    Aligning large vertebrate genomes that are structurally complex poses a variety of problems not encountered on smaller scales. Such genomes are rich in repetitive elements and contain multiple segmental duplications, which increases the difficulty of identifying true orthologous SNA segments in alignments. The sizes of the sequences make many alignment algorithms designed for comparing single proteins extremely inefficient when processing large genomic intervals. We integrated both local and global alignment tools and developed a suitemore » of programs for automatically aligning large vertebrate genomes and identifying conserved non-coding regions in the alignments. Our method uses the BLAT local alignment program to find anchors on the base genome to identify regions of possible homology for a query sequence. These regions are postprocessed to find the best candidates which are then globally aligned using the AVID global alignment program. In the last step conserved non-coding segments are identified using VISTA. Our methods are fast and the resulting alignments exhibit a high degree of sensitivity, covering more than 90% of known coding exons in the human genome. The GenomeVISTA software is a suite of Perl programs that is built on a MySQL database platform. The scheduler gets control data from the database, builds a queve of jobs, and dispatches them to a PC cluster for execution. The main program, running on each node of the cluster, processes individual sequences. A Perl library acts as an interface between the database and the above programs. The use of a separate library allows the programs to function independently of the database schema. The library also improves on the standard Perl MySQL database interfere package by providing auto-reconnect functionality and improved error handling.« less

    Comparison of DNA sequences from different species is a fundamental method for identifying functional elements in genomes. Here we describe the VISTA family of tools created to assist biologists in carrying out this task. Our first VISTA server at http://www-gsd.lbl.gov/VISTA/ was launched in the summer of 2000 and was designed to align long genomic sequences and visualize these alignments with associated functional annotations. Currently the VISTA site includes multiple comparative genomics tools and provides users with rich capabilities to browse pre-computed whole-genome alignments of large vertebrate genomes and other groups of organisms with VISTA Browser, submit their own sequences of interest to several VISTA servers for various types of comparative analysis, and obtain detailed comparative analysis results for a set of cardiovascular genes. We illustrate capabilities of the VISTA site by the analysis of a 180 kilobase (kb) interval on human chromosome 5 that encodes for the kinesin family member3A (KIF3A) protein.

    In 1998, carbon tetrachloride was found above the maximum contaminant level (MCL) of 5 {micro}g/L in groundwater from one private livestock well at Sylvan Grove, Kansas, by the Kansas Department of Health and Environment (KDHE). The 1998 KDHE sampling was conducted under the U.S. Department of Agriculture (USDA) private well sampling program. The Commodity Credit Corporation (CCC), a USDA agency, operated a grain storage facility in Sylvan Grove from 1954 to1966. Carbon tetrachloride is the contaminant of primary concern at sites associated with former CCC/USDA grain storage operations. Sylvan Grove is located in western Lincoln County, approximately 60 mi west of Salina (Figure 1.1). To determine whether the former CCC/USDA facility at Sylvan Grove is a potential contaminant source and its possible relationship to the contamination in groundwater, the CCC/USDA has agreed to conduct an investigation, in accordance with the Intergovernmental Agreement between the KDHE and the Farm Service Agency (FSA) of the USDA. This Work Plan presents historical data related to previous investigations, grain storage operations, local private wells and public water supply (PWS) wells, and local geologic and hydrogeologic conditions at Sylvan Grove. The findings from a review of all available documents are discussed in Section 2. On the basis of the analyses of historical data, the following specific technical objectives are proposed for the site investigation at Sylvan Grove: (1) Evaluate the potential source of carbon tetrachloride at the former CCC/USDA facility; (2) Determine the relationship of potential contamination (if present) at the former CCC/USDA facility to contamination identified in 1998 in groundwater samples from one private well to the west; and (3) Delineate the extent of potential contamination associated with the former CCC/USDA facility. The detailed scope of work is outlined in Section 3. The results of the proposed work will provide the basis for determining

    VISTA is a comprehensive suite of programs and databases developed by and hosted at the Genomics Division of Lawrence Berkeley National Laboratory. They provide information and tools designed to facilitate comparative analysis of genomic sequences. Users have two ways to interact with the suite of applications at the VISTA portal. They can submit their own sequences and alignments for analysis (VISTA servers) or examine pre-computed whole-genome alignments of different species. A key menu option is the Enhancer Browser and Database at http://enhancer.lbl.gov/. The VISTA Enhancer Browser is a central resource for experimentally validated human noncoding fragments with gene enhancer activity as assessed in transgenic mice. Most of these noncoding elements were selected for testing based on their extreme conservation with other vertebrates. The results of this enhancer screen are provided through this publicly available website. The browser also features relevant results by external contributors and a large collection of additional genome-wide conserved noncoding elements which are candidate enhancer sequences. The LBL developers invite external groups to submit computational predictions of developmental enhancers. As of 10/19/2009 the database contains information on 1109 in vivo tested elements - 508 elements with enhancer activity.

