Enzymatic protective mechanism against lipid peroxidation damage to lungs of ozone-exposed rats
Abstract
Rats were fed a vitamin E-deficient diet containing 1 to 1500 mg dl-..cap alpha..-tocopherol acetate per kilogram and then exposed to 0.7 or 0.8 ppM O/sub 3/ for 5 or 7 days. Lung homogenates were analyzed for enzyme activities. O/sub 3/ elevated thiobarbituric acid (TBA) test dreactants, primarily malonaldehyde produced by lipid peroxidation, as well as activities of GSH peroxidase, G-6-P dehydrogenase, and GSH reductase. The first three effects were partially inhibited by increasing dietary tocopherol (log basis) but the last was not affected. Compensation mechanism (homeostasis) to lipid peroxidation is: (a) increase in GSH peroxidase (increased lipid peroxide catabolism), (b) increase in GSH reductase (provides more GSH for (a)), and (c) increase in G-6-P dehydrogenase (provides more Nadph.
