    Spirochaeta caldaria Pohlschroeder et al. 1995 is an obligately anaerobic, spiral-shaped bac- terium that is motile via periplasmic flagella. The type strain, H1T, was isolated in 1990 from cyanobacterial mat samples collected at a freshwater hot spring in Oregon, USA, and is of in- terest because it enhances the degradation of cellulose when grown in co-culture with Clos- tridium thermocellum. Here we provide a taxonomic re-evaluation for S. caldaria based on phylogenetic analyses of 16S rRNA sequences and whole genomes, and propose the reclassi- fication of S. caldaria and two other Spirochaeta species as members of the emended genus Treponema. Whereas genera such as Borrelia and Sphaerochaeta possess well-distinguished genomic features related to their divergent lifestyles, the physiological and functional ge- nomic characteristics of Spirochaeta and Treponema appear to be intermixed and are of little taxonomic value. The 3,239,340 bp long genome of strain H1T with its 2,869 protein-coding and 59 RNA genes is a part of the Genomic Encyclopedia of Bacteria and Archaea project.

    The 8.2 kyr event is the largest abrupt climatic change recorded in the last 10,000 years, and is widely hypothesized to have been triggered by the release of thousands of kilometers cubed of freshwater into the North Atlantic Ocean. Using a high-resolution (1/6°) global, ocean-ice circulation model we present an alternative view that freshwater discharged from glacial Lake Agassiz would have remained on the continental shelf as a narrow, buoyant, coastal current, and would have been transported south into the subtropical North Atlantic. The pathway we describe is in contrast to the conceptual idea that freshwater from this lake outburst spread over most of the sub-polar North Atlantic, and covered the deep, open-ocean, convection regions. This coastally confined freshwater pathway is consistent with the present-day routing of freshwater from Hudson Bay, as well as paleoceanographic evidence of this event. Using a coarse-resolution (2.6°) version of the same model, we demonstrate that the previously reported spreading of freshwater across the sub-polar North Atlantic results from the inability of numerical models of this resolution to accurately resolve narrow coastal flows, producing instead a diffuse circulation that advects freshwater away from the boundaries. To understand the climatic impact of freshwater released in the past or future (e.g. Greenland and Antarctica), the ocean needs to be modeled at a resolution sufficient to resolve the dynamics of narrow, coastal buoyant flows.

  8. Effects on Freshwater Organisms of Magnetic Fields Associated with Hydrokinetic Turbines

    Underwater cables will be used to transmit electricity between turbines in an array (interturbine cables), between the array and a submerged step-up transformer (if part of the design), and from the transformer or array to shore. All types of electrical transmitting cables (as well as the generator itself) will emit EMF into the surrounding water. The electric current will induce magnetic fields in the immediate vicinity, which may affect the behavior or viability of animals. Because direct electrical field emissions can be prevented by shielding and armoring, we focused our studies on the magnetic fields that are unavoidably induced by electric current moving through a generator or transmission cable. These initial experiments were carried out to evaluate whether a static magnetic field, such as would be produced by a direct current (DC) transmitting cable, would affect the behavior of common freshwater fish and invertebrates.

    This document is the first annual report for the study titled 'Ecology of Juvenile Salmonids in Shallow Tidal Freshwater Habitats in the Vicinity of the Sandy River Delta in the Lower Columbia River'. Hereafter, we refer to this research as the Tidal Freshwater Monitoring (TFM) Study. The study is part of the research, monitoring, and evaluation effort developed by the Action Agencies (Bonneville Power Administration, U.S. Army Corps of Engineers, U.S. Bureau of Reclamation) in response to obligations arising from the Endangered Species Act as a result of operation of the Federal Columbia River Power System (FCRPS). The project is performed under the auspices of the Northwest Power and Conservation Council's Columbia Basin Fish and Wildlife Program. The goal of the 2007-2009 Tidal Freshwater Monitoring Study is to answer the following questions: In what types of habitats within the tidal freshwater area of the lower Columbia River and estuary (LCRE; Figure 1) are yearling and subyearling salmonids found, when are they present, and under what environmental conditions?1 And, what is the ecological importance2 of shallow (0-5 m) tidal freshwater habitats to the recovery of Upper Columbia River spring Chinook salmon and steelhead and Snake River fall Chinook salmon? Research in 2007 focused mainly on the first question, with fish stock identification data providing some indication of Chinook salmon presence at the variety of habitat types sampled. The objectives and sub-objectives for the 2007 study were as follows: (1) Habitat and Fish Community Characteristics-Provide basic data on habitat and fish community characteristics for yearling and subyearling salmonids at selected sites in the tidal freshwater reach in the vicinity of the Sandy River delta.   10. Potential Risks of Freshwater Aquifer Contamination with Geosequestration

    Substantial leakage of CO{sub 2} from deep geological strata to shallow potable aquifers is likely to be rare, but chemical detection of potential leakage nonetheless remains an integral component of any safe carbon capture and storage system. CO{sub 2} that infiltrates an unconfined freshwater aquifer will have an immediate impact on water chemistry by lowering pH in most cases and by altering the concentration of total dissolved solids. Chemical signatures in affected waters provide an important opportunity for early detection of leaks. In the presence of CO{sub 2}, trace elements such as Mn, Fe, and Ca can increase by an order of magnitude or more above control concentrations within 100 days. Therefore, these and other elements should be monitored along with pH as geochemical markers of potential CO{sub 2} leaks. Dissolved inorganic carbon and alkalinity can also be rapidly responsive to CO{sub 2} and are stable indicators of a leak. Importantly, such changes may be detectable long before direct changes in CO{sub 2} are observed. The experimental results also suggest that the relative severity of the impact of leaks on overlying drinking-water aquifers should be considered in the selection of CO{sub 2} sequestration sites. One primary selection criteria should be metal and metalloid availability, such as uranium and arsenic abundance, to carefully monitor chemical species that could trigger changes above maximum contaminant levels (MCLs). Overall, the risks of leakage from underground CO{sub 2} storage are real but appear to be manageable if systems are closely monitored.