    In 2007, the U.S. Department of Energy joined the California Energy Commission in funding a project to begin to examine the technical, economic and institutional (policy and regulatory) aspects of energy-efficient community development. That research project was known as the Chula Vista Research Project for the host California community that co-sponsored the initiative. The researches proved that the strategic integration of the selected and economically viable buildings energy efficiency (EE) measures, photovoltaics (PV), distributed generation (DG), and district cooling can produce significant reductions in aggregate energy consumption, peak demand and emissions, compared to the developer/builder's proposed baseline approach. However, the central power plant emission reductions achieved through use of the EE-DG option would increase local air emissions. The electric and natural gas utility infrastructure impacts associated with the use of the EE and EE-PV options were deemed relatively insignificant while use of the EE-DG option would result in a significant reduction of necessary electric distribution facilities to serve a large-scale development project. The results of the Chula Vista project are detailed in three separate documents: (1) Energy-Efficient Community Development in California; Chula Vista Research Project report contains a detailed description of the research effort and findings. This includes the methodologies, and tools used and the analysis of the efficiency, economic and emissions impacts of alternative energy technology and community design options for two development sites.     A series of integral effect tests (IETs) was performed by the Korea Atomic Energy Research Inst. (KAERI) using the VISTA integral test loop (VISTA-ITL) as a small-scale IET program. Among them this paper presents major results acquired from the safety-related IETs with the VISTA-ITL facility for the SMART design. Three small-break loss-of-coolant accident (SBLOCA) tests of safety injection system (SIS) line break, shutdown cooling system (SCS) line break and pressurizer safety valve (PSV) line break were successfully performed and the transient characteristics of a complete loss of flowrate (CLOF) was simulated properly with the VISTA-ITL facility.     DOE`s Federal Energy Management Program supports efforts to reduce energy use and associated expenditures within the federal sector; one such effort, the New Technology Demonstration Program (NTDP)(formerly the Test Bed Demonstration program), seeks to evaluate new energy saving US technologies and secure their more timely adoption by the federal government. This report describes the field evaluation conducted to examine the performance of a 15-ton natural-gas-engine- driven, split-system, air-conditioning unit. The unit was installed at a multiple-use building at Willow Grove Naval Air Station, a regular and reserve training facility north of Philadelphia, and its performance was monitored under the NTDP.

    Identifying transcriptional regulatory elements represents a significant challenge in annotating the genomes of higher vertebrates. We have developed a computational tool, rVISTA, for high-throughput discovery of cis-regulatory elements that combines transcription factor binding site prediction and the analysis of inter-species sequence conservation. Here, we illustrate the ability of rVISTA to identify true transcription factor binding sites through the analysis of AP-1 and NFAT binding sites in the 1 Mb well-annotated cytokine gene cluster1 (Hs5q31; Mm11). The exploitation of orthologous human-mouse data set resulted in the elimination of 95 percent of the 38,000 binding sites predicted upon analysis of the human sequence alone, while it identified 87 percent of the experimentally verified binding sites in this region.

    Orange County Florida completed the construction of a 20 kilowatt Solar Demonstration and Research Facility in March 2015. The system was constructed at the Orange County/University of Florida Cooperative Extension Center whose electric service address is 6021 South Conway Road, Orlando, Florida 32802. The Solar Demonstration and Research Facility is comprised of 72 polycrystalline photovoltaic modules and 3 inverters which convert direct current from the solar panels to alternating current electricity. Each module produces 270 watts of direct current power, for a total canopy production of just under 20,000 watts. The solar modules were installed with a fixed tilt of 5 degrees and face south, toward the equator to maximize the amount of sunlight captures. Each year, the electricity generated by the solar array will help eliminate 20 metric tons of carbon dioxide emissions as well as provide covered parking for staff and visitors vehicles. The solar array is expected to generate 27,000 kilowatt hours of electricity annually equating to an estimated $266 savings in the monthly electric bill, or $3,180 annually for the Orange County/University of Florida Cooperative Extension Center. In addition to reducing the electric bill for the Extension Center, Orange County’s solar array also takes advantage of a rebate incentive offered by the local utility, Orlando Utility Commission, which provided a meter that measures the amount of power produced by the solar array. The local utility company’s Solar Photovoltaic Production Incentive will pay Orange County $0.05 per kilowatt hour for the power that is produced by the solar array. This incentive is provided in addition to Net Metering benefits, which is an effort to promote the use of clean, renewable energy on the electric grid. The Photovoltaic Solar Demonstration and Research Facility also serves an educational tool to the public; the solar array is tied directly into a data logger that provides real time power

