAR) reanalysis and the European Centre for Medium Range Weather Forecast Reanalysis (ERA). The data used are the monthly mean wind fields from 1979 to 1988. The kinetic energy is decomposed into the divergent (DKE) and rotational (RKE) components and emphasis is placed on examining the former. The two reanalysis data sets show reasonable agreement that is best for the rotational kinetic energy. The largest difference in the divergent kinetic energy occurs during the northern summer. As might be expected, the two analyses are closet in regions where there are sufficient observations such that the effect of the model used in the assimilation cycle are minimized. The observed RKE show only a slight seasonal cycle with a maximum occuring during the northern winter. The DKE, on the other hand, has a very pronounced seasonal cycle with maxima at the solsticial seasons and minima during the equinoctial seasons. The model results show a very large spread in the magnitudes of the RKE and DKE although the models all evince a seasonal variation in phase with that observed. The median values of the seasonal cycle of RKE and DKE for the models are usually superior to those of any individual model. Results are also presented for simulation following the AMIP protocol but using updated versions of the original AMIP entries. In most cases these new integrations show better agreement with the observations.

  19. Non-Abelian Bremsstrahlung and Azimuthal Asymmetries in High Energy p+A Reactions

    DOE PAGES-Beta [OSTI]

Here we apply the GLV reaction operator solution to the Vitev-Gunion-Bertsch (VGB) boundary conditions to compute the all-order in nuclear opacity non-abelian gluon bremsstrahlung of event- by-event uctuating beam jets in nuclear collisions. We evaluate analytically azimuthal Fourier moments of single gluon, vmore » $$M\\atop{n}$$ {1}, and even number 2ℓ gluon, v$$M\\atop{n}$$ {2ℓ} inclusive distributions in high energy p+A reactions as a function of harmonic $n$, target recoil cluster number, $M$, and gluon number, 2ℓ, at RHIC and LHC. Multiple resolved clusters of recoiling target beam jets together with the projectile beam jet form Color Scintillation Antenna (CSA) arrays that lead to character- istic boost non-invariant trapezoidal rapidity distributions in asymmetric B+A nuclear collisions. The scaling of intrinsically azimuthally anisotropic and long range in η nature of the non-Abelian bremsstrahlung leads to vn moments that are similar to results from hydrodynamic models, but due entirely to non-Abelian wave interference phenomena sourced by the fluctuating CSA. Our analytic non-flow solutions are similar to recent numerical saturation model predictions but differ by predicting a simple power-law hierarchy of both even and odd vn without invoking kT factorization. A test of CSA mechanism is the predicted nearly linear η rapidity dependence of the vn(kTη). Non- Abelian beam jet bremsstrahlung may thus provide a simple analytic solution to Beam Energy Scan (BES) puzzle of the near $$\\sqrt{s}$$ independence of vn(pT) moments observed down to 10 AGeV where large-x valence quark beam jets dominate inelastic dynamics. Recoil bremsstrahlung from multiple independent CSA clusters could also provide a partial explanation for the unexpected similarity of vn in p(D) + A and non-central A + A at same dN=dη multiplicity as observed at RHIC and LHC.« less

  10. Unreviewed Disposal Question Evaluation: Impact of New Information since 2008 PA on Current Low-Level Solid Waste Operations

    SciTech Connect (OSTI)

    2014-10-06

Solid low-level waste disposal operations are controlled in part by an E-Area Low-Level Waste Facility (ELLWF) Performance Assessment (PA) that was completed by the Savannah River National Laboratory (SRNL) in 2008 (WSRC 2008). Since this baseline analysis, new information pertinent to disposal operations has been identified as a natural outcome of ongoing PA maintenance activities and continuous improvement in model simulation techniques (Flach 2013). An Unreviewed Disposal Question (UDQ) Screening (Attachment 1) has been initiated regarding the continued ability of the ELLWF to meet Department of Energy (DOE) Order 435.1 performance objectives in light of new PA items and data identified since completion of the original UDQ Evaluation (UDQE). The present UDQE assesses the ability of Solid Waste (SW) to meet performance objectives by estimating the influence of new information items on a recent sum-of-fractions (SOF) snapshot for each currently active E-Area low-level waste disposal unit. A final SOF, as impacted by this new information, is projected based on the assumptions that the current disposal limits, Waste Information Tracking System (WITS) administrative controls, and waste stream composition remain unchanged through disposal unit operational closure (Year 2025). Revision 1 of this UDQE addresses the following new PA items and data identified since completion of the original UDQE report in 2013: New Kd values for iodine, radium and uranium; Elimination of cellulose degradation product (CDP) factors; Updated radionuclide data; Changes in transport behavior of mobile radionuclides; Potential delay in interim closure beyond 2025; and Component-in-grout (CIG) plume interaction correction. Consideration of new information relative to the 2008 PA baseline generally indicates greater confidence that PA performance objectives will be met than indicated by current SOF metrics. For SLIT9, the previous prohibition of non-crushable containers in revision 0

