    Human cell lines are often used for in vitro biotransformation and transport studies of drugs. In vivo, genetic polymorphisms have been identified in drug-metabolizing enzymes and ABC-drug transporters leading to altered enzyme activity, or a change in the inducibility of these enzymes. These genetic polymorphisms could also influence the outcome of studies using human cell lines. Therefore, the aim of our study was to pharmacogenotype four cell lines frequently used in drug metabolism and transport studies, HepG2, IGROV-1, CaCo-2 and LS180, for genetic polymorphisms in biotransformation enzymes and drug transporters. The results indicate that, despite the presence of some genetic polymorphisms, no real effects influencing the activity of metabolizing enzymes or drug transporters in the investigated cell lines are expected. However, this characterization will be an aid in the interpretation of the results of biotransformation and transport studies using these in vitro cell models.

    There are many examples of groups of proteins that have similar function, but the determinants of functional specificity may be hidden by lack of sequencesimilarity, or by large groups of similar sequences with different functions. Transporters are one such protein group in that the general function, transport, can be easily inferred from the sequence, but the substrate specificity can be impossible to predict from sequence with current methods. In this paper we describe a linguistic-based approach to identify functional patterns from groups of unaligned protein sequences and its application to predict multi-drug resistance transporters (MDRs) from bacteria. We first show that our method can recreate known patterns from PROSITE for several motifs from unaligned sequences. We then show that the method, MDRpred, can predict MDRs with greater accuracy and positive predictive value than a collection of currently available family-based models from the Pfam database. Finally, we apply MDRpred to a large collection of protein sequences from an environmental microbiome study to make novel predictions about drug resistance in a potential environmental reservoir.

Even highly experienced diplomats are often advised to attend training sessions on this topic. Intercultural preparedness training promises to be especially relevant and useful for professionals working in the field of nuclear nonproliferation, including in the application of international nuclear safeguards. This paper outlines the fundamental philosophies underlying a training program that will benefit professionals in the nuclear arena, whether practitioners of nonproliferation or other sub-fields relying on international cooperation and collaboration, and how such a training program might be implemented efficiently.

    This report provides technical information on air sampling that will be useful for facilities following the recommendations in the NRC`s Regulatory Guide 8.25, Revision 1, ``Air sampling in the Workplace.`` That guide addresses air sampling to meet the requirements in NRC`s regulations on radiation protection, 10 CFR Part 20. This report describes how to determine the need for air sampling based on the amount of material in process modified by the type of material, release potential, and confinement of the material. The purposes of air sampling and how the purposes affect the types of air sampling provided are discussed. The report discusses how to locate air samplers to accurately determine the concentrations of airborne radioactive materials that workers will be exposed to. The need for and the methods of performing airflow pattern studies to improve the accuracy of air sampling results are included. The report presents and gives examples of several techniques that can be used to evaluate whether the airborne concentrations of material are representative of the air inhaled by workers. Methods to adjust derived air concentrations for particle size are described. Methods to calibrate for volume of air sampled and estimate the uncertainty in the volume of air sampled are described. Statistical tests for determining minimum detectable concentrations are presented. How to perform an annual evaluation of the adequacy of the air sampling is also discussed.

    Calorie restriction (CR) is one of the most effective anti-aging interventions in mammals. A modern theory suggests that aging results from a decline in detoxification capabilities and thus accumulation of damaged macromolecules. The present study aimed to determine how short-term CR alters mRNA profiles of genes that encode metabolism and detoxification machinery in the liver. Male C57BL/6 mice were fed CR (0, 15, 30, or 40%) diets for one month, followed by mRNA quantification of 98 xenobiotic processing genes (XPGs) in the liver, including 7 uptake transporters, 39 phase-I enzymes, 37 phase-II enzymes, 10 efflux transporters, and 5 transcription factors. In general, 15% CR did not alter mRNAs of most XPGs, whereas 30 and 40% CR altered over half of the XPGs (32 increased and 29 decreased). CR up-regulated some phase-I enzymes (fold increase), such as Cyp4a14 (12), Por (2.3), Nqo1 (1.4), Fmo2 (5.4), and Fmo3 (346), and numerous number of phase-II enzymes, such as Sult1a1 (1.2), Sult1d1 (2.0), Sult1e1 (33), Sult3a1 (2.2), Gsta4 (1.3), Gstm2 (1.3), Gstm3 (1.7), and Mgst3 (2.2). CR feminized the mRNA profiles of 32 XPGs in livers of male mice. For instance, CR decreased the male-predominantly expressed Oatp1a1 (97%) and increased the female-predominantly expressed Oatp1a4 (11). In conclusion, short-term CR alters the mRNA levels of over half of the 98 XPGs quantified in livers of male mice, and over half of these alterations appear to be due to feminization of the liver. - Highlights: Utilized a graded CR model in male mice The mRNA profiles of xenobiotic processing genes (XPGs) in liver were investigated. CR up-regulates many phase-II enzymes. CR tends to feminize the mRNA profiles of XPGs.