    In a field evaluation conducted for the US Department of Energy (DOE) Office of Federal Energy Management Program (FEMP), the Pacific Northwest Laboratory (PNL) examined the performance of a new US energy-related technology under the FEMP Test Bed Demonstration Program. The technology was a 15-ton natural gas engine driven roof top air conditioning unit. Two such units were installed on a naval retail building to provide space conditioning to the building. Under the Test Bed Demonstration Program, private and public sector interests are focused to support the installation and evaluation of new US technologies in the federal sector. Participating in this effort under a Cooperative Research and Development Agreement (CRADA) with DOE were the American Gas Cooling Center, Philadelphia Electric Company, Thermo King Corporation, and the US Naval Air Station at Willow Grove, Pennsylvania. Equipment operating and service data as well as building interior and exterior conditions were secured for the 1992 cooling season. Based on a computer assessment of the building using standard weather data, a comparison was made with the energy and operating costs associated with the previous space conditioning system. Based on performance during the 1992 cooling season and adjusted to a normal weather year, the technology will save the site $6,000/yr in purchased energy costs. An additional $9,000 in savings due to electricity demand ratchet charge reductions will also be realized. Detailed information on the technology, the installation, and the results of the technology test are provided to illustrate the advantages to the federal sector of using this technology. A history of the CRADA development process is also reported.

  17. VISTA -- A Vehicle for Interplanetary Space Transport Application Powered by Inertial Confinement Fusion

    Inertial Confinement Fusion (ICF) is an ideal technology to power self-contained single-stage piloted (manned) spacecraft within the solar system because of its inherently high power/mass ratios and high specific impulses (i.e., high exhaust velocities). These technological advantages are retained when ICF is utilized with a magnetic thrust chamber, which avoids the plasma thermalization and resultant degradation of specific impulse that are unavoidable with the use of mechanical thrust chambers. We started with Rod Hyde's 1983 description of an ICF-powered engine concept using a magnetic thrust chamber, and conducted a more detailed systems study to develop a viable, realistic, and defensible spacecraft concept based on ICF technology projected to be available in the first half of the 21st century. The results include an entirely new conical spacecraft conceptual design utilizing near-existing radiator technology. We describe the various vehicle systems for this new concept, estimate the missions performance capabilities for general missions to the planets within the solar system, and describe in detail the performance for the baseline mission of a piloted roundtrip to Mars with a 100-ton payload. For this mission, we show that roundtrips totaling {ge}145 days are possible with advanced DT fusion technology and a total (wet) spacecraft mass of about 6000 metric tons. Such short-duration missions are advantageous to minimize the known cosmic-radiation hazards to astronauts, and are even more important to minimize the physiological deteriorations arising from zero gravity. These ICF-powered missions are considerably faster than those available using chemical or nuclear-electric-propulsion technologies with minimum-mass vehicle configurations. VISTA also offers onboard artificial gravity and propellant-based shielding from cosmic rays, thus reducing the known hazards and physiological deteriorations to insignificant levels.     The experimental detection of the sharp lines of the (e{sup +}e{sup -}) Puzzle is viewed as a struggle against Doppler broadening. Gedanken experiments which are realistic in zeroth order of detail are analyzed to show that the ORANGE and EPOS/I geometries select narrower slices of a Doppler broadened line than spherically inclusive (APEX and EPOS/II -like) apparati. Roughly speaking, the latter require event-by-event Doppler reconstruction simply to regain an even footing with the former. This suggests that APEX` or EPOS/II`s coincident pair distributions must be statistically superior to those of EPOS/I or ORANGE in order to support a comparable inference about sharp structure. Under such circumstances, independent alternative data is invaluable. Therefore, a corroboration of Sakai`s 330.1 keV (< 3 keV wide) electron line in few MeV e{sup +} or e{sup -} bombardments of U and Th targets could prove crucial.

    Phototoxic fluorescent proteins represent a sparse group of genetically encoded photosensitizers that could be used for precise light-induced inactivation of target proteins, DNA damage, and cell killing. Only two such GFP-based fluorescent proteins (FPs), KillerRed and its monomeric variant SuperNova, were described up to date. We present a crystallographic study of their two orange successors, dimeric KillerOrange and monomeric mKiller-Orange, at 1.81 and 1.57 Å resolution, respectively. They are the first orange-emitting protein photosensitizers with a tryptophan-based chromophore (Gln65-Trp66-Gly67). Same as their red progenitors, both orange photosensitizers have a water-filled channel connecting the chromophore to the β-barrel exterior and enabling transport of ROS. In both proteins, Trp66 of the chromophore adopts an unusual trans-cis conformation stabilized by H-bond with the nearby Gln159. This trans-cis conformation along with the water channel was shown to be a key structural feature providing bright orange emission and phototoxicity of both examined orange photosensitizers.