  11. Freshwater Availability and Constraints on Thermoelectric Power Generation in the Southeast U.S.

    There is a myriad of uses to which our country's freshwater supply is currently committed. Together with increasing quantities of consumption, there are growing constraints on water availability. In our future there will be two elements of consumption at the forefront of concern: availability and efficiency. Availability of freshwater is the most important of these and is the subject of this report. To use water efficiently, we must first have it. Efficiency is key to ensuring availability for future needs. As population grows and economic and technology demands increase - especially for thermoelectric power - needs for freshwater will also increase. Thus, using our limited supplies of freshwater must be done as efficiently as possible. Thermoelectric generating industry is the largest user of our nation's water resources, including fresh, surface, ground, and saline water. Saline water use accounts for approximately 30% of thermoelectric use, while the remaining 70% is from freshwater sources. The U.S. Geological Survey (USGS) estimates that thermoelectric generation accounts for roughly 136,000 million gallons per day (MGD), or 39% of freshwater withdrawals. This ranks slightly behind agricultural irrigation as the top source of freshwater withdrawals in the U.S. in 2000. For Americans to preserve their standard of living and maintain a thriving economy it is essential that greater attention be paid to freshwater availability in efforts to meet energy demands - particularly for electric power. According to projections by the Energy Information Administration's (EIA) Annual Energy Outlook 2006 (AEO 2006) anticipated growth of thermoelectric generating capacity will be 22% between 2005 and 2030. In the 2007 Report, EIA estimates that capacity to grow from approximately 709 GW in 2005 to 862 GW in 20303. These large increases in generating capacity will result in increased water demands by thermoelectric power plants and greater competition over water between the

    For freshwater production from open-cycle OTEC systems, the suitability of commercially available mist eliminators is examined. The mist eliminators are characterized in terms of their liquid collection efficiencies, allowable vapor velocities at the onset of reentrainment, and pressure losses. Suitable design modifications can be projected to allow steam velocities of up to 35 m/s, with a corresponding parasitic power loss of less than 5% of the gross potential of an open-cycle OTEC power system.

    The tidal freshwater monitoring (TFM) project reported herein is part of the research, monitoring, and evaluation effort developed by the Action Agencies (Bonneville Power Administration, the U.S. Army Corps of Engineers [USACE], and the U.S. Bureau of Reclamation) in response to obligations arising from the Endangered Species Act (ESA) as a result of operation of the Federal Columbia River Power System. The project is being performed under the auspices of the Northwest Power and Conservation Council's Columbia Basin Fish and Wildlife Program (Project No. 2005-001-00). The research is a collaborative effort among the Pacific Northwest National Laboratory, the Oregon Department of Fish and Wildlife, the National Marine Fisheries Service, and the University of Washington. The overarching goal of the TFM project is to bridge the gap in knowledge between tidal freshwater habitats and the early life history attributes of migrating salmon. The research questions include: In what types of habitats within the tidal freshwater area of the Columbia River are juvenile salmon found, when are they present, and under what environmental conditions? What is the ecological contribution of shallow (0-5 m) tidal freshwater habitats to the recovery of ESA-listed salmon in the Columbia River basin? Field data collection for the TFM project commenced in June 2007 and since then has continued monthly at six to nine sites in the vicinity of the Sandy River delta (river kilometer 192-208). While this report includes summary data spanning the 19-month period of study from June 2007 through December 2008, it highlights sampling conducted during calendar year 2008. Detailed data for calendar year 2007 were reported previously.   18. Potential nanotechnology applications for reducing freshwater consumption at coal fired power plants : an early view.

    This report was funded by the U.S. Department of Energy's (DOE's) National Energy Technology Laboratory (NETL) Existing Plants Research Program, which has an energy-water research effort that focuses on water use at power plants. This study complements the overall research effort of the Existing Plants Research Program by evaluating water issues that could impact power plants. A growing challenge to the economic production of electricity from coal-fired power plants is the demand for freshwater, particularly in light of the projected trends for increasing demands and decreasing supplies of freshwater. Nanotechnology uses the unique chemical, physical, and biological properties that are associated with materials at the nanoscale to create and use materials, devices, and systems with new functions and properties. It is possible that nanotechnology may open the door to a variety of potentially interesting ways to reduce freshwater consumption at power plants. This report provides an overview of how applications of nanotechnology could potentially help reduce freshwater use at coal-fired power plants. It was developed by (1) identifying areas within a coal-fired power plant's operations where freshwater use occurs and could possibly be reduced, (2) conducting a literature review to identify potential applications of nanotechnology for facilitating such reductions, and (3) collecting additional information on potential applications from researchers and companies to clarify or expand on information obtained from the literature. Opportunities, areas, and processes for reducing freshwater use in coal-fired power plants considered in this report include the use of nontraditional waters in process and cooling water systems, carbon capture alternatives, more efficient processes for removing sulfur dioxide and nitrogen oxides, coolants that have higher thermal conductivities than water alone, energy storage options, and a variety of plant inefficiencies, which, if improved

  19. Evaluation of Common Angling-Induced Sources of Epithelial Damage for Popular Freshwater Sport Fish using Fluorescein

    Angling is a popular recreational activity across the globe and a large proportion of fish captured by anglers are released due to voluntary or mandatory catch-and-release practices. The handling associated with hook removal and return of the fish to their environment can cause physical damage to the epidermal layer of the fish which may affect the condition and survival of released fish. This study investigated possible sources of epithelial damage associated with several different handling methods (i.e. landing net types, interactions with different boat floor surfaces, tournament procedures) commonly used in recreational angling for two popular freshwater sport fish species, largemouth bass (Micropterus salmoides) and northern pike (Esox lucius). Epithelial damage was examined using fluorescein, a non-toxic dye, which has been shown to detect latent epithelial damage. Northern pike exhibited extensive epithelial damage after exposure to several of the induced treatments (i.e., interaction with a carpeted surface, knotted nylon net, and line rolling) but relatively little epithelial damage when exposed to others (i.e., knotless rubber nets, smooth boat surfaces, or lip gripping devices). Largemouth bass did not show significant epithelial damage for any of the treatments, with the exception of fish caught in a semi-professional live release tournament. The detection of latent injuries using fluorescein can be an important management tool as it provides visual examples of potential damage that can be caused by different handling methods. Such visualizations can be used to encourage fish friendly angler behaviour and enhance the survival and welfare of released fish. It can also be used to test new products that are intended to or claim to reduce injury to fish that are to be released. Future research should evaluate the relationship between different levels of epithelial damage and mortality across a range of environmental conditions.

  20. The occurrence of freshwater limestones in the Upper Pennsylvanian and lower Permian of the northern Appalachian basin

    Freshwater limestones comprise a significant portion of the Upper Pennsylvanian and Lower Permian strata of the northern Appalachian basin. In some areas, the Monogahela Group consists of as much as 50% freshwater limestones. These limestones are clayey to silty, iron-bearing, sparsely fossiliferous, micritic, and thinly bedded or nodular and are presumed to be lacustrine in origin. They are part of a recurring sequence of beds that include coal, sandstone, shale/clay, and marine limestone. The initial goals of this study are to develop a stratigraphic framework for these freshwater limestones and to determine, their lateral continuity, thickness variability, and relation to other beds in this section, in particular the coals, on a regional scale. The Pittsburgh coal and Ames limestone are very persistent regionally and are used as a datum. Regional correlations show that possibly 20 freshwater limestone beds are persistent and widespread; some cover at least 5,000 mi{sup 2} in eastern Ohio, western Pennsylvania, northern West Virginia and western Maryland. Cross sections and fence diagrams constructed from drill logs and measured sections indicate that the thickness of these limestone beds varies laterally and between beds, ranging from less than 1 to 150 ft thick. Correlations also show stratigraphic and paleogeographic relationships between the coals and freshwater limestones. Many of the limestones underlie coal beds, directly in some places. The coals and limestones are better developed in the northern part of the Appalachian basin. These associations may hold significant clues to the depositional and the paleoclimatic setting of the freshwater limestones and the coals.

