The analysis shows that a flexible unit strategy that provides a suite of design solutions tailored to the characteristics of the immobilized low-activity waste will provide a disposal system that best meets the program goals of reducing the environmental, health, and safety impacts; meeting the schedule milestones; and minimizing the life-cycle cost of the program.

Higher activity was observed in the MgTi{sub 4}(PO{sub 4}){sub 6}- and CaTi{sub 4}(PO{sub 4}){sub 6}-containing glass ceramics. -- Abstract: Glass ceramics were prepared by heat-treating MOTiO{sub 2}P{sub 2}O{sub 5} (M = Mg, Ca, Sr and Ba) and R{sub 2}OTiO{sub 2}P{sub 2}O{sub 5}SiO{sub 2} (R = Li, Na and K) glasses, and their photocatalytic activity was investigated. The crystalline phases precipitated in the glasses were only Nasicon-type crystals, MTi{sub 4}(PO{sub 4}){sub 6} or RTi{sub 2}(PO{sub 4}){sub 3}. Decomposition experiments of both methylene blue (MB) and acetaldehyde showed that the glass ceramics exhibited effective photocatalytic activity. The activity did not depend on the radius of the M{sup 2+} or R{sup +} ion, and higher activity was observed in the MgTi{sub 4}(PO{sub 4}){sub 6} and CaTi{sub 4}(PO{sub 4}){sub 6} precipitated glass ceramics.

    Both hospital- and community-acquired Staphylococcus aureus infections have become major health concerns in terms of morbidity, suffering and cost. Trimethoprim-sulfamethoxazole (TMP-SMZ) is an alternative treatment for methicillin-resistant S. aureus (MRSA) infections. However, TMP-resistant strains have arisen with point mutations in dihydrofolate reductase (DHFR), the target for TMP. A single point mutation, F98Y, has been shown biochemically to confer the majority of this resistance to TMP. Using a structure-based approach, we have designed a series of novel propargyl-linked DHFR inhibitors that are active against several trimethoprim-resistant enzymes. We screened this series against wild-type and mutant (F98Y) S. aureus DHFR and found that several are active against both enzymes and specifically that the meta-biphenyl class of these inhibitors is the most potent. In order to understand the structural basis of this potency, we determined eight high-resolution crystal structures: four each of the wild-type and mutant DHFR enzymes bound to various propargyl-linked DHFR inhibitors. In addition to explaining the structure-activity relationships, several of the structures reveal a novel conformation for the cofactor, NADPH. In this new conformation that is predominantly associated with the mutant enzyme, the nicotinamide ring is displaced from its conserved location and three water molecules complete a network of hydrogen bonds between the nicotinamide ring and the protein. In this new position, NADPH has reduced interactions with the inhibitor. An equilibrium between the two conformations of NADPH, implied by their occupancies in the eight crystal structures, is influenced both by the ligand and the F98Y mutation. The mutation induced equilibrium between two NADPH-binding conformations may contribute to decrease TMP binding and thus may be responsible for TMP resistance.

    The population of LPS molecules on Salmonella montevideo that bind C3 during alternative pathway activation in serum was studied. LPS molecules of Salmonella are composed of lipid A:core oligosaccharide (one copy per molecule), substituted by an O-polysaccharide (O-PS) side chain, which is a linear polymer of 0 to >60 O-antigen repeat units containing mannose. A mutant of S. montevideo called SL5222 that inserts galactose only into core oligosaccharide and mannose only into O-antigen sub-units was grown with (/sup 3/H)mannose and (/sup 14/C)galactose, so that LPS molecules bearing large numbers of O-antigen subunits have high /sup 3/H to /sup 14/C ratios, whereas molecules with few O-antigen subunits have lower /sup 3/H to /sup 14/C ratios. The authors postulate that preferential attachment of C3 to long-chain LPS in SL5222 results because long-chain LPS molecules sterically hinder shorter chain LPS molecules from macromolecules. This study provides direct proof that the O-PS of LPS sterically hinders access of large molecules to the outer membrane and indicates that the LPS coat of these bacteria functions as a barrier against large protein molecules.

