Title: Role of zinc, copper and cadmium in the regulation of aryl sulfotransferase IV activity in rat liver

Purified rat liver aryl sulfotransferase IV (AST IV) was found to be inhibited in vitro by Zn, Cu, Cd and Tb. Cu, Zn and Cd were all strongly inhibitory at very low concentrations. Cu, Zn and Cd at 2.5{mu}M to 0.025{mu}M could decrease the enzyme activity to {le} 50% Tb also showed a slight inhibitory effect on AST IV activity. In rat liver cytosols, Zn, Cu and Cd at 25{mu}M to .25{mu}M also decreased sulfotransferase activity to {le} 50% of controls. In order to asses the possible effects of these metals in vivo on liver AST IV activity, studies of increased dietary exposure to Zn, Cu and Cd were conducted. Male Sprague-Dawley rats were fed one of twelve diets varied in Zn, Cu and Cd content. The effect of chronic high, Cu and Cd ingestion and short term Zn administration were examined. In the chronic experiment, Cu deficiency resulted in reduction in liver Cu, and high Cu resulted in higher accumulation in liver Cu, but Zn content in diet had less effect in reduction or accumulation in liver tissue Zn. AST IV activity was only slightly decreased or normal by the chronic ingestion of Zn, Cu and Cd. The acute administration of Zn caused a slight decrease of liver AST IV activity.
