Title: Ozone toxicity effects on normal cultured lung cells and combined effects of ozone and antitumor drugs on normal and tumor cells

Abstract

A system was developed for investigating the effects of ozone (O/sub 3/) on cultured cells. Numerous factors were evaluated in designing this in vitro system and several factors, including composition of the liquid phase, depth of liquid covering cells and gas-liquid reaction products, were found to contribute significantly to the toxicity of O/sub 3/ for cultured rat lung fibroblasts (F-cells). Cell injury caused by different in vitro O/sub 3/ exposure methods was shown to be mediated by different toxic species. Cell injury, was mediated primarily by H/sub 2/O/sub 2/ with some contribution by O/sub 2//sup -/ and OH - depending upon the exposure method. Serum was found to protect from many of these O/sub 3/ reaction products by mechanisms involving protein-bound and free sulfhydryls. Combined treatment with O/sub 3/ and antitumor drugs was investigated for potential selective toxicity for tumor cells. Of the drugs tested, only vitamin K/sub 3/ (Vit K) produced a synergistic increase in toxicity for Walker rat carcinoma (WRC) cells when administered prior to or during O/sub 3/ exposure.