  12. Classical and alternative macrophage activation in the lung following ozone-induced oxidative stress

    Ozone is a pulmonary irritant known to cause oxidative stress, inflammation and tissue injury. Evidence suggests that macrophages play a role in the pathogenic response; however, their contribution depends on the mediators they encounter in the lung which dictate their function. In these studies we analyzed the effects of ozone-induced oxidative stress on the phenotype of alveolar macrophages (AM). Exposure of rats to ozone (2 ppm, 3 h) resulted in increased expression of 8-hydroxy-2?-deoxyguanosine (8-OHdG), as well as heme oxygenase-1 (HO-1) in AM. Whereas 8-OHdG was maximum at 24 h, expression of HO-1 was biphasic increasing after 3 h and 4872 h. Cleaved caspase-9 and beclin-1, markers of apoptosis and autophagy, were also induced in AM 24 h post-ozone. This was associated with increased bronchoalveolar lavage protein and cells, as well as matrix metalloproteinase (MMP)-2 and MMP-9, demonstrating alveolar epithelial injury. Ozone intoxication resulted in biphasic activation of the transcription factor, NF?B. This correlated with expression of monocyte chemotactic protein?1, inducible nitric oxide synthase and cyclooxygenase?2, markers of proinflammatory macrophages. Increases in arginase-1, Ym1 and galectin-3 positive anti-inflammatory/wound repair macrophages were also observed in the lung after ozone inhalation, beginning at 24 h (arginase-1, Ym1), and persisting for 72 h (galectin-3). This was associated with increased expression of pro-surfactant protein-C, a marker of Type II cell proliferation and activation, important steps in wound repair. These data suggest that both proinflammatory/cytotoxic and anti-inflammatory/wound repair macrophages are activated early in the response to ozone-induced oxidative stress and tissue injury. -- Highlights: ?     We have identified a G to A substitution in the 5{prime} donor splice site of intron 18 of one COL1A1 allele in two unrelated families with osteogenesis imperfecta (OI) type I. A third OI type I family has a G to A substitution at the identical position in intron 48 of one COL1A1 allele. Both mutations abolish normal splicing and lead to reduced steady-state levels of mRNA from the mutant COL1A1 allele. The intron 18 mutation leads to both exon 18 skipping in the mRNA and to utilization of a single alternative splice site near the 3{prime} end of exon 18. The latter results in deletion of the last 8 nucleotides of exon 18 from the mRNA, a shift in the translational reading-frame, and the creation of a premature termination codon in exon 19. Of the potential alternative 5{prime} splice sites in exon 18 and intron 18, the one utilized has a surrounding nucleotide sequence which most closely resembles that of the natural splice site. Although a G to A mutation was detected at the identical position in intron 48 of one COL1A1 allele in another OI type I family, nine complex alternative splicing patterns were identified by sequence analysis of cDNA clones derived from fibroblast mRNA from this cell strain. All result in partial or complete skipping of exon 48, with in-frame deletions of portions of exons 47 and/or 49. The different patterns of RNA splicing were not explained by their sequence homology with naturally occuring 5{prime} splice sites, but rather by recombination between highly homologous exon sequences, suggesting that we may not have identified the major splicing alternative(s) in this cell strain. Both G to A mutations result in decreased production of type I collagen, the common biochemical correlate of OI type I.

    We present black hole masses and accretion rates for 182 Type 1 active galactic nuclei (AGNs) in COSMOS. We estimate masses using the scaling relations for the broad H {beta}, Mg II, and C IV emission lines in the redshift ranges 0.16 < z < 0.88, 1 < z < 2.4, and 2.7 < z < 4.9. We estimate the accretion rate using an Eddington ratio L{sub I}/L{sub Edd} estimated from optical and X-ray data. We find that very few Type 1 AGNs accrete below L{sub I} /L{sub Edd} {approx} 0.01, despite simulations of synthetic spectra which show that the survey is sensitive to such Type 1 AGNs. At lower accretion rates the broad-line region may become obscured, diluted, or nonexistent. We find evidence that Type 1 AGNs at higher accretion rates have higher optical luminosities, as more of their emission comes from the cool (optical) accretion disk with respect to shorter wavelengths. We measure a larger range in accretion rate than previous works, suggesting that COSMOS is more efficient at finding low accretion rate Type 1 AGNs. However, the measured range in accretion rate is still comparable to the intrinsic scatter from the scaling relations, suggesting that Type 1 AGNs accrete at a narrow range of Eddington ratio, with L{sub I} /L{sub Edd} {approx} 0.1.

Within the limitations of the model, the study presents the first attempt to produce synthetic (simulated) dynamical spectrum of the type III radio bursts in the fully kinetic plasma model. The latter is based on 1.5D non-zero pitch angle (non-gyrotropic) electron beam that is an alternative to the plasma emission classical mechanism for which two spatial dimensions are needed.

    We report that Mycobacterium tuberculosis and Staphylococcus aureus secrete virulence factors via type VII protein secretion (T7S), a system that intriguingly requires all of its secretion substrates for activity. To gain insights into T7S function, we used structural approaches to guide studies of the putative translocase EccC, a unique enzyme with three ATPase domains, and its secretion substrate EsxB. The crystal structure of EccC revealed that the ATPase domains are joined by linker/pocket interactions that modulate its enzymatic activity. EsxB binds via its signal sequence to an empty pocket on the C-terminal ATPase domain, which is accompanied by an increasemore » in ATPase activity.     Macrophages adapt their response to micro-environmental signals. While Th1 cytokines promote pro-inflammatory M1 macrophages, Th2 cytokines promote an 'alternative' anti-inflammatory M2 macrophage phenotype. Peroxisome proliferator-activated receptors (PPARs) are ligand-activated transcription factors expressed in macrophages where they control the inflammatory response. It has been shown that PPAR{gamma} promotes the differentiation of monocytes into anti-inflammatory M2 macrophages in humans and mice, while a role for PPAR{beta}/{delta} in this process has been reported only in mice and no data are available for PPAR{alpha}. Here, we show that in contrast to PPAR{gamma}, expression of PPAR{alpha} and PPAR{beta}/{delta} overall does not correlate with the expression of M2 markers in human atherosclerotic lesions, whereas a positive correlation with genes of lipid metabolism exists. Moreover, unlike PPAR{gamma}, PPAR{alpha} or PPAR{beta}/{delta} activation does not influence human monocyte differentiation into M2 macrophages in vitro. Thus, PPAR{alpha} and PPAR{beta}/{delta} do not appear to modulate the alternative differentiation of human macrophages.

