In order to improve the understanding of the nature of the strongly bound cisplatin on DNA, the reactivity of a large number of complexes of cis-(NH/sub 3/)/sub 2/Pt/sup II/ with the model nucleobases, 9-ethylguanine, 9-methyladenine, 1-methylcytisine, 1-methylthymine, and i-methyluracil, toward a large excess of cyanide was studied. The behavior of Pt-nucleobase complexes toward CN/sup -/ is compared with that of simple Pt-amine complexes, and reactions of thiourea with two selected nucleobase complexes is reported. The relevance of these findings with respect to substitution reactions of Pt-nucleobase complexes and the nature of the tightly DNA-bound Pt, which cannot be removed by excess KCN, is discussed.
