    The United States Department of Energy's (DOE) Atmospheric Radiation Measurement (ARM) Program began operations in its Tropical Western Pacific (TWP) locale in October 1996 when the first Atmospheric Radiation and Cloud Station (ARCS) began collecting data on Manus Island in Papua New Guinea (PNG). Two years later, in November 1998, a second ARCS began operations on the island of Nauru in the Central Pacific. Now a third ARCS has begun collecting data in Darwin, Australia. The Manus, Nauru, and Darwin sites are operated through collaborative agreements with the PNG National Weather Service, The Nauru Department of Industry and Economic Development (IED), and the Australian Bureau of Meteorology's (BOM) Special Services Unit (SSU) respectively. All ARM TWP activities in the region are coordinated with the South Pacific Regional Environment Programme (SPREP) based in Apia, Samoa. The Darwin ARM site and its role in the ARM TWP Program are discussed.

    A cloud properties and radiative heating rates dataset is presented where cloud properties retrieved using lidar and radar observations are input into a radiative transfer model to compute radiative fluxes and heating rates at three ARM sites located in the Tropical Western Pacific (TWP) region. The cloud properties retrieval is a conditional retrieval that applies various retrieval techniques depending on the available data, that is if lidar, radar or both instruments detect cloud. This Combined Remote Sensor Retrieval Algorithm (CombRet) produces vertical profiles of liquid or ice water content (LWC or IWC), droplet effective radius (re), ice crystal generalized effective size (Dge), cloud phase, and cloud boundaries. The algorithm was compared with 3 other independent algorithms to help estimate the uncertainty in the cloud properties, fluxes, and heating rates (Comstock et al. 2013). The dataset is provided at 2 min temporal and 90 m vertical resolution. The current dataset is applied to time periods when the MMCR (Millimeter Cloud Radar) version of the ARSCL (Active Remotely-Sensed Cloud Locations) Value Added Product (VAP) is available. The MERGESONDE VAP is utilized where temperature and humidity profiles are required. Future additions to this dataset will utilize the new KAZR instrument and its associated VAPs.

The operations are supported by government agencies in each host country. Covering the area roughly between 10 degrees N and 10 degrees S of the equator and from 130 degrees E to 167 degrees E, the TWP locale includes a region that plays a large role in the interannual variability observed in the global climate system. More than 250,000 TWP data sets from 1996 to the present reside in the ARM Archive. Begin at the TWP information page for links or access data directly from the ARM Archive at http://www.archive.arm.gov/. Users will need to register for a password, but all files are then free for viewing or downloading. The ARM Archive physically resides at the Oak Ridge National Laboratory.

    Single column models (SCM) are useful testbeds for investigating the parameterisation schemes of numerical weather prediction and climate models. The usefulness of SCM simulations are limited, however, by the accuracy of the best-estimate large-scale data prescribed. One method to address this uncertainty is to perform ensemble simulations of the SCM. This study first derives an ensemble of large-scale data for the Tropical Warm Pool International Cloud Experiment (TWP-ICE) based on an estimate of a possible source of error in the best-estimate product. This data is then used to carry out simulations with 11 SCM and 2 cloud-resolving models (CRM). Best-estimate simulations are also performed. All models show that moisture related variables are close to observations and there are limited differences between the best-estimate and ensemble mean values. The models, however, show different sensitivities to changes in the forcing particularly when weakly forced. The ensemble simulations highlight important differences in the moisture budget between the SCM and CRM. Systematic differences are also apparent in the ensemble mean vertical structure of cloud variables. The ensemble is further used to investigate relations between cloud variables and precipitation identifying large differences between CRM and SCM. This study highlights that additional information can be gained by performing ensemble simulations enhancing the information derived from models using the more traditional single best-estimate simulation.

    This study documents the characteristics of the large-scale structures and diabatic heating and drying profiles observed during the Tropical Warm Pool–International Cloud Experiment (TWP-ICE), which was conducted in January–February 2006 in Darwin during the northern Australian monsoon season. The examined profiles exhibit significant variations between four distinct synoptic regimes that were observed during the experiment. The active monsoon period is characterized by strong upward motion and large advective cooling and moistening throughout the entire troposphere, while the suppressed and clear periods are dominated by moderate midlevel subsidence and significant low- to midlevel drying through horizontal advection. The midlevel subsidence andmore » horizontal dry advection are largely responsible for the dry midtroposphere observed during the suppressed period and limit the growth of clouds to low levels. During the break period, upward motion and advective cooling and moistening located primarily at midlevels dominate together with weak advective warming and drying (mainly from horizontal advection) at low levels. The variations of the diabatic heating and drying profiles with the different regimes are closely associated with differences in the large-scale structures, cloud types, and rainfall rates between the regimes. Strong diabatic heating and drying are seen throughout the troposphere during the active monsoon period while they are moderate and only occur above 700 hPa during the break period. The diabatic heating and drying tend to have their maxima at low levels during the suppressed periods. Furthermore, the diurnal variations of these structures between monsoon systems, continental/coastal, and tropical inland-initiated convective systems are also examined.« less

    The impact of oceanic convection on its environment and the relationship between the characteristics of the convection and the resulting cirrus characteristics is still not understood. An intense airborne measurement campaign combined with an extensive network of ground-based observations is being planned for the region near Darwin, Northern Australia, during January-February, 2006, to address these questions. The Tropical Warm Pool International Cloud Experiment (TWP-ICE) will be the first field program in the tropics that attempts to describe the evolution of tropical convection, including the large scale heat, moisture, and momentum budgets, while at the same time obtaining detailed observations of cloud properties and the impact of the clouds on the environment. The emphasis will be on cirrus for the cloud properties component of the experiment. Cirrus clouds are ubiquitous in the tropics and have a large impact on their environment but the properties of these clouds are poorly understood. A crucial product from this experiment will be a dataset suitable to provide the forcing and testing required by cloud-resolving models and parameterizations in global climate models. This dataset will provide the necessary link between cloud properties and the models that are attempting to simulate them.

    Ten 3D cloud-resolving model (CRM) simulations and four 3D limited area model (LAM) simulations of an intense mesoscale convective system observed on January 23-24, 2006 during the Tropical Warm Pool International Cloud Experiment (TWP-ICE) are compared with each other and with observations and retrievals from a scanning polarimetric radar, co-located UHF and VHF vertical profilers, and a Joss-Waldvogel disdrometer in an attempt to explain published results showing a low bias in simulated stratiform rainfall. Despite different forcing methodologies, similar precipitation microphysics errors appear in CRMs and LAMs with differences that depend on the details of the bulk microphysics scheme used. One-moment schemes produce too many small raindrops, which biases Doppler velocities low, but produces rain water contents (RWCs) that are similar to observed. Two-moment rain schemes with a gamma shape parameter (?) of 0 produce excessive size sorting, which leads to larger Doppler velocities than those produced in one-moment schemes, but lower RWCs than observed. Two moment schemes also produce a convective median volume diameter distribution that is too broad relative to observations and thus, may have issues balancing raindrop formation, collision coalescence, and raindrop breakup. Assuming a ? of 2.5 rather than 0 for the raindrop size distribution improves one-moment scheme biases, and allowing ? to have values greater than 0 may improve two-moment schemes. Under-predicted stratiform rain rates are associated with under-predicted ice water contents at the melting level rather than excessive rain evaporation, in turn likely associated with convective detrainment that is too high in the troposphere and mesoscale circulations that are too weak. In addition to stronger convective updrafts than observed, limited domain size prevents a large, well-developed stratiform region from developing in CRMs, while a dry bias in ECMWF analyses does the same to the LAMs.

