Purification of Apase-1 sup 1 and retrieval of the nematode resistance gene
This patent describes a method to purify Apase-1{sup 1}. It comprises subjecting a crude extract from tomato cells to ammonium sulfate precipitation at 55--85% saturation; desalting the precipitate; subjecting the desalted preparation to anion exchange chromatography and collecting fractions containing Apase-1{sup 1}; desalting the fractions; subjecting the desalted preparation to chromatography on hydroxyapatite and collecting fractions containing Apase-1{sup 1}; desalting the fractions obtained; subjecting the desalted preparation to affinity chromatography and recovering the Apase-1{sup 1} fractions. Apase-1{sup 1} prepared by this method. A purified Apase-1{sup 1} protein having an apparent molecular weight on SDS-PAGE of 31 kd and an amino acid composition.