    Phototoxic fluorescent proteins represent a sparse group of genetically encoded photosensitizers that could be used for precise light-induced inactivation of target proteins, DNA damage, and cell killing. Only two such GFP-based fluorescent proteins (FPs), KillerRed and its monomeric variant SuperNova, were described up to date. We present a crystallographic study of their two orange successors, dimeric KillerOrange and monomeric mKiller-Orange, at 1.81 and 1.57 Å resolution, respectively. They are the first orange-emitting protein photosensitizers with a tryptophan-based chromophore (Gln65-Trp66-Gly67). Same as their red progenitors, both orange photosensitizers have a water-filled channel connecting the chromophore to the β-barrel exterior and enablingmore » transport of ROS. In both proteins, Trp66 of the chromophore adopts an unusual trans-cis conformation stabilized by H-bond with the nearby Gln159. This trans-cis conformation along with the water channel was shown to be a key structural feature providing bright orange emission and phototoxicity of both examined orange photosensitizers.« less

  2. Performance and evaluation of gas engine driven rooftop air conditioning equipment at the Willow Grove (PA) Naval Air Station. Interim report, 1992 cooling season

    In a field evaluation conducted for the US Department of Energy (DOE) Office of Federal Energy Management Program (FEMP), the Pacific Northwest Laboratory (PNL) examined the performance of a new US energy-related technology under the FEMP Test Bed Demonstration Program. The technology was a 15-ton natural gas engine driven roof top air conditioning unit. Two such units were installed on a naval retail building to provide space conditioning to the building. Under the Test Bed Demonstration Program, private and public sector interests are focused to support the installation and evaluation of new US technologies in the federal sector. Participating in this effort under a Cooperative Research and Development Agreement (CRADA) with DOE were the American Gas Cooling Center, Philadelphia Electric Company, Thermo King Corporation, and the US Naval Air Station at Willow Grove, Pennsylvania. Equipment operating and service data as well as building interior and exterior conditions were secured for the 1992 cooling season. Based on a computer assessment of the building using standard weather data, a comparison was made with the energy and operating costs associated with the previous space conditioning system. Based on performance during the 1992 cooling season and adjusted to a normal weather year, the technology will save the site $6,000/yr in purchased energy costs. An additional $9,000 in savings due to electricity demand ratchet charge reductions will also be realized. Detailed information on the technology, the installation, and the results of the technology test are provided to illustrate the advantages to the federal sector of using this technology. A history of the CRADA development process is also reported.

  4. Biohydrogen production from rotten orange with immobilized mixed culture: Effect of immobilization media for various composition of substrates

    Enriched–immobilized mixed culture was utilized to produce biohydrogen in mesophilic condition under anaerobic condition using rotten orange as substrate. The process was conducted in batch reactors for 100 hours. Microbial cultures from three different sources were subject to a series of enrichment and immobilized in two different types of media, i.e. calcium alginate (CA, 2%) and mixture of alginate and activated carbon (CAC, 1:1). The performance of immobilized culture in each media was tested for biohydrogen production using four different substrate compositions, namely orange meat (OM), orange meat added with peel (OMP), orange meat added with limonene (OML), and mixture of orange meat and peel added with limonene (OMPL). The results show that, with immobilized culture in CA, the variation of substrate composition gave significant effect on the production of biohydrogen. The highest production of biohydrogen was detected for substrate containing only orange meet, i.e. 2.5%, which was about 3-5 times higher than biohydrogen production from other compositions of substrate. The use of immobilized culture in CAC in general has increased the hydrogen production by 2-7 times depending on the composition of substrate, i.e. 5.4%, 4.8%, 5.1%, and 4.4% for OM, OMP, OML, and OMPL, respectively. The addition of activated carbon has eliminated the effect of inhibitory compounds in the substrate. The major soluble metabolites were acetic acid, propionic acid, and butyric acid.

    The purpose of this report is to present this Consultant`s findings regarding the nature and extent of the mineral estate of the United States at National Petroleum Reserve No. 2 (NPR-2), Buena Vista Hills Field, Kern County, California. Determination of the mineral estate is a necessary prerequisite to this Consultant`s calculation of estimated future cash flows attributable to said estate, which calculations are presented in the accompanying report entitled ``Phase II Final Report, Study of Alternatives for Future Operations of the Naval Petroleum and Oil Shale Reserves, NPR-2, California.`` This Report contains a discussion of the leases in effect at NPR-2 and subsequent contracts affecting such leases. This Report also summarizes discrepancies found between the current royalty calculation procedures utilized at NPR-2 and those procedures required under applicable agreements and regulations. Recommendations for maximizing the government`s income stream at NPR-2 are discussed in the concluding section of this Report.

The resulting product was physically characterized by X-ray powder diffraction, scanning electron microscopy, transmission electron microscope, etc. The as-fabricated SnO{sub 2} NRs exhibited excellent photocatalytic degradation of the methyl orange with high degradation efficiency of 99.3% with only 60 min ultra violet light irradiation. Meanwhile, the 1D SnO{sub 2} NRs exhibited intriguing photostability after four recycles.