    The Tropical Warm Pool - International Cloud Experiment (TWP-ICE) provided high quality model forcing and observational datasets through which detailed model and observational intercomparisons could be performed. In this first of a two part study, precipitation and cloud structures within nine cloud-resolving model simulations are compared with scanning radar reflectivity and satellite infrared brightness temperature observations during an active monsoon period from 19 to 25 January 2006. Most simulations slightly overestimate volumetric convective rainfall. Overestimation of simulated convective area by 50% or more in several simulations is somewhat offset by underestimation of mean convective rain rates. Stratiform volumetric rainfall is underestimated by 13% to 53% despite overestimation of stratiform area by up to 65% because stratiform rain rates in every simulation are much lower than observed. Although simulations match the peaked convective radar reflectivity distribution at low levels, they do not reproduce the peaked distributions observed above the melting level. Simulated radar reflectivity aloft in convective regions is too high in most simulations. 29 In stratiform regions, there is a large spread in model results with none resembling 30 observed distributions. Above the melting level, observed radar reflectivity decreases 31 more gradually with height than simulated radar reflectivity. A few simulations produce 32 unrealistically uniform and cold 10.8-?m infrared brightness temperatures, but several 33 simulations produce distributions close to observed. Assumed ice particle size 34 distributions appear to play a larger role than ice water contents in producing incorrect 35 simulated radar reflectivity distributions aloft despite substantial differences in mean 36 graupel and snow water contents across models. 37

    Platelet-associated C3 (PA-C3) was measured with a quantitative immunofluorescence assay. With this assay, PA-C3 levels were determined for 78 normal volunteers, 30 patients with immune thrombocytopenic purpura (ITP), and 20 patients with nonimmune thrombocytopenias. Platelet-associatd IgG (PA-lgG) levels were also measured with our standard quantitative immunofluorescence assay. All patients with nonimmune thrombocytopenias and ITP in remission had normal PA-C3 levels. Twenty-four patients with active ITP wre classified into 3 groups: 9 (38%) with increased PA-IgG and normal PA-C3 levels, 10 (42%) with elevated PA-C3 and PA-IgG levels, and 5 (20%) with increased PA-C3 values only. A direct correlation was found between PA-C3 and PA-IgG levels. PA-IgG levels were higher in the group of patients with elevated PA-C3 levels than in those with normal values. Platelet survival studies showed reduced survival times of 1.5-2.5 days for the 5 patients with elevated PA-C3 levels only. Elevated PA-C3 levels returned to normal in 7 ITP patients whose platelet counts increased in response to corticosteriod therapy or to splenectomy. Therefore, PA-C3 and PA-IgG assays can be used to identify patients with ITP, to follow their response to therapy, and to classify them into immunologic subgroups similar to red cell classifiation by Coombs' testing in immune hemolytic anemia.

    Faceted browsing is a common technique for exploring collections where the data can be grouped into a number of pre-defined categories, most often generated from textual metadata. Historically, faceted browsing has been applied to a single data type such as text or image data. However, typical collections contain multiple data types, such as information from web pages that contain text, images, and video. Additionally, when browsing a collection of images and video, facets are often created based on the metadata which may be incomplete, inaccurate, or missing altogether instead of the actual visual content contained within those images and video. In this work we address these limitations by presenting MultiFacet, a faceted browsing interface that supports multiple data types. MultiFacet constructs facets for images and video in a collection from the visual content using computer vision techniques. These visual facets can then be browsed in conjunction with text facets within a single interface to reveal relationships and phenomena within multimedia collections. Additionally, we present a use case based on real-world data, demonstrating the utility of this approach towards browsing a large multimedia data collection.

    Spontaneous emission measurements are discussed for the Swings transitions of the C3 radical in laser-generated graphite plasma, and the spectroscopy of the C3 radical in carbon vapor and plasma is summarized. A review is given of some theoretical calculations and emission spectroscopic investigations are presented. Time-averaged, laser-induced optical breakdown spectra arereported from Nd:YAG laser generated graphite microplasma. In 200-300 Torr of argon and helium, and depending on the specific experimental configuration,a weak emission continuum is observed centered at 400 nm when using a laser fluence of typically1 J/cm2. Such continua were not detected in our previous experiments using focused laser radiation. The possibilities for the origin of this continuum are considered.

    Ten 3D cloud-resolving model (CRM) simulations and four 3D limited area model (LAM) simulations of an intense mesoscale convective system observed on January 23-24, 2006 during the Tropical Warm Pool International Cloud Experiment (TWP-ICE) are compared with each other and with observed radar reflectivity fields and dual-Doppler retrievals of vertical wind speeds in an attempt to explain published results showing a high bias in simulated convective radar reflectivity aloft. This high bias results from ice water content being large, which is a product of large, strong convective updrafts, although hydrometeor size distribution assumptions modulate the size of this bias. Snow reflectivity can exceed 40 dBZ in a two-moment scheme when a constant bulk density of 100 kg m-3 is used. Making snow mass more realistically proportional to area rather than volume should somewhat alleviate this problem. Graupel, unlike snow, produces high biased reflectivity in all simulations. This is associated with large amounts of liquid water above the freezing level in updraft cores. Peak vertical velocities in deep convective updrafts are greater than dual-Doppler retrieved values, especially in the upper troposphere. Freezing of large rainwater contents lofted above the freezing level in simulated updraft cores greatly contributes to these excessive upper tropospheric vertical velocities. Strong simulated updraft cores are nearly undiluted, with some showing supercell characteristics. Decreasing horizontal grid spacing from 900 meters to 100 meters weakens strong updrafts, but not enough to match observational retrievals. Therefore, overly intense simulated updrafts may partly be a product of interactions between convective dynamics, parameterized microphysics, and large-scale environmental biases that promote different convective modes and strengths than observed.

  3. Analysis of Cloud-resolving Simulations of a Tropical Mesoscale Convective System Observed during TWP-ICE: Vertical Fluxes and Draft Properties in Convective and Stratiform Regions

    We analyze three cloud-resolving model simulations of a strong convective event observed during the TWP-ICE campaign, differing in dynamical core, microphysical scheme or both. Based on simulated and observed radar reflectivity, simulations roughly reproduce observed convective and stratiform precipitating areas. To identify the characteristics of convective and stratiform drafts that are difficult to observe but relevant to climate model parameterization, independent vertical wind speed thresholds are calculated to capture 90% of total convective and stratiform updraft and downdraft mass fluxes. Convective updrafts are fairly consistent across simulations (likely owing to fixed large-scale forcings and surface conditions), except that hydrometeor loadings differ substantially. Convective downdraft and stratiform updraft and downdraft mass fluxes vary notably below the melting level, but share similar vertically uniform draft velocities despite differing hydrometeor loadings. All identified convective and stratiform downdrafts contain precipitation below ~10 km and nearly all updrafts are cloudy above the melting level. Cold pool properties diverge substantially in a manner that is consistent with convective downdraft mass flux differences below the melting level. Despite differences in hydrometeor loadings and cold pool properties, convective updraft and downdraft mass fluxes are linearly correlated with convective area, the ratio of ice in downdrafts to that in updrafts is ~0.5 independent of species, and the ratio of downdraft to updraft mass flux is ~0.5-0.6, which may represent a minimum evaporation efficiency under moist conditions. Hydrometeor loading in stratiform regions is found to be a fraction of hydrometeor loading in convective regions that ranges from ~10% (graupel) to ~90% (cloud ice). These findings may lead to improved convection parameterizations.

    The human complement system plays an essential role in innate and adaptive immunity by marking and eliminating microbial intruders. Activation of complement on foreign surfaces results in proteolytic cleavage of complement component 3 (C3) into the potent opsonin C3b, which triggers a variety of immune responses and participates in a self-amplification loop mediated by a multi-protein assembly known as the C3 convertase. The human pathogen Staphylococcus aureus has evolved a sophisticated and potent complement evasion strategy, which is predicated upon an arsenal of potent inhibitory proteins. One of these, the staphylococcal complement inhibitor (SCIN), acts at the level of the C3 convertase (C3bBb) and impairs downstream complement function by trapping the convertase in a stable but inactive state. Previously, we have shown that SCIN binds C3b directly and competitively inhibits binding of human factor H and, to a lesser degree, that of factor B to C3b. Here, we report the co-crystal structures of SCIN bound to C3b and C3c at 7.5 and 3.5 {angstrom} limiting resolution, respectively, and show that SCIN binds a critical functional area on C3b. Most significantly, the SCIN binding site sterically occludes the binding sites of both factor H and factor B. Our results give insight into SCIN binding to activated derivatives of C3, explain how SCIN can recognize C3b in the absence of other complement components, and provide a structural basis for the competitive C3b-binding properties of SCIN. In the future, this may suggest templates for the design of novel complement inhibitors based upon the SCIN structure.

