Title: Influence of Rhodococcus equi on the respiratory burst of resident alveolar macrophages from horses

Abstract

Rhodococcus equi is the etiologic agent of a devastating pneumonia of sporadic incidence in foals. The purpose of this study was to evaluate the influence of R. equi on the superoxide anion production, measured spectrophotometrically as the reduction of cytochrome C, and hexose monophosphate shunt activity, measured by /sup 14/CO/sub 2/ liberation from /sup 14/C-1-D-glucose, of alveolar macrophages from horses. Alveolar macrophages were harvested from 6 anesthetized, healthy, light-breed, adult horses by bronchoalveolar lavage. Following a randomized complete block design, the suspension of cells was divided into aliquots of 10/sup 6/ viable alveolar macrophages which were exposed to 1, 10 or 100 g. of opsonized R. equi or opsonized zymosan A at 37 C for 2 hours. In this study the respiratory burst of equine alveolar macrophages was only evidenced by the hexose monophosphate shunt activity and superoxide anion was not coincidentally produced. Rhodococcus equi did not adversely affect that response. The insignificant superoxide anion production by the alveolar macrophages suggests that this may not be a significant oxygen metabolite in those cells.

  • Pulmonary alveolar macrophages (PAM), lung cells which inactivate bacteria, and, lung lavage fluid, were investigated following in vivo exposure to O/sub 3/. PAM isolated from Sprague-Dawley female rats exposed to 1.4 ppm O/sub 3/ for 3 hours exhibited a decrease in PAM superoxide anion radical (O/sub 2/) production (as measured by the superoxide dismutase inhibitable reduction of ferricytochrone c), phagocytosis (as measured by latex bead ingestion), membrane lipid fluidity (as measured by fluorescence polarization), and number of available sulfhydryl groups on the PAM plasma membrane (as measured by reaction between carboxypyridine disulfide and plasma membrane sulfhydryl groups). Incubation of controlmore » rat PAM with lung lavage fluid (LLF) from rats exposed to 3.0 ppm O/sub 3/ for 3 hours exhibited decreases in the same parameters, as compared to PAM incubated with LLF from control rats.« less
  • Components of the respiratory burst oxidase reside in intracellular membranes of the tertiary granules in resting cells, yet oxidase activity in the activated cells occurs at the neutrophil surface. The role of degranulation in activation of the neutrophil respiratory burst was therefore investigated. Surface labeling experiments were carried out on resting and activated neutrophils using three impermeant labeling methods. Activated neutrophils labeled with (/sup 35/S) diazobenzene sulfonic acid showed a fourfold higher specific radioactivity than resting neutrophils. Similar results were obtained with the pyridoxal phosphate/borotritide labeling method. On the other hand, little difference in labeling was seen using the periodate/borotritidemore » method which detects the carbohydrate of glycoproteins. These results suggest that either a large amount of protein, or a highly reactive protein becomes exposed upon activation. Resting, activated, and enucleated cells were labeled using the (/sup 125/I) lactoperoxidase method, then subjected to polyacrylamide gel electrophoresis. Autoradiograms of these gels showed that two proteins of about 75 and 45 kD, are labeled at the external surface of enucleated and activated cells but not resting cells.« less
  • No effect of the inhibitor on the relative rates of synthesis of the precursor form of either enzyme was observed with either (/sup 35/S)methionine or 2-(/sup 3/H)mannose as the labeled tracer. On the other hand, processing of ..beta..-galactosidase and ..beta..-glucuronidase was markedly altered by swainsonine as manifested by a number of differences in the processed forms. (1) The altered forms had slightly greater electrophoretic mobility than normal forms. (2) Endo-..beta..-N-acetylglucosaminidase H digestion decreased the molecular sizes of the altered forms much more than those of the normal forms. (3) On isoelectric separation of the charge isoforms of ..beta..-glucuronidase, the threemore » most negatively charged forms of the normal processed enzyme were absent in the altered material. This difference was eliminated by neuraminidase treatment of the processed form. These findings are consistent with a blockage by swainsonine of the removal of the ..cap alpha..-1,3 and ..cap alpha..-1,6 linked mannose residues that occurs in normal processing.« less
  • This study explores perceptions of risks to health (POR), risk taking behavior (RTB), and associated illness from the perspectives of medical anthropology and epidemiology. Specific objectives included investigation of the following: (1) general POR: (2) POR from indoor air pollution (IAP) and parental usage of IAP sources; (3) the relationship between POR and RTB; factors affecting POR and RTB; (4) the construction of risk domains; (5) sources of intracultural variation in POR, RTB, and the partitioning of risk domains; (6) patterns of indoor heating in central Michigan; and (7) respiratory illness symptoms (RIS) in children from exposure to indoor heatingmore » with a woodburning stove (WBS). POR and RTB were investigated in random and ethnographic samples, RIS in a random sample with an internal comparison group, and indoor heating in a random sample. Interview schedules were developed for each phase of inquiry. The instrument used to investigate POR and RTB includes both qualitative and quantitative scales for POR assessment. Findings of this study provide an ethnography of risk, an inferential analysis of POR/RTB, and a preliminary assessment of risks of RIS from heating with WBS.« less
