CHANGES IN SERUM AND TISSUE ENZYMES UNDER THE INFLUENCE OF IONIZING RADIATIONS. 2. HISTOCHEMICAL ENZYME STUDIES IN SKIN, LIVER, AND INTESTINE OF WHOLE-BODY IRRADIATED RATS (in German)
Frozen tissue sections were obtained 2 hr to 30 days after whole-body x irradiation of rats with 800 r doses to ascertain changes in lactic dehydrogenase (LDH), diphosphopyridine nucleotide (DPN) diaphorase, and succinic dehydrogenase (SDH) activitles. In some rats a 2 x 2 cm lead shield was placed over the abdomen during exposure to shield part of the intestine. LDH and SDH activities, which generally changed in parallel in various tissues, showed a marked drop in unshielded intestine and a complete disappearance by the 3rd day. By the 30th day complete restoration of these activities was present. No changes in their activities were noted in shielded intestine. In the liver, which was not shielded, the depletion of all three enzymes was greater in the absence of intestinal shielding, which suggests that toxic products from irradiated intestine augments liver injury, since in shielded rats the liver enzyme changes were less pronounced. Enzyme changes in skin were similar in the two groups; DPN diaphorase was especially depleted in skin after irradiation but returned to normal within 30 days. The enzyme changes were most marked in the stratum spinosum of the epidermis and hair follicle epithelium. Fibroblasts showed only a transient enzyme depletion, which was followed by a compensatory supranormal rise in activity in some cells in the regenerative phase. The 50-day mortality rates were 50% in the shielded and 10% in the unshielded rats, indicating that substances liberated into the circulation from the radiation-injured intestine may contribute to the lethal action of radiation.
