Abstract

Tritium, with its low energy beta-emission produces high-resolution autoradiograms (ARG). H{sup 3}TDR is chemically stable, labels desoxyribonucleic acid (DNA) and thereafter is non-exchangeable. The DNA is diluted only by new DNA at the time of chromosomal replications. The intensity of the label is thus diminished by successive cell divisions. The behaviour of the labelled population is apparently consistent with the Taylor-Woods-Hughes (TWH) model of chromosome replication. Intuitively one expects to observe radiation toxicity when radio-active elements have been incorporated into DNA. The toxicity has been considered in respect to the properties of tritium and influence of chromosomal replication (TWH model), and studied empirically in mammalian testes and lymphocytes. After doses satisfactory for serial autoradiographic studies, radiation injury or perturbations in cell proliferation have not been demonstrated. To date normal hemopoiesis, acute and chronic leukaemias, multiple myeloma and selected solid tumours have been studied in man and animals. Serial data on the fraction of cells labelled and the rate of diminution in the intensity of the label have been collected. These data will be presented. These problems will be discussed in principle and practice. It has been established tentatively that growth rate for some normal proliferating organ systems in steady state equilibrium is greater than the growth rate in many expanding tumours. (author) [French] Le tritium, emetteur beta de faible energie, permet d'obtenir des autoradiogrammes de haute resolution. La thymidine tritiee est chimiquement stable; apres avoir marque l'acide desoxyribonucleique (ADN), elle n'est plus echangeable. L'ADN n'est dilue que par un nouvel ADN, au moment du dedoublement chromosomique. L'intensite de l'indicateur se trouve ainsi diminuee par les divisions cellulaires successives. Le comportement de la population marquee est apparemment conforme au modele de dedoublement chromosomique de Taylor, Woods et Hughes (TWH). Intuitivement, on s'attend a observer une radiotoxicite lorsque des elements radioactifs ont ete incorpores a l'ADN. .Les auteurs ont examine la toxicite eu egard aux proprietes du tritium et a l'influence des dedoublements chromosomiques (modele TWH) et ils l'ont etudiee empiriquement dans les testicules et les lymphocytes de mammiferes. Apres irradiation a des doses suffisantes pour des etudes autoradiographiques en serie, aucune radiolesion ni perturbation dans la proliferation des cellules n'a ete constatee. Jusqu'a present, les auteurs ont etudie l'hematopoieese, les leucemies aiguee et chronique, le myelome multiple et diverses tumeurs solides, chez l'homme et les animaux. Ils presentent dans leur memoire des series de donnees recueillies sur le fractionnement des cellules marquees et le rythme de decroissance de l'intensite de l'indicateur. L'interpretation de ces donnees est compliquee du fait qu'il est difficile d'identifier les cellules - d'etablir des limites morphologiques entre les stades successifs de la proliferation cellulaire depuis la cellule initiale jusqu'a la progeniture non sujette a division - et aussi du fait qu'il est impossible de determiner si une cellule qui n'est pas en etat de mitose ou de synthese d'ADN reste capable d'une nouvelle division. Les auteurs examinent ces problemes aux points de vue theorique et pratique. Ils ont etabli provisoirement que le rythme de croissance est plus rapide chez certains systemes organiques a proliferation normale en equilibre stable que chez de nombreuses tumeurs en expansion. La {sup 3}HTDR es quimicamente estable, marca el acido desoxirribonucleico (ADN) y no experimenta intercambio ulterior. El ADN solo es diluido por nuevo ADN en el momento de producirse las duplicaciones cromosomaticas. Por tanto, la intensidad de la marcacion disminuye a medida que tienen lugar las sucesivas divisiones celulares. Al parecer, el comportamiento de la poblacion marcada es compatible con el modelo Taylor-Woods-Hughes (TWH) de duplicacion cromosomatica. La intuicion sugiere que el ADN adquiere radiotoxicidad cuando se le incorporan elementos radiactivos. Se ha estudiado la toxicidad en relacion con las propiedades del tritio y la influencia de la duplicacion cromosomatica (modelo TWH), y empiricamente en testiculos y linfocitos de mamiferos. Despues de administrar dosis suficientes para poder realizar estudios autorradiograficos en serie, no se han observado radiolesiones ni perturbaciones en la proliferacion celular. Hasta la fecha se han estudiado, tanto en el hombre como en animales, la hematopoyesis normal, leucemias agudas y cronicas, mieloma multiple y determinados tumores solidos. Se ha reunido una serie de datos sobre la fraccion de las celulas marcadas y la velocidad de disminucion de la intensidad de la marcacion. En la memoria se presentan esos datos. Su interpretacion resulta complicada a consecuencia de las restricciones impuestas por la identificacion citologica y de la determinacion de limites morfologicos entre fases sucesivas de la proliferacion celular pasando de la celula originaria a la progenie que no sufre division, asi como de nuestra incapacidad para determinar si una celuia que no este en estado de mitosis o en fase de sintesis de ADN conserva su capacidad para volver a dividirse. En la memoria se examinan los aspectos teorico y practico de estos problemas. Se ha establecido provisionalmente que la velocidad de crecimiento correspondiente a algunos conjuntos organicos de proliferacion normal en un estado de regimen estacionario es superior a la velocidad de crecimiento en muchos tumores expansivos. Posle vvedenija dozy, neobhodimoj dlja obychnyh radio- avtograficheskih issledovanij, v processe razmnozhenija kletok ne nabljudalos' ni radiacionnogo porazhenija, ni kakih-libo drugih narushenij. Do nastojashhego vremeni na cheloveke i zhivotnyh byli izucheny javlenija normal'nogo krovotvore- nija, ostroj i hronicheskoj lejkomii, mnogostoronnej mielomy i nekotoryh tverdyh opuholej.