  5. Regulation of the activity of platelet-bound C3 convertase of the alternative pathway of complement by platelet factor H

    The alternative pathway of complement is regulated on the surface of homologous blood cells at the C3 amplification step by the membrane protein decay-accelerating factor, as well as by the plasma protein factor H. The authors have reported elsewhere that platelets from patients with paroxysmal nocturnal hemoglobinuria regulate the activity of the C3 convertase C3bBb, even though they lack decay-accelerating factor. They now report that normal human platelets contain factor H, which was released from the platelet in response to complement deposition or thrombin stimulation. Factor H was localized to the platelet ..cap alpha.. granules by immunocytochemical techniques. As determined by a solid-phase radioimmunoassay, thrombin-stimulated platelets released approx. 54 ng of factor H per 10/sup 8/ platelets. The release of factor H in response to complement or thrombin was inhibited by treating the platelets with metabolic inhibitors. Such inhibition resulted in a 3-fold increase in the activity of C3bBb. Platelets that released factor H bound only half as many molecules of radiolabeled factor B to platelet-bound C3b than platelets that could not release factor H. Treatment of platelets with anti-decay-accelerating factor antibody had no effect on the activity of C3bBb unless the release of factor H was blocked. Therefore, human platelets have a unique mechanism for the regulation of the alternative pathway of complement.

    We report a sizable class of type 1 active galactic nuclei (AGNs) with unusually weak near-infrared (1-3 {mu}m) emission in the XMM-COSMOS type 1 AGN sample. The fraction of these 'hot-dust-poor' AGNs increases with redshift from 6% at low redshift (z < 2) to 20% at moderate high redshift (2 < z < 3.5). There is no clear trend of the fraction with other parameters: bolometric luminosity, Eddington ratio, black hole mass, and X-ray luminosity. The 3 {mu}m emission relative to the 1 {mu}m emission is a factor of 2-4 smaller than the typical Elvis et al. AGN spectral energy distribution (SED), which indicates a 'torus' covering factor of 2%-29%, a factor of 3-40 smaller than required by unified models. The weak hot dust emission seems to expose an extension of the accretion disk continuum in some of the source SEDs. We estimate the outer edge of their accretion disks to lie at (0.3-2.0) x 10{sup 4} Schwarzschild radii, {approx}10-23 times the gravitational stability radii. Formation scenarios for these sources are discussed.

    Demonstrating that a waste form produced by a given immobilization process is chemically and physically durable as well as compliant with disposal facility acceptance criteria is critical to the success of a waste treatment program, and must be pursued in conjunction with the maturation of the waste processing technology. Testing of waste forms produced using differing scales of processing units and classes of feeds (simulants versus actual waste) is the crux of the waste form qualification process. Testing is typically focused on leachability of constituents of concern (COCs), as well as chemical and physical durability of the waste form. A principal challenge regarding testing immobilized low-activity waste (ILAW) forms is the absence of a standard test suite or set of mandatory parameters against which waste forms may be tested, compared, and qualified for acceptance in existing and proposed nuclear waste disposal sites at Hanford and across the Department of Energy (DOE) complex. A coherent and widely applicable compliance strategy to support characterization and disposal of new waste forms is essential to enhance and accelerate the remediation of DOE tank waste. This paper provides a background summary of important entities, regulations, and considerations for nuclear waste form qualification and disposal. Against this backdrop, this paper describes a strategy for meeting and demonstrating compliance with disposal requirements emphasizing the River Protection Project (RPP) Integrated Disposal Facility (IDF) at the Hanford Site and the fluidized bed steam reforming (FBSR) mineralized low-activity waste (LAW) product stream.

  8. Cost-effectiveness of controlling emissions for various alternative-fuel vehicle types, with vehicle and fuel price subsidies estimated on the basis of monetary values of emission reductions

    Emission-control cost-effectiveness is estimated for ten alternative-fuel vehicle (AFV) types (i.e., vehicles fueled with reformulated gasoline, M85 flexible-fuel vehicles [FFVs], M100 FFVs, dedicated M85 vehicles, dedicated M100 vehicles, E85 FFVS, dual-fuel liquefied petroleum gas vehicles, dual-fuel compressed natural gas vehicles [CNGVs], dedicated CNGVs, and electric vehicles [EVs]). Given the assumptions made, CNGVs are found to be most cost-effective in controlling emissions and E85 FFVs to be least cost-effective, with the other vehicle types falling between these two. AFV cost-effectiveness is further calculated for various cases representing changes in costs of vehicles and fuels, AFV emission reductions, and baseline gasoline vehicle emissions, among other factors. Changes in these parameters can change cost-effectiveness dramatically. However, the rank of the ten AFV types according to their cost-effectiveness remains essentially unchanged. Based on assumed dollars-per-ton emission values and estimated AFV emission reductions, the per-vehicle monetary value of emission reductions is calculated for each AFV type. Calculated emission reduction values ranged from as little as $500 to as much as $40,000 per vehicle, depending on AFV type, dollar-per-ton emission values, and baseline gasoline vehicle emissions. Among the ten vehicle types, vehicles fueled with reformulated gasoline have the lowest per-vehicle value, while EVs have the highest per-vehicle value, reflecting the magnitude of emission reductions by these vehicle types. To translate the calculated per-vehicle emission reduction values to individual AFV users, AFV fuel or vehicle price subsidies are designed to be equal to AFV emission reduction values. The subsidies designed in this way are substantial. In fact, providing the subsidies to AFVs would change most AFV types from net cost increases to net cost decreases, relative to conventional gasoline vehicles.

