Title: Sex differences in cadmium accumulation in a laboratory population of a wild British insectivore, Sorex araneux

Abstract

A series of feeding trials using laboratory reared common shrews Sorex araneus were undertaken to examine the accumulation of Cd in a known trace metal accumulator. A significant sex difference in Cd accumulation in the liver and kidney was identified. The results are discussed in relation to data on traditional laboratory animals and field studies on wild populations of small mammals. 9 references, 3 figures.

  • The authors have previously described ten cases of astrocytoma (three WHO grade II, four grade III and four grade IV) with seemingly contradictory results on immunohistochemical analysis of the p53 protein and molecular genetic analysis of the p53 gene. Fixed, embedded tissues from these cases were immunohistochemically positive with the PAb 1801 antibody, which supposedly implies the presence of mutant protein. These ten cases, however, did not have mutations in exons 5 through 8 of the p53 gene, the conserved regions in which almost all human mutations have been described. The authors suggested that these cases might either represent overexpressionmore » of wild-type p53 protein (since the PAb 1801 antibody reacts with both wild-type and mutant p53 protein) or mutations in less conserved regions of the gene. To investigate these possibilities further, they performed single strand conformational polymorphism analysis and DNA sequencing on p53 exons 4, 9 and 10 in the nine cases with available DNA, since rare mutations have been noted at these loci. None of the cases showed alterations, making it highly unlikely that these tumors harbor mutations in exons of the p53 gene. They also performed immunohistochemistry on frozen sections from seven available tumors, using the mutant-specific antibody PAb 240 in addition to PAb 1801. All tumors continued to show positive staining with PAb 1801, but only one tumor reacted with PAb 240. The results support the hypothesis that the accumulated p53 protein in most cases is wild-type. Because the product of the MDM2 oncogene can bind to wild-type p53 protein, and because MDM2 amplification has recently been demonstrated in human tumors, the authors evaluated MDM2 amplification in the nine astrocytomas with available DNA. Using slot blot analysis with a 96-base pair, PCR-generated probe to the first exon of the MDM2 gene, they were unable to show MDM2 gene amplification in these tumors or in other assayed astrocytomas.« less
  • In vivo cadmium with copper was investigated using mice exposed to various combinations of Cd and Cu concentrations in drinking water. Animals were sacrificed at 0, 3, 6, 9 and 12 wk and brain, liver, kidney and spleen digests analyzed for cadmium, copper, iron and zinc by flame or flameless atomic absorption spectrophotometry. Tissue Zn, Fe and Cu levels were not altered by Cu or Cd additions to drinking water. Although a dose related increase in Cd was observed in kidney and liver throughout the experiment, a significant decrease was observed in Cd concentrations of these 2 tissues when 100more » ppM Cu was added to 5 ppM Cd in drinking water but not when Cd levels were increased to 25 ppM or when Cu was reduced to 25 ppM. In order to determine the intracellular location of the increased Cd in these tissues, nuclei, microsomes, mitochondria and soluble proteins were isolated from liver and analyzed for Cd and Cu. Cadmium levels in the soluble protein fraction of liver of animals administered 5 ppM Cd were approx. 3-fold greater than that for the same Cd dose when Cu was added. The same was the case for the low molecular weight fraction (TCA supernatant) of liver. At the same time, Cu increased in both the total soluble protein and low mol wt protein fractions of Cd exposed animals when Cu was added. These results suggest an in vivo replacement of Cu for Cd in induced metallothionein-like Cu and Cd binding proteins. Copper at relatively low levels may play an important role in the prevention of accumulation of Cd in liver and kidney; however, this interaction appears to be dependent on a specific Cd:Cu ratio of metal intake.« less
  • Previous work in this laboratory has shown that heavy metal contamination is commonplace in fields and gardens in areas where lead was once mined in Britain. Cadmium and Pb uptake by radish was investigated in field garden plots in northeast Wales (Halkyn Mountain), Derbyshire and the Mendips, including the village of Shipham. Here limestone is the host rock for mineralization and soils tend to have a near neutral reaction. Plots were also established in the non-limestone mining area of Ceredigion where soils are generally acid. The edible root of radish accumulated significant amounts of metals in each area. EDTA provedmore » a useful extractant for estimating plant-available Cd and Pb. There was some evidence that increasing pH decreased the availability of Pb but the relationship for Cd was more complex being non-linear. The Halkyn area has the severest Pb problem and the Mendips the severest Cd problem.« less