  13. Characterization of the third component of complement (C3) after activation by cigarette smoke

    Activation of lung complement by tobacco smoke may be an important pathogenetic factor in the development of pulmonary emphysema in smokers. We previously showed that cigarette smoke can modify C3 and activate the alternative pathway of complement in vitro. However, the mechanism of C3 activation was not fully delineated in these earlier studies. In the present report, we show that smoke-treated C3 induces cleavage of the alternative pathway protein, Factor B, when added to serum containing Mg-EGTA. This effect of cigarette smoke is specific for C3 since smoke-treated C4, when added to Mg-EGTA-treated serum, fails to activate the alternative pathway and fails to induce Factor B cleavage. Smoke-modified C3 no longer binds significant amounts of (/sup 14/C)methylamine (as does native C3), and relatively little (/sup 14/C)methylamine is incorporated into its alpha-chain. Thus, prior internal thiolester bond cleavage appears to have occurred in C3 activated by cigarette smoke. Cigarette smoke components also induce formation of noncovalently associated, soluble C3 multimers, with a Mr ranging from 1 to 10 million. However, prior cleavage of the thiolester bond in C3 with methylamine prevents the subsequent formation of these smoke-induced aggregates. These data indicate that cigarette smoke activates the alternative pathway of complement by specifically modifying C3 and that these modifications include cleavage of the thiolester bond in C3 and formation of noncovalently linked C3 multimers.

    Carbenes of platinum and palladium, PtC3 and PdC3 , were generated in the gas phase through laser vaporization of a metal target in the presence of a low concentration of a hydrocarbon precursor undergoing supersonic expansion. Rotational spectroscopy and abinitio calculations confirm that both molecules are linear. The geometry of PtC3 was accurately determined by fitting to the experimental moments of inertia of twenty-six isotopologues. In conclusion, the results are consistent with the proposal of an autogenic isolobal relationship between O, Au+ , and Ptatoms.

  16. Halogenation and proteolysis of complement component C3 on Salmonella typhimurium during phagocytosis by human neutrophils

    We examined the fate of C component C3 on the surface of Salmonella typhimurium during ingestion by human neutrophils. Initial experiments showed that C3 fragments and C3-acceptor complexes were the major serum ligands which were surface iodinated by canine myeloperoxidase on serum-incubated rough and smooth isolates of S. typhimurium. In contrast, labeled C3 was not identified when the same organisms were ingested by neutrophils in the presence of 125I-Na, a situation previously shown to iodinate particulate targets via the neutrophil myeloperoxidase-halide-H2O2 system. Pretreatment of neutrophils before phagocytosis with the lipid-soluble protease inhibitor diisopropylfluorophosphate (DFP), but not with other protease inhibitors (p-nitrophenylguanidinobenzoate, leupeptin, pepstatin), substantially blocked proteolysis of 125I-C3 on S. typhimurium strain RG108 during ingestion by neutrophils. Purification of neutrophil phagosomes containing S. typhimurium-bearing 125I-C3 showed that DFP but no other protease inhibitors blocked proteolysis of 125I-C3 within phagosomes. Iodinated C3-acceptor complexes were identified by immunoprecipitation from the detergent-insoluble fraction of phagosomes prepared from DFP-treated cells ingesting S. typhimurium in the presence of 125I-Na. These results show that C3 fragments on the surface of S. typhimurium are the major serum ligands which are halogenated and degraded by proteolysis during phagocytosis by human neutrophils, and suggest that the majority of proteolysis on the ingested target occurs within the neutrophil phagosome.

    Crithidia fasciculata, an insect trypanosomatid is readily lysed by normal human serum at concentrations as low as 3%. Lysis occurs in the presence of Mg+2-EGTA and is antibody independent, indicating that the alternative pathway of complement activation is involved. Analysis of (131I)C3 deposition on C. fasciculata cells using C8-deficient serum, revealed that about 4 x 10(5) C3 molecules bound to each cell. Most of the C3 was bound to cells as C3b, part of it forming high molecular weight complexes, which could be dissociated by methylamine treatment at alkaline pH. To characterize the C3 acceptors on C. fasciculata, surface-iodinated cells were incubated with C8D or heat-inactivated serum, extracted and immunoprecipitated with anti-C3 or anti-arabinogalactan antisera. Analysis of the immunoprecipitated material on SDS gels showed high-molecular weight components, which disappeared after methylamine treatment, giving rise to a component of 200 kDa molecular size. This 200-kDa component corresponded to a purified arabinogalactan complex, which was immunoprecipitated from labeled cell extracts, without incubation with C8D, using anti-arabinogalactan antibodies. These results suggest that the arabinogalactan glycoconjugate is a C3 acceptor in C. fasciculata during complement activation. Purified arabinogalactan complexes were able to inactivate C3 in vitro. Solubilization in KOH to cleave the peptide moiety rendered it unable to inactivate C3. Apparently, the aggregated state of the purified arabinogalactan component at the cell surface is important for C3 deposition and activation.

    The project, ''Reduction of U(VI) and toxic metals by Desulfovibrio cytochrome c3'', is designed to obtain spectroscopic information for or against a functional interaction of cytochrome c3 and uranium in the whole cells. That is, is the cytochrome c3 the uranium reductase? Our approach has been to start with purified cytochrome and determine any unique spectral disturbances during electron flow to U(VI). Then we will attempt to identify these signals emanating from cells actively reducing uranium. This project is being carried out in collaboration with Dr. William Woodruff at the Los Alamos National Laboratory where the spectral experiments are being carried out.

  6. Physiological and growth responses of C3 and C4 plants at the Pleistocene glacial maximum

    A C3 plant (Abutilon theophrasti) and a C4 plant (Amaranthus retroflexus) were grown from seed in the Duke University Phytotron under four CO2 concentrations (15 Pa, below the Pleistocene minimum), 27 Pa (pre-industrial), 35 Pa (current), and 70 Pa (future) to examine photosynthetic, growth and reproduction responses of annual plants to historic and future levels of CO2. Net photosynthesis and growth were greatly inhibited at 15 Pa and greatly stimulated at 70 Pa. in the C3 Abutilon but only slightly affected in the C4 Amaranthus. Flower bud initiation was not affected by CO2 treatment in either species but all flower buds in 15 Pa CO2 aborted in the C3 within two days of appearance while no inhibition of reproduction was observed at low CO2 in the C4. Differences in physiology, growth and reproduction to the low levels of atmospheric CO2 of the Pleistocene suggest that competitive interactions of C3 and C4 annuals have changed through geologic time. A major question concerning the survival and evolution of obligate C3 annuals during the CO2 minima of the Pleistocene is raised by the results of this study.

    The central objective of our proposed research was twofold: 1) to investigate the structure-function relationship of Desulfovibrio desulfuricans (now Desulfovibrio alaskensis G20) cytochrome c3 with uranium and 2) to elucidate the mechanism for uranium reduction in vitro and in vivo. Physiological analysis of a mutant of D. desulfuricans with a mutation of the gene encoding the type 1 tetraheme cytochrome c3 had demonstrated that uranium reduction was negatively impacted while sulfate reduction was not if lactate were the electron donor. This was thought to be due to the presence of a branched pathway of electron flow from lactate leading to sulfate reduction. Our experimental plan was to elucidate the structural and mechanistic details of uranium reduction involving cytochrome c3.

    Complete active space self-consistent calculations of the energy and dipole moment functions were carried out for C3 in its electronic ground state. The absorption coefficient between 0.7 and 75 microns is calculated on the basis of the vibrational band strength of transitions between the 800 lowest states with v less than or equal to (4, 39, 3). In cool carbon stars with a high C/O ratio, C3 is found to be the most prevalent of the known opacity sources. 49 refs.

    The new C3 Gore Excluder delivery system enables both up/downward and rotational repositioning of the device before complete deployment. This contributes to more precise proximal landing and permits facilitation of the contralateral gate cannulation. During separate deployment, the position of the ipsilateral limb can also be readjusted. We have used the modified C3 delivery system in more than 50 patients, and in most cases, we were able to utilize the repositioning options of the device to achieve optimal fixation and sealing. However, we present a case where our attempt to readjust the position of the ipsilateral limb led to upward migration of the main body with coverage of the left renal artery origin. The latter was secured with a bare stent implantation.

  16. Atmospheric formation and removal of C3-C5 peroxyacyl nitrates

    The C3-C5 peroxyacyl nitrates RC(O)OONO{sub 2} (R=Et, n-Pr, i-Pr, n-Bu, i-Bu, sec-Bu, t-Bu, Ch{sub 2}=CH- and CH{sub 2}=C(CH{sub 3})-) have been synthesized and prepared in situ and have been characterized by electron capture gas chromatography. Their thermal decomposition rates have been measured and are similar to that of PAN (R = CH{sub 3}). Carbonyl products have been identified and the corresponding reaction mechanisms have been outlined. Ambient levels of several peroxyacyl nitrates (R =CH{sub 3}, Et, n-Pr and CH{sub 2}=(CH{sub 3})-) have been measured. The results are discussed with focus on atmospheric hydrocarbons as precursors to C3-C5 peroxyacyl nitrates in the atmosphere.

