OXIDATIVE PHOSPHORYLATION AND THE OPTICAL DENSITY OF MITOCHONDRIA IN THE NORMAL AND REGENERATING LIVER OF RATS SUBJECTED TO A WHOLE BODY DOSE OF X-RAYS (in Russian)
It is well known that tissue characterized by a high degree of mitotic activity is generally more sensitive to radiation than is the same tissue in a state of relative rest. About 2/3 of the liver by weight was extirpated from male rats subjected to a 1000 r dose of x-rays. The mitochondria were isolated from the liver by centrifugation at 10,000 g for 15 minutes. The oxygen requirement, determined on a Warburg manometer, and the phosphate release on incubation with yeast hexokinase were expressed as a ratio of P: O (ratio of atoms). The ratio served as a measure of oxidative phosphorylation (OP) in the tissue. The level of oxidative phosphorylation in the mitcchondria for rats 24 hours after irradiation was lower with a noticeable decrease in phosphate esterification than it was for normal intact rats. The experiments showed that whole body irradation during the period of recuperation (48 hours or more after the partial hepatectomy) led to a sharper suppression of oxidative phosphorylation than in experiments where the whole body dose of 1000 r was administered before the recuperative period (24 hours or less after the operation). It was found that the mitochondria, isolated 72 hours aiter the operation, in animals which had been irradiated 48 hours after the operation, showed a lower optical density in 0.125M KCl or in a phosphate buffer than did a suspension of mitochondria isolated from the same animals before recuperation and irradation. Damage to the structure of the mitochondria was also evident from the fact that the swelled mitochondria did not contract as readily as did the mitochondria from the controls on adding ATP and MgCl/sub 2/ to the suspension. These data indicate that radiation administered during an active period of regeneration (72 hours after the operation) damages cell structure of mitochondria more than does radiation administered before period of active growth (24 hrs after the operation).