    Mammalian cysteine dioxygenase type 1 (CDO1) is an essential enzyme for taurine biosynthesis and the biodegradation of toxic cysteine. As previously suggested, Cdo1 may be a marker of liposarcoma progression and adipogenic differentiation, but the role of Cdo1 in adipogenesis has yet been reported. In this study, we found that the expression of Cdo1 is dramatically elevated during adipogenic differentiation of 3T3-L1 pre-adipocytes and mouse bone marrow-derived mesenchymal stem cells (mBMSCs). Conversely, knockdown of Cdo1 inhibited expression of adipogenic specific genes and lipid droplet formation in 3T3-L1 cells and mBMSCs. Mechanistically, we found Cdo1 interacted with Pparγ in response to adipogenic stimulus. Further, depletion of Cdo1 reduced the recruitment of Pparγ to the promoters of C/EBPα and Fabp4. Collectively, our finding indicates that Cdo1 may be a co-activator of Pparγ in adipogenesis, and may contribute to the development of disease associated with excessive adipose tissue. - Highlights: • Cdo1expression is highly up-regulated during adipogenic differentiation of 3T3-L1 and mBMSCs. • Depletion of Cdo1 inhibited expression of adipogenic specific genes and lipid droplet formation. • Cdo1interacts with Pparγ during adipogenesis. • Knockdown of Cdo1 inhibited Pparγ binding to the promoters of C/EBPα and Fabp4.

    Alternative splicing in Mo-MuLV recruits a splice donor site, SD', within the gag that is required for optimal replication in vitro. Remarkably, this SD' site was also found to be utilized for production of oncogenic gag-myb fusion RNA in 100% of murine-induced myeloid leukemia (MML) in pristane-treated BALB/c mice. Therefore, we investigated the influence of silent mutations of SD' in this model. Although there was no decrease in the overall incidence of disease, there was a decrease in the incidence of myeloid leukemia with a concomitant increase in lymphoid leukemia. Importantly, there was a complete lack of myeloid tumors associated with 5' insertional mutagenic activation of c-myb, suggesting the specific requirement of the SD' site in this mechanism.

  11. THE INTEGRAL HIGH-ENERGY CUT-OFF DISTRIBUTION OF TYPE 1 ACTIVE GALACTIC NUCLEI

    In this Letter we present the primary continuum parameters, the photon index Γ, and the high-energy cut-off E {sub c} of 41 type-1 Seyfert galaxies extracted from the International Gamma-Ray Astrophysics Laboratory (INTEGRAL) complete sample of active galactic nuclei (AGNs). We performed broadband (0.3-100 keV) spectral analysis by simultaneously fitting the soft and hard X-ray spectra obtained by XMM and INTEGRAL/IBIS-Swift/BAT, respectively, in order to investigate the general properties of these parameters, in particular their distribution and mean values. We find a mean photon index of 1.73 with a standard deviation of 0.17 and a mean high-energy cut-off of 128 keV with a standard deviation of 46 keV for the whole sample. This is the first time that the cut-off energy is constrained in such a large number of AGNs. We have 26 measurements of the cut-off, which corresponds to 63% of the entire sample, distributed between 50 and 200 keV. There are a further 11 lower limits mostly below 300 keV. Using the main parameters of the primary continuum, we have been able to obtain the actual physical parameters of the Comptonizing region, i.e., the plasma temperature kT {sub e} from 20 to 100 keV and the optical depth τ < 4. Finally, with the high signal-to-noise ratio spectra starting to come from NuSTAR it will soon be possible to better constrain the cut-off values in many AGNs, allowing the determination of more physical models and thus better understand the continuum emission and geometry of the region surrounding black holes.

  12. Hydrogen-Bond Networks: Strengths of Different Types of Hydrogen Bonds and An Alternative to the Low Barrier Hydrogen-Bond Proposal

    We report quantifying the strengths of different types of hydrogen bonds in hydrogen bond networks (HBNs) via measurement of the adiabatic electron detachment energy of the conjugate base of a small covalent polyol model compound (i.e., (HOCH2CH2CH(OH)CH2)2CHOH) in the gas phase and the pKa of the corresponding acid in DMSO. The latter result reveals that the hydrogen bonds to the charged center and those that are one solvation shell further away (i.e., primary and secondary) provide 5.3 and 2.5 pKa units of stabilization per hydrogen bond in DMSO. Computations indicate that these energies increase to 8.4 and 3.9 pKa units in benzene and that the total stabilizations are 16 (DMSO) and 25 (benzene) pKa units. Calculations on a larger linear heptaol (i.e., (HOCH2CH2CH(OH)CH2CH(OH)CH2)2CHOH) reveal that the terminal hydroxyl groups each contribute 0.6 pKa units of stabilization in DMSO and 1.1 pKa units in benzene. All of these results taken together indicate that the presence of a charged center can provide a powerful energetic driving force for enzyme catalysis and conformational changes such as in protein folding due to multiple hydrogen bonds in a HBN.

    Background: The mitogen-activated protein kinases (MAPKs) pathway is critical for cellular signaling, and proteins such as phosphatases that regulate this pathway are important for normal tissue development. Based on our previous work on dual specificity phosphatase-5 (DUSP5), and its role in embryonic vascular development and disease, we hypothesized that mutations in DUSP5 will affect its function. Results: In this study, we tested this hypothesis by generating full-length glutathione-S-transferase-tagged DUSP5 and serine 147 proline mutant (S147P) proteins from bacteria. Light scattering analysis, circular dichroism, enzymatic assays and molecular modeling approaches have been performed to extensively characterize the protein form and function.more » We demonstrate that both proteins are active and, interestingly, the S147P protein is hypoactive as compared to the DUSP5 WT protein in two distinct biochemical substrate assays. Furthermore, due to the novel positioning of the S147P mutation, we utilize computational modeling to reconstruct full-length DUSP5 and S147P to predict a possible mechanism for the reduced activity of S147P.     During vertical testing of SNS superconducting RF cavities, a lot of electron activity was observed. This could be evidence of in-cell multipacting, cell to cell electron mitigation or near-axis dark current. To study the potential for these electron activities in SRF cavities, a multi-purpose electron tracking code (FishPact) was developed based on the Poisson/Superfish Field solvers. Electron trajectories between the impacts in both medium-{beta} and high-{beta} cavities are tracked under varying accelerating gradient (E{sub acc}), RF phases, and emission locations. It is found that the high beta cavities show a greater propensity for potential sources of dark current than their medium beta counterparts. It is further proposed that field emission induced multipacting is a potential, though rare, source of further Q{sub 0} degradation.