    New data are reported from the operation of a 2-liter C3F8 bubble chamber in the 2100 meter deep SNOLAB underground laboratory, with a total exposure of 211.5 kg-days at four different recoil energy thresholds ranging from 3.2 keV to 8.1 keV. These data show that C3F8 provides excellent electron recoil and alpha rejection capabilities at very low thresholds, including the rst observation of a dependence of acoustic signal on alpha energy. Twelve single nuclear recoil event candidates were observed during the run. The candidate events exhibit timing characteristics that are not consistent with the hypothesis of a uniform time distribution, and no evidence for a dark matter signal is claimed. These data provide the most sensitive direct detection constraints on WIMP-proton spin-dependent scattering to date, with signicant sensitivity at low WIMP masses for spin-independent WIMP-nucleon scattering.

    To address the relationship between the apolipoprotein A5 and C3 genes, we generated independent lines of mice that either over-expressed or completely lacked both genes. We report both lines display normal triglyceride concentrations compared to over-expression or deletion of either gene alone. Together, these data support that APOA5 and APOC3 independently influence plasma triglyceride concentrations but in an opposing manner.

    Sera of 72 lead workers and of 53 reference subjects were examined for levels of immunoglobulins IgM, IgG, and IgA, and complement C3 by radial immunodiffusion. Salivary IgA levels were determined in 33 lead workers and 40 reference subjects. On the average the lead workers had lower serum complement C3 and immunoglobulin levels, as well as lower salivary IgA levels, than the reference subjects. A significant negative correlation was found between blood lead concentrations (PbB) and the serum levels of complement C3 and IgG in the group of lead workers, as well as in the total population examined. However, a significant positive correlation was observed between PbB and serum IgA in the group of lead workers. The results obtained in this study are discussed in relation to numerous reports in the literature showing that lead exerts adverse effects on the immune system in animals.

    The PICO-2L C3F8 bubble chamber search forWeakly Interacting Massive Particle (WIMP) dark matter was operated in the SNOLAB underground laboratory at the same location as the previous CF3I lled COUPP-4kg detector. Neutron calibrations using photoneutron sources in C3F8 and CF3I lled calibration bubble chambers were performed to verify the sensitivity of these target uids to dark matter scattering. This data was combined with similar measurements using a low-energy neutron beam at the University of Montreal and in situ calibrations of the PICO-2L and COUPP-4kg detectors. C3F8 provides much greater sensitivity to WIMP-proton scattering than CF3I in bubble chamber detectors. PICO-2L searched for dark matter recoils with energy thresholds below 10 keV. Radiopurity assays of detector materials were performed and the expected neutron recoil background was evaluated to be 1.6+0:3

    Cobra venom factor (CVF) is a functional analog of human complement component C3b, the active fragment of C3. Similar to C3b, in human and mammalian serum, CVF binds factor B, which is then cleaved by factor D, giving rise to the CVFBb complex that targets the same scissile bond in C3 as the authentic complement convertases C4bC2a and C3bBb. Unlike the latter, CVFBb is a stable complex and an efficient C5 convertase. We solved the crystal structure of CVF, isolated from Naja naja kouthia venom, at 2.6 {angstrom} resolution. The CVF crystal structure, an intermediate between C3b and C3c, lacks the TED domain and has the CUB domain in an identical position to that seen in C3b. The similarly positioned CUB and slightly displaced C345c domains of CVF could play a vital role in the formation of C3 convertases by providing important primary binding sites for factor B.

    The population of LPS molecules on Salmonella montevideo that bind C3 during alternative pathway activation in serum was studied. LPS molecules of Salmonella are composed of lipid A:core oligosaccharide (one copy per molecule), substituted by an O-polysaccharide (O-PS) side chain, which is a linear polymer of 0 to >60 O-antigen repeat units containing mannose. A mutant of S. montevideo called SL5222 that inserts galactose only into core oligosaccharide and mannose only into O-antigen sub-units was grown with (/sup 3/H)mannose and (/sup 14/C)galactose, so that LPS molecules bearing large numbers of O-antigen subunits have high /sup 3/H to /sup 14/C ratios, whereas molecules with few O-antigen subunits have lower /sup 3/H to /sup 14/C ratios. The authors postulate that preferential attachment of C3 to long-chain LPS in SL5222 results because long-chain LPS molecules sterically hinder shorter chain LPS molecules from macromolecules. This study provides direct proof that the O-PS of LPS sterically hinders access of large molecules to the outer membrane and indicates that the LPS coat of these bacteria functions as a barrier against large protein molecules.

  9. Dark matter search results from the PICO-2L C$_3$F$_8$ bubble chamber

    New data are reported from the operation of a 2 liter C3F8 bubble chamber in the SNOLAB underground laboratory, with a total exposure of 211.5 kg days at four different energy thresholds below 10 keV. These data show that C3F8 provides excellent electron-recoil and alpha rejection capabilities at very low thresholds. The chamber exhibits an electron-recoil sensitivity of < 3.5 × 10–10 and an alpha rejection factor of > 98.2%. These data also include the first observation of a dependence of acoustic signal on alpha energy. Twelve single nuclear recoil event candidates were observed during the run. The candidate eventsmore » exhibit timing characteristics that are not consistent with the hypothesis of a uniform time distribution, and no evidence for a dark matter signal is claimed. Lastly, these data provide the most sensitive direct detection constraints on WIMP-proton spin-dependent scattering to date, with significant sensitivity at low WIMP masses for spin-independent WIMP-nucleon scattering.« less

  10. Structural features of the human C3 gene: Intron/exon organization, transcriptional start site, and promoter region sequence

    The third component of human complement (C3) is a key molecule in the activation of the complement cascade. C3 cDNA fragments were used to identify seven cosmid clones that covered all but 1 kilobase pair (kb) of the C3 gene. The remainder of the gene was cloned by using the polymerase chain reaction. These clones were used to identify the interon/exon boundaries and to map the gene. The C3 gene is 42 kb in length and comprises 41 exons ranging in size from 52 to 213 base pairs (bp). The transcription start site was identified by primer extension, and approximately 1 kb of DNA upstream of this site was sequenced. Putative TATA and CAAT boxes were identified along with a number of regions that shared homology with known regulatory sequences. These include responsive elements for interferon-{gamma}, interleukin-6, nuclear factor kB, estrogen, glucocorticoids and thyroid hormone. Several of these agents have been shown to affect C3 synthesis and mRNA levels. The sizes of the exons in C3 were compared to those of C4 and {alpha}{sub 2}-macroglobulin ({alpha}{sub 2}M). Thirty-nine of 41 exons in C4 were found to be of similar size to the analogous ones in C3, and two-thirds of those in {alpha}{sub 2}M were also similarly sized, supporting the hypothesis that these genes arose from a common ancestor.

  19. The Role of the Tetraheme Cytochrome c3 in Desulfovibrio vulgaris Hildenborough Metabolism

    The role of tetraheme cytochrome c3 (CycA) in the metabolism of the sulfate-reducing bacterium Desulfovibrio vulgaris Hildenborough (DvH) was investigated by deletion of the cycA gene using a marker-exchange deletion strategy. A highly abundant periplasmic cytochrome, CycA has the important function of transferring electrons from periplasmic hydrogenases (Hyd, Hyn, Hys) to transmembrane complexes which transport the electrons to the cytoplasm where sulfate is reduced. Previous studies have indicated that during its interaction with periplasmic hydrogenases, CycA is also involved in the reduction of toxic metals. Growth of the cycA mutant strain on lactate as the electron donor and sulfate as the terminal electron acceptor showed that, despite its abundance, CycA is not essential for DvH growth. However, the rate of growth of the mutant strain was significantly lower, and the extent of growth less, than rates and extents of growth of the wild type and complement strains on lactate/sulfate medium. This indicates that a portion of the electrons generated from cytoplasmic lactate oxidation are transported by CycA for energy production, possibly in a hydrogen cycling mechanism employed to generate ATP. Failure of the mutant strain to grow on either formate or H2, with sulfate or sulfite as electron acceptors, further indicated that CycA may be the only redox partner of periplasmic hydrogenases. The cycA mutant strain also did not grow as well as either the wild type or complement strains on medium supplemented with pyruvate/sulfate. Final growth on pyruvate/sulfate was comparable, but the mutant grew more slowly than the wild type and complement strains. Interestingly, the mutant grew better than the wild type or complement strains on pyruvate alone, possibly due to the release of H2 and/or CO2 in concentrations which may be somewhat inhibitory to wild type growth.