  1. Modeling Cd partitioning in oxic lake sediments and Cd concentrations in a freshwater bivalve

    The author measured cadmium concentrations in the biotic (soft tissues of a freshwater bivalve) and abiotic (oxic sediments and overlying water, diagenetic iron and manganese oxides) compartments of a large number of lakes. The lakes encompass a large range in pH and in Cd concentrations, and are distributed over a geographical area of about 350,000 km{sup 2}. The distribution of Cd between the sediments and the overlying water can be described by sorption to a limited number of sedimentary phases; apparent binding constants of Cd to these phases can be determined from in situ measurements. This approach is somewhat similar to that of ``Trace Metal Equilibrium Partitioning`` which has been considered by the US EPA for the development of sediment quality criteria. The author shows that the binding constants, together with the abundance of the sorbing phases, total sedimentary cadmium concentration and water pH, can be used to predict successfully cadmium accumulation in the freshwater bivalve.

    The TFM study was designed to investigate the ecology and early life history of juvenile salmonids within shallow (<5 m) tidal freshwater habitats of the LCRE. We started collecting field data in June 2007. Since then, monthly sampling has occurred in the vicinity of the Sandy River delta (rkm 192ā€“208) and at other sites and times in lower river reaches of tidal freshwater (rkm 110 to 141). This report provides a comprehensive synthesis of data covering the field period from June 2007 through April 2010.

Trachemys scripta (Agassiz) and Chelydra serpentina (Linnaeus) were selected for comparison based on species abundance and differences in food habits and sediment contact. A review of the literature on contaminants in turtles and results of preliminary surveys conducted at the field sites, which are included in this study, were used to direct and focus this research project. White Oak Lake, a settling basin for low-level radioactive and nonradioactive contaminants, and Bearden Creek Embayment, an uncontaminated reference site upriver, were used as study sites in the investigation of turtles as indicators of chemical contamination. Turtles were analyzed for concentrations of strontium-90, cesium-137, cobalt 60, and mercury in specific target tissues, and for single-stranded DNA breaks, a non-specific indicator of possible exposure to genotoxic agents in the environment. 133 refs., 2 figs., 15 tabs.

    Coal-fired power plants consume huge quantities of water, and in some water-stressed areas, power plants compete with other users for limited supplies. Extensive use of coal to generate electricity is projected to continue for many years. Faced with increasing power demands and questionable future supplies, industries and governments are seeking ways to reduce freshwater consumption at coal-fired power plants. As the United States investigates various freshwater savings approaches (e.g., the use of alternative water sources), other countries are also researching and implementing approaches to address similar - and in many cases, more challenging - water supply and demand issues. Information about these non-U.S. approaches can be used to help direct near- and mid-term water-consumption research and development (R&D) activities in the United States. This report summarizes the research, development, and deployment (RD&D) status of several approaches used for reducing freshwater consumption by coal-fired power plants in other countries, many of which could be applied, or applied more aggressively, at coal-fired power plants in the United States. Information contained in this report is derived from literature and Internet searches, in some cases supplemented by communication with the researchers, authors, or equipment providers. Because there are few technical, peer-reviewed articles on this topic, much of the information in this report comes from the trade press and other non-peer-reviewed references. Reducing freshwater consumption at coal-fired power plants can occur directly or indirectly.   11. Literature review of the concentration ratios of selected radionuclides in freshwater and marine fish

    Concentration ratios (CR's) used for modeling the uptake and food chain transport of radionuclides in fish have usually been conservative; that is, at the high end of reported values. This practice ensures that the dose to the consumer of contaminated fish will not be underestimated. In many models, however, conservative values have been used for all variables that have any uncertainty associated with them. As a result the dose to the consumer is overestimated. Realistic CR values need to be developed to establish model parameters that will accurately reflect tissue burdens in fish and resulting dose rates to consumers. This report reviews and summarizes published literature on the uptake and distribution of stable and radioactive isotopes of 26 elements. Based on this review, we have made recommendations on CR values to be used for modeling the accumulation of radionuclides in fish. Our recommendations are compared with CR values reported in other publications. A generic discussion of abiotic and biotic factors that influence CR values is provided so that CR values may be adjusted based on site-specific characteristics of the fishes habitat. Recommended CR values for freshwater fish and for marine fish are listed. Although this report emphasizes radionuclides, it is applicable to stable elements as well.

Furthermore, a ten-fold decrease in flow rate from 11 cubic meters per sec (cms) in 1987 to one cms in 1988 was recorded in the marsh followed by a shift to drier wetland communities. It is known that numerous marine and freshwater organisms are sensitive to electrical and magnetic fields, often depending on them for such diverse activities as prey location and navigation (DOE 2009; Normandeau et al. 2011). Despite the wide range of aquatic organisms that are sensitive to EMF and the increasing numbers of underwater electrical transmitting cables being installed in rivers and coastal waters, little information is available to assess whether animals will be attracted, repelled, or unaffected by these new sources of EMF. This knowledge gap is especially significant for freshwater systems, where electrosensitive organisms such as paddlefish and sturgeon may interact with electrical transmission cables. We carried out a series of laboratory experiments to test the sensitivity of freshwater fish and invertebrates to the levels of EMF that are expected to be produced by HK projects in rivers. In this context, EM fields are likely to be emitted primarily by generators in the water column and by transmission cables on or buried in the substrate. The HK units will be located in areas of high-velocity waters that are used as only temporary habitats for most riverine species, so long-term exposure of fish and benthic invertebrates to EMF is unlikely. Rather, most aquatic organisms will be briefly exposed to the fields as they drift downstream or migrate upstream. Because the exposure of most aquatic organisms to EMF in a river would be relatively brief and non-lethal, we focused our investigations on detecting behavioral effects. For example, attraction to the EM fields could result in prolonged exposures to the fields or the HK rotor. On the other hand, avoidance reactions might hinder upstream migrations of fish. The experiments reported here are a continuation of studies begun in FY 2010, which focused on the potential effects of static magnetic fields on snails, clams, and fathead minnows (Cada et al. 2011

