Tobacco vein mottling virus (TVMV) is a member of the Potyviridae, one of the largest families of plant viruses. The TVMV genome is translated into a single large polyprotein that is subsequently processed by three virally encoded proteases. Seven of the nine cleavage events are carried out by the NIa protease. Its homolog from the tobacco etch virus (TEV) is a widely used reagent for the removal of affinity tags from recombinant proteins. Determination of the 5'-terminal sequence of tobacco vein mottling virus RNA and mapping of the genome-linked protein

The nature of the 5'-terminus of the potyvirus tobacco vein mottling virus RNA (TVMV) was investigated. Recombinant plasmids containing cDNA fragments from the 5'-terminal region of TVMV RNA were constructed and used to transform Escherichia coli. Clones containing the desired plasmids were selected by hybridization. To localize the point of attachment of the genome-linked protein (VPg), /sup 125/I-labeled TVMV RNA was partially hydrolyzed and used as hybridization probe against the plasmid containing 5'-terminal sequence and four previously described plasmids containing TVMV cDNA. X-ray crystallographic structure of a complex between a synthetic protease of human immunodeficiency virus 1 and a substrate-based hydroxyethylamine inhibitor

The structure of a crystal complex of the chemically synthesized protease of human immunodeficiency virus 1 with a heptapeptide-derived inhibitor bound in the active site has been determined. The sequence of the inhibitor JG-365 is Ac-Ser-Leu-Asn-Phe-{psi} (CH(OH)CH{sub 2}N)-Pro-Ile-Val-OMe; the K{sub i} is 0.24 nM. The hydroxyethylamine moiety, in place of the normal scissile bond of the substrate, is believed to mimic a tetrahedral reaction intermediate.
