The precise quantitative analysis of biomass sugars is a very important step in the conversion of biomass feedstocks to fuels and chemicals. However, the most accurate method of biomass sugar analysis is based on the gas chromatography analysis of derivatized sugars either as alditol acetates or trimethylsilanes. The derivatization method is time consuming but the alternative high-performance liquid chromatography (HPLC) method cannot resolve most sugars found in biomass hydrolysates. We have demonstrated for the first time that by careful manipulation of the HPLC mobile phase, biomass monomeric sugars (arabinose, xylose, fructose, glucose, mannose, and galactose) can be analyzed quantitatively and there is excellent baseline resolution of all the sugars. This method was demonstrated for standard sugars, pretreated corn stover liquid and solid fractions. Our method can also be used to analyze dimeric sugars (cellobiose and sucrose).

  5. Characterization of Sugar Insensitive (sis) Mutants of Arabidopsis

    SciTech Connect (OSTI)

    Gibson, Susan I.

    2009-06-08

Despite the fact that soluble sugar levels have been postulated to play an important role in the control of a wide variety of plant metabolic and developmental pathways, the mechanisms by which plants respond to soluble sugar levels remain poorly understood. Plant responses to soluble sugar levels are also important in bioenergy production, as plant sugar responses are believed to help regulate both carbon fixation and carbon partitioning. For example, accumulation of soluble sugars, such as sucrose and glucose, in source tissues leads to feedback inhibition of photosynthesis, thereby decreasing rates of carbon fixation. Soluble sugar levels can also affect sink strengths, affecting the rates of accumulation of carbon-based compounds into both particular molecular forms (e.g. carbohydrates versus lipids versus proteins) and particular plant organs and tissues. Mutants of Arabidopsis that are defective in the ability to respond to soluble sugar levels were isolated and used as tools to identify some of the factors involved in plant sugar response. These sugar insensitive (sis) mutants were isolated by screening mutagenized seeds for those that were able to germinate and develop relatively normal shoot systems on media containing 0.3 M glucose or 0.3 M sucrose. At these sugar concentrations, wild-type Arabidopsis germinate and produce substantial root systems, but show little to no shoot development. Twenty-eight sis mutants were isolated during the course of four independent mutant screens. Based on a preliminary characterization of all of these mutants, sis3 and sis6 were chosen for further study. Both of these mutations appear to lie in previously uncharacterized loci. Unlike many other sugar-response mutants, sis3 mutants exhibit a wild-type or near wild-type response in all phytohormone-response assays conducted to date. The sis6-1 mutation is unusual in that it appears to be due to overexpression of a gene, rather than representing a loss of function mutation. Characterization of mutant and wild-type plants has revealed that sugars inhibit breakdown of seed storage lipids. In addition, high concentrations of exogenous sugars largely eliminate the development of mature chloroplasts by developing seedlings. Affymetrix GeneChip experiments have revealed that expression of many plant genes is partially regulated by sugar levels, with approximately two percent of genes exhibiting alterations in steady-state mRNA levels in response to changing sugar concentrations. Ultimately, a better understanding of plant sugar responses may allow improvements in rates of carbon fixation and manipulation of carbon partitioning. These improvements will be needed to help make production of energy from biomass more economically attractive.

The invention includes a method of treating a solid acid catalyst. After exposing the catalyst to a mixture containing a sugar alcohol, the catalyst is washed with an organic solvent and is then exposed to a second reaction mixture. The invention includes a process for production of anhydrosugar alcohol. A solid acid catalyst is provided to convert sugar alcohol in a first sample to an anhydrosugar alcohol. The catalyst is then washed with an organic solvent and is subsequently utilized to expose a second sample. The invention includes a method for selective production of an anhydrosugar. A solid acid catalyst is provided within a reactor and anhydrosugar alcohol is formed by flowing a starting sugar alcohol into the reactor. The acid catalyst is then exposed to an organic solvent which allows a greater amount of additional anhydrosugar to be produced than would occur without exposing the acid catalyst to the organic solvent.