  13. Effect of Bubbles on Liquid Nitrogen Breakdown in Plane-Plane Electrode Geometry From 100-250 kPa

    SciTech Connect (OSTI)

Sauers, Isidor; James, David Randy; Tuncer, Enis; Polyzos, Georgios; Pace, Marshall O

    2011-01-01

Liquid nitrogen (LN(2)) is used as the cryogen and dielectric for many high temperature superconducting, high voltage applications. When a quench in the superconductor occurs, bubbles are generated which can affect the dielectric breakdown properties of the LN(2). Experiments were performed using plane-plane electrode geometry where bubbles were introduced into the gap through a pinhole in the ground electrode. Bubbles were generated using one or more kapton heaters producing heater powers up to 30 W. Pressure was varied from 100-250 kPa. Breakdown strength was found to be relatively constant up to a given heater power and pressure at which the breakdown strength drops to a low value depending on the pressure. After the drop the breakdown strength continues to drop gradually at higher heater power. This is particularly illustrated at 100 kPa. After the drop in breakdown strength the breakdown is believed to be due to the formation of a vapor bridge. Also the heater power at which the breakdown strength changes from that of LN(2) to that of gaseous nitrogen increases with increasing pressure. The data can provide design constraints for high temperature superconducting fault current limiters (FCLs) so that the formation of a vapor bridge can be suppressed or avoided.

  19. Using FEP's List and a PA Methodology for Evaluating Suitable Areas for the LLW Repository in Italy

    SciTech Connect (OSTI)

    Risoluti, P.; Ciabatti, P.; Mingrone, G.

    2002-02-26

In Italy following a referendum held in 1987, nuclear energy has been phased out. Since 1998, a general site selection process covering the whole Italian territory has been under way. A GIS (Geographic Information System) methodology was implemented in three steps using the ESRI Arc/Info and Arc/View platforms. The screening identified approximately 0.8% of the Italian territory as suitable for locating the LLW Repository. 200 areas have been identified as suitable for the location of the LLW Repository, using a multiple exclusion criteria procedure (1:500,000), regional scale (1:100.000) and local scale (1:25,000-1:10,000). A methodology for evaluating these areas has been developed allowing, along with the evaluation of the long term efficiency of the engineered barrier system (EBS), the characterization of the selected areas in terms of physical and safety factors and planning factors. The first step was to identify, on a referenced FEPs list, a group of geomorphological, geological, hydrogeological, climatic and human behavior caused process and/or events, which were considered of importance for the site evaluation, taking into account the Italian situation. A site evaluation system was established ascribing weighted scores to each of these processes and events, which were identified as parameters of the new evaluation system. The score of each parameter is ranging from 1 (low suitability) to 3 (high suitability). The corresponding weight is calculated considering the effect of the parameter in terms of total dose to the critical group, using an upgraded AMBER model for PA calculation. At the end of the process an index obtained by a score weighted sum gives the degree of suitability of the selected areas for the LLW Repository location. The application of the methodology to two selected sites is given in the paper.

  20. Development of Xe and Kr empirical potentials for CeO2, ThO2, UO2 and PuO2, combining DFT with high temperature MD

    Cooper, M. W. D.; Kuganathan, N.; Burr, P. A.; Rushton, M. J. D.; Grimes, R. W.; Stanek, C. R.; Andersson, D. A.