    The authors recently showed that culture-derived metacyclic trypomastigotes (CMT), but not epimastigotes (Epi), of the Miranda 99 strain of Trypanosoma cruzi evade lysis by the human alternative complement pathway because of inefficient binding of factor B to complement component C3b on the parasite surface. These results suggested that CMT and tissue-culture-derived trypomastigotes (TCT), which also activate the alternative pathway poorly, might produce a molecule capable of interfering with factor B binding to C3b. They now demonstrate that CMT and TCT lysates, as well as molecules spontaneously shed from CMT and TCT but not Epi, accelerate decay of /sup 125/I-labeled factor Bb from the alternative-pathway C3 convertase (C3bBb) assembled on zymosan or Epi and also accelerate decay of the classical-pathway C3 convertase (C4b2a) on sheep erythrocytes. Parasites metabolically labeled with (/sup 35/S)methionine spontaneously shed a limited number of radioactive components, ranging in molecular mass from 86 to 155 kDa for trypomastigotes and 25 to 80 kDa for Epi. Decay-accelerating activity within supernatants is inactivated by papain and is coeluted with /sup 35/S-containing polypeptides on FPLC anion-exchange chromatography, suggesting that the active constituents are protein molecules. Molecules with decay-accelerating activity may explain the developmentally regulated resistance to complement-mediated lysis in infective and vertebrate stages for T. cruzi life cycle.

    The MBPT (2) procedure with the 6-31g (asterisk) basis set was used to study nearly linear carbon chains. The theoretical vibrational frequencies of the molecules C3 through C9 are presented and, for C3 through C6, compared to experimental stretching frequencies and their (C-13)/(C-12) isotopomers. Predictions for C7, C8, and C9 stretching frequencies are calculated by directly scaling the theoretical frequencies with factors derived from experimental-to-theoretical ratios known for the smaller molecules. 28 refs.

  3. Structural homologies of component C5 of human complement with components C3 and C4 by neutron scattering

    The complement component C5 is one of a family of structurally related plasma proteins that includes components C3 and C4. Activation of C5 is the initial step in the formation of the membrane attack complex of complement. Analysis of the solution structure of C5 and comparisons with similar analyses of the structures of C3 and C4 are reported here. Neutron solution scattering gave an Mr for C5 of 201,000, which demonstrates that C5 is monomeric in solution. The radius of gyration RG of C5 at infinite contrast is 4.87 nm and corresponds to an elongated structure. The longest length of C5 was determined to be at least 15-16 nm from three calculations on the basis of the RG, the scattering intensity at zero angle I(0), and the indirect transformation of the scattering curve into real space. Comparison of the RG and contrast variation data and indirect transformations of the scattering curves for C3, C4, and C5 show that these have very similar structures. Comparisons of the C5 scattering curve with Debye small-sphere models previously employed for C4 and C3 show that good curve fits could be obtained. Unlike previous studies that have suggested significant differences, these experiments indicate that, while C5 differs from C3 and C4 in its activation and inactivation pathways, significant structural homology exists between the native proteins, as might be predicted from their high (and similar) sequence homology.

    Combined hyperlipidemia (CHL) is a common disorder of lipidmetabolism that leads to an increased risk of cardiovascular disease. Thelipid profile of CHL is characterised by high levels of atherogeniclipoproteins and low levels of high-density-lipoprotein-cholesterol.Apolipoprotein (APO) A5 is a newly discovered gene involved in lipidmetabolism located within 30kbp of the APOA1/C3/A4 gene cluster. Previousstudies have indicated that sequence variants in this cluster areassociated with increased plasma lipid levels. To establish whethervariation at the APOA5 gene contributes to the transmission of CHL, weperformed linkage and linkage disequilibrium (LD) tests on a large cohortof families (n=128) with familial CHL (FCHL). The linkage data producedevidence for linkage of the APOA1/C3/A4/A5 genomic interval to FCHL (NPL= 1.7, P = 0.042). The LD studies substantiated these data. Twoindependent rare alleles, APOA5c.56G and APOC3c.386G of this gene clusterwere over-transmitted in FCHL (P = 0.004 and 0.007, respectively), andthis was associated with a reduced transmission of the most commonAPOA1/C3/A4/A5 haplotype (frequency 0.4425) to affected subjects (P =0.013). The APOA5c.56G allele was associated with increased plasmatriglyceride levels in FCHL probands, whereas the second, andindependent, APOC3c.386G allele was associated with increased plasmatriglyceride levels in FCHL pedigree founders. Thus, this allele (or anallele in LD) may mark a quantitative trait associated with FCHL, as wellas representing a disease susceptibility locus for the condition. Thisstudy establishes that sequence variation in the APOA1/C3/A4/A5 genecluster contributes to the transmission of FCHL in a substantialproportion of affected families, and that these sequence variants mayalso contribute to the lipid abnormalities of the metabolic syndrome,which is present in up to 40 percent of persons with cardiovasculardisease.

    Serum levels of cardiac troponins serve as biomarkers of myocardial injury. However, troponins are released into the serum only after damage to cardiac tissue has occurred. Here, we report development of a mouse model of doxorubicin (DOX)-induced chronic cardiotoxicity to aid in the identification of predictive biomarkers of early events of cardiac tissue injury. Male B6C3F{sub 1} mice were administered intravenous DOX at 3 mg/kg body weight, or an equivalent volume of saline, once a week for 4, 6, 8, 10, 12, and 14 weeks, resulting in cumulative DOX doses of 12, 18, 24, 30, 36, and 42 mg/kg, respectively. Mice were sacrificed a week following the last dose. A significant reduction in body weight gain was observed in mice following exposure to a weekly DOX dose for 1 week and longer compared to saline-treated controls. DOX treatment also resulted in declines in red blood cell count, hemoglobin level, and hematocrit compared to saline-treated controls after the 2nd weekly dose until the 8th and 9th doses, followed by a modest recovery. All DOX-treated mice had significant elevations in cardiac troponin T concentrations in plasma compared to saline-treated controls, indicating cardiac tissue injury. Also, a dose-related increase in the severity of cardiac lesions was seen in mice exposed to 24 mg/kg DOX and higher cumulative doses. Mice treated with cumulative DOX doses of 30 mg/kg and higher showed a significant decline in heart rate, suggesting drug-induced cardiac dysfunction. Altogether, these findings demonstrate the development of DOX-induced chronic cardiotoxicity in B6C3F{sub 1} mice. -- Highlights: ? 24 mg/kg was a cumulative cardiotoxic dose of doxorubicin in male B6C3F{sub 1} mice. ? Doxorubicin-induced hematological toxicity was in association with splenomegaly. ? Doxorubicin induced severe testicular toxicity in B6C3F{sub 1} male mice.

    SAR development of indole-based palm site inhibitors of HCV NS5B polymerase exemplified by initial indole lead 1 (NS5B IC{sub 50} = 0.9 {micro}M, replicon EC{sub 50} > 100 {micro}M) is described. Structure-based drug design led to the incorporation of novel heterocyclic moieties at the indole C3-position which formed a bidentate interaction with the protein backbone. SAR development resulted in leads 7q (NS5B IC{sub 50} = 0.032 {micro}M, replicon EC{sub 50} = 1.4 {micro}M) and 7r (NS5B IC{sub 50} = 0.017 {micro}M, replicon EC{sub 50} = 0.3 {micro}M) with improved enzyme and replicon activity.