  6. Low-Temperature Catalytic Process To Produce Hydrocarbons From Sugars

    DOE Patents [OSTI]

Cortright, Randy D. (Madison, WI); Dumesic, James A. (Verona, WI)

    2005-11-15

Disclosed is a method of producing hydrogen from oxygenated hydrocarbon reactants, such as methanol, glycerol, sugars (e.g. glucose and xylose), or sugar alcohols (e.g. sorbitol). The method takes place in the condensed liquid phase. The method includes the steps of reacting water and a water-soluble oxygenated hydrocarbon in the presence of a metal-containing catalyst. The catalyst contains a metal selected from the group consisting of Group VIIIB transitional metals, alloys thereof, and mixtures thereof. The disclosed method can be run at lower temperatures than those used in the conventional steam reforming of alkanes.

  7. Engineering assessment of inactive uranium mill tailings: Monument Valley Site, Monument Valley, Arizona

    SciTech Connect (OSTI)

    Not Available

    1981-10-01

Ford, Bacon and Davis Utah Inc. has reevalated the Monument Valley site in order to revise the March 1977 engineering assessment of the problems resulting from the existence of radioactive uranium mill tailings at Monument Valley, Arizona. This engineering assessment has included the preparation of topographic maps, the performance of core drillings and radiometric measurements sufficient to determine areas and volumes of tailings and radiation exposure of individuals and nearby populations, the investigations of site hydrology and meteorology, and the evaluation and costing of alternative corrective actions. Radon gas released from the 1.1 million tons of tailings at the Monument Valley site constitutes the most significant environmental impact, although windblown tailings and external gamma radiation also are factors. The four alternative actions presented in this engineering assessment range from millsite decontamination with the addition of 3 m of stabilization cover material (Option I), to removal of the tailings to remote disposal sites and decontamination of the tailings site (Options II through IV). Cost estimates for the four options range from about $6,600,000 for stabilization in-place, to about $15,900,000 for disposal at a distance of about 15 mi. Three principal alternatives for reprocessing the Monument Valley tailings were examined: heap leaching; Treatment at an existing mill; and reprocessing at a new conventional mill constructed for tailings reprocessing. The cost of the uranium recovery is economically unattractive.

  11. Restricting lignin and enhancing sugar deposition in secondary cell walls enhances monomeric sugar release after low temperature ionic liquid pretreatment

DOE Public Access Gateway for Energy & Science Beta (PAGES Beta)

Scullin, Chessa; Cruz, Alejandro G.; Chuang, Yi -De; Simmons, Blake A.; Loque, Dominique; Singh, Seema

    2015-07-04

Lignocellulosic biomass has the potential to be a major source of renewable sugar for biofuel production. Before enzymatic hydrolysis, biomass must first undergo a pretreatment step in order to be more susceptible to saccharification and generate high yields of fermentable sugars. Lignin, a complex, interlinked, phenolic polymer, associates with secondary cell wall polysaccharides, rendering them less accessible to enzymatic hydrolysis. Herein, we describe the analysis of engineered Arabidopsis lines where lignin biosynthesis was repressed in fiber tissues but retained in the vessels, and polysaccharide deposition was enhanced in fiber cells with little to no apparent negative impact on growth phenotype.

    DOE Patents [OSTI]

Dumesic, James A.; Luterbacher, Jeremy S.

    2015-06-02

A process to produce an aqueous solution of carbohydrates that contains C6-sugar-containing oligomers, C6 sugar monomers, C5-sugar-containing oligomers, C5 sugar monomers, or any combination thereof is presented. The process includes the steps of reacting biomass or a biomass-derived reactant with a solvent system including a lactone and water, and an acid catalyst. The reaction yields a product mixture containing water-soluble C6-sugar-containing oligomers, C6-sugar monomers, C5-sugar-containing oligomers, C5-sugar monomers, or any combination thereof. A solute is added to the product mixture to cause partitioning of the product mixture into an aqueous layer containing the carbohydrates and a substantially immiscible organic layer containing the lactone.