  15. The comparative kinetics of Ca, Sr, And Ra in a freshwater turtle, Trachemys scripta

    The accumulation of {sup 45}Ca, {sup 47}Ca, {sup 88}Sr, and {sup 226}Ra was studied in the yellow-bellied slider, a common freshwater turtle of the Southeastern US. The author was particularly interested in testing the hypothesis of competitive inhibition, a concept whereby decreasing the intake of a stable dietary element increases the absorption and retention of chemically similar radionuclides. He established four specific hypotheses and examined the processes of absorption and elimination as a function of stable dietary calcium (2 and 20 mg g{sup {minus}1}), season (summer, fall, winter and spring), and age and sex of the animals (hatchlings, juveniles, adult males, adult females, and gravid females). Turtles were gavaged with radionuclides and the gamma-emitting isotopes were detected during serial whole-body counts performed on the live animals for up to 480 d. The analysis of the beta-emitting {sup 45}Ca was accomplished by chemical separation procedures. Data were fit to a two-component exponential retention model by nonlinear regression. The 10-fold reduction in dietary Ca did not affect the elimination rate constants, and increased the assimilation of Sr and Ra only within juveniles. For all animals the absorption of Ca was significantly greater than Sr, and likewise, Sr was greater than Ra. Mean absorptions were generally higher than values reported for other organisms. Unlike many other organisms, absorption rates did not decline at maturity. He suspects that high Ca demands in constructing and maintaining the massive shell, necessitated by the turtle's survival strategy, may contribute to the high absorption, as well as the lack of a decline at maturity. Elimination rate constants were greatest in the summer and declined to levels that were generally not distinguishable from zero in the spring, winter, and fall seasons.

In addition, turtles from White Oak Lake contained a significantly greater amount of DNA damage than those from the reference site. Although this suggests greater exposure of White Oak Lake turtles to genotoxic agents, further studies are needed to establish the cause of the enhanced amount of single-stranded breaks. Interspecific comparisons of the turtles from White Oak Lake indicated that diet may play a significant role in the exposure of turtles to certain contaminants. No difference was detected between the concentrations of {sup 90}Sr, {sup 137}Cs, {sup 60}Co between the two species.

  17. Laboratory Studies of the Short-term Responses of Freshwater Fish to Electromagnetic Fields

    Hydrokinetic energy technologies are being proposed as an environmentally preferred means of generating electricity from river and tidal currents. Among the potential issues that must be investigated in order to resolve environmental concerns are the effects on aquatic organisms of electromagnetic fields created by underwater generators and transmission cables. The behavioral responses of common freshwater fishes to static and variable electromagnetic fields (EMF) that may be emitted by hydrokinetic projects were evaluated in laboratory experiments. Various fish species were exposed to either static (DC) EMF fields created by a permanent bar magnet or variable (AC) EMF fields created by a switched electromagnet for 48 h, fish locations were recorded with a digital imaging system, and changes in activity level and distribution relative to the magnet position were quantified at 5-min intervals. Experiments with fathead minnows, redear sunfish, striped bass, lake sturgeon, and channel catfish produced mixed results. Except for fathead minnows there was no effect on activity level. Only redear sunfish and channel catfish exhibited a change in distribution relative to the position of the magnet with an apparent attraction to the EMF source. In separate experiments, rapid behavioral responses of paddlefish and lake sturgeon to onset of the AC field were recorded with high-speed video. Paddlefish did not react to a variable, 60-Hz magnetic field like that which would be emitted by an AC generator or cable, but lake sturgeon consistently responded to the variable, AC-generated magnetic field with a variety of altered swimming behaviors. These results will be useful for determining under what circumstances cables or generators need to be positioned to minimize interactions with sensitive species.

    Relative abundance profiles of bacterial populations measured by sequencing DNA or RNA of marker genes can widely differ. These differences, made apparent when calculating ribosomal RNA:DNA ratios, have been interpreted as variable activities of bacterial populations. However, inconsistent correlations between ribosomal RNA:DNA ratios and metabolic activity or growth rates have led to a more conservative interpretation of this metric as the cellular protein synthesis potential (PSP). Little is known, particularly in freshwater systems, about how PSP varies for specific taxa across temporal and spatial environmental gradients and how conserved PSP is across bacterial phylogeny. Here, we generated 16S rRNA genemoreĀ Ā» sequencing data using simultaneously extracted DNA and RNA from fractionated (free-living and particulate) water samples taken seasonally along a eutrophic freshwater estuary to oligotrophic pelagic transect in Lake Michigan. In contrast to previous reports, we observed frequent clustering of DNA and RNA data from the same sample. Analysis of the overlap in taxa detected at the RNA and DNA level indicated that microbial dormancy may be more common in the estuary, the particulate fraction, and during the stratified period. Across spatiotemporal gradients, PSP was often conserved at the phylum and class levels. PSPs for specific taxa were more similar across habitats in spring than in summer and fall. This was most notable for PSPs of the same taxa when located in the free-living or particulate fractions, but also when contrasting surface to deep, and estuary to Lake Michigan communities. Our results show that community composition assessed by RNA and DNA measurements are more similar than previously assumed in freshwater systems. Furthermore, the similarity between RNA and DNA measurements and taxa-specific PSPs that drive community-level similarities are conditional on spatiotemporal factors.Ā«Ā less

Although actively swimming or passively drifting animals may collide with any of the physical structures associated with hydrokinetic devices, turbine rotors are the most likely sources for risk of strike or significant collision (DOE 2009). It is also possible that during a close encounter with a HK device no physical contact will be made between the device and the organism, either because the animal avoids the device by successfully changing its direction of movement, or by successfully evading any moving parts of the device. Oak Ridge National Laboratory (ORNL) has been funded by the US Department of Energy (DOE) Waterpower Program to evaluate strike potential and consequences for Marine and Hydrokinetic (MHK) technologies in rivers and estuaries of the United States. We will use both predictive models and laboratory/field experiments to evaluate the likelihood and consequences of strike at HK projects in rivers. Efforts undertaken at ORNL address three objectives: (1) Assess strike risk for marine and freshwater organisms; (2) Develop experimental procedures to assess the risk and consequences of strike; and (3) Conduct strike studies in experimental flumes and field installations of hydrokinetic devices.     Mercury (Hg) bioavailability and geochemical cycling is affected by its partitioning between the aqueous and particulate phases. We applied X-ray fluorescence (XRF) microprobes to directly visualize and quantify the spatial localization of Hg and its correlations with other elements of interest on suspended particles from a Hg contaminated freshwater system. Up to 175 Ī¼g gā€“1 Hg is found on suspended particles. Mercury is heterogeneously distributed among phytoplankton (e.g., diatoms) and mineral particles that are rich in iron oxides and natural organic matter (NOM), possibly as Hg-NOM-iron oxide ternary complexes. The diatom-bound Hg is mostly found on outer surfaces of themoreĀ Ā» cells, suggesting passive sorption of inorganic Hg on diatoms.     Radioactive wastes discharged from establishments involved in the use of radioisotopes such as nuclear-powered industries, tracer research and nuclear medicine are a potential public health hazard. Such wastes contain radionuclides, particularly Iodine-131 ({sup 131}I), produced in fission with a yield of about 3%. Radionuclides in waste waters are known to be taken up by molluscs such as mussels, oysters, and clams. This study aims to determine the uptake of {sup 131}I from algal phytoplankton (Chroococcus dispersus) fed to the freshwater clam Corbicula manilensis as well as the organ/tissue distribution. The results will be compared with a previous study on {sup 131}I uptake from water by the same clams.

