Abstract
The relationship between cell viability and DNA-single and double-strand breaks induced by UV-reductone treatment of E.coli AB1157 and AB1886 cells was studied. A molecular and quantitative analysis was done by sedimentation in sucrose gradients. Bacteriophage T4-DNA was submitted to similar conditions. The two bacterial strains showed the same kinetics of single strand-breaks induction by reductone, in agreement with the hypothesis that the repair of these lesion is independent of uvr A uvr B genes product. This paper suggests that the `UV-sensibilization` to reductone action is linked to a synergistic process between reductone induced single breaks and the enzymatic incisions accumulated by modification in the excision repair, during the treatment.