    2009-03-22

This study considered assessing the feasibility of developing small scale hydro-electric power from seven major tributaries within the Hoopa Valley Indian Reservation of Northern California (http://www.hoopa-nsn.gov/). This study pursued the assessment of seven major tributaries of the Reservation that flow into the Trinity River. The feasibility of hydropower on the Hoopa Valley Indian Reservation has real potential for development and many alternative options for project locations, designs, operations and financing. In order to realize this opportunity further will require at least 2-3 years of intense data collection focusing on stream flow measurements at multiple locations in order to quantify real power potential. This also includes on the ground stream gradient surveys, road access planning and grid connectivity to PG&E for sale of electricity. Imperative to this effort is the need for negotiations between the Hoopa Tribal Council and PG&E to take place in order to finalize the power rate the Tribe will receive through any wholesale agreement that utilizes the alternative energy generated on the Reservation.

DOE Data Explorer [Office of Scientific and Technical Information (OSTI)]

In 2011 and 2012, the Ganges Valley Aerosol Experiment (GVAX) began in the Ganges Valley region of India. The objective was to obtain measurements of clouds, precipitation, and complex aerosols to study their impact on cloud formation and monsoon activity in the region. During the Indian Ocean Experiment (INDOEX) field studies, aerosols from the Ganges Valley region were shown to affect cloud formation and monsoon activity over the Indian Ocean. The complex field study used the ARM Mobile Facility (AMF) to measure radiative, cloud, convection, and aerosol characteristics over the mainland. The resulting data set captured pre-monsoon to post-monsoon conditions to establish a comprehensive baseline for advancements in the study of the effects of atmospheric conditions of the Ganges Valley.

  14. Ward Valley status report: Science versus politics. Which will win?

    SciTech Connect (OSTI)

    1996-10-01

The State of California has issued a license to US Ecology, Inc. to construct and operate a disposal facility for low-level radioactive waste (LLRW) at the remote, arid Ward Valley site in the Mojave Desert. The license and certification of the associated environmental documentation have been upheld by the California courts. The Ward Valley license is the first and, so far, only license to be issued for a new LLRW disposal facility pursuant to the Low-Level Radioactive Waste Policy Act enacted in 1980 and amended in 1985. However, the dates of construction and operation of the disposal facility are uncertain because the federal government has refused to sell land in Ward Valley to the State of California for the site of the Southwestern Compact`s regional disposal facility. The Clinton Administration`s repeated excuses for delaying the land transfer, and the circumstances of these delays, indicate that prospects for success of the Ward Valley project, and perhaps the Policy Act itself, depend on the outcome of a battle between science and politics. In view of these delays by the administration, Congressional action to Transfer the Ward Valley lands to California will serve both state and federal goals for safe disposal of LLRW.

  15. Feasibility of converting a sugar beet plant to fuel ethanol production

    SciTech Connect (OSTI)

    Hammaker, G S; Pfost, H B; David, M L; Marino, M L

    1981-04-01

This study was performed to assess the feasibility of producing fuel ethanol from sugar beets. Sugar beets are a major agricultural crop in the area and the beet sugar industry is a major employer. There have been some indications that increasing competition from imported sugar and fructose sugar produced from corn may lead to lower average sugar prices than have prevailed in the past. Fuel ethanol might provide an attractive alternative market for beets and ethanol production would continue to provide an industrial base for labor. Ethanol production from beets would utilize much of the same field and plant equipment as is now used for sugar. It is logical to examine the modification of an existing sugar plant from producing sugar to ethanol. The decision was made to use Great Western Sugar Company's plant at Mitchell as the example plant. This plant was selected primarily on the basis of its independence from other plants and the availability of relatively nearby beet acreage. The potential feedstocks assessed included sugar beets, corn, hybrid beets, and potatoes. Markets were assessed for ethanol and fermentation by-products saleability. Investment and operating costs were determined for each prospective plant. Plants were evaluated using a discounted cash flow technique to obtain data on full production costs. Environmental, health, safety, and socio-economic aspects of potential facilities were examined. Three consulting engineering firms and 3 engineering-construction firms are considered capable of providing the desired turn-key engineering design and construction services. It was concluded that the project is technically feasible. (DMC)

