Abstract
For successful biological wast water treatment informations about the specific enzyme systems involved in the detoxification of the xenobiotics are of great importance. Mineralisation of methyl-substituted aromatics can mainly be achieved by microorganisms with the ability to induce the meta-cleavage of the key-intermediate catechol, as otherwise - in the case of ortho-fission of the armoatic ring - degradation will stop. Pseudomonas sp. T7 isolated as a toluidine-degrading bacterium mineralised all three isomers of the toluidines as single substrates as well as in mixtures. In contrast to other isolated microorganisms, the extraordinary capability of strain T7 of additionally detoxifying aniline, phenol and cresols was shown to be enabled by both the specific meta-cleavage pathway and the broad substrate specificity of the initial enzymes (toluidine oxygenase and phenol hydroxylase). Due to the ability of a complete and simultaneous degradation of xenobiotic compounds and the wide substrate utilisation spectrum strain T7 might be of special interest for the detoxification of mixtures of these hazardeous zenobiotic compounds. Aufgrund dieser Tatsache liefern enzymatische Untersuchungen, z.B. die Catecholdioxygenasen betreffend, wertvolle primaere Informationen ueber den zu erwartenden Verlauf des Abbaus, ob naemlich in dem Stamm der ortho-Spaltungsweg (ueber Catechol-1,2-dioxygenase, C12D) oder der meta-Spaltungsweg (ueber Catechol-2,3-dioxygenase, C23D) induziert wird. Der von uns aus Xenobiotika-belasteten Boeden isolierte Bakterienstamm Pseudomonas sp. T7 zeigte die Faehigkeit, die drei Isomere des Toluidins als Einzelsubstanzen und auch als Gemisch zu mineralisieren. Zusaetzlich zum Abbau der Toluidine kann dieser Stamm auch Anilin, Phenol und die drei Kresole detoxifizieren und weist somit ein im Vergleich zu anderen von uns isolierten Staemmen aussergewoehnlich bereits Abbaupotential auf. Dies konnte aufgrund enzymatischer Untersuchungen einerseits auf die stammspezifische Induktion des meta-Spaltungsweges und zusaetzlich auf eine breite Substratspezifitaet der entsprechenden Primaerenzyme (Phenolydroxylase bzw. Anilinoxygenase) zurueckgefuehrt werden. (orig.)}