    2016-08-23

In this study, the development of embedded atom method (EAM) many-body potentials for actinide oxides and associated mixed oxide (MOX) systems has motivated the development of a complementary parameter set for gas-actinide and gas-oxygen interactions. A comprehensive set of density functional theory (DFT) calculations were used to study Xe and Kr incorporation at a number of sites in CeO2, ThO2, UO2 and PuO2. These structures were used to fit a potential, which was used to generate molecular dynamics (MD) configurations incorporating Xe and Kr at 300 K, 1500 K, 3000 K and 5000 K. Subsequent matching to the forces predictedmore » by DFT for these MD configurations was used to refine the potential set. This fitting approach ensured weighted fitting to configurations that are thermodynamically significant over a broad temperature range, while avoiding computationally expensive DFT-MD calculations. The resultant gas potentials were validated against DFT trapping energies and are suitable for simulating combinations of Xe and Kr in solid solutions of CeO2, ThO2, UO2 and PuO2, providing a powerful tool for the atomistic simulation of conventional nuclear reactor fuel UO2 as well as advanced MOX fuels.« less

The alpha-emitters {sup 225}Ac and {sup 213}Bi are of great interest for alpha-radioimmunotherapy which uses radioisotopes attached to cancer-seeking antibodies to efficiently treat various types of cancers. Both radioisotopes are daughters of the long-lived {sup 229}Th(t{sub 1/2} = 7880y). {sup 229}Th can be produced by proton irradiation of {sup 232}Th and {sup 230}Th, either directly or through production of isobars that beta-decay into {sup 229}Th. To obtain excitation functions, {sup 232}Th and {sup 230}Th have been irradiated at the On-Line Test Facility at the Holifield Radioactive Ion Beam Facility at ORNL. Benchmark tests conducted with Cu and Ni foils show very good agreement with literature results. The experiments with thorium targets were focused on the production of {sup 229}Pa and its daughter {sup 225}Ac from both {sup 232}Th and {sup 230}Th. Differential cross-sections for production of {sup 229}Pa and other Pa isotopes have been obtained.

  15. Ganodermanontriol (GDNT) exerts its effect on growth and invasiveness of breast cancer cells through the down-regulation of CDC20 and uPA

Highlights: Black-Right-Pointing-Pointer Ganodermanontriol (GDNT), a Ganoderma mushroom alcohol, inhibits growth of breast cancer cells. Black-Right-Pointing-Pointer CDC20 is over-expressed in tumors but not in the tumor surrounding tissue in breast cancer patients. Black-Right-Pointing-Pointer GDNT inhibits expression of CDC20 in breast cancer cells. Black-Right-Pointing-Pointer GDNT inhibits cell adhesion, cell migration and cell invasion of breast cancer cells. Black-Right-Pointing-Pointer GDNT inhibits secretion of uPA and down-regulates expression of uPAR in breast cancer cells. -- Abstract: Ganoderma lucidum is a medicinal mushroom that has been recognized by Traditional Chinese Medicine (TCM). Although some of the direct anticancer activities are attributed to the presence of triterpenes-ganoderic and lucidenic acids-the activity of other compounds remains elusive. Here we show that ganodermanontriol (GDNT), a Ganoderma alcohol, specifically suppressed proliferation (anchorage-dependent growth) and colony formation (anchorage-independent growth) of highly invasive human breast cancer cells MDA-MB-231. GDNT suppressed expression of the cell cycle regulatory protein CDC20, which is over-expressed in precancerous and breast cancer cells compared to normal mammary epithelial cells. Moreover, we found that CDC20 is over-expressed in tumors when compared to the tissue surrounding the tumor in specimens from breast cancer patients. GDNT also inhibited invasive behavior (cell adhesion, cell migration, and cell invasion) through the suppression of secretion of urokinase-plasminogen activator (uPA) and inhibited expression of uPA receptor. In conclusion, mushroom GDNT is a natural agent that has potential as a therapy for invasive breast cancers.

The cladding from one additional Zircaloy-2 clad fuel rod from the pressurized water reactor at Shippingport, Pa. was destructively examined for corrosion film thickness and hydrogen accumulation. These additional examinations were conducted primarily to determine whether or not the hydrogen pickup ratio (..delta..H/..delta..O) increased with increasing neutron exposure, as had been suggested by the results from earlier studies on these fuel rods. The current results indicate that the hydrogen pickup ratio for Zircaloy-2 does not change with increasing neutron exposure and suggest that some of the earlier reported data may be anomolous.

