Abstract

Tritium-labelled progesterone, as a precursor of the 17-OH-corticosteroids, was used to study the biosynthesis of placental and adrenal steroids. Homogenates from cattle adrenals were incubated with H{sup 3}-progesterone for 3 h in an O{sub 2} atmosphere. Then corticosteroids were extracted according to Meyer's method and chromatographically prepared in Forisil columns and delta 4-3-ketosteroids were localized on chromatographic paper. The latter was soaked in a nuclear emulsion (Ilford K2 ''gel form'') and was kept at 4 Degree-Sign C for 21 d, allowing the formation of H{sup 3}-cortisone, H{sup 3}-cortisol and of other steroidal-compounds while the photographic development was made evident. The same incubation technique was used in the research on placental homogenates which revealed the radioactivity of recently produced 11-OH-corticoids by means of a gas-filling counter of the single chromatographic spots. Ces chromatographies ont ete ensuite immergees dans une emulsion nucleaire (Ilford K 2 'gel form') et maintenues pendant 21 jours a une temperature de 4 Degree-Sign C pour permettre l'identification de cortisone tritiee, de Cortisol tritie et d'autres steroiedes au cours du processus photographique. On a utilise la meme methode d'incubation pour la recherche sur des homogenats placentaires; l'examen des taches chromatographiques a l'aide d'un compteur a atmosphere gazeuse a revele la radioactivite des 11-hydroxycorticoiedes recemment obtenus. (author) [Spanish] Los autores han realizado estudios sobre la biosintesis de los esteroides suprarrenales y placentarios empleando progesterona marcada con tritio como precursor de los 17 hidroxicorticosteroides. Incubaron durante tres horas, en atmosfera de O{sub 2} , homogeneizados de capsulas suprarrenales de vacunos junto con progesterona tritiada. Seguidamente extrajeron los corticosteroides por el metodo de Meyer y los separaron por cromatografia en columna de Florisil, determinando los {Delta}-4-3-cetosteroides por cromatografia sobre papel. Sumergieron estas ultimas en una emulsion nuclear (Ilford K2 ''gel form'') y los mantuvieron durante 21 dias a 4 Degree-Sign C para lograr la identificacion de cortisona y Cortisol tritiados y de otros esteroides durante el proceso fotografico. Para los homogeneizados de placenta emplearon la misma tecnica que, con ayuda de un contador de gas, permitio detectar en las manchas del cromatograma la radiactividad de los 11-hidroxicorticoides de formacion reciente. Zatem izvlekalis' korti- kosteroidy po metodu Mejera i prigotavlivalis' hromatograficheski v florizi- lovyh kolonkah i na hromatograficheskoj bumage lokalizovalis' del'ta-4-3- ketosteroidy. Poslednjaja propityvalas' jadernoj jemul'siej (Ilford K 2 'forma gelja') i vyderzhivalas' pri temperature 4 Degree-Sign S v techenie 21 dnja, chto obespechivalo for- mirovanie {sup 3}N-kortizona, {sup 3}N-kortizola i drugih steroidal'nyh soedinenij, kogda stalo ochevidnym fotograficheskoe projavlenie. Podobnyj metod inkubacii ispol'zovalsja v issledovanijah placentnyh gomogenatov, kotorye obnaruzhivajut radioaktivnost' nedavno poluchennyh 11-ON- kortikoidov, opredelennuju na odinochnyh hromatograficheskih pjatnah s pomoshh'ju zapolnennogo gazom schetchika.
