Abstract
Labelled ..cap alpha..-D-glucan separated from photoautotrophic organisms is introduced into a reaction with 5 to 25% methanolic trifluoroacetic acid in the presence of 2-4% water related to the weight of the methanol. The reaction takes place in a closed system at a temperature of 85 to 100 degC over a period of 4 to 24 hours. Examples are given of the preparation of methyl-..cap alpha..-D-glucopyranoside under various reaction conditions. The use of trifluoroacetic acid at acidic methanolysis of ..cap alpha..-D-glucan is kinder than the use of the usual mineral acids. The processing of the mixture following the reaction is simpler, too - trifluoroacetic acid may be removed by vacuum distillation. Another advantage of the method is the reduction of losses connected with the decomposition of D-glucose and the product and with the processing of the reaction mixture. Labelled methanol may be used instead of labelled ..cap alpha..-D-glucan.