    1994-11-01

Divergence and convergence centers at 200 hPa and mean sea level pressure (MSLP) cyclones were located every 6 hr for a 10-yr general circulation model (GCM) simulation with the ECMWF (Cycle 36) for the boreal winters from 1980 to 1988. The simulation used the observed monthly mean sea surface temperature (SST) for the decade. Analysis of the frequency, location, and strength of these centers and cyclones gives insight into the dynamical response of the model to the varying SST. The results indicate that (1) the model produces reasonable climatologies of upper-level divergence and MSLP cyclones; (2) the model distribution of anomalies of divergence and convergence centers and MSLP cyclones is consistent with observations for the 1982-83 and 1986-87 El Nifio events; (3) the tropical Indian Ocean is the region of greatest divergence activity and interannual variability in the model; (4) the variability of the divergence centers is greater than that of the convergence centers; (5) strong divergence centers occur chiefly over the ocean in the midlatitudes but are more land-based in the tropics, except in the Indian Ocean; and (6) locations of divergence and convergence centers can be a useful tool for the intercomparison of global atmospheric simulations.

    1994-12-01

Urokinase-type plasminogen activator (u-PA) is a 54-kDa glycoprotein that catalyzes the conversion of plasminogen to plasmin, a broad-specificity protease responsible for the degradation of fibrin clots and extracellular matrix components. The u-PA protein consists of three individual modules: a growth factor domain (GFD), a kringle, and a serine protease domain. The amino terminal fragment (ATF) includes the GFD-responsible for u-PA binding to its receptor-and the kringle domains. This protein was expressed and uniformly {sup 15}N-and {sup 15}N/{sup 13}C-labeled in mammalian cells by methods that will be described. In addition, we present the three-dimensional structure of ATF that was derived from 1299 NOE-derived distance restraints along with the {phi} angle and hydrogen bonding restraints. Although the individual domains in the structures were highly converged, the two domains are structurally independent. The overall structures of the individual domains are very similar to the structures of homologous proteins. However, important structural differences between the growth factor domain of u-PA and other homologous proteins were observed in the region that has been implicated in binding the urokinase receptor. These results may explain, in part, why other growth factors show no appreciable affinity for the urokinase receptor.

Purpose: Assess tumor-growth delay and survival in a mouse model of prostate cancer treated with tumor-targeting gold nanoparticles (AuNPs) and proton therapy. Methods: We first examined the accumulation of targeting nanoparticles within prostate tumors by imaging AuNPs with ultrasound-guided photoacoustics at 24h after the intravenous administration of goserelin-conjugated AuNPs (gAuNP) in three mice. Nanoparticles were also imaged at the cellular level with TEM in PC3 cells incubated with gAuNP for 24h. Pegylated AuNPs (pAuNP) were also imaged in vivo and in vitro for comparison. PC3 cells were then implanted subcutaneously in nude mice; 51mice with 810mm tumors were included. AuNPs were injected intravenously at 0.2%w/w final gold concentration 24h before irradiation. A special jig was designed to facilitate tumor irradiation perpendicular to the proton beam. Proton energy was set to 180MeV, the radiation field was 1818cm{sup 2}, and 9cm or 13.5cm thick solid-water compensators were used to position the tumors at either the beam entrance (BE) or the SOBP. Physical doses of 5Gy were delivered to all tumors on a patient beam line at MD Anderson's Proton Therapy Center. Results: The photoacoustic experiment reveled that our nanoparticles leak from the tumor-feeding vasculature and accumulate within the tumor volume over time. Additionally, TEM images showed gAuNP are internalized in cancer cells, accumulating within the cytoplasm, whereas pAuNP are not. Tumor-growth was delayed by 11 or 32days in mice receiving gAuNP irradiated at the BE or the SOBP, relative to proton radiation alone. Survival curves (ongoing experiment) reveal that gAuNPs improved survival by 36% or 74% for tumors irradiated at the BE or SOBP. Conclusion: These important, albeit preliminary, in vivo findings reveal nanoparticles to be potent sensitizers to proton therapy. Further, conjugation of AuNPs to tumor-specific antigens that promote enhanced cellular internalization improved both tumor

Sediments found in the New York/New Jersey Harbor are widely contaminated with organic and inorganic compounds of anthropogenic origin. As a result, the environmental health of the Harbor has deteriorated and the efficient operation of the Port compromised by difficulties in disposing of sediments resulting from maintenance and improvements of navigational channels. Knowledge of the properties of the sediments on a micro-scale is useful in understanding the transport of contaminants through the environment, for developing effective methods for sediment decontamination, and for subsequent beneficial use of the cleaned sediments. We have investigated several properties of these sediments using synchrotron radiation techniques. These include computed microtomography using absorption and fluorescence contrast mechanisms, x-ray microscopy, microbeam x-ray fluorescence, and Fourier Transform Infrared Spectroscopy (FTIR) for measurements of microstructure, distribution of metals on individual sediment particles, and chemical forms of the contaminants on a micrometer scale. Typical results obtained with these techniques are presented.