The present invention provides a process for preparation of low molecular weight polyols from high molecular weight polyols in a hydrogenolysis reaction under elevated temperature and hydrogen pressure. The process comprises providing in a reaction mixture the polyols, a base, and a metal catalyst prepared by depositing a transition metal salt on an inert support, reducing the metal salt to the metal with hydrogen, and passivating the metal with oxygen, and wherein the catalyst is reduced with hydrogen prior to the reaction. In particular, the process provides for the preparation of glycerol, propylene glycol, and ethylene glycol from sugar alcohols such as sorbitol or xylitol. In a preferred process, the metal catalyst comprises ruthenium which is deposited on an alumina, titania, or carbon support, and the dispersion of the ruthenium on the support increases during the hydrogenolysis reaction.

An accurate simulation of the evolution of the atmospheric boundary layer is very important, as the evolution of the boundary layer sets the stage for many weather phenomena, such as deep convection. Over mountain areas the evolution of the boundary layer is particularly complex, due to the nonlinear interaction between boundary layer turbulence and thermally-induced mesoscale wind systems, such as the slope and valley winds. As the horizontal resolution of operational forecasts progresses to finer and finer resolution, more and more of the thermally-induced mesoscale wind systems can be explicitly resolved, and it is very timely to document the current state-of-the-art of mesoscale models at simulating the coupled evolution of the mountain boundary layer and the valley wind system. In this paper we present an intercomparison of valley wind simulations for an idealized valley-plain configuration using eight state-of-the-art mesoscale models with a grid spacing of 1 km. Different sets of three-dimensional simulations are used to explore the effects of varying model dynamical cores and physical parameterizations. This intercomparison project was conducted as part of the Terrain-induced Rotor Experiment (T-REX; Grubisic et al., 2008).

Above-normal precipitation and surface-water runoff, which have been generally related to the cyclic recurrence of the El Nino-Southern Oscillation, have produced modern ephemeral lakes in the closed-basin Death Valley watershed. This study evaluates the regional hydroclimatic relations between precipitation, runoff, and lake transgressions in the Death Valley watershed. Recorded precipitation, runoff, and spring discharge data for the region are used in conjunction with a closed-basin, lake-water-budget equation to assess the relative contributions of water from these sources to modern lakes in Death Valley and to identify the requisite hydroclimatic changes for a late Holocene perennial lake in the valley. As part of the Yucca Mountain Site Characterization Program, an evaluation of the Quaternary regional paleoflood hydrology of the potential nuclear-waste repository site at Yucca Mountain, Nevada, was planned. The objectives of the evaluation were (1) to identify the locations and investigate the hydraulic characteristics of paleofloods and compare these with the locations and characteristics of modern floods, and (2) to evaluate the character and severity of past floods and debris flows to ascertain the potential future hazards to the potential repository during the pre-closure period (US Department of Energy, 1988). This study addresses the first of these objectives, and the second in part, by assessing and comparing the sizes, locations, and recurrence rates of modern, recorded (1962--83) floods and late Holocene paleofloods for the 8,533-mi{sup 2}, closed-basin, Death Valley watershed with its contributing drainage basins in the Yucca Mountain site area.

Disclosed in the present invention is a Zymomonas integrant and derivatives of these integrants that posses the ability to ferment pentose into ethanol. The genetic sequences encoding for the pentose-fermenting enzymes are integrated into the Zymomonas in a two-integration event of homologous recombination and transposition. Each operon includes more than one pentose-reducing enzyme encoding sequence. The integrant in some embodiments includes enzyme sequences encoding xylose isomerase, xylulokinase, transketolase and transketolase. The Zymomonas integrants are highly stable, and retain activity for producing the pentose-fermenting enzyme for between 80 to 160 generations. The integrants are also resistant to acetate inhibition, as the integrants demonstrate efficient ethanol production even in the presence of 8 up to 16 grams acetate per liter media. These stably integrated sequences provide a unique Zymomonas that may then be used for the efficient conversion of pentose sugars (xylose, arabinose) to ethanol. Method of using the Zymomonas integrants and derivatives thereof in production of ethanol from cellulosic feedstock is also disclosed. The invention also provides a method for preparing a Zymomonas integrant as part of the present invention. The host Zymomonas strain found particularly useful in the creation of these compositions and methods is Zymomonas mobilis 31821.