    A key advantage of using microalgae for biofuel production is the ability of some algal strains to thrive in waters unsuitable for conventional crop irrigation such as saline groundwater or seawater. Nonetheless, the availability of sustainable water supplies will provide significant challenges for scale-up and development of algal biofuels. We conduct a limited techno-economic assessment based on the availability of freshwater, saline groundwater, and seawater for use in open pond algae cultivation systems. We explore water issues through GIS-based models of algae biofuel production, freshwater supply, and cost models for supplying seawater and saline groundwater. We estimate that combined, within the coterminous US these resources can support production on the order of 9.46E+7 m3 yr-1 (25 billion gallons yr-1) of renewable biodiesel. Achievement of larger targets requires the utilization of less water efficient sites and relatively expensive saline waters. Geographically, water availability is most favorable for the coast of the Gulf of Mexico and Florida peninsula, where evaporation relative to precipitation is moderate and various saline waters are economically available. As a whole, barren and scrub lands of the southwestern US have limited freshwater supplies so accurate assessment of alternative waters is critical.

    Hydraulic fracturingā€”the injection of pressurized fluid, often water, to increase recovery of oil or gasā€”has become increasingly popular in combination with horizontal drilling. Hydraulic fracturing improves production from a well, but requires a significant amount of water to do so and could put pressure on existing water resources, especially in water-stressed areas. To supply water needs, some water rights holders sell or lease their water resources to oil and gas producers in an informal water market. These transactions enable the opportunity for cross-sectoral investments, by which the energy sector either directly or indirectly provides the capital for water efficiency improvementsmoreĀ Ā» in the agricultural sector as a mechanism to increase water availability for other purposes, including oil and gas production. In this analysis, we employ an original water and cost model to evaluate the water market in Texas and the potential for cross-sectoral collaboration on water efficiency improvements through a case study of the Lower Rio Grande Valley in Texas. Furthermore, we find that if irrigation efficiency management practices were fully implemented, between 420 and 800 million m3 of water could be spared per year over a ten year period, potentially enabling freshwater use in oil and gas production for up to 26,000 wells, while maintaining agricultural productivity and possibly improving water flows to the ecosystem.Ā«Ā less

  10. Comparison of two freshwater turtle species as monitors of radionuclide and chemical contamination: DNA damage and residue analysis

    Two species of turtles that occupy different ecological niches were compared for their usefulness as monitors of freshwater ecosystems where both low-level radioactive and nonradioactive contaminants are present. The pond slider (Trachemys scripta) and common snapping turtle (Chelydra serpentina) were analyzed for the presence of [sup 90]Sr, [sup 137]Cs, [sup 60]Co, and Hg, radionuclides and chemicals known to be present at the contaminated site, and single-strand breaks in liver DNA. The integrity of the DNA was examined by the alkaline unwinding assay, a technique that detects strand breaks as a biological marker of possible exposure to genotoxic agents. This measure of DNA damage was significantly increased in both species of turtles at the contaminated site compared with turtles of the same species at a reference site, and shows that contaminant-exposed populations were under more severe genotoxic stress than those at the reference site. The level of strand breaks observed at the contaminated site was high and in the range reported for other aquatic species exposed to deleterious concentrations of genotoxic agents such as chemicals and ionizing radiation. Statistically significantly higher concentrations of radionuclides and Hg were detected in the turtles from the contaminated area. Mercury concentrations were significantly higher in the more carnivorous snapping turtle compared with the slider; however, both species were effective monitors of the contaminants.

  11. Phospholipid fatty acid biomarkers in a freshwater periphyton community exposed to uranium: discovery by non-linear statistical learning

    Phospholipid fatty acids (PLFA) have been widely used to characterize environmental microbial communities, generating community profiles that can distinguish phylogenetic or functional groups within the community. The poor specificity of organism groups with fatty acid biomarkers in the classic PLFA-microorganism associations is a confounding factor in many of the statistical classification/clustering approaches traditionally used to interpret PLFA profiles. In this paper we demonstrate that non-linear statistical learning methods, such as a support vector machine (SVM), can more accurately find patterns related to uranyl nitrate exposure in a freshwater periphyton community than linear methods, such as partial least squares discriminant analysis. In addition, probabilistic models of exposure can be derived from the identified lipid biomarkers to demonstrate the potential model-based approach that could be used in remediation. The SVM probability model separates dose groups at accuracies of ~87.0%, ~71.4%, ~87.5%, and 100% for the four groups; Control (non-amended system), low-dose (amended at 10 µg U L-1), medium dose (amended at 100 µg U L-1), and high dose (500 µg U L-1). The SVM model achieved an overall cross-validated classification accuracy of ~87% in contrast to ~59% for the best linear classifier.

    Mercury (Hg) bioavailability and geochemical cycling is affected by its partitioning between the aqueous and particulate phases. We applied a synchrotron-based X-ray fluorescence (XRF) microprobe to visualize and quantify directly the spatial localization of Hg and its correlations with other elements of interest on suspended particles from a Hg-contaminated freshwater system. Up to 175 Ī¼g gāˆ’1 Hg is found on suspended particles, but less than 0.01% is in the form of methylmercury. Mercury is heterogeneously distributed among phytoplankton (e.g., diatoms) and mineral particles that are rich in iron oxides and natural organic matter (NOM). The diatom-bound Hg is mostly foundmoreĀ Ā» on outer surfaces of the cells, suggesting passive sorption of Hg on diatoms. Our results indicate that localized sorption of Hg onto suspended particles, including diatoms and NOM-coated oxide minerals, may play an important role in affecting the partitioning, reactivity, and biogeochemical cycling of Hg in natural aquatic environments.Ā«Ā less

  13. Hydraulic Geometry and Microtopography of Tidal Freshwater Forested Wetlands and Implications for Restoration, Columbia River, U.S.A.

