Title: The effect of centaurein on interferon-{gamma} expression and Listeria infection in mice

Abstract

We previously found that centaurein enhanced IFN-{gamma} transcription in T cells. Here, we demonstrate that centaurein increased the IFN-{gamma} expression in T and NK cells and the serum IFN-{gamma} level in mice. Centaurein elevated the transcription of T-bet but not GATA-3, which is consistent with its effect on that of IFN-{gamma} but not IL-4. Additionally, centaurein effectively protected mice against Listeria infection. Moreover, centaurein per se or in combination with antibiotics could treat Listeria infection. Our mechanistic studies suggest that centaurein augments IFN-{gamma} expression via a transcriptional up-regulation of T-bet and that centaurein protects against or treats Listeria infection via a modulation of IFN-{gamma} expression.

  • As a model for the study of human atypical mycobacterial disease, the basis for the prolonged mycobacteriosis in mice infected with Mycobacterium intracellulare was studied. Two weeks after i.v. injection of mycobacteria, peritoneal macrophages were found to be activated, as indicated by their capacity to produce large amounts of superoxide anion (O/sub 2//sup -/) in response in phorbol myristate acetate (PMA) or viable M. intracellulare. However, 4 wk after infection, despite the continued presence of large numbers of mycobacteria in the spleen, macrophages from infected animals produced low amounts of O/sub 2//sup -/. Additional investigation showed that macrophages from infectedmore » animals produced large amounts of prostaglandin E/sub 2/ (PGE/sub 2/) when stimulated by mycobacterial antigens. In vitro, such concentrations of PGE/sub 2/ inhibited uptake of (/sup 3/H)thymidine by stimulated spleen lymphocytes from infected animals. The results support the concept that interaction between the host and M. intracellulare is modulated profoundly by PG and suggest that administration of agents that directly promote macrophage activation can enhance resistance to infection by this organism.« less
  • Previously, we have reported that thalidomide can modulate the immune responses in female B6C3F1 mice. Furthermore, thalidomide immunomodulation increased primary host resistance to intravenously infected Listeria monocytogenes. The present study was intended to evaluate the mechanisms underlying the enhanced host resistance to L. monocytogenes by focusing on the neutrophils. Female B6C3F1 mice were treated intraperitoneally with thalidomide (100 mg/kg) for 15 days. Exposure to thalidomide increased the numbers of neutrophils in the spleens and livers of L. monocytogenes-infected mice when compared to the L. monocytogenes-infected control mice. Additionally, the percentage of neutrophils was also significantly increased after Thd treatment inmore » L. monocytogenes-infected mice. Further studies using antibodies to deplete corresponding cells indicated that thalidomide-mediated increase in primary host resistance (both the moribundity and colony counts in the liver and spleen) to L. monocytogenes infection was due to its effect on neutrophils but not CD8{sup +} T cells or NK cells. Finally, Thd exposure also increased host resistance to secondary host resistance to L. monocytogenes infection, and depletion of neutrophils abolished the protective effect. In conclusion, thalidomide enhanced host resistance to both primary and secondary L. monocytogenes infections by a neutrophil-related mechanism in female B6C3F1 mice.« less
  • The present study was undertaken to elucidate the mechanism for the antifibrotic effect of interferon gamma (IFN-{gamma}) in the bleomycin (BL)-mouse model of lung fibrosis. The expression of transforming growth factor (TGF-{beta}) and procollagen I and III and their mRNAs was investigated in the BL-mouse model of lung fibrosis with and without IFN-{gamma} treatment by Northern and slot blot analyses. Temporal changes in the content of procollagen and TGF-{beta} mRNAs in the lungs of mice receiving saline or BL by intratracheal route, with and without IFN-{gamma} treatment by intramuscular route, were quantitated. The level of TGF-{beta} mRNA increased rapidly andmore » peaked at day 5, whereas the levels of mRNAs for procollagens {alpha}{sub 1}(I) and {alpha}{sub 1}(III) peaked at 10 days after BL instillation. The peak levels of these mRNAs in BL-treated animals were five- to seven-fold higher than those of the control. BL treatment also increased the hydroxyproline content significantly from 3 to 14 days as compared to the corresponding saline control groups. A maximal increase to 447 {mu}g/lung from 223 {mu}g/lung in saline control was obtained at 10 days after instillation. Daily treatment with IFN-{gamma} markedly reduced the BL-induced increases in the mRNA levels of TGF-{beta}, and procollagen {alpha}{sub 1}(I) and {alpha}{sub 1}(III) without any effect on the lung level of {beta}-actin mRNA. IFN-{gamma} treatment also caused significant reduction in the BL-induced increase in the lung hydroxyproline content from 417 to 283 {mu}g/lung at 7 days and from 447 to 264 {mu}g/lung at 10 days. It may be concluded from the findings of the present study that the cellular mechanisms for the antifibrotic effect of IFN-{gamma} in the BL-mouse model of lung fibrosis are to initially downregulate the BL-induced overexpression of TFG-{beta} mRNA, and subsequently procollagen mRNAs, leading to a decreased collagen content. 42 refs., 7 figs.« less
  • Freshly isolated normal human T lymphocytes constitutively expressed receptors for interferon-alpha (IFN-alpha) and IFN-gamma. Upon activation, the number of IFN-alpha receptors increased, paralleling the increases in cell size to give a nearly constant density of IFN-alpha receptors. In contrast, activation of T lymphocytes with phytohemagglutinin (PHA), concanavalin A (ConA), or phorbol myristate acetate (PMA) for 18 h decreased the specific binding of radiolabeled (Cys-Tyr-Cys) rIFN-gamma to acid-washed cells. Scatchard analysis demonstrated that the decreased binding was due to a reduction in the number of high affinity IFN-gamma receptors. Moreover, resting T lymphocytes had a single class of high-affinity IFN-gamma receptors,more » whereas the activated cells appeared to have both high- and lower-affinity IFN-gamma binding sites. When normalized for differences in cell size, the number of high-affinity IFN-gamma receptors per unit cell surface area was approximately twofold lower on activated than on resting T lymphocytes. At least part of the decrease was due to receptor downregulation by endogenously produced IFN-gamma, since monoclonal antibodies to IFN-gamma prevented a large portion of the PHA-induced decrease in IFN-gamma receptors.« less