    California is currently faced with some critical decisions about water resource infrastructure development in highly urbanized regions, whose outcome will dictate the future long-term viability of plentiful water. Among these is developing and safely implementing the reuse of advanced treated waste water. One of the most reliable strategies for this water resource is its indirect reuse via groundwater recharge and storage, with particular emphasis on supplementing annual water demand or during drought relief. The Orange County Water District (District) is currently implementing the first phase of a large-scale water reuse project that will advance-treat up to 60 million gallons per day of waste water and recharge it into existing percolation basins in the Forebay region of the Orange County groundwater basin. In order for the District to protect public health, the fate and potability of this recharged waste water needs to be understood. In particular, the direction and rates of flow into underlying aquifers need to be characterized so that changes in water quality can be quantified between the recharge basins and points of production. Furthermore, to ensure compliance to California Department of Health Services (DHS) draft regulations, the direction and rate of recharged waste water from these basins need to be understood to sufficient detail that small mixtures can be delineated in monitoring and production wells. Under proposed DHS guidelines, consumptive use of recycled water is permissive only if its residence time in an aquifer exceeds a specified six-month time-frame. DHS guidelines also limit the percentage of recycled water at production wells. However, attaining such detail using current hydrogeological and computer-assisted modeling tools is either cost-prohibitive or results in uncertainties too large to achieve regulatory confidence.     In this work, methyl orange (MO) was effectively removed from aqueous solution with the calcined product of hydrothermal synthesized Mg/Fe layered double hydroxide (Mg/Fe-LDH). The structure, composition, morphology and textural properties of the Mg/Fe-LDH before and after adsorption were characterized by X-ray diffraction, Fourier transformation infrared spectroscopy, transmission electron microscopy, nitrogen adsorption apparatus and X-ray photoelectron spectroscopy. It was confirmed that MO had been absorbed by calcined Mg/Fe-LDH which had strong interactions with MO. The adsorption of MO onto the Mg/Fe-LDH was systematically investigated by batch tests. The adsorption capacity of the Mg/Fe-LDH toward MO was found to be 194.9 mg • g{sup −1}. Adsorption kinetics and isotherm studies revealed that the adsorption of MO onto Mg/Fe-LDH was a spontaneous and endothermic process. These results indicate that Mg/Fe-LDH is a promising material for the removal of MO.

  10. Mass spectrometry footprinting reveals the structural rearrangements of cyanobacterial orange carotenoid protein upon light activation

    The orange carotenoid protein (OCP), a member of the family of blue light photoactive proteins, is required for efficient photoprotection in many cyanobacteria. Photoexcitation of the carotenoid in the OCP results in structural changes within the chromophore and the protein to give an active red form of OCP that is required for phycobilisome binding and consequent fluorescence quenching. We characterized the light-dependent structural changes by mass spectrometry-based carboxyl footprinting and found that an α helix in the N-terminal extension of OCP plays a key role in this photoactivation process. Although this helix is located on and associates with the outside of the β-sheet core in the C-terminal domain of OCP in the dark, photoinduced changes in the domain structure disrupt this interaction. We propose that this mechanism couples light-dependent carotenoid conformational changes to global protein conformational dynamics in favor of functional phycobilisome binding, and is an essential part of the OCP photocycle.

    The influence of some parameters (pH, salt concentration) on scandium precipitation with oxalic acid in the presence of calcium oxalate as collector has been examined. Scandium precipitates quantitatively as an oxalate at pH 0.8-1.2 in 0.5-1.0 M NH/sub 4/NO/sub 3/. A spectrophotometric method is proposed for determining scandium with Xylenol Orange combined with preseparation from interfering elements by successive precipitation with inositolhexaphosphoric acid and oxalic acid. The method was applied for scandium determination in samples of complex compositions.

    The dissociation of methyl orange was investigated by spectrophotometry in aqueous solutions from 10 to 90°C and by quantum chemical calculations. Combined chemometric and thermodynamic analyses of the spectrophotometric data were used to simultaneously extract the thermodynamic stabilities and the spectrophotometric attributes of the dominant methyl orange species in solutions containing less than 20.00 mmol kg-1 perchloric acid and submicromolal concentrations of methyl orange. The analyses revealed the presence of only one monomeric deprotonated and one monomeric protonated species. The spectra did not reveal any evidence for the presence of tautomeric equilibria between the protonated azo and ammonium species in the experimental range studied. Thermodynamic analyses of the temperature dependent dissociation constants showed the reactions to be endothermic and enthalpy driven reaction with increasing acidity and increasing temperature. All molar absorption coefficients in the 275-375 nm range can be adequately reproduced in the 10-90°C range with a set of Gauss-Lorentz parameters and used to predict the absorption spectra for any desired condition. The dominant features of the spectrophotometric attributes of the methyl orange species could also be retrieved in Time Dependent-Density Functional Theory (TD-DFT) calculations. Topological analyses of the electron density also revealed the formation of a dihydrogen bond between the azo proton and an adjacent phenyl ring hydridic hydrogen which increases the stability of the azo molecules relative to the ammonium molecule.