    The hydrologic reconnection of tidal channels, riverine floodplains, and main stem channels are among responses by ecological restoration practitioners to the increasing fragmentation and land conversion occurring in coastal and riparian zones. Design standards and monitoring of such ecological restoration depend upon the characterization of reference sites that vary within and among regions. Few locales, such as the 235 km tidal portion of the Columbia River on the West Coast U.S.A., remain in which the reference conditions and restoration responses of tidal freshwater forested wetlands on temperate zone large river floodplains can be compared. This study developed hydraulic geometry relationships for Picea sitchensis (Sitka spruce) dominated tidal forests (swamps) in the vicinity of Grays Bay on the Columbia River some 37 km from the Pacific Coast using field surveys and Light Detection and Ranging (LiDAR) data. Scaling relationships between catchment area and the parameters of channel cross-sectional area at outlet and total channel length were comparable to tidally influenced systems of San Francisco Bay and the United Kingdom. Dike breaching, culvert replacement, and tide gate replacement all affected channel cross-sectional geometry through changes in the frequency of over-marsh flows. Radiocarbon dating of buried wood provided evidence of changes in sedimentation rates associated with diking, and restoration trajectories may be confounded by historical subsidence behind dikes rendering topographical relationships with water level incomparable to reference conditions. At the same time, buried wood is influencing the development of channel morphology toward characteristics resembling reference conditions. Ecological restoration goals and practices in tidal forested wetland regions of large river floodplains should reflect the interactions of these controlling factors.

    The Hanford Reach of the Columbia River is the last unimpounded section of the river and contains substrate characteristics (cobble, gravel, sand/silt) suitable for many of the native freshwater mussels known to exist in the Pacific Northwest. Information concerning the native mussel species composition, densities, and distributions in the mainstem of the Columbia River is limited. Under funding from the U.S. Department of Energy Richland Operations Office (DOE-RL), Pacific Northwest National Laboratory conducted an assessment of the near-shore habitat on the Hanford Reach. Surveys conducted in 2004 as part of the Ecological Monitoring and Compliance project documented several species of native mussels inhabiting the near-shore habitat of the Hanford Reach. Findings reported here may be useful to resource biologists, ecologists, and DOE-RL to determine possible negative impacts to native mussels from ongoing near-shore remediation activities associated with Hanford Site cleanup. The objective of this study was to provide an initial assessment of the species composition, densities, and distribution of the freshwater mussels (Margaritiferidae and Unionidae families) that exist in the Hanford Reach. Researchers observed and measured 201 live native mussel specimens. Mussel density estimated from these surveys is summarized in this report with respect to near-shore habitat characteristics including substrate size, substrate embeddedness, relative abundance of aquatic vegetation, and large-scale geomorphic/hydrologic characteristics of the Hanford Reach.

    A tornado type wind turbine has a vertically disposed wind collecting tower with spaced apart inner and outer walls and a central bore. The upper end of the tower is open while the lower end of the structure is in communication with a wind intake chamber. An opening in the wind chamber is positioned over a turbine which is in driving communication with an electrical generator. An opening between the inner and outer walls at the lower end of the tower permits radially flowing air to enter the space between the inner and outer walls while a vertically disposed opening in the wind collecting tower permits tangentially flowing air to enter the central bore. A porous portion of the inner wall permits the radially flowing air to interact with the tangentially flowing air so as to create an intensified vortex flow which exits out of the top opening of the tower so as to create a low pressure core and thus draw air through the opening of the wind intake chamber so as to drive the turbine.

    The research conducted as part of this project contributes significantly to the understanding of the microbes and their activities involved in methane metabolism in freshwater lake sediments and in the environment in a more global sense. Significant new insights have been gained into the identity of the species that are most active in methane oxidation. New concepts have been developed based on the new data on how these organisms metabolize methane, impacting not only environmental microbiology but also biotechnology, including biotechnology of next generation biofuels. Novel approaches have been developed for studying functional microbial communities, via holistic approaches, such as metagenomics, metatrancriptomics and metabolite analysis. As a result, a novel outlook has been obtained at how such communities operate in nature. Understanding methane-oxidizing communities in lakes and other environments is of significant benefit to the public, in terms of methane emission mitigation and in terms of potential biotechnological applications.

The LocationType object is included in the diorama-app package in the diorama::types namespace. Abstractly, the object is intended to capture the full time history of the location of an object or reference point. For example, a location may be speci ed as a near-Earth orbit in terms of a two-line element set, in which case the location type is capable of propagating the orbit both forward and backward in time to provide a location for any given time. Alternatively, the location may be speci ed as a xed set of geodetic coordinates (latitude, longitude, and altitude), in which case the geodetic location of the object is expected to remain constant for all time.     This project investigated possible mechanisms by which melt-water pulses can induce abrupt change in the Atlantic Meridional Overturning Circulation (AMOC) magnitude. AMOC magnitude is an important ingredient in present day climate. Previous studies have hypothesized abrupt reduction in AMOC magnitude in response to influxes of glacial melt water into the North Atlantic. Notable fresh-water influxes are associated with the terminus of the last ice age. During this period large volumes of melt water accumulated behind retreating ice sheets and subsequently drained rapidly when the ice weakened sufficiently. Rapid draining of glacial lakes into the North Atlantic is a possible origin of a number of paleo-record abrupt climate shifts. These include the Younger-Dryas cooling event and the 8,200 year cooling event. The studies undertaken focused on whether the mechanistic sequence by which glacial melt-water impacts AMOC, which then impacts Northern Hemisphere global mean surface temperature, is dynamically plausible. The work has implications for better understanding past climate stability. The work also has relevance for todayā€™s environment, in which high-latitude ice melting in Greenland appears to be driving fresh water outflows at an accelerating pace.

    We compare the host galaxies of 902 supernovae (SNe), including SNe Ia, SNe II, and SNe Ibc, which are selected by cross-matching the Asiago Supernova Catalog with the Sloan Digital Sky Survey (SDSS) Data Release 7. We selected an additional 213 galaxies by requiring the light fraction of spectral observations to be >15%, which could represent well the global properties of the galaxies. Among these 213 galaxies, 135 appear on the Baldwin-Phillips-Terlevich diagram, which allows us to compare the hosts in terms of whether they are star-forming (SF) galaxies, active galactic nuclei (AGNs; including composites, LINERs, and Seyfert 2s) or absorption-line galaxies (Absorps; i.e., their related emission lines are weak or non-existent). The diagrams related to the parameters D{sub n}(4000), H?{sub A}, stellar masses, star formation rates (SFRs), and specific SFRs for the SNe hosts show that almost all SNe II and most of the SNe Ibc occur in SF galaxies, which have a wide range of stellar masses and low D{sub n}(4000). The SNe Ia hosts as SF galaxies following similar trends. A significant fraction of SNe Ia occurs in AGNs and absorption-line galaxies, which are massive and have high D{sub n}(4000). The stellar population analysis from spectral synthesis fitting shows that the hosts of SNe II have a younger stellar population than hosts of SNe Ia. These results are compared with those of the 689 comparison galaxies where the SDSS fiber captures less than 15% of the total light. These comparison galaxies appear biased toward higher 12+log(O/H) (?0.1 dex) at a given stellar mass. Therefore, we believe the aperture effect should be kept in mind when the properties of the hosts for different types of SNe are discussed.