Suggesting CB2R as a target to treat diseases with lipid dysregulation. -- Abstract: Abnormal fatty acid oxidation has been associated with obesity and type 2 diabetes. At the transcriptional level, peroxisome proliferator-activated receptor-gamma coactivator 1? (PGC-1?) has been reported to strongly increase the ability of hormone nuclear receptors PPAR? and ERR? to drive transcription of fatty acid oxidation enzymes. In this study, we report that a specific agonist of the type 2 cannabinoid receptor (CB2R) can lead to fatty acid oxidation through the PGC-1? pathway. We have found that CB2R is expressed in differentiated C2C12 myotubes, and that use of the specific agonist trans-caryophyllene (TC) stimulates sirtuin 1 (SIRT1) deacetylase activity by increasing the phosphorylation of cAMP response element-binding protein (CREB), thus leading to increased levels of PGC-1? deacetylation. This use of TC treatment increases the expression of genes linked to the fatty acid oxidation pathway in a SIRT1/PGC-1?-dependent mechanism and also drastically accelerates the rate of complete fatty acid oxidation in C2C12 myotubes, neither of which occur when CB2R mRNA is knocked down using siRNA. These results reveal that activation of CB2R by a selective agonist promotes lipid oxidation through a signaling/transcriptional pathway. Our findings imply that pharmacological manipulation of CB2R may provide therapeutic possibilities to treat metabolic diseases associated with lipid dysregulation.

    Since 1970, solid waste with TRU or U-233 contamination in excess of 10 ..mu..Ci per kilogram of waste has been stored in a retrievable fashion at ORNL, such as in ss drums, concrete casks, and ss-lined wells. This report describes the results of a study performed to identify and evaluate alternatives for management of this waste and of the additional waste projected to be stored through 1995. The study was limited to consideration of the following basic strategies: Strategy 1: Leave waste in place as is; Strategy 2: Improve waste confinement; and Strategy 3: Retrieve waste and process for shipment to a Federal repository. Seven alternatives were identified and evaluated, one each for Strategies 1 and 2 and five for Strategy 3. Each alternative was evaluated from the standpoint of technical feasibility, cost, radiological risk and impact, regulatory factors and nonradiological environmental impact.

    We report herein the functional expression of an Fe-type nitrile hydratase (NHase) without the co-expression of an activator protein or the Escherichia coli chaperone proteins GroES/EL. Soluble protein was obtained when the {alpha}- and {beta}-subunit genes of the Fe-type NHase Comamonas testosteroni Ni1 (CtNHase) were synthesized with optimized E. coli codon usage and co-expressed. As a control, the Fe-type NHase from Rhodococcus equi TG328-2 (ReNHase) was expressed with (ReNHase{sup +Act}) and without (ReNHase{sup -Act}) its activator protein, establishing that expression of a fully functional, metallated ReNHase enzyme requires the co-expression of its activator protein, similar to all other Fe-type NHase enzymes reported to date, whereas the CtNHase does not. The X-ray crystal structure of CtNHase was determined to 2.4 {angstrom} resolution revealing an {alpha}{beta} heterodimer, similar to other Fe-type NHase enzymes, except for two important differences. First, two His residues reside in the CtNHase active site that are not observed in other Fe-type NHase enzymes and second, the active site Fe(III) ion resides at the bottom of a wide solvent exposed channel. The solvent exposed active site, along with the two active site histidine residues, are hypothesized to play a role in iron incorporation in the absence of an activator protein.

    This paper compares a small onsite wet storage pool to a dry cask storage facility in order to determine what type of spent fuel storage alternatives would best serve the utilities in consideration of the Nuclear Waste Policy Act of 1982. The Act allows the DOE to provide a total of 1900 metric tons (MT) of additional spent fuel storage capacity to utilities that cannot reasonably provide such capacity for themselves. Topics considered include the implementation of the Act (DOE away-from reactor storage), the Act's impact on storage needs, and an economic evaluation. The Waste Act mandates schedules for the determination of several sites, the licensing and construction of a high-level waste repository, and the study of a monitored retrievable storage facility. It is determined that a small wet pool storage facility offers a conservative and cost-effective approach for many stations, in comparison to dry cask storage.

    The Department of Energy is preparing a Programmatic Environmental Impact Statement (PEIS) for the Environmental Restoration and Waste Management Program. The PEIS has been divided into an Environmental Restoration section and a Waste Management section. Each section has a unique set of alternatives. This paper will focus on the waste management alternatives and analysis. The set of alternatives for waste management has been divided into waste categories. These categories are: high-level waste, transuranic waste, low-level waste, low-level mixed waste, greater-than-class C and low-level waste from commercial sources, hazardous waste, and spent nuclear fuel. This paper will discuss the alternatives and analytical approach that will be used to evaluate these alternatives for the low-level waste section. Although the same alternatives will be considered for all waste types, the analysis will be performed separately for each waste type. In the sections that follow, information will be provided on waste management configurations, the analysis of waste management alternatives, waste types and locations, facility and transportation activities, the facility and transportation impacts assessment, and the compilation of impacts.