We show that inversion symmetry breaking together with spin-orbit coupling leads to coupled spin and valley physics in monolayer MoS2 and group-VI dichalcogenides, making possible controls of spin and valley in these 2D materials. The spin-valley coupling at the valence band edges suppresses spin and valley relaxation, as flip of each index alone is forbidden by the 0.1 eV valley contrasting spin splitting. Valley Hall and spin Hall effects coexist in both electron-doped and hole-doped systems. Optical interband transitions have frequency-dependent polarization selection rules which allow selective photoexcitation of carriers with various combination of valley and spin indices. Photo-induced spin Hall and valley Hall effects can generate long lived spin and valley accumulations on sample boundaries. The physics discussed here provides a route towards the integration of valleytronics and spintronics in multi-valley materials with strong spin-orbit coupling and inversion symmetry breaking.

The Monument Valley Uranium Mill Tailings Remedial Action (UMTRA) Project site in Cane Valley is a former uranium mill that has undergone surface remediation in the form of tailings and contaminated materials removal. Contaminated materials from the Monument Valley (Arizona) UMTRA Project site have been transported to the Mexican Hat (Utah) UMTRA Project site for consolidation with the Mexican Hat tailings. Tailings removal was completed in February 1994. Three geologic units at the site contain water: the unconsolidated eolian and alluvial deposits (alluvial aquifer), the Shinarump Conglomerate (Shinarump Member), and the De Chelly Sandstone. Water quality analyses indicate the contaminant plume has migrated north of the site and is mainly in the alluvial aquifer. An upward hydraulic gradient in the De Chelly Sandstone provides some protection to that aquifer. This water sampling and analysis plan recommends sampling domestic wells, monitor wells, and surface water in April and September 1994. The purpose of sampling is to continue periodic monitoring for the surface program, evaluate changes to water quality for site characterization, and provide data for the baseline risk assessment. Samples taken in April will be representative of high ground water levels and samples taken in September will be representative of low ground water levels. Filtered and nonfiltered samples will be analyzed for plume indicator parameters and baseline risk assessment parameters.

This report documents the results of a geologic study of the area between the underground-nuclear-explosion testing areas on Pahute Mesa, in the northwesternmost part of the Nevada Test Site, and the springs in Oasis Valley, to the west of the Test Site. The new field data described in this report are also presented in a geologic map that is a companion product(Fridrich and others, 1999) and that covers nine 7.5-minute quadrangles centered on Thirsty Canyon SW, the quadrangle in which most of the Oasis Valley springs are located. At the beginning of this study, published detailed maps were available for 3 of the 9 quadrangles of the study area: namely Thirsty Canyon (O'Connor and others, 1966); Beatty (Maldonado and Hausback, 1990); and Thirsty Canyon SE (Lipman and others, 1966). Maps of the last two of these quadrangles, however, required extensive updating owing to recent advances in understanding of the regional structure and stratigraphy. The new map data are integrated in this re port with new geophysical data for the Oasis Valley area, include gravity, aeromagnetic, and paleomagnetic data (Grauch and others, 1997; written comm., 1999; Mankinen and others, 1999; Hildenbrand and others, 1999; Hudson and others, 1994; Hudson, unpub. data).

In monolayer group-VI transition metal dichalcogenides, charge carriers have spin and valley degrees of freedom, both associated with magnetic moments. On the other hand, the layer degree of freedom in multilayers is associated with electrical polarization. Here we show that transition metal dichalcogenide bilayers offer an unprecedented platform to realize a strong coupling between the spin, valley and layer pseudospin of holes. Such coupling gives rise to the spin Hall effect and spin-dependent selection rule for optical transitions in inversion symmetric bilayer and leads to a variety of magnetoelectric effects permitting quantum manipulation of these electronic degrees of freedom. Oscillating electric and magnetic fields can both drive the hole spin resonance where the two fields have valley-dependent interference, making an interplay between the spin and valley as information carriers possible for potential valley-spintronic applications. We show how to realize quantum gates on the spin qubit controlled by the valley bit.