    Mn + 1AXn (MAX) phase materials based on Ti–Al–C have been irradiated at 400 °C (673 K) with fission neutrons to a fluence of 2 × 1025 n/m2 (E > 0.1 MeV), corresponding to ~ 2 displacements per atom (dpa). We report preliminary results of microcracking in the Al-containing MAX phase, which contained the phases Ti3AlC2 and Ti5Al2C3. Equibiaxial ring-on-ring tests of irradiated coupons showed that samples retained 10% of pre-irradiated strength. Volumetric swelling of up to 4% was observed. Phase analysis and microscopy suggest that anisotropic lattice parameter swelling caused microcracking. Lastly, variants of titanium aluminum carbide may bemore » unsuitable materials for irradiation at light water reactor-relevant temperatures.« less

  12. The structure of C2b, a fragment of complement component C2 produced during C3 convertase formation

    The crystal structure of C2b has been determined at 1.8 Å resolution, which reveals the arrangement of its three complement control protein (CCP) modules. A model for complement component C2 is presented and its conformational changes during the C3-convertase formation are also discussed. The second component of complement (C2) is a multi-domain serine protease that provides catalytic activity for the C3 and C5 convertases of the classical and lectin pathways of human complement. The formation of these convertases requires the Mg{sup 2+}-dependent binding of C2 to C4b and the subsequent cleavage of C2 by C1s or MASP2, respectively. The crystal structure of full-length C2 is not yet available, although the structure of its C-terminal catalytic segment C2a has been determined. The crystal structure of the N-terminal segment C2b of C2 determined to 1.8 Å resolution presented here reveals the arrangement of its three CCP domains. The domains are arranged differently compared with most other CCP-domain assemblies, but their arrangement is similar to that found in the Ba part of the full-length factor B structure. The crystal structures of C2a, C2b and full-length factor B are used to generate a model for C2 and a discussion of the domain association and possible interactions with C4b during formation of the C4b–C2 complex is presented. The results of this study also suggest that upon cleavage by C1s, C2a domains undergo conformational rotation while bound to C4b and the released C2b domains may remain folded together similar to as observed in the intact protein.

    Antibody-sensitized sheep erythrocytes were used as a model to determine the effects of therapeutic immune serum globulin (ISG) preparations on the ability of this particulate activator to fix C3 and initiate hemolysis. Both standard and intravenous forms of ISG inhibit uptake of /sup 125/I-C3, presumably by competing for the deposition of ''nascent'' C3b molecules onto the erythrocytes. Both forms of ISG also inhibit hemolytic activity of whole serum or purified complement components. The inhibition appears to be a specific property of IgG itself, since similar inhibition was not caused by equivalent concentrations of human serum albumin, and was not affected by the buffer in which the ISG was dissolved. Interference with C3 uptake onto antibody-sensitized platelets and/or inhibition of hemolytic complement activity could contribute to the efficacy of high dose intravenous ISG in idiopathic thrombocytopenic purpura.

  14. Regulation of the activity of platelet-bound C3 convertase of the alternative pathway of complement by platelet factor H

    The alternative pathway of complement is regulated on the surface of homologous blood cells at the C3 amplification step by the membrane protein decay-accelerating factor, as well as by the plasma protein factor H. The authors have reported elsewhere that platelets from patients with paroxysmal nocturnal hemoglobinuria regulate the activity of the C3 convertase C3bBb, even though they lack decay-accelerating factor. They now report that normal human platelets contain factor H, which was released from the platelet in response to complement deposition or thrombin stimulation. Factor H was localized to the platelet ..cap alpha.. granules by immunocytochemical techniques. As determined by a solid-phase radioimmunoassay, thrombin-stimulated platelets released approx. 54 ng of factor H per 10/sup 8/ platelets. The release of factor H in response to complement or thrombin was inhibited by treating the platelets with metabolic inhibitors. Such inhibition resulted in a 3-fold increase in the activity of C3bBb. Platelets that released factor H bound only half as many molecules of radiolabeled factor B to platelet-bound C3b than platelets that could not release factor H. Treatment of platelets with anti-decay-accelerating factor antibody had no effect on the activity of C3bBb unless the release of factor H was blocked. Therefore, human platelets have a unique mechanism for the regulation of the alternative pathway of complement.

    The Institute for Defense Analyses (IDA) has collected and analyzed information on network technology that is relevant to Battle Management Command, Control, and Communications (BM/C3). This memorandum report represents a program plan that will provide the SDIO BM C3 directorate with administrative and technical insight into network technology. This program plan focuses on C3 network concepts and provides information and analysis to the SDIO to be used in formulating budget requirements for FY 1988 and beyond. Based upon analysis of network requirements and ongoing programs, recommendations have been made for research areas that should be funded, including both the continuation of current work and the initiation of new tasks. While emphasis here is on the SDIO-funded tasks, other relevant government-sponsored and commercial efforts, such as packet radio networks, also have been examined.

  7. Biochemical bases for the biogeography of C3/C4 grasses and implications for changing distributions since the last glacial maximum

    Differential distributions of C3 and C4 grass taxa correlate with geographic and climatic factors. A simple model based on the temperature dependence of the photosynthetic quantum yield of C3 plants and the lack of response of the C4 quantum yield to temperature is used to predict the global distribution of C4 grasses at current atmospheric CO2 concentrations and climate. The model predicts a cross over temperature at which the quantum yield responses intersect; at temperatures above the cross over point C4 grasses are favored over C3. The cross over temperature is about 22{degrees}C at current atmospheric CO2 concentrations. Using this criterion an accurate 1x1 degree map of C4 grass dominance over C3 grasses is produced from climatological mean monthly temperatures. Accuracy is improved by considering the co-occurrence of sufficient rainfall for growth during the months warm enough for C4 dominance. Rising temperatures and CO2 concentrations since the last glacial maximum (LGM) are expected to have an impact on past C4 grass distributions. We have used climate generated by the NCAR CCM to predict the extent of climatic regions favoring C4 over C3 since the LGM. Though low temperatures favor C3 photosynthesis, the low CO2 concentrations in the past more than off-set this effect. The extent of C4 favorable climates are predicted to have been greater during the LGM and have shrunk since then. The model does not take into account important biotic factors such as competition for light and herbivory or abiotic factors such as fire frequency that can affect the dominance of grasslands over other vegetation types.

    The effect that ionizing radiation has in vitro on Fc and C3 receptors was evaluated at various doses and measured by means of erythrocytes coated with antibody (EA) and erythrocytes coated with antibody and complement (EAC) rosettes on human peripheral blood leukocytes (PBL) and on mouse bone marrow cells (BMC) and PBL. We found that the number of cells with either EA and EAC rosettes decreased as the radiation doses increased, and that they were almost absent when the highest doses were employed. We obtained evidence that albumin is a natural source of radio-protection for Fc and C3 receptors, and we showed that by increasing the amount of this molecule we could completely protect receptors for EA and EAC in vitro. Finally, the possible therapeutic value of the administration of human albumin to patients undergoing radiotherapy is discussed.

    The crystallization and results of multiwavelength anomalous diffraction studies of a recombinant C3-inhibitory fragment of Efb from S. aureus are reported. The extracellular fibrinogen-binding protein (Efb) of Staphylococcus aureus is a multifunctional virulence factor capable of potent inhibition of complement component-3 (C3) activity in addition to its previously described fibrinogen-binding properties. A truncated recombinant form of Efb (Efb-C) that binds C3 has been overexpressed and purified and has been crystallized using the hanging-drop vapor-diffusion technique. Crystals of native Efb-C grew in the tetragonal space group P4{sub 3} (unit-cell parameters a = b = 59.53, c = 46.63 Å) with two molecules in the asymmetric unit and diffracted well beyond 1.25 Å limiting Bragg spacing. To facilitate de novo phasing of the Efb-C crystals, two independent site-directed mutants were engineered in which either residue Ile112 or Val140 was replaced with methionine and crystals isomorphous to those of native Efb-C were reproduced using a seleno-l-methionine-labeled form of each mutant protein. Multiwavelength anomalous diffraction (MAD) data were collected on both mutants and analyzed for their phasing power toward solution and refinement of a high-resolution Efb-C crystal structure.