    This report presents the first compilation by the Energy Information Administration (EIA) of information on alternatives to gasoline and diesel fuel. The purpose of the report is: (1) to provide background information on alternative transportation fuels and replacement fuels compared with gasoline and diesel fuel, and (2) to furnish preliminary estimates of alternative transportation fuels and alternative fueled vehicles as required by the Energy Policy Act of 1992 (EPACT), Title V, Section 503, ``Replacement Fuel Demand Estimates and Supply Information.`` Specifically, Section 503 requires the EIA to report annually on: (1) the number and type of alternative fueled vehicles in existence the previous year and expected to be in use the following year, (2) the geographic distribution of these vehicles, (3) the amounts and types of replacement fuels consumed, and (4) the greenhouse gas emissions likely to result from replacement fuel use. Alternative fueled vehicles are defined in this report as motorized vehicles licensed for on-road use, which may consume alternative transportation fuels. (Alternative fueled vehicles may use either an alternative transportation fuel or a replacement fuel.)     We have conducted an experiment to explore an alternative non-contact method of measuring the Inconel target window temperature. This experiment involves using a standard color camera to observe the visible light emitted from the Inconel target window at high heat in order to estimate the window temperature. The safety limit to prevent target window failure is 700 °C and therefore we need a reliable and accurate method of measuring temperature especially in the range of 600 °C to 700 °C if it is to replace the IR camera. In this temperature range the window will emit a significant amount of black body radiation within the visible range and hence the idea of using a color camera. The goal is to see if the shift in window color (determined by the RBG pixel values of the camera) as the target window is heated to 700 °C can be calibrated to the temperature.The reasons for exploring this as an alternative to an IR are camera are: significant cost reduction and potentially less complicated to calibrate.

    Active sites may be regarded as layers of residues, whereby the residues that interact directly with substrate also interact with residues in a second shell and these in turn interact with residues in a third shell. These residues in the second and third layers may have distinct roles in maintaining the essential chemical properties of the first-shell catalytic residues, particularly their spatial arrangement relative to the substrate binding pocket, and their electrostatic and dynamic properties. The extent to which these remote residues participate in catalysis and precisely how they affect first-shell residues remains unexplored. To improve our understanding of the roles of second- and third-shell residues in catalysis, we used THEMATICS to identify residues in the second and third shells of the Co-type nitrile hydratase from Pseudomonas putida (ppNHase) that may be important for catalysis. Five of these predicted residues, and three additional, conserved residues that were not predicted, have been conservatively mutated, and their effects have been studied both kinetically and structurally. The eight residues have no direct contact with the active site metal ion or bound substrate. These results demonstrate that three of the predicted second-shell residues ({alpha}-Asp164, {beta}-Glu56, and {beta}-His147) and one predicted third-shell residue ({beta}-His71) have significant effects on the catalytic efficiency of the enzyme. One of the predicted residues ({alpha}-Glu168) and the three residues not predicted ({alpha}-Arg170, {alpha}-Tyr171, and {beta}-Tyr215) do not have any significant effects on the catalytic efficiency of the enzyme.

    The 'Cosmic Evolution Survey' (COSMOS) enables the study of the spectral energy distributions (SEDs) of active galactic nuclei (AGNs) because of the deep coverage and rich sampling of frequencies from X-ray to radio. Here we present an SED catalog of 413 X-ray (XMM-Newton)-selected type 1 (emission line FWHM > 2000 km s{sup -1}) AGNs with Magellan, SDSS, or VLT spectrum. The SEDs are corrected for Galactic extinction, broad emission line contributions, constrained variability, and host galaxy contribution. We present the mean SED and the dispersion SEDs after the above corrections in the rest-frame 1.4 GHz to 40 keV, and show examples of the variety of SEDs encountered. In the near-infrared to optical (rest frame {approx}8 {mu}m-4000 A), the photometry is complete for the whole sample and the mean SED is derived from detections only. Reddening and host galaxy contamination could account for a large fraction of the observed SED variety. The SEDs are all available online.

    Protease activity within nascently released human immunodeficiency virus type 1 (HIV-1) particles is responsible for the cleavage of the viral polyproteins Gag and Gag-Pol into their constituent parts, which results in the subsequent condensation of the mature conical core surrounding the viral genomic RNA. Concomitant with viral maturation is a conformational change in the packaged viral RNA from a loosely associated dimer into a more thermodynamically stable form. In this study we used suboptimal concentrations of two protease inhibitors, lopinavir and atazanavir, to study their effects on Gag polyprotein processing and on the properties of the RNA in treated virions. Analysis of the treated virions demonstrated that even with high levels of inhibition of viral infectivity (IC{sub 90}), most of the Gag and Gag-Pol polyproteins were processed, although slight but significant increases in processing intermediates of Gag were detected. Drug treatments also caused a significant increase in the proportion of viruses displaying either immature or aberrant mature morphologies. The aberrant mature particles were characterized by an electron-dense region at the viral periphery and an electron-lucent core structure in the viral center, possibly indicating exclusion of the genomic RNA from these viral cores. Intriguingly, drug treatments caused only a slight decrease in overall thermodynamic stability of the viral RNA dimer, suggesting that the dimeric viral RNA was able to mature in the absence of correct core condensation.

