Abstract

Insects generally have been found to require a dietary source of sterol for normal larval growth and metamorphosis. Our work has pointed to two additional physiological roles for sterols in the housefly, Musca domestica L.: (1) A dietary source of sterol is essential for sustained viable egg production in the female fly; on a sterol-deficient diet eggs are produced but hatch and viability are low. (2) Cholesterol is also involved in the mobilization and utilization of nutrient reserves associated with the initiation of ovarian maturation in the female fly. The quantitative sterol requirements for the above physiological processes and the metabolic conversions that occur during growth, metamorphosis and reproduction have been studied in this insect, using C{sup 14}- and H{sup 3}-labelled sterols in conjunction with a variety of analytical tools, including reverse isotope dilution, gasliquid chromatography and spectroscopy, and employing aseptic rearing techniques and semi-defined larval and Adult diets. Both C{sup 14}-cholesterol and H{sup 3}-{beta}- sitosterol have been used as a source df sterol in either the larval or the adult diet of the house fly, and the pattern of utilization and metabolism was found to be almost identical for these two sterols. The utilization and fate of the 'sparing sterol' has been investigated using C{sup 14} cholestanol, and the metabolism of the minute quantity of essential cholesterol is currently under study using high-specific-activity C{sup 14} cholesterol. - Other species of insects, including the German cockroach (Blattella germanica), have been examined in relation to the patterns of utilization and the metabolic pathways for sterols found in the house-fly. (author) [French] On a determine que, de facon generale, les insectes ont besoin de sterols dans leur regime alimentaire pour que leurs larves se developpent et se metamorphosent normalement. Les travaux de l'auteur ont fait apparaitre deux autres fonctions physiologiques des sterols chez la mouche domestique (Musca domestica L.): 1. il est indispensable que le regime alimentaire de la femelle contienne des sterols pour que celle-ci ponde regulierement des oeufs viables; s'il y a carence de sterols, la femelle continue a pondre mais seul un faible pourcentage des oeufs parviennent a eclosion et sont viables; 2. le cholesterol joue egalement un role dans l'utilisation des reserves nutritives dont s'accompagne le debut du developpement des ovaires. On a etudie chez cet insecte les quantites de sterols qui sont necessaires pour que les processus physiologiques ci-dessus se deroulent, ainsi que les transformations metaboliques qui se produisent au cours du developpement, de la metamorphose et de la reproduction. Cette etude a ete faite a l'aide de sterols marques par {sup 14}C et {sup 3}H, en appliquant diverses methodes analytiques: dilution isotopique inversee, chromatographie a phase gazeuse et phase liquide, spectroscopie; on a eu recours a des methodes d'elevage en milieu aseptique et on a etabli des regimes alimentaires semi-controles pour les larves et les adultes. Le cholesterol marque par {sup 14}C et le sitosterol-{beta} marque par {sup 3}H ont ete utilises comme sources de sterols dans le regime alimentaire de la mouche domestique aux stades larvaire et adulte; il s'est revele que l'utilisation et le metabolisme de ces deux sterols etaient a peu pres identiques. Toutefois, on n'a pas observe de transformation du sitosterol- {beta} en cholesterol. Dans le regime alimentaire des larves on a egalement fait entrer du cholesterol en quantite inferieure aux besoins minima et on y a ajoute des ''sterols pauvres'' comme le cholestanol, qui ne satisfont que partiellement les besoins en sterols de l'insecte. L'utilisation et le sort du ''sterol pauvre'' ont ete etudies a Taide de cholestanol marque par {sup 14}C; a l'aide de cholesterol marque par {sup 14}C d'activite specifique elevee, on poursuit les recherches sur le metabolisme de la microquantite de cholesterol indispensable. On a etudie chez d'autres especes d'insectes, notamment la blatte allemande (Blattela germanica), l'utilisation et les voies metaboliques des sterols et on les a comparees a celles qui ont ete observees chez la mouche domestique. (author) [Spanish] Se ha descubierto que en general los insectos necesitan consumir habitualmente esterol para que el desarrollo de las larvas y las metamorfosis se efectuen en condiciones normales. El presente trabajo se refiere a otras dos funciones fisiologicas del esterol en la mosca comun (Musca domestica L.), que pueden concretarse del modo siguiente: 1) Las hembras necesitan consumir habitualmente esterol para poder producir huevos fertiles; en caso de consumo insuficiente, loe huevos son de escasa fertilidad y viabilidad; 2) El colesterol intervienne tambien en el mecanismo de utilizacion de las reservas nutritivas en la mosca hembra cuando se inicia la maduracion ovarica. La cantidad de esterol necesaria para el proceso fisiologico indicado y las transformaciones metabolicas que se producen durante el desarrollo, la metamorfosis y la reproduccion han sido estudiadas en este insecto empleando esteroles marcados con {sup 14}C y {sup 3}H; al mismo tiempo se ha hecho uso de diversos procedimientos de analisis, entre ellos la dilucion isotopica reciproca, la cromatografia gas-liquido y la espectroscopia, y se han utilizado procedimientos de cria en medio aseptico y dietas semidefinidas de larvas y adultos. Como fuente de esterol en la dieta de los individuos adultos o de las larvas de mosca comun, se emplearon el colesterol-{sup 14}C y el sitosterol-{beta}-{sup 3}H, habiendo sido casi identicos en ambos casos el proceso de asimilacion y metabolismo de estos esteroles. Sin embargo, no hubo transformacion apreciable de {beta}-sitosterol en colesterol. En las dietas de las larvas se utilizaron tambien cantidades de colesterol inferiores al minimo necesario, combinadas con ''esteroles de ahorro''', como el colestanol, que pueden suministrar solo una parte del esterol que necesita el insecto. La asimilacion y el destino final del ''esterol de ahorro'' se han estudiado empleando colestanol-{sup 14}C; tambien se esta estudiando el metabolismo de la-minuscula cantidad de colesterol esencial, empleando para ello colesterol-{sup 14}C de elevada actividad especifica.