    Ag-SrTiO{sub 3} nanotube arrays were successfully prepared for the degradation of methyl orange (MO) under ultraviolet irradiation. In order to form highly ordered SrTiO{sub 3} nanotube arrays, the preparation of TiO{sub 2} nanotube arrays by anodic oxidation of titanium foil in different electrolytes was investigated. The selected organic solvents in electrolytes include glycerol, dimethyl sulfoxide and glycol. The results indicate that the morphology of TiO{sub 2} nanotube arrays prepared in glycol containing ammonium fluoride electrolyte is more regular. Then SrTiO{sub 3} nanotube arrays were synthesized by a hydrothermal method using TiO{sub 2} nanotube arrays as the precursor. In order to further improve the photocatalytic activity of SrTiO{sub 3} nanotube arrays, Ag nanoparticles were loaded on SrTiO{sub 3} nanotube arrays by two sets of experiments. The loaded Ag results in an enhancement of photocatalytic activity of SrTiO{sub 3} nanotube arrays. Moreover, the effect of pH on the photocatalytic degradation of MO was also studied. - Graphical abstract: Ag-SrTiO{sub 3} nanotube arrays were successfully prepared. Coral-like Au/ZnO exhibited superior photocatalytic activity to degrade Orange II. - Abstract: A porous coral-like zinc oxide (c-ZnO) photocatalyst was synthesized by the hydrothermal method. The coral-like structure was obtained by precipitating Zn{sub 4}(CO{sub 3})(OH){sub 6}H{sub 2}O (ZnCH), which forms nanosheets that aggregate together to form microspheres with the coral-like structure. X-ray diffraction (XRD) studies indicate that after heating at 550 C the ZnCH microspheres can be converted to ZnO microspheres with a morphology similar to that of ZnCH microspheres. Thermogravimetric analysis (TGA) shows this conversion takes place at approximately 260 C. A simple electrostatic self-assembly method has been employed to uniformly disperse Au nanoparticles (1 wt.%) on the ZnO surface. In this procedure ?-D-glucose was used to stabilize the Au nanoparticles. Scanning electron microscope images indicate that the diameter of coral-like ZnO microspheres (c-ZnO) is about 8 ?m. X-ray diffraction reveals that the ZnO is highly crystalline with a wurtzite structure and the Au metallic particles have an average size of about 13 nm. X-ray photoelectron spectroscopic (XPS) studies have confirmed the presence of ZnO and also showed that the Au is present in the metallic state. The photocatalytic degradation of Orange II dye, with either ultraviolet or visible light, is faster on Au/c-ZnO than on c-ZnO.

In this context, carbon nanotubes/TiO{sub 2} (CNTs/TiO{sub 2}) composites were prepared using multi-walled CNTs (MWCNTs), titanium (IV) propoxide and commercial TiO{sub 2} (P25{sup }) as titanium oxide sources, to degrade the methyl orange dye in solution through photocatalyst activity using UV irradiation. The composites were prepared by solution processing followed by thermal treatment at 400, 500 and 600 C. The heterojunction between nanotubes and TiO{sub 2} was confirmed by XRD, specific surface area. The coating morphology was observed with SEM and TEM.

    [Mg{sub 1-x} Al{sub x}(OH){sub 2}][(NO{sub 3}){sub x}, nH{sub 2}O] Layered Double Hydroxide (LDH) sorbents with variable Mg/Al molar (R=(1-x)/x) ratios were investigated for adsorption of azo dye, orange II (OII) at various pH and temperature conditions. Mg{sub 2}AlNO{sub 3} displays the highest adsorption capacity with 3.611 mmol of OII per gram of Mg{sub 2}AlNO{sub 3} at 40 {sup o}C. Adsorption isotherms have been fitted using the Langmuir model and free energy of adsorption ({Delta}G{sup o}), enthalpy ({Delta}H{sup o}) and entropy ({Delta}S{sup o}) were calculated. The experimental values for {Delta}G{sup o} in temperature range between 10 and 40 {sup o}C were found to be negative indicating that a spontaneous process occurred. Positive calculated enthalpy values, characteristic of an endothermic process were found. Characterization of solids (PXRD, FTIR, UV-vis, TGA/DTA, adsorption isotherm BET analysis, SEM and Zetametry) before and after adsorption showed that adsorption proceeds in two steps. Systematic structural (X-ray diffraction (XRD)), morphological (field emission scanning electron microscopy (FE-SEM) and transmission electron microscopic (TEM)), BET surface area and diffuse reflectance spectral (DRS) analyses were carried out to characterize the formed product. The effects of experimental parameters on the microstructural and morphological behavior of Bi{sub 2}O{sub 3}/CeO{sub 2} composites were discussed. The semiconductor material as obtained was evaluated as a heterogeneous photocatalyst for the photolytic degradation of an azo dye in water and the degradation was found to follow pseudo-first-order rate kinetics. The study compares the rate constants of the new hybrid catalyst to a pristine Bi{sub 2}O{sub 3} and CeO{sub 2}, which has excellent catalytic properties under visible light irradiation for the degradation of Orange II dye.

