Altered distribution of (R)-quinuclidinyl I-125 4-iodobenzilate after whole body fission neutron or cobalt-60 irradiation
Two groups of male rats were exposed to either 600 rad fission neutrons or cobalt-60 irradiation. At specific times after irradiation the rats were injected with 10 ..mu..Ci of 4-IQNB and sacrificed one hour later. Significant increase in small intestinal (SI) and stomach (ST) uptake was observed immediately after neutron irradiation out to 6 days (SI 1.30-1.28;ST 0.46-0.44 as compared to control values (SI 0.96, ST 0.29). There was no significant increase in uptake of 4-IQNB in the small intestine and stomach for the Co-60 irradiated group until day 10 (SI 1.73, ST 0.43). The left ventricular uptake was significantly increased for the neutron group only at day 4 (1.28) and for the Co-60 group only at day 10 (1.22) compared to the control value (0.53). No significant differences in uptake were observed in the corpus striatum or cerebellum. Although other investigators found enhanced cholinergic response in the brain and heart after irradiation, the authors did not find altered mAChR binding for 4-IQNB in the brain. The altered biodistribution of 4-IQNB in the SI and ST may be a result of altered muscarinic receptor interactions and/or blood flow. It has been observed by other investigators that whole body x-irradiation produced increased cholinergic activity in the heart and the brain. The objective of this study was to determine the relationship of the increase in cholinergic activity and muscarinic receptor mAChR binding for (R)-quinuclidinyl I-/sup 125/ 4-iodobenzilate (4-IQNB) in these organs and to study the distribution of 4-IQNB in other organs after irradiation.
