Rapid and efficient capillary column supercritical fluid chromatography with mass spectrometric detection
Capillary column supercritical fluid chromatography (SFC) using small diameter columns (approx. 50 ..mu..m i.d.) has been demonstrated to provide efficient high-resolution separations. Microparticle packed column SFC has previously been demonstrated to allow very rapid moderate resolution separations, but the large pressure drop required by this approach does not allow effective pressure programming for fast separations to be obtained with capillary column SFC using high-pressure programming rates of up to 50 bar/min and provide advantages compared to packed column methods when the components to be separated span a wide molecular weight range. This approach complements the high-resolution capability of capillary SFC obtained using longer columns and provides increased sensitivity and narrow, nearly constant peak widths ( less than or equal to 6 s), which are ideally compatible with mass spectrometry (MS). Detection limits were found to be approx. 0.1 pg for a series of biphenyl compounds using single ion monitoring. The application of capillary SFC/MS for both rapid and high-resolution separations is demonstrated for standard mixtures and a complex coal tar extract.