An effort is underway at Pacific Northwest National Laboratory (PNNL) to develop a fundamental and general framework to foster the science and technology needed to support real-time monitoring of early degradation in materials used in the production of nuclear power. The development of such a capability would represent a timely solution to the mounting issues operators face with materials degradation in nuclear power plants. The envisioned framework consists of three primary and interconnected “thrust” areas including 1) microstructural science, 2) behavior assessment, and 3) monitoring and predictive capabilities. A brief state-of-the-art assessment for each of these core technology areas is discussed in the paper.

    1991-09-20

The 40-acre Chemsol site is a former solvent recovery and waste reprocessing facility in Piscataway Township, Middlesex County, New Jersey. Land use in the area is predominantly commercial and residential, with an onsite marshy area that may be considered a wetlands. The site overlies a bedrock aquifer that is used as a regional drinking water source. Between 1980 and 1990, sampling of residential wells indicated the presence of organic contaminants and PCBs. The Record of Decision (ROD) provides an interim remedy to restrict the offsite migration of highly contaminated ground water. The primary contaminants of concern affecting the ground water are VOCs including benzene, toluene, and xylenes; other organics including pesticides and phenols; and metals including arsenic, chromium, and lead. The selected remedial action for this interim remedy includes installing a ground water collection trench; installing three ground water extraction wells to a depth of 130 feet; constructing an onsite treatment plant and treating contaminated ground water using air stripping, biological filtration, and activated carbon adsorption.

Beginning in 1969, Evor Phillips used the site for hauling activities until leasing the property to North American Metal and Chemical Company (NAMCC) in 1971. Silver recovery operations were conducted at the site by NAMCC. Additionally, the Naval Ammunition Depot Earle sent 2,000 gallons of drummed spent potassium hydroxide to the NAMCC. The ROD addresses an interim remedy for contaminated ground water at the site and a final remedy for the drum disposal areas as OU1. The primary contaminants of concern affecting the debris, and ground water are VOCs, including benzene, TCE, PCE, and toluene; other organics; and inorganics.

The 200-acre Kin-Buc Landfill consists of several inactive disposal areas and is located in Edison Township, Middlesex County, New Jersey. Land use in the area is predominantly industrial and commercial, with some residences within 2 miles north of the site. No drinking water supply wells are located within a 2-mile radius of the site. As a result of an oil spill in 1976, EPA conducted an investigation of the property. In 1980, clean-up activities were initiated under the Clean Water Act and included removal, treatment, and disposal of leachate and drummed waste. The ROD addresses a final remedy for OU2 consisting of the sediment and groundwater in the Edmonds Creek wetlands area, Mill Brook/Martins Creek, Mound B, and the low-lying area. The primary contaminants of concern affecting the sediment and ground water are VOCs, including benzene and xylenes; other organics, including PAHs, PCBs, and pesticides; and metals, including arsenic and lead. The selected remedy for the site are included.

