Abstract

Desiodation of thyroid hormones results in the formation of iodides and of thyronine (T{sub 0}), the description of which has been based on tissue sections or on a thyroxine desiodase preparation. We have provided direct evidence of this process: 3,5,3'-triiodothyronine and 3,3',5'-triiodothyronine, each tritiumlabelled at alpha-beta and at 3, result - inter alia, in the kidney - in the formation of T{sub 0}. The identification of T{sub 0} as a natural derivative of thyroid hormones justified the study of its tissue metabolism; T{sub 0} which was tritium-labelled at 3,5 was incubated in-vitro with sections of kidney and muscle. By liquid scintillation radiochromatogryphy and by radioautography we identified: (1) tyrosine, whose formation is evidence of the rupture of the diphenylether bridge of T{sub 0}; (2) 3'-hydroxythyronine and 3,4-dihydroxyphenylalanine, whose presence shows that 0-hydroxylation occurs in the catabolism of the cycles; (3) thyroacetic acid and p-hydroxyphenylacetic acid, whose appearance is in keeping with the general scheme of alpha-aminated acid degradation. (author) [Spanish] La eliminacion de yodo de las hormonas tiroideas da origen a yoduros y a tironina (T{sub 0}) que el autor ha caracterizado en cortes de tejido o en un preparado de tiroxinodesyodasa. El autor aporta una prueba directa de este proceso: la 3, 5, 3'-triyodotironina y la 3, 3', 5'-triyodotironina, marcadas, respectivamente, en {alpha}, {beta} y en 3 con {sup 3}H conducen a la formacion de T{sub 0} y de otros compuestos en el rinon. La identificacion de la T{sub 0} como derivado natural de las hormonas tiroideas justificaba el estudio de su metabolismo en los tejidos; el autor incubo T{sub 0}, marcada en 3 y 5 con {sup 3}H, junto con cortes de rinon y de musculo. Por radiocromatografia con contador de centelleo liquido y por autorradiografia logro determinar: 1) Tirosina, cuya formacion constituye una prueba de la ruptura del puente difenileter de la T{sub 0}. 2) 3'-hydroxitironina y 3,4-dihidroxifenilalanina, compuestos cuya presencia indica que en el catabolismo de los cielos intervienen procesos de O-hidroxilacion. 3) Los acidos tiroacetico y p-hidroxifenilacetico, cuya aparicion concuerda con el esquema general de la degradacion de los a-aminoacidos. Privoditsja neposredst- vennoe dokazatel'stvo jetogo processa: 3,5 i 3's trehiodistym tironinom i 3. 3' i 5 s trehiodistym tironinom mechennyh sootvetstvenno i al'fa i beta luchami i 3 mechennogo tritiem, vedut, mezhdu prochim, k obrazovaniju v pochke tironina. Identifikacija tironina v kachestve estestvennogo produkta shhitovidnyh gormnov podtverzhdaet issledovanie ego tkanevogo metabolizma; mechennyj tri- tiem tironin v 3 i 5 byl poluchen v laboratornyh uslovijah (in-vitro) putem razrezov pochki i myshcy. Harakternye osobennosti opredeljalis' zhidkoscintil- -ljacionnym radiohromatografom radioavtografiej : 1. Tirozin, obrazovanie kotorogo svidetel'stvuet o razryve svjazujushhego (difeniletornogo) mosta tironina. 2. 3' s gidroksitironinom i 3 i 4 s bigidroksifenilalaninom, nalichie koto- ryh illjustriruet uchastie processa d'gidroksilacii v katabolizme ciklov. 3. Tiroacetichnye i rgidroksifenilaceticheskie kisloty, pojavdenie kotoryh otvechaet obshhej sheme ponizhenija al'fa- amidirovannyh kislot. (author)}