    We present a new photometric identification technique for SN 1991bg-like type Ia supernovae (SNe Ia), i.e., objects with light curve characteristics such as later primary maxima and the absence of a secondary peak in redder filters. This method is capable of selecting this sub-group from the normal type Ia population. Furthermore, we find that recently identified peculiar sub-types such as SNe Iax and super-Chandrasekhar SNe Ia have photometric characteristics similar to 91bg-like SNe Ia, namely, the absence of secondary maxima and shoulders at longer wavelengths, and can also be classified with our technique. The similarity of these different SN Ia sub-groups perhaps suggests common physical conditions. This typing methodology permits the photometric identification of peculiar SNe Ia in large upcoming wide-field surveys either to study them further or to obtain a pure sample of normal SNe Ia for cosmological studies.

    Osaka Gas Co. has been developing a planar type SOFC (OG type SOFC) which has a suitable structure for stacking. Murata Mfg. Co. has begun to develop the OG type SOFC stack through joint program since 1993. Figure 1 shows OG type cell structure. Because each cell is sustained by cell holders acting air manifold, the load of upper cell is not put on the lower cells. Single cell is composed of 3-layered membrane and LaCrO{sub 3} separator. 5 single cells are mounted on the cell holder, connected with Ni felt electrically, and bonded by glassy material sealant. We call the 5-cell stack a unit. Stacking 13 units, we succeeded 870 W generation in 1993. But the power density was low, 0.11 Wcm{sup -2} because of crack in the electrolyte and gas leakage at some cells.

    For the past 6 years, the US Department of Energy (DOE) Office of Facility Safety Analysis (EH-32) has contracted Westinghouse Hanford Company (WHC) to conduct compliance testing on DOE Type A packagings. The packagings are tested for compliance with the U.S. Department of Transportation (DOT) Specification 7A, general packaging, Type A requirements. The DOE has shared the Type A packaging information throughout the nuclear materials transportation community. During testing, there have been recurring areas of packaging design that resulted in testing delays and/or initial failure. The lessons learned during the testing are considered a valuable resource. DOE requested that WHC share this resource. By sharing what is and can be encountered during packaging testing, individuals will hopefully avoid past mistakes.

    To use type Ia supernovae as standard candles for cosmologywe need accurate broadband magnitudes. In practice the observed magnitudemay differ from the ideal magnitude-redshift relationship either throughintrinsic inhomogeneities in the type Ia supernova population or throughobservational error. Here we investigate how we can choose filterbandpasses to reduce the error caused by both these effects. We find thatbandpasses with large integral fluxes and sloping wings are best able tominimise several sources of observational error, and are also leastsensitive to intrinsic differences in type Ia supernovae. The mostimportant feature of a complete filter set for type Ia supernovacosmology is that each bandpass be a redshifted copy of the first. Wedesign practical sets of redshifted bandpasses that are matched totypical high resistivity CCD and HgCdTe infra-red detector sensitivities.These are designed to minimise systematic error in well observedsupernovae, final designs for specific missions should also considersignal-to-noise requirements and observing strategy. In addition wecalculate how accurately filters need to be calibrated in order toachieve the required photometric accuracy of future supernova cosmologyexperiments such as the SuperNova-Acceleration-Probe (SNAP), which is onepossible realisation of the Joint Dark-Energy mission (JDEM). We considerthe effect of possible periodic miscalibrations that may arise from theconstruction of an interference filter.

    A compact fast analyzer of the rotary cuvette type is provided for simultaneously determining concentrations in a multiplicity of discrete samples using either absorbance or fluorescence measurement techniques. A rigid, generally rectangular frame defines optical passageways for the absorbance and fluorescence measurement systems. The frame also serves as a mounting structure for various optical components as well as for the cuvette rotor mount and drive system. A single light source and photodetector are used in making both absorbance and fluorescence measurements. Rotor removal and insertion are facilitated by a swing-out drive motor and rotor mount. BACKGROUND OF THE INVENTION The invention relates generally to concentration measuring instruments and more specifically to a compact fast analyzer of the rotary cuvette type which is suitable for making either absorbance or fluorescence measurements. It was made in the course of, or under, a contract with the U.S. Atomic Energy Commission.

    The nature of eolian-dune reservoirs is strongly influenced by stratification types (in decreasing order of quality: grain-flow, grain-fall, wind-ripple deposits) and their packaging by internal bounding surfaces. These are, in turn, a function of dune surface processes and migration behavior, allowing for predictive models of reservoir behavior. Migrating, simple crescentic dunes produce tabular bodies consisting mainly of grain-flow cross-strata, and form the best, most predictable reservoirs. Reservoir character improves as both original dune height and preserved set thickness increase, because fewer grain-fall deposits and a lower percentage of dune-apron deposits occur in the cross-strata, respectively. It is probable that many linear and star dunes migrate laterally, leaving a blanket of packages of wind ripple laminae reflecting deposition of broad, shifting aprons. This is distinct from models generated by freezing large portions of these dunes in place. Trailing margins of linear and star dunes are prone to reworking by sand-sheet processes that decrease potential reservoir quality. The occurrence of parabolic dunes isolated on vegetated sand sheets results in a core of grain-flow and grain-fall deposits surrounded by less permeable and porous deposits. Compound crescentic dunes, perhaps the most preservable dune type, may yield laterally (1) single sets of cross-strate, (2) compound sets derived from superimposed simple dunes, or (3) a complex of diverse sets derived from superimposed transverse and linear elements.

    Previous analyses of a microbial metagenome from uranium and nitric-acid contaminated groundwater (FW106) showed significant environmental effects resulting from the rapid introduction of multiple contaminants. Effects include a massive loss of species and strain biodiversity, accumulation of toxin resistant genes in the metagenome and lateral transfer of toxin resistance genes between community members. To better understand these results in an ecological context, a second metagenome from a pristine groundwater system located along the same geological strike was sequenced and analyzed (FW301). It is hypothesized that FW301 approximates the ancestral FW106 community based on phylogenetic profiles and common geological parameters; however, even if is not the case, the datasets still permit comparisons between healthy and stressed groundwater ecosystems. Complex carbohydrate metabolism has been almost entirely lost in the stressed ecosystem. In contrast, the pristine system encodes a wide diversity of complex carbohydrate metabolism systems, suggesting that carbon turnover is very rapid and less leaky in the healthy groundwater system. FW301 encodes many (~;;160+) carbon monoxide dehydrogenase genes while FW106 encodes none. This result suggests that the community is frequently exposed to oxygen from aerated rainwater percolating into the subsurface, with a resulting high rate of carbon metabolism and CO production. When oxygen levels fall, the CO then serves as a major carbon source for the community. FW301 appears to be capable of CO2 fixation via the reductive carboxylase (reverse TCA) cycle and possibly acetogenesis, activities; these activities are lacking in the heterotrophic FW106 system which relies exclusively on respiration of nitrate and/or oxygen for energy production. FW301 encodes a complete set of B12 biosynthesis pathway at high abundance suggesting the use of sodium gradients for energy production in the healthy groundwater community. Overall comparative analysis suggests that the introduction of contaminants is accompanied by a decrease in biodiversity, loss of nutrient cycling, less metabolic diversity, increased respiration and dominance by r-strategists. These results are consistent with trends predicted for stressed ecosystems.