    Excavation of a Superfund site, the Ralph Gray Truncking Company located in Westminster Orange County, California was anticipated to release sulfur dioxide and other chemicals. The California Department of Health Services, under cooperative agreement with the Agency for Toxic Substances and Disease Registry, conducted a surveillance study to assess whether illnesses were associated with cleanup activities. A panel primarily composed of more sensitive persons (n = 36) was selected to report daily respiratory symptoms and odors. Exposures included sulfur dioxide (SO{sub 2}) measurements and daily tonnage of waste removed. Analysis used Conditional Likelihood Regression and Generalized Estimating Equations (GEE) methods. Levels of SO{sub 2} were generally higher than usual ambient air, at times exceeding levels which can cause health effects among asthmatics in laboratory settings. Wheeze and cough were significantly associated with tonnage of waste removed, especially on days when the highest amounts of waste were removed. Upper respiratory symptoms were found to be associated with SO{sub 2}, and weak relationships were found with nausea and burning nose and SO{sub 2}.

  17. Correlation of Process Data and Electrochemical Noise to Assess Kraft Digester Corrosion: Spring Grove Experiment

    Electrochemical noise (ECN) probes were deployed in a carbon steel continuous kraft digester at four locations and at one location in the bottom cone of the associated flash tank. The probes consisted of carbon steel electrodes, representing the vessel construction material, and 309LSi stainless steel overlay electrodes, representing the weld overlay repair in a portion of the vessel. Current and potential noise, the temperature at each probe location, and the value of about 32 process parameters (flow rates, liquor chemistry, etc.) were monitored continuously for a period of almost one year. Historical vessel inspection data and post-test evaluation of the probe components were used to assess/compare ECN corrosion activity with physical changes in wall thickness and corrosion patterns on the digester shell. In addition, attempts were made to correlate ECN activity from each electrode type with process parameters. The results indicate the high general corrosion rates of steel observed just below the extraction screens--on the order of 35 mils/y for the past few years--accelerated further during the period of probe deployment. The maximum wastage of steel (normalized to one full year exposure) was about 85 mils/y at the ring 6N probe just below the extraction screens. Consistent with recent historical observations, the steel corrosion rate at the ring 6S probe--at the same elevation but directly across the digester from ring 6N--was significantly lower at about 50 mils/y. Just prior to probe deployment, the digester shell below the extraction screens was overlaid with 309LSi stainless steel, which was observed to be essentially immune to corrosion at this location. While the ECN probes detected differences in electrochemical behavior between steel probes and between 309LSi probes at rings 6N and 6S, there was only poor quantitative correlation of current sums with actual corrosion rates at these locations. A significant contribution of redox reactions on both steel and stainless steel electrodes appears to complicate ECN interpretation for all the probes, but particularly at the ring 6 locations. With the exception of start-up and shutdown activity, including brief upsets for ''hanging columns'' or brief maintenance periods, no correlation was observed between tracked process variables and ECN activity. Based on the ECN data, re-pressurization of the digester and resumption of chip/liquor feed after a brief disruption results in very aggressive corrosion events.

The as-prepared phosphors were characterized by X-ray powder diffraction (XRD), fluorescence spectroscopy, and spectroradiometry. XRD showed that all prepared samples exhibited a monoclinic Rb{sub 2}CaP{sub 2}O{sub 7} phase. Fluorescence spectroscopy showed that the photoluminescence efficiency of Rb{sub 2}Ca{sub 1−x}P{sub 2}O{sub 7}:Eu{sub x}{sup 2+} phosphors increased with increasing Eu{sup 2+} concentration until x = 0.006, then decreased at higher concentrations, due to a concentration quenching effect. The thermal activation energy was also measured to be 0.40 eV. Furthermore, a phosphor-converted LED (pc-LED) coated with Rb{sub 2}Ca{sub 0.994}P{sub 2}O{sub 7}:Eu{sub 0.006}{sup 2+} was fabricated, which exhibited bright orange emission under a forward bias, from 200 to 300 mA. The color rendering index (CRI) of pc-LED coated with Rb{sub 2}Ca{sub 0.994}P{sub 2}O{sub 7}:Eu{sub 0.006}{sup 2+} was higher than the CRI of pc-LED coated with commercial red phosphor, due to the broad emission band of Rb{sub 2}CaP{sub 2}O{sub 7}:Eu{sup 2+} phosphor. In applying with three-band pc-LEDs, moreover, white pc-LED using Rb{sub 2}CaP{sub 2}O{sub 7}:Eu{sup 2+} phosphor had a higher CRI, than using commercial phosphor. These results indicated that Rb{sub 2}CaP{sub 2}O{sub 7}:Eu{sup 2+} phosphor could be a good candidate for a near-UV based w-LED.