Particle physics aims to understand the universe around us. The Standard Model of particle physics describes the basic structure of matter and forces, to the extent we have been able to probe thus far. However, it leaves some big questions unanswered. Some are within the Standard Model itself, such as why there are so many fundamental particles and why they have different masses. In other cases, the Standard Model simply fails to explain some phenomena, such as the observed matter-antimatter asymmetry in the universe, the existence of dark matter and dark energy, and the mechanism that reconciles gravity with quantum mechanics. These gaps lead us to conclude that the universe must contain new and unexplored elements of Nature. Most of particle and nuclear physics is directed towards discovering and understanding these new laws of physics. These questions are best pursued with a variety of approaches, rather than with a single experiment or technique. Particle physics uses three basic approaches, often characterized as exploration along the cosmic, energy, and intensity frontiers. Each employs different tools and techniques, but they ultimately address the same fundamental questions. This allows a multi-pronged approach where attacking basic questions from different angles furthers knowledge and provides deeper answers, so that the whole is more than a sum of the parts. A coherent picture or underlying theoretical model can more easily emerge, to be proven correct or not. The intensity frontier explores fundamental physics with intense sources and ultra-sensitive, sometimes massive detectors. It encompasses searches for extremely rare processes and for tiny deviations from Standard Model expectations. Intensity frontier experiments use precision measurements to probe quantum effects. They typically investigate very large energy scales, even higher than the kinematic reach of high energy particle accelerators. The science addresses basic questions, such as: Are there new sources of CP violation? Is there CP violation in the leptonic sector? Are neutrinos their own antiparticles? Do the forces unify? Is there a weakly coupled hidden sector that is related to dark matter? Do new symmetries exist at very high energy scales? To identify the most compelling science opportunities in this area, the workshop Fundamental Physics at the Intensity Frontier was held in December 2011, sponsored by the Office of High Energy Physics in the US Department of Energy Office of Science. Participants investigated the most promising experiments to exploit these opportunities and described the knowledge that can be gained from such a program. The workshop generated much interest in the community, as witnessed by the large and energetic participation by a broad spectrum of scientists. This document chronicles the activities of the workshop, with contributions by more than 450 authors. The workshop organized the intensity frontier science program along six topics that formed the basis for working groups: experiments that probe (i) heavy quarks, (ii) charged leptons, (iii) neutrinos, (iv) proton decay, (v) light, weakly interacting particles, and (vi) nucleons, nuclei, and atoms. The conveners for each working group included an experimenter and a theorist working in the field and an observer from the community at large. The working groups began their efforts well in advance of the workshop, holding regular meetings and soliciting written contributions. Specific avenues of exploration were identified by each working group. Experiments that study rare strange, charm, and bottom meson decays provide a broad program of measurements that are sensitive to new interactions. Charged leptons, particularly muons and taus, provide a precise probe for new physics because the Standard Model predictions for their properties are very accurate. Research at the intensity frontier can reveal CP violation in the lepton sector, and elucidate whether neutrinos are their own antiparticles. A very weakly coupled hidden-sector that may comprise the dark matter in the universe could be discovered. The search for proton decay can probe the unification of the forces with unprecedented reach and test sacrosanct symmetries to very high scales. Detecting an electric dipole moment for the neutron, or neutral atoms, could establish a clear signal for new physics, while limits on such a measurement would place severe constraints on many new theories. This workshop marked the first instance where discussion of these diverse programs was held under one roof. As a result, it was realized that this broad effort has many connections; a large degree of synergy exists between the different areas and they address similar questions. Results from one area were found to be pertinent to experiments in another domain.

Here, the presumptive function for alpha-tocopherol (αtoc) in membranes is to protect polyunsaturated lipids against oxidation. Although the chemistry of the process is well established, the role played by molecular structure that we address here with atomistic molecular-dynamics simulations remains controversial. The simulations were run in the constant particle NPT ensemble on hydrated lipid bilayers composed of SDPC (1-stearoyl-2-docosahexaenoylphosphatidylcholine, 18:0-22:6PC) and SOPC (1-stearoyl-2-oleoylphosphatidylcholine, 18:0-18:1PC) in the presence of 20 mol % αtoc at 37°C. SDPC with SA (stearic acid) for the sn-1 chain and DHA (docosahexaenoic acid) for the sn-2 chain is representative of polyunsaturated phospholipids, while SOPC with OA (oleic acid) substituted for the sn-2 chain serves as a monounsaturated control. Solid-state 2H nuclear magnetic resonance and neutron diffraction experiments provide validation. The simulations demonstrate that high disorder enhances the probability that DHA chains at the sn-2 position in SDPC rise up to the bilayer surface, whereby they encounter the chromanol group on αtoc molecules. This behavior is reflected in the van der Waals energy of interaction between αtoc and acyl chains, and illustrated by density maps of distribution for acyl chains around αtoc molecules that were constructed. An ability to more easily penetrate deep into the bilayer is another attribute conferred upon the chromanol group in αtoc by the high disorder possessed by DHA. By examining the trajectory of single molecules, we found that αtoc flip-flops across the SDPC bilayer on a submicrosecond timescale that is an order-of-magnitude greater than in SOPC. Our results reveal mechanisms by which the sacrificial hydroxyl group on the chromanol group can trap lipid peroxyl radicals within the interior and near the surface of a polyunsaturated membrane. At the same time, water-soluble reducing agents that regenerate αtoc can access the chromanol group when it locates at the surface.

Future federal legislation and/or regulation may further limit CO 2 emissions from U.S. power generation. Efforts to control CO 2 emissions from this sector are under- way through the development of carbon capture and storage (CCS) technologies.