    The Alternative Fuels for Vehicles Fleet Demonstration Program (AFV-FDP) was a multiyear effort to collect technical data for use in determining the costs and benefits of alternative-fuel vehicles (AFVs) in typical applications in New York State. This report, Volume 2, includes 13 appendices to Volume 1 that expand upon issues raised therein. Volume 1 provides: (1) Information about the purpose and scope of the AFV-FDP; (2) A summary of AFV-FDP findings organized on the basis of vehicle type and fuel type; (3) A short review of the status of AFV technology development, including examples of companies in the State that are active in developing AFVs and AFV components; and (4) A brief overview of the status of AFV deployment in the State. Volume 3 provides expanded reporting of AFV-FDP technical details, including the complete texts of the brochure Garage Guidelines for Alternative Fuels and the technical report Fleet Experience Survey Report, plus an extensive glossary of AFV terminology.     c-type cytochromes play essential roles in many biological activities of both prokaryotic and eukaryotic cells, including electron transfer, enzyme catalysis and induction of apoptosis. We report a novel enrichment strategy for identifying c-type heme-containing peptides that uses non-activated IMAC resin. The strategy demonstrated at least seven-fold enrichment for heme-containing peptides digested from a cytochrome c protein standard, and quantitative linear performance was also assessed for heme-containing peptide enrichment. Heme-containing peptides extracted from the periplasmic fraction of Shewanella oneidensis MR-1 were further identified using higher-energy collisional dissociation tandem mass spectrometry. The results demonstrated the applicability of this enrichment strategy to identify c-type heme-containing peptides from a highly complex biological sample, and at the same time, confirmed the periplasmic localization of heme-containing proteins during suboxic respiration activities of S. oneidensis MR-1.

    The National Renewable Energy Laboratory (NREL) is a U.S. Department of Energy (DOE) national laboratory; this project was funded by DOE. One of NREL`s missions is to objectively evaluate the performance, emissions, and operating costs of alternative fuel vehicles so fleet managers can make informed decisions when purchasing them. Alternative fuels have made greater inroads into the transit bus market than into any other. Each year, the American Public Transit Association (APTA) surveys its members on their inventory and buying plans. The latest APTA data show that about 4% of the 50,000 transit buses in its survey run on an alternative fuel. Furthermore, 1 in 5 of the new transit buses that members have on order are alternative fuel buses. This program was designed to comprehensively and objectively evaluate the alternative fuels in use in the industry.

    This technical memorandum estimates the effects of alternative contractual commitments that may be initiated by the Western Area Power Administration`s Salt Lake City Area Office. It also studies hydropower operational restrictions at the Salt Lake City Area Integrated Projects in combination with these alternatives. Power marketing and hydropower operational effects are estimated in support of Western`s Electric Power Marketing Environmental Impact Statement (EIS). Electricity production and capacity expansion for utility systems that will be directly affected by alternatives specified in the EIS are simulated. Cost estimates are presented by utility type and for various activities such as capacity expansion, generation, long-term firm purchases and sales, fixed operation and maintenance expenses, and spot market activities.     We present new mid-infrared imaging data for three Type-1 Seyfert galaxies obtained with T-ReCS on the Gemini-South Telescope at subarcsecond resolution. Our aim is to enlarge the sample studied in a previous work to compare the properties of Type-1 and Type-2 Seyfert tori using clumpy torus models and a Bayesian approach to fit the infrared (IR) nuclear spectral energy distributions. Thus, the sample considered here comprises 7 Type-1, 11 Type-2, and 3 intermediate-type Seyferts. The unresolved IR emission of the Seyfert 1 galaxies can be reproduced by a combination of dust heated by the central engine and direct active galactic nucleus (AGN) emission, while for the Seyfert 2 nuclei only dust emission is considered. These dusty tori have physical sizes smaller than 6 pc radius, as derived from our fits. Unification schemes of AGN account for a variety of observational differences in terms of viewing geometry. However, we find evidence that strong unification may not hold and that the immediate dusty surroundings of Type-1 and Type-2 Seyfert nuclei are intrinsically different. The Type-2 tori studied here are broader, have more clumps, and these clumps have lower optical depths than those of Type-1 tori. The larger the covering factor of the torus, the smaller the probability of having a direct view of the AGN, and vice versa. In our sample, Seyfert 2 tori have larger covering factors (C{sub T} = 0.95 {+-} 0.02) and smaller escape probabilities (P{sub esc} = 0.05% {+-} {sup 0.08}{sub 0.03}%) than those of Seyfert 1 (C{sub T} = 0.5 {+-} 0.1; P{sub esc} = 18% {+-} 3%). All the previous differences are significant according to the Kullback-Leibler divergence. Thus, on the basis of the results presented here, the classification of a Seyfert galaxy as a Type-1 or Type-2 depends more on the intrinsic properties of the torus rather than on its mere inclination toward us, in contradiction with the simplest unification model.

    Perovskite-type transition-metal oxides have a wide variety of physical properties and triggered intensive research on functional devices in the form of heteroepitaxial junctions. However, there is a missing component that is a p-type conventional band semiconductor. LaRhO{sub 3} (LRO) is one of very few promising candidates having its bandgap between filled t{sub 2g} and empty e{sub g} of Rh in low-spin state, but there has been no report on the synthesis of large-size single crystals or thin films. Here, we report on the junction properties of single-crystalline thin films of LRO grown on (110) oriented Nb-doped SrTiO{sub 3} substrates. The external quantum efficiency of the photo-electron conversion exceeds 1% in the visible-light region due to the wide depletion layer and long diffusion length of minority carriers in LRO. Clear indication of p-type band semiconducting character in a perovskite oxide of LRO will pave a way to explore oxide electronics of perovskite heterostructures.