    Reddish orange-emitting 0.948(K{sub 0.5}Na{sub 0.5})NbO{sub 3}-0.052LiSbO{sub 3}-xmol%Sm{sub 2}O{sub 3} (KNN-5.2LS-xSm{sub 2}O{sub 3}) lead-free piezoelectric ceramics with good piezoelectric properties were fabricated in this study, and the photoluminescence and electrical properties of the ceramics were systematically studied. Results showed that Sm{sub 2}O{sub 3} substitution into KNN-5.2LS induces a phase transition from the coexistence of orthorhombic and tetragonal phases to a pseudocubic phase and shifts the polymorphic phase transition (PPT) to below room temperature. The temperature stability and fatigue resistance of the modified ceramics were significantly improved by Sm{sub 2}O{sub 3} substitution. The KNN-5.2LS ceramic with 0.4 mol. % Sm{sub 2}O{sub 3} exhibited temperature-independent properties (25–150 °C), fatigue-free behavior (up to 10{sup 6} cycles), and good piezoelectric properties (d{sub 33}{sup * }= 230 pm/V, d{sub 33} = 176 pC/N, k{sub p} = 35%). Studies on the photoluminescence properties of the samples showed strong reddish-orange emission upon blue light excitation; these emission intensities were strongly dependent on the doping concentration and sintering temperature. The 0.4 mol. % Sm{sub 2}O{sub 3}-modified sample exhibited temperature responses over a wide temperature range of 10–443 K. The maximum sensing sensitivity of the sample was 7.5 × 10{sup −4} K at 293 K, at which point PPT occurred. A relatively long decay lifetime τ of 1.27–1.40 ms and a large quantum yield η of 0.17–0.19 were obtained from the Sm-modified samples. These results suggest that the KNN-5.2LS-xSm{sub 2}O{sub 3} system presents multifunctional properties and significant technological potential in novel multifunctional devices.

    This work reports the synthesis at relatively low temperatures of a highly emissive AlN:Mn2+ emitter. Though the AlN matrix shows an emission peak at a similar position to the emission peak observed for AlN:Mn product, the Mn-containing species generates red emission by a different mechanism, which was supported by the emission life time studies. The PLQY of the AlN:Mn emitter was measured to be 82%, the highest ever reported on a RE free-based phosphor.

    The operator of the Energy Department's Savannah River weapons plant has at least 1,800 more employees than it needs, and could save $400 million over a five-year period by cutting its staff accordingly, a DOE inspector general study says. Most of the boat - 1,206 employees - was attributed to excessive numbers of managers, with the inspector general concluding that Westinghouse Savannah River Co. had roughly twice as many layers of management than two other DOE weapons contractors. The study also concluded that Westinghouse in fiscal year 1992 significantly understated its actual staffing levels in reports to DOE, failing to disclose 1,765 full-time employees or the equivalent hours worked. Through such underreporting Westinghouse was able to [open quotes]circumvent staffing ceilings established by the department,[close quotes] the study added. Overall, DOE Inspector General John Layton said Westinghouse's staff levels substantially exceeded those needed for efficient operation of the South Carolina nuclear weapons facility. Layton based his analysis on efficiency standards attained by other DOE weapons plant contractors, such as Martin Marietta Energy Systems at DOE's Oak Ridge, Tenn., plant and EG G Rocky Flats, as well as widely utilized worker performance requirements used by the Navy and private sector companies that perform work similar to that done at Savannah River.

    Optical properties of Sm{sup 3+} doped different fluorophosphate glasses have been synthesized and discussed. The J- O intensity parameters Ω{sub λ} (λ= 2, 4, 6) from absorption spectra have been evaluated. No sharp edges are found in the absorption spectra, which confirm amorphous nature of present glass matrices. Various radiative parameters have been obtained from luminescence spectra for excited states to corresponding {sup 4}G{sub 5/2}→{sup 6}H{sub 5/2}, {sup 6}H{sub 7/2}, {sup 6}H{sub 9/2} and {sup 6}H{sub 11/2} transitions. The nature of decay curve analysis was performed for the {sup 4}G{sub 5/2} level. These glasses are expected to give interesting application in field of optical devices.

    Eu2+-activated phosphors are being widely used in illuminations and displays. Some of these phosphors feature an extremely broad and red-shifted Eu2+ emission band; however, convincing explanation of this phenomenon is lacking. Here we report a new localized/delocalized emitting state of Eu2+ ions in a new hexagonal EuAl2O4 phosphor whose Eu2+ luminescence exhibits a very large bandwidth and an extremely large Stokes shift. At 77 K, two luminescent sites responsible for 550 nm and 645 nm broadband emissions are recognized, while at room temperature only the 645 nm emission band emits. The 645 nm emission exhibits a typical radiative lifetime ofmore » 1.27 μs and an unusually large Stokes shift of 0.92 eV. We identify the 645 nm emission as originating from a new type of emitting state whose composition is predominantly that of localized 4f65d character but which also contains a complementary component with delocalized conduction-band-like character. This investigation gives new insights into a unique type of Eu2+ luminescence in solids whose emission exhibits both a very large bandwidth and an extremely large Stokes shift.« less