    The determination of the physical speciation of aluminum in water samples by filtration through polycarbonate membranes proved feasible; control experiments revealed neither contamination nor analyte loss. Treatment of sample filtrates with a fractionally loaded Chelex 100 ion-exchange resin (>75% H/sup +/-form) allows one to distinguish between different forms of aluminum on the basis of their kinetic and thermodynamic properties. Monomeric hydroxo- and fluoroaluminum complexes exchanged readily (>85% after 30 min), as did low molecular weight polynuclear species. Under similar conditions, forms of Al associated with fulvic and humic acids of natural origin exchanged much more slowly (<5% after 30 min, at an Al:dissolved organic carbon atomic ratio of approx. 1:155). Before photooxidation, the filterable aluminum present in natural waters exhibited intermediate behavior (0-50% exchange after 30 min); after UV irradiation the nonexchangeable Al fraction had practically disappeared (90-96% exchange after 30 min), suggesting that the major portion of the nonexchangeable Al initially present was associated with organic matter.

    We present the first large-scale effort of creating composite spectra of high-redshift type Ia supernovae (SNe Ia) and comparing them to low-redshift counterparts. Through the ESSENCE project, we have obtained 107 spectra of 88 high-redshift SNe Ia with excellent light-curve information. In addition, we have obtained 397 spectra of low-redshift SNe through a multiple-decade effort at Lick and Keck Observatories, and we have used 45 ultraviolet spectra obtained by HST/IUE. The low-redshift spectra act as a control sample when comparing to the ESSENCE spectra. In all instances, the ESSENCE and Lick composite spectra appear very similar. The addition of galaxy light to the Lick composite spectra allows a nearly perfect match of the overall spectral-energy distribution with the ESSENCE composite spectra, indicating that the high-redshift SNe are more contaminated with host-galaxy light than their low-redshift counterparts. This is caused by observing objects at all redshifts with similar slit widths, which corresponds to different projected distances. After correcting for the galaxy-light contamination, subtle differences in the spectra remain. We have estimated the systematic errors when using current spectral templates for K-corrections to be {approx}0.02 mag. The variance in the composite spectra give an estimate of the intrinsic variance in low-redshift maximum-light SN spectra of {approx}3% in the optical and growing toward the ultraviolet. The difference between the maximum-light low and high-redshift spectra constrain SN evolution between our samples to be < 10% in the rest-frame optical.

    Dermal fibroblasts from most individuals with osteogenesis imperfecta (OI) type I produce about half the normal amount of type I procollagen, as a result of decreased synthesis of one of its constituent chains, pro[alpha](I). To test the hypothesis that decreased synthesis of pro[alpha](I) chains results from mutations in the COL1A1 gene, the authors used primer extension with nucleotide-specific chain termination to measure the contribution of individual COL1A1 alleles to the mRNA pool in fibroblasts from affected individuals. A polymorphic Mn/I restriction endonuclease site in the 3'-untranslated region of COL1A1 was used to distinguish the transcripts of the two alleles in heterozygous individuals. Twenty-three individuals from 21 unrelated families were studied. In each case there was marked diminution in steady-state mRNA levels from one COL1A2 allele. Loss of an allele through deletion or rearrangement was not the cause of the diminished COL1A1 mRNA levels. Primer extension with nucleotide-specific chain termination allows identification of the mutant COL1A1 allele in cell strains that are heterozygous for an expressed polymorphism. It is applicable to sporadic cases, to small families, and to large families in whom key individuals are uninformative at the polymorphic sites used in linkage analysis, making it a useful adjunct to the biochemical screening of collagenous proteins for OI. 40 refs., 3 figs., 1 tab.

    A new n-type semiconducting diamond is disclosed, which is doped with n-type dopant atoms. Such diamond is advantageously formed by chemical vapor deposition from a source gas mixture comprising a carbon source compound for the diamond, and a volatile hot wire filament for the n-type impurity species, so that the n-type impurity atoms are doped in the diamond during its formation. A corresponding chemical vapor deposition method of forming the n-type semiconducting diamond is disclosed. The n-type semiconducting diamond of the invention may be usefully employed in the formation of diamond-based transistor devices comprising pn diamond junctions, and in other microelectronic device applications.

    Production of nuclei heavier than the target is treated for projectile-target combinations used in cold fusion reactions leading to superheavy nuclei. These products are related to transfer-type or to asymmetry-exit-channel quasifission reactions. The production of isotopes in the transfer-type reactions emitting of {alpha} particles with large energies is discussed.

    Silver doped ZnO films have been grown on sapphire (0001) substrates by pulsed laser deposition. Hall measurements indicate that p-type conductivity is realized for the films deposited at 500ā€‰Ā°C and 750ā€‰Ā°C. Transmission electron microscopy images show more obvious and higher density of stacking faults (SFs) present in the p-type ZnO films as compared to the n-type films. Top view and cross sectional photoluminescence of the n- and p-type samples revealed free excitonic emission from both films. A peak at 3.314ā€‰eV, attributed to SF emission, has been observed only for the n-type sample, while a weak neutral acceptor peak observed at 3.359ā€‰eV in the p-type film. The SF emission in the n-type sample suggests localization of acceptor impurities nearby the SFs, while lack of SF emission for the p-type sample indicates the activation of the Ag acceptors in ZnO.

  16. Bucket-type steam traps removed in $82K retrofit

    A retrofit of 481 mostly failed steam traps at Martin Marietta's Aerospace Division should reduce steam costs by $70,000 and require little or no maintenance. Payback should occur within 14 months. The new traps include orifice, bellow-type thermostatic, and float-type traps that have few or no moving parts. Lack of maintenance was responsible for the poor performance of the bucket traps that were replaced, although manufacturers of the bucket traps disagree that replacement of certain parts is necessary every six months. The author describes the design and operation of each type of trap.

    Solar type III radio bursts and their production mechanisms have been intensively studied in both theory and observation and are believed to be the most important signatures of electron acceleration in active regions. Recently, Wu et al. proposed that the electron-cyclotron maser emission (ECME) driven by an energetic electron beam could be responsible for producing type III bursts and pointed out that turbulent Alfvén waves can greatly influence the basic process of ECME via the oscillation of these electrons in the wave fields. This paper investigates effects of homochromous Alfvén waves (HAWs) on ECME driven by electron beams. Our results show that the growth rate of the O-mode wave will be significantly modulated by HAWs. We also discuss possible application to the formation of fine structures in type III bursts, such as so-called solar type IIIb radio bursts.
