Title: pH-sensitive hydrogels

Abstract

This work dealt with the diffusant release from the polyvinyl-pyrrolidone-polyacrylic acid (PVP-PAA) semi-interpenetrating network (semi-IPN) film when the film was placed in separate aqueous dissolution media with various pH values. The pH effect on the swelling behavior of the film and the rates of diffusant release from the film were studied. The PVP-PAA semi-IPN films and the PVP-PAA complexes were prepared from photopolymerization of the mixture of PVP and acrylic acid, in the presence of benzin methyl ether. The PVP-PAA complexes were characterized by means of DSC and FT-IR. The PVP-PAA semi-IPN films with various percentages of a crosslinking agent were investigated. The study of pH effect on the swelling of the semi-IPN film was carried out in 0.1 N HCl solution, pH 3.0 and 6.0 buffers. The swelling rate of PVP-PAA semi-IPN film in pH 6.0 buffer was much higher than the rates in 0.1 N HCl and pH 3.0 buffer. The chemical to be released from the film was incorporated during the film preparation and the diffusant used was either caffeine (hydrogen-bonding-acceptor) or salicylamide (hydrogen-bonding-donator). The diffusant release from the PVP-PAA semi-IPN in 0.1 N HCl solution and in the phosphate buffers with various pH values was investigated.more » Release rate was faster in high pH media. The chemical valve function of the PVP-PAA semi-IPN film in controlling release rate was studied by alternating the dissolution medium between 0.1N HCl solution and pH 6.0 buffer. Consistently, the release rate increased when the dissolution medium was changed from 0.1 N HCl solution to pH 6.0 buffer, and the rate dropped while the medium was was switched from pH 6.0 buffer to 0.1 N HCl solution. Finally, the effects of the type of crosslinking agent, the percentage of crosslinking agent used, and the molar ratio of PVP/AA on the diffusant release from the PVP-PAA semi-IPN film were explored.« less