    account for individual traveler response to the dynamic transportation environment. In contrast, TRANSIMS provides disaggregated information that more explicitly represents the complex nature of humans interacting with the transportation system. It first generates a synthetic population that represents individuals and their households in the metropolitan region in a statistically valid way. The demographic makeup and spatial distribution of this synthetic population is derived from census data so that it matches that of the region’s real population. From survey data, a model is built of household and individual activities that may occur at home, in the workplace, school or shopping centers, for example. Trip plans including departure times, travel modes, and specific routes are created for each individual to get to his or her daily activities. TRANSIMS then simulates the movement of millions of individuals, following their trip plans throughout the transportation network, including their use of vehicles such as cars or buses, on a second-by-second basis. The virtual travel in TRANSIMS mimics the traveling and driving behavior of real people in the metropolitan region. The interactions of individual vehicles produce realistic traffic dynamics from which analysts can judge to performance of the transportation sysime and estimate vehicle emissions. Los Alamos, in cooperation with the Department of Transportation, Federal HIghway Administration and the local Metropolitan Planning Offices, has done TRANSIMS micro-simulations of auto traffic patterns in these two urban areas and completed associated scenario-based studies.« less

    Heavy ion beam transport through the containment chamber plays a crucial role in all heavy ion fusion (HIF) scenarios. Here, several parameters are used to characterize the operating space for HIF beams; transport modes are assessed in relation to evolving target/accelerator requirements; results of recent relevant experiments and simulations of HIF transport are summarized; and relevant instabilities are reviewed. All transport options still exist, including (1) vacuum ballistic transport, (2) neutralized ballistic transport, and (3) channel-like transport. Presently, the European HIF program favors vacuum ballistic transport, while the US HIF program favors neutralized ballistic transport with channel-like transport as an alternate approach. Further transport research is needed to clearly guide selection of the most attractive, integrated HIF system.

Alphabetically listed, each entry covers a specific chemical, standards and recommendations for exposure, its physical properties, and its toxic and hazard rating. A CAS, RTECS and DOT number identifies each substance; standards and recommendations cover its OSHA Permissible Exposure Limit, ACGIH Threshold Limit Value, MAKS, and DOT hazard classification. Physical properties include form, color, and odor. Here too is additional identifying information such as chemical formulas and well-known synonyms. The Toxic and Hazard Review summarizes hazards associated with the chemicals, revealing their: acute, immediate, chronic, and delayed effects; toxic or hazardous decomposition products; flammable and explosive properties; and incompatible materials and instabilities.     Pharmacokinetic drug interactions can lead to serious adverse events or decreased drug efficacy. The evaluation of a new molecular entity's (NME's) drug-drug interaction potential is an integral part of risk assessment during drug development and regulatory review. Alteration of activities of enzymes or transporters involved in the absorption, distribution, metabolism, or excretion of a new molecular entity by concomitant drugs may alter drug exposure, which can impact response (safety or efficacy). The recent Food and Drug Administration (FDA) draft drug interaction guidance ( (http://www.fda.gov/downloads/Drugs/GuidanceComplianceRegulatoryInformation/Guidances/ucm072101.pdf)) highlights the methodologies and criteria that may be used to guide drug interaction evaluation by industry and regulatory agencies and to construct informative labeling for health practitioner and patients. In addition, the Food and Drug Administration established a 'Drug Development and Drug Interactions' website to provide up-to-date information regarding evaluation of drug interactions ( (http://www.fda.gov/Drugs/DevelopmentApprovalProcess/DevelopmentResources/DrugInteractionsLabeling/ucm080499.htm)). This review summarizes key elements in the FDA drug interaction guidance and new scientific developments that can guide the evaluation of drug-drug interactions during the drug development process.

    WHC-CM-1-6 is the primary guidance for radiological control at Westinghouse Hanford Company (WHC). It was written to implement DOE N 5480.6 ``US Department of Energy Radiological Control Manual`` as it applies to programs at Hanford which are now overseen by WHC. As such, it complies with Title 10, Part 835 of the Code of Federal Regulations. In addition to WHC-CM-1-6, there is HSRCM-1, the ``Hanford Site Radiological Control Manual`` and several Department of Energy (DOE) Orders, national consensus standards, and reports that provide criteria, standards, and requirements for workplace air sampling programs. This document provides a summary of these, as they apply to WHC facility workplace air sampling programs. This document also provides an evaluation of the compliance of Tank Farms` workplace air sampling program to the criteria, standards, and requirements and documents compliance with the requirements where appropriate. Where necessary, it also indicates changes needed to bring specific locations into compliance.

    The purpose of this Technical Assessment is to satisfy HSRCM-1, ''Hanford Site Radiological Control Manual'' Article 551.4 for a documented study of facility Workplace Air Monitoring (WAM) programs. HSRCM-1 is the primary guidance for radiological control at Waste Management Federal Services of Hanford, Inc. (WMH). The HSRCM-1 complies with Title 10. Part 835 of the Code of Federal Regulations (10CFR835). This document provides an evaluation of the compliance of the Waste Receiving and Processing facility (WRAP) WAM program to the criteria standards, requirements, and documents compliance with the requirements where appropriate. Where necessary, it also indicates changes needed to bring specific locations into compliance.