    This invention relates to an electromechanical energy converter with inertial energy storage. The device, a single phase, two or multi-pole alternator with stationary field coils, and a rotating armature is provided. The rotor itself may be of laminated steel for slower pulses or for faster pulses should be nonmagnetic and electrically nonconductive in order to allow rapid penetration of the field as the armature coil rotates. The armature coil comprises a plurality of power generating conductors mounted on the rotor. The alternator may also include a stationary or counterrotating compensating coil to increase the output voltage thereof and to reduce the internal impedance of the alternator at the moment of peak outout. As the machine voltage rises sinusoidally, an external trigger switch is adapted to be closed at the appropriate time to create the desired output current from said alternator to an external load circuit, and as the output current passes through zero a self-commutating effect is provided to allow the switch to disconnect the generator from the external circuit.

    Even though peat is a low-heating value and low-bulk density fossil fuel which in its natural state contains over 80 percent moisture, it can be an economical alternative to coal, and fuel oil, as is the case in Iceland and Finland for direct combustion applications. This is because of the relative ease with which peat can be harvested, and the generally low sulfur and ash content of peat. Recent studies show that peat also has very favorable characteristics for conversion to synthetic fuels. Tests show that on the basis of chemistry and kinetics, peat is a better raw material than coal for production of synthetic fuels. Recent estimates also show that conversion of peat to high-Btu gas (>950 Btu/scf) is competitive with other alternatives of synthetic high-Btu gas. Therefore, peat can be an economical energy alternative depending upon location of peat deposits, region of energy need, scale of operation and cost of other energy alternatives.

    The commercialization of alternative fuels is analyzed. Following a synopsis of US energy use, the concept of commercialization, the impacts of supply shortages and demand inelasticity upon commercialization, and the status of alternative fuels commercialization to date in the US are discussed. The US energy market is viewed as essentially numerous submarkets. The interrelationship among these submarkets precludes the need to commercialize for a specific fuel/use. However, the level of consumption, the projected growth in demand, and the inordinate dependence upon foreign fuels dictate that additional fuel supplies in general be brought to the US energy marketplace. Commercialization efforts encompass a range of measures designed to accelerate the arrival of technologies or products in the marketplace. As discussed in this paper, such a union of willing buyers and willing sellers requires that three general conditions be met: product quality comparable to existing products; price competitiveness; and adequate availability of supply. Product comparability presently appears to be the least problematic of these three requirements. Ethanol/gasoline and methanol/gasoline blends, for example, demonstrate the fact that alternative fuel technologies exist. Yet price and availability (i.e., production capacity) remain major obstacles. Given inelasticity (with respect to price) in the US and abroad, supply shortages - actual or contrived - generate upward price pressure and should make once-unattractive alternative fuels more price competitive. It is noted, however, that actual price competitiveness has been slow to occur and that even with price competitiveness, the lengthy time frame needed to achieve significant production capacity limits the near-term impact of alternative fuels.

    Since the 1960's, the Department of Transportation Specification (DOT Spec) 6M packages have been used extensively for transportation of Type B quantities of radioactive materials between Department of Energy (DOE) facilities, laboratories, and productions sites. However, due to the advancement of packaging technology, the aging of the 6M packages, and variability in the quality of the packages, the DOT implemented a phased elimination of the 6M specification packages (and other DOT Spec packages) in favor of packages certified to meet federal performance requirements. DOT issued the final rule in the Federal Register on October 1, 2004 requiring that use of the DOT Specification 6M be discontinued as of October 1, 2008. A main driver for the change was the fact that the 6M specification packagings were not supported by a Safety Analysis Report for Packaging (SARP) that was compliant with Title 10 of the Code of Federal Regulations part 71 (10 CFR 71). Therefore, materials that would have historically been shipped in 6M packages are being identified as contents in Type B (and sometimes Type A fissile) package applications and addenda that are to be certified under the requirements of 10 CFR 71. The requirements in 10 CFR 71 include that the Safety Analysis Report for Packaging (SARP) must identify the maximum radioactivity of radioactive constituents and maximum quantities of fissile constituents (10 CFR 71.33(b)(1) and 10 CFR 71.33(b)(2)), and that the application (i.e., SARP submittal or SARP addendum) demonstrates that the external dose rate (due to the maximum radioactivity of radioactive constituents and maximum quantities of fissile constituents) on the surface of the packaging (i.e., package and contents) not exceed 200 mrem/hr (10 CFR 71.35(a), 10 CFR 71.47(a)).     In addition to promoting energy efficiency, China has actively pursued alternative energy development as a strategy to reduce its energy demand and carbon emissions. One area of particular focus has been to raise the share of alternative energy in China’s rapidly growing electricity generation with a 2020 target of 15% share of total primary energy. Over the last ten years, China has established several major renewable energy regulations along with programs and subsidies to encourage the growth of non-fossil alternative energy including solar, wind, nuclear, hydro, geothermal and biomass power as well as biofuels and coal alternatives. This study thus seeks to examine China’s alternative energy in terms of what has and will continue to drive alternative energy development in China as well as analyze in depth the growth potential and challenges facing each specific technology. This study found that despite recent policies enabling extraordinary capacity and investment growth, alternative energy technologies face constraints and barriers to growth. For relatively new technologies that have not achieved commercialization such as concentrated solar thermal, geothermal and biomass power, China faces technological limitations to expanding the scale of installed capacity. While some alternative technologies such as hydropower and coal alternatives have been slowed by uneven and often changing market and policy support, others such as wind and solar PV have encountered physical and institutional barriers to grid integration. Lastly, all alternative energy technologies face constraints in human resources and raw material resources including land and water, with some facing supply limitations in critical elements such as uranium for nuclear, neodymium for wind and rare earth metals for advanced solar PV.