  • The author has characterized pertussis toxin-sensitive G proteins in the nervous systems of the gastropod mollusc Aplysia and the cephalopod Loligo using ({sup 32}P)ADP-ribosylation and immunoblotting with G protein specific antisera. As in vertebrates, this class of G protein is associated with membranes and enriched in nervous tissue in Aplysia. Analysis of dissected Aplysia ganglia reveal that it is enriched in neuropil, a region containing most of the central nervous system synapses. Because both Aplysia and Loligo synaptosomes are enriched in pertussis toxin-sensitive G proteins, it is likely that they are found in synaptic terminals. Fractionation of Aplysia synaptosomes intomore » membrane and vesicle fractions reveals that, although the majority of G protein is recovered in the plasma membrane fraction, a small proportion is recovered in the vesicle fraction. He shows that G proteins are on intracellular membranes by ADP-ribosylating extruded axoplasm with pertussis toxin. A plausible explanation for vesicular localization of G protein in axoplasm is that G proteins are transported to terminals on vesicles. He has shown, using ligature experiments with Aplysia connectives and temperature block experiments in the giant axon of Loligo, that G proteins move by anterograde fast axonal transport. Injection of pertussis toxin into the identified Aplysia neuron L10 blocks histamine-induced presynaptic inhibition of transmitter release. This suggests that pertussis toxin sensitive G proteins play a role in modulating transmitter release at synaptic terminals. In the giant synapse of Loligo, he presents preliminary data that demonstrates that the activation of G proteins in the presynaptic terminal results in decreased transmitter release.« less
  • The determination of level populations and detailed population mechanisms in dense plasmas has become an increasingly important problem in atomic physics. In the present work, the density variation of line intensities and level populations in aluminum K-shell and molybdenum and silver L-shell emission spectra have been measured from high-powered, laser-produced plasmas. For each case, the density dependence of the observed line emission is due to the effect of high frequency electron-ion collisions on metastable levels. The density dependent line intensities vary greatly in laser-produced plasmas and can be used to extract detailed information concerning the population kinetics and level populationsmore » of the ions. In aluminum, the electron density dependence of various helium-like line intensity ratios were measured. Standard collisional radiative equilibrium models fail to account for the observed density dependence measured for the He{sub {alpha}}/IC ratio. Instead, a quasi-steady state atomic model based on a purely recombining plasma is shown to accurately predict the measured density dependence. This same recombining plasma calculation successfully models the density dependence of the high-n He{sub {gamma}}/He{sub {beta}} and He{sub {delta}}/He{sub {beta}} helium-like resonance line intensity ratios. The density dependence of metastable levels of neon-like ions is also measured. Detailed neon-like theoretical atomic model calculations are shown to correctly predict the observed density sensitivity of the electric quadrupole to dipole-allowed resonance line x/3A ratio. The measured density dependence of the 3A/3G resonance line ratio does not agree with theory.« less
  • In this study, the authors investigate the effects of elasticity and nonsingular stress terms on the asymptotic plane-stress mode 1 crack-tip fields for pressure-sensitive materials under small scale yielding. Perfectly plastic behavior is assumed and plastic dilatancy is introduced by the normality flow rule. A closed-form general asymptotic solution for singular centered fan sectors is given as a function of mu which is a pressure sensitivity parameter introduced in the yield condition. When elastic perfectly plastic behavior is considered, the results of finite element computations show the existence of elastic sectors bordering the stress-free crack faces. The near-tip stresses ofmore » the finite element results agree well with those of the corresponding asymptotic analysis. The angular spans of the elastic and plastic sectors vary with the value of mu. They also vary, for a specific pressure sensitivity parameter, with the value of the nonsingular stress term (T-stress) in the asymptotic expansion of the linear elastic crack-tip stresses, which is the normal stress in the direction of the crack line. In particular, the elastic sector span is in general shown to decrease with increasing T-stress. The parameter mu as well as the nonsingular stress term also have significant effects on the sizes and shapes of the plastic zones. The contribution of hydrostatic stress in the yield criterion for this class of pressure-sensitive materials extends the boundary of the plastic zone much farther in front of the crack-tip than that for incompressible Mises materials. Also, as the T-stress increases, the height of the plastic zone increases substantially for materials with a large pressure sensitivity. In zirconia ceramics, the dilatational phase transformation strain from tetragonal to monoclinic phase is limited to 4 percent. This condition is also considered in this study.« less
  • Artificial Neural Networks (ANN) are parallel distributed models which are capable of performing nonlinear modeling and adaptation without making functional assumptions. This thesis presents a non-fully connected ANN model with cascaded training strategy for forecasting weather sensitive electric loads. The proposed model is capable of forecasting the hourly load for an entire week and has been implemented on real load data. The average absolute forecast errors of the one hour ahead forecast in the Winter and Summer test cases are shown to be 3.42% and 3.19% respectively; the average absolute forecast errors at peak in both test cases are shownmore » to be 1.19% and 3.45%. The model is superior to the conventional statistical method in model life cycle length, degree of recursiveness and accuracy. With non-fully connected network structure, the network training time is reduced and the degree of robustness is enhanced. By applying the cascaded learning strategy, the network performance is improved. General guidelines in constructing ANN forecasting models are developed. Intensive illustrations are also provided to explain the fundamental behavior of the ANN models in time series applications.« less
  • This project investigated the effects of dihydropyridine calcium channel blockers on behavioral and electrophysiological aspects of ethanol withdrawal. The effects of the dihydropyridine (+)-PN 200-110, on changes in neuronal function during ethanol withdrawal, were compared with effects on changes caused by the GABAergic convulsant drug bicuculline. Behavioral correlates of ethanol withdrawal were measured in two strains of mice using a rating of handling-induced convulsions. Concurrent chronic treatment with ethanol and the dihydropyridine calcium channel blockers ([plus minus])-nitrendipine, ([plus minus])-nimodipine or ([plus minus])-PN 200-110 prevented withdrawal-induced increased in convulsive behavior. This effect was dose dependent. The duration of chronic treatment withmore » calcium channel blocker affected the degree of protection against increases in convulsive behavior seen during ethanol withdrawal. Concurrent chronic treatment with ethanol, and the mixed calcium channel activator/blocker ([plus minus])-BAY K 8644, prevented ethanol withdrawal-induced increases in convulsive behavior. Single acute injections of nitrendipine immediately on cessation of chronic treatment with ethanol, or 2h later, reduced withdrawal-induced increases in convulsive behavior in a dose-dependent manner throughout the 12h test period. Slices isolated from mice after chronic ethanol treatment showed a complex, time-dependent pattern of changes in the above measurements, culminating in epileptiform discharges seen from 4h to 7h into withdrawal.« less