    In this study, two variants of Methylobacterium extorquens AM1 demonstrated a trade-off between growth rate and biomass yield. In addition, growth rate and biomass yield were also affected by supplementation of growth medium with different amounts of cobalt. The metabolism changes relating to these growth phenomena as well as the trade-off were investigated in this study. 13C metabolic flux analysis was used to generate a detailed central carbon metabolic flux map with both absolute and normalized flux values. As a result, the major differences between the two variants occurred at the formate node as well as within C3-C4 inter-conversion pathways.more » Higher relative fluxes through formyltetrahydrofolate ligase, phosphoenolpyruvate carboxylase, and malic enzyme led to higher biomass yield, while higher relative fluxes through pyruvate kinase and pyruvate dehydrogenase led to higher growth rate. These results were then tested by phenotypic studies on three mutants (null pyk, null pck mutant and null dme mutant) in both variants, which agreed with the model prediction. In this study, 13C metabolic flux analysis for two strain variants of M. extorquens AM1 successfully identified metabolic pathways contributing to the trade-off between cell growth and biomass yield. Phenotypic analysis of mutants deficient in corresponding genes supported the conclusion that C3-C4 inter-conversion strategies were the major response to the trade-off.« less

  17. A CHRONIC INHALATION STUDY OF METHYL BROMIDE TOXICITY IN B6C3F1 MICE. (FINAL REPORT TO THE NATIONAL TOXICOLOGY PROGRAM)

    This report provides a detailed account of a two year chronic inhalation study of methyl bromide toxicity in B6C3Fl mice conducted for the National Toxicology Program. Mice were randomized into three dose groups (10, 33 and 100 ppm methyl bromide) and one control group (0 ppm) per sex and exposed 5 days/week, 6 hours/day, for a total of 103 weeks. Endpoints included body weight; clinical signs and mortality, and at 6, 15 and 24 months of exposure, animals were sacrificed for organ weights, hematology and histopathology. In addition, a subgroup of animals in each dosage group was monitored for neurobehavioral and neuropathological changes. After only 20 weeks of exposure, 48% of the males and 12% of the females in the 100 ppm group had died. Exposures were terminated in that group and the surviving mice were observed for the duration of the study. Exposure of B6C3Fl mice to methyl bromide, even for only 20 weeks, produced significant changes in growth rate, mortality, organ weights and neurobehavioral functioning. These changes occurred in both males and females, but were more pronounced in males.

    This IDA paper was prepared at the request of the Strategic Defense Initiative Organization. The paper documents findings of an evaluation on the capabilities of certain computer software/computer-aided software engineering (CASE) tools to provide computer-aided graphic design of Battle Management/C3 for the SDIO. Each tool (of the five selected on the basis of the best available at this time), was installed at IDA. After training by vendor tool staff, an IDA team, using a hands-on design experience determined the merits of the tools for SDI application. A comparative summary of the tools is given relative to envisaged SDI requirements and an extensive questionnaire is answered for each.

    A superposition of the monochromatic charts transmitted by Vega 2's three-channel spectrometer during its approach of Comet P/Halley in March, 1986, has yielded composite images of the inner coma with moderate spatial resolution which cover a sector of angle 50 deg converging to the nucleus. Images of the CN, C3, C2 Delta-v = 1 and C2 Delta-v = 0 emissions show evidence of two well-contrasted jets, one of which is in the direction of the sun while the other lies in the perpendicular direction; they are separated by a valley-shaped low-intensity zone. Radial and transverse profiles of the emissions are presented. 28 refs.

  20. The states of carbon and nitrogen atoms after photodissociation of CN, CH, CH(+), C2, C3, and CO in comets

    The photodissociation of carbon compounds by solar UV radiation at a heliocentric distance of 1 AU is examined, comparing published observational data with the predictions of theoretical models and results from laboratory experiments. It is shown that species other than CO, including CN, CH, CH(+), C2, and C3, can contribute to the observed brightness of the VUV lines of C I (156.1, 165.7, and 193.1 nm) and C II (133.5 nm) in comet comae. CN photodissociation is also found to produce metastable 2D0 and 2P0 N I atoms, possibly leading (at heliocentric distances less than 0.25 AU) to 143.9-nm emission via resonance fluorescence. 37 refs.

    We report the design, synthesis, biological evaluation, and the X-ray crystal structure of a novel inhibitor bound to the HIV-1 protease. Various C3-functionalized cyclopentanyltetrahydrofurans (Cp-THF) were designed to interact with the flap Gly48 carbonyl or amide NH in the S2-subsite of the HIV-1 protease. We investigated the potential of those functionalized ligands in combination with hydroxyethylsulfonamide isosteres. Inhibitor 26 containing a 3-(R)-hydroxyl group on the Cp-THF core displayed the most potent enzyme inhibitory and antiviral activity. Our studies revealed a preference for the 3-(R)-configuration over the corresponding 3-(S)-derivative. Inhibitor 26 exhibited potent activity against a panel of multidrug-resistant HIV-1 variants. A high resolution X-ray structure of 26-bound HIV-1 protease revealed important molecular insight into the ligand-binding site interactions.

    The Eva South Morrow Sand Unit is located in western Texas County, Oklahoma. The field produces from an upper Morrow sandstone, termed the Eva sandstone, deposited in a transgressive valley-fill sequence. The field is defined as a combination structural stratigraphic trap; the reservoir lies in a convex up -dip bend in the valley and is truncated on the west side by the Teepee Creek fault. Although the field has been a successful waterflood since 1993, reservoir heterogeneity and compartmentalization has impeded overall sweep efficiency. A 4.25 square mile high-resolution, three component three-dimensional (3C3D) seismic survey was acquired in order to improve reservoir characterization and pinpoint the optimal location of a new horizontal producing well, the ESU 13-H.

    This report documents a series of tests used to assess the proposed air sampling location in the Hanford Tank Waste Treatment and Immobilization Plant (WTP) Lab C3V (LB-S1) exhaust stack with respect to the applicable criteria regarding the placement of an air sampling probe. Federal regulations require that an air sampling probe be located in the exhaust stack in accordance with the criteria of American National Standards Institute/Health Physics Society (ANSI/HPS) N13.1-1999, Sampling and Monitoring Releases of Airborne Radioactive Substances from the Stack and Ducts of Nuclear Facilities. These criteria address the capability of the sampling probe to extract a sample that represents the effluent stream.

    Studies at the Inhalation Toxicology Research Institute (ITRI) have explored the species-specific uptake and metabolism of benzene. Results have shown that metabolism is dependent on both dose and route of administration. Of particular interest were shifts in the major metabolic pathways as a function of exposure concentration. In these studies, B6C3F{sub 1} mice were exposed to increasing levels of benzene by either gavage or inhalation. As benzene internal dose increased, the relative amounts of muconic acid and hydroquinone decreased. In contrast, the relative amount of catechol increased with increasing exposure. These results show that the relative levels of toxic metabolites are a function of exposure level. Based on these results and assuming a linear relationship between exposure concentration and levels of bone marrow metabolites, it would be difficult to detect an elevation of any phenolic metabolites above background after occupational exposures to the OSHA Permissible Exposure Limit of 1 ppm benzene.

It is a homodimeric protein that belongs to the S100 protein family. The receptor for advanced glycation end products (RAGE) has been shown to play a role in the progression of various disease conditions, such as diabetes and immune/inflammatory disorders. Information regarding the association of RAGE with S100 proteins at a molecular level is useful to understand the diversity of the RAGE signaling pathways. In this report, biomolecular NMR techniques were utilized for the resonance assignment of the C3S mutation in human S100A6 and characterizing its interaction with the RAGE V domain. Further binding affinity between S100A6m and the RAGE V domain was determined by isothermal titration calorimetric studies. HADDOCK was used to generate a heterotetramer model of the S100A6m–RAGE V domain complex. This model provides an important insights into the S100–RAGE cellular signaling pathway.

  6. Murine natural resistance to Trypanosoma lewisi involves complement component C3 and radiation-resistant, silica dust-sensitive effector cells

    The phenomenon of natural (innate) resistance is exemplified by the solid resistance of mice to infection with the rat-specific Trypanosoma lewisi. We provide more information about the mechanism of resistance in this model system. Resistance was not diminished in aged mice or reduced by ionizing radiation or splenectomy. There was no difference in resistance of C5-deficient mice (B10.D2/oSn and AKR) compared with their normocomplementemic counterparts (B10.D2/nSn and C57BL/6). Treatment of mice with cobra venom factor resulted in greatly prolonged survival of T. lewisi in mice. Combined treatment of mice with cobra venom factor and silica dust resulted in not only prolonged survival but also considerable intravascular growth of the parasite and resultant death of the mice. T. lewisi cells from irradiated donor rats, or after surface coat removal with trypsin, were eliminated by inoculated mice more efficiently than were parasites acquired from normal rats. Incubation of trypanosomes obtained from irradiated rats in normal rat serum restored their resistance to rapid elimination. From the results of these studies and other recent investigations, we have concluded that murine resistance to T. lewisi involves activation and binding of C3b by uncoated trypanosomes and concomitant participation of radiation-resistant, silica dust-sensitive effector cells (probably macrophages and neutrophils), and rat plasma proteins, integrated into the trypanosome surface coat, stabilize the coat, make it less susceptible to removal when the trypanosome is present in the mouse bloodstream, and thus delay exposure of underlying molecules (receptors) that activate murine C3.

  7. Haplotypes in the APOA1-C3-A4-A5 gene cluster affect plasma lipids in both humans and baboons

    Genetic studies in non-human primates serve as a potential strategy for identifying genomic intervals where polymorphisms impact upon human disease-related phenotypes. It remains unclear, however, whether independently arising polymorphisms in orthologous regions of non-human primates leads to similar variation in a quantitative trait found in both species. To explore this paradigm, we studied a baboon apolipoprotein gene cluster (APOA1/C3/A4/A5) for which the human gene orthologs have well established roles in influencing plasma HDL-cholesterol and triglyceride concentrations. Our extensive polymorphism analysis of this 68 kb gene cluster in 96 pedigreed baboons identified several haplotype blocks each with limited diversity, consistent with haplotype findings in humans. To determine whether baboons, like humans, also have particular haplotypes associated with lipid phenotypes, we genotyped 634 well characterized baboons using 16 haplotype tagging SNPs. Genetic analysis of single SNPs, as well as haplotypes, revealed an association of APOA5 and APOC3 variants with HDL cholesterol and triglyceride concentrations, respectively. Thus, independent variation in orthologous genomic intervals does associate with similar quantitative lipid traits in both species, supporting the possibility of uncovering human QTL genes in a highly controlled non-human primate model.

  8. Spectroscopic Evidence for a High-Spin Br-Fe(IV)-Oxo Intermediate in the alpha-Ketoglutarate-Dependent Halogenase CytC3 from Streptomyces

    The complex of the mononuclear non-heme halogenase CytC3 from Streptomyces, Fe(II), {alpha}-ketoglutarate, bromide, and the substrate l-2-aminobutyryl-S-CytC2 reacts with O2 to form a reaction intermediate. Variable-field, freeze-quench Mossbauer spectroscopy reveals this intermediate to be a mixture of two high-spin Fe(IV) complexes in an approximate 3.7/1 ratio. Freeze-quench Fe K-edge X-ray absorption spectroscopy provides further insight into the structure of this intermediate. A short 1.62-Angstroms interaction between the Fe and one of its ligands is attributed to the Fe(IV)-oxo group, and a 2.43-Angstroms interaction is assigned to the Fe-Br interaction. A significantly longer Fe-Br separation (2.53 Angstroms) is observed in the reactant complex, consistent with lower valency of the Fe in the reactant complex. This intermediate is the first example for a Br-Fe(IV)-oxo complex in a protein and provides evidence for a unifying mechanism for Fe(II) and {alpha}-ketoglutarate-dependent dioxygenases and halogenases.

  9. Ectopic bone formation and chondrodysplasia in transgenic mice carrying the rat C3(1)/T{sub AG} fusion gene

    Transgenic mice expressing the SV40 large T-antigen (T{sup AG}) under the regultory control of the hormone-responsive rat C3(1) prostatein promoter develop unusual bone and cartilage lesions, as well as ectopic bone and cartilage formation. Two lines of transgenic animals have been propagated in which the expression of the transgene in chondrocytes results in a mild to moderate generalized disorganization of cartilage growth which appears to affect multiple tissues, including the trachea, ear pinna and articular cartilage. The epiphyseal plates are also affected with normal architecture of the zones of proliferation and maturation, but marked elongation of the zone of hypertrophy. Immunocytochemistry demonstrates that expression of T{sup AG} is limited to the zone of hypertropny in the epiphyseal plates, suggesting that the chondrocytes become hormone-responsive at this particular stage of differentiation. Normal mineralization and trabecular formation in long bone appears to occur. Ectopic bone and cartilage formation occurs in the foot pads of the fore- and hind- feet over the course of several months. This is preceded by proliferation of sweat gland epithelial cells followed by the appearance of nodules of cartilage and bone. The nodules are closely associated with proliferating epithelium but are not contiguous with bony structures normally found in the feet. The roles of BMP`s, growth factors, oncogenes and hormones in the development of these lesions will be presented. These transgenic animals may provide new insights into hormone-responsiveness of chondrocytes, as well as factors involved in the processes of bone and cartilage differentiation and growth. These transgenic animals may serve as a useful model for human heterotopic bone formation.

    Mono specific stands of the C3 sedge, Scirpus olneyi, and the C4 grass, Spartina patens, were exposed to normal ambient or elevated CO2 (normal ambient + 340 ul/l) in open top chambers throughout two growing seasons. Photosynthesis in the C3 sedge measured in elevated CO2 or low O2 (air with 1% O2) was greater at all light levels than photosynthesis measured in normal ambient CO2. Plants grown in ambient or ambient + 340 ul/l CO2 showed an identical photosynthetic response when tested at elevated CO2 with no evidence of acclimation to elevated CO2. No differences in the photosynthetic response to light were noted for the C4 species at any of the gas mixtures tested. However, nighttime dark respiration rates decreased in both the C3 and C4 species when grown in elevated CO2.

    In this study, lignocellulosic materials provide an attractive replacement for food-based crops used to produce ethanol. Understanding the interactions within the cell wall is vital to overcome the highly recalcitrant nature of biomass. One factor imparting plant cell wall recalcitrance is lignin, which can be manipulated by making changes in the lignin biosynthetic pathway. In this study, eucalyptus down-regulated in expression of cinnamate 4-hydroxylase (C4H, EC 1.14.13.11) or p-coumaroyl quinate/shikimate 3'-hydroxylase (C3'H, EC 1.14.13.36) were evaluated for cell wall composition and reduced recalcitrance.

    A new, very sensitive search for interstellar triatomic carbon has resulted in upper limits for a few diffuse clouds of order 10 to the 10th/sq cm, or about 10 to the -11th with respect to hydrogen. These limits are consistent with recent cold diffuse cloud chemistry models, but may be in conflict with shocked cloud models such as those invoked to explain CH(+) abundances. The results mmay also argue against linear carbon-chain molecules as carriers of the diffuse interstellar bands. 30 references.

    Simultaneous lactate production and extraction have been previously demonstrated in the myocardium in patients with coronary artery disease. To quantitate this lactate production and determine its source, dual carbon-labeled isotope experiments were performed. L-(1,2,3-/sup 13/C3) lactate and D-(6-/sup 14/C) glucose were infused in 10 patients with significant coronary artery disease. Metabolic samples were obtained at rest and during atrial pacing. Despite net chemical myocardial lactate extraction in the 10 patients at rest and no evidence of clinical ischemia, the L-(1,2,3-/sup 13/C3) lactate analysis demonstrated that lactate was being released by the myocardium. During atrial pacing, seven patients did not develop clinical symptoms of ischemia, and the chemical lactate analysis showed net lactate extraction. However, tracer analysis demonstrated that there was a significant increase in the lactate released during atrial pacing. In these seven patients, circulating glucose was the source of 23 of the lactate released at rest, and there was no significant change during pacing. The remaining three patients had mild chest pain and net chemical lactate production during pacing. In these patients, the contribution of glucose to lactate production increased significantly during pacing-induced clinical ischemia from 25 to 67%. Thus, dual carbon-labeled isotopic experiments are powerful tools for investigating myocardial metabolic pathways.

    Plants containing thujone have widespread use and hence have significant human exposure. α-Thujone caused seizures in rodents following gavage administration. We investigated the toxicokinetics of α-thujone in male and female F344/N rats and B6C3F1 mice following intravenous and gavage administration of α-thujone or a mixture of α- and β-thujone (which will be referred to as α,β-thujone). Absorption of α-thujone following gavage administration was rapid without any dose-, species-, sex- or test article-related effect. Absolute bioavailability of α-thujone following administration of α-thujone or α,β-thujone was generally higher in rats than in mice. In rats, females had higher bioavailability than males following administration of either test article although a sex difference was not observed in mice. C{sub max} and AUC{sub ∞} increased greater than proportional to the dose in female rats following administration of α-thujone and in male and female mice following administration of α,β-thujone suggesting possible saturation of elimination kinetics with increasing dose. Dose-adjusted AUC{sub ∞} for male and female rats was 5- to 15-fold and 3- to 24-fold higher than mice counterparts following administration of α-thujone and α,β-thujone, respectively (p-value < 0.0001 for all comparisons). Following both intravenous and gavage administration, α-thujone was distributed to the brains of rats and mice with females, in general, having higher brain:plasma ratios than males. These data are in support of the observed toxicity of α-thujone and α,β-thujone where females were more sensitive than males of both species to α-thujone-induced neurotoxicity.
