    A combined license (COL) application is required by the Nuclear Regulatory Commission (NRC) for all proposed nuclear plants. The information requirements for a COL application are set forth in 10 CFR 52.79, “Contents of Applications; Technical Information in Final Safety Analysis Report.” An applicant for a modular high temperature gas-cooled reactor (HTGR) must develop and submit for NRC review and approval a COL application which conforms to these requirements. The technical information necessary to allow NRC staff to evaluate a COL application and resolve all safety issues related to a proposed nuclear plant is detailed and comprehensive. To this, Regulatory Guide (RG) 1.206, “Combined License Applications for Nuclear Power Plants” (LWR Edition), was developed to assist light water reactor (LWR) applicants in incorporating and effectively formatting required information for COL application review (Ref. 1). However, the guidance prescribed in RG 1.206 presumes a LWR design proposal consistent with the systems and functions associated with large LWR power plants currently operating under NRC license.

    ETS, Inc., a pollution consulting firm with headquarters in Roanoke, Virginia, has developed a dry, limestone-based flue gas desulfurization (FGD) system. This SO{sub 2} removal system, called Limestone Emission Control (LEC), can be designed for installation on either new or existing coal-fired boilers. In the LEC process, the SO{sub 2} in the flue gas reacts with wetted granular limestone that is contained in a moving bed. A surface layer of principally calcium sulfate (CaSO{sub 4}) is formed on the limestone. Periodic removal of this surface layer by mechanical agitation allows high utilization of the limestone granules. The primary goal of the current study is the demonstration of the techno/economic capability of the LEC system as a post-combustion FGD process capable of use in both existing and future coal-fired boiler facilities burning high-sulfur coal. A nominal 5,000 acfm LEC pilot plant has been designed, fabricated and installed on the slipstream of a 70,000 pph stoker boiler providing steam to Ohio University`s Athens, Ohio campus. The pilot plant was normally operated on the slipstream of the Ohio Univ. boiler plant flue gas, but also had the capability of operating at higher inlet SO{sub 2} concentrations (typically equivalent to 3-1/2% sulfur coal) than those normally available from the flue gas slipstream. This was accomplished by injecting SO{sub 2} gas into the slipstream inlet. The pilot plant was instrumented to provide around-the-clock operation and was fully outfitted with temperature, SO{sub 2}, gas flow and pressure drop monitors.

    The research described in this product was performed in part in the Environmental Molecular Sciences Laboratory, a national scientific user facility sponsored by the Department of Energy's Office of Biological and Environmental Research and located at Pacific Northwest National Laboratory. The (TiO?)n clusters and their anions for n ) 1-4 have been studied with coupled cluster theory [CCSD(T)] and density functional theory (DFT). For n > 1, numerous conformations are located for both the neutral and anionic clusters, and their relative energies are calculated at both the DFT and CCSD(T) levels. The CCSD(T) energies are extrapolated to the complete basis set limit for the monomer and dimer and calculated up to the triple-? level for the trimer and tetramer. The adiabatic and vertical electron detachment energies of the anionic clusters to the ground and first excited states of the neutral clusters are calculated at both levels and compared with the experimental results. The comparison allows for the definitive assignment of the groundstate structures of the anionic clusters. Anions of the dimer and tetramer are found to have very closely lying conformations within 2 kcal/mol at the CCSD(T) level, whereas that of the trimer does not. In addition, accurate clustering energies and heats of formation are calculated for the neutral clusters and compared with the available experimental data. Estimates of the titanium-oxygen bond energies show that they are stronger than the group VIB transition metal-oxygen bonds except for tungsten. The atomization energies of these clusters display much stronger basis set dependence than the clustering energies. This allows the calculation of more accurate heats of formation for larger clusters on the basis of calculated clustering energies.

    The overall goal of the project was to define the cellular processes that underlie embryo development in plants at a mechanistic level. Our studies focused on a critical transcriptional regulator, Arabidopsis LEAFY COTYLEDON (LEC1), that is necessary and sufficient to induce processes required for embryo development. Because LEC1 regulates lipid accumulation during the maturation phase of embryo development, information about LEC1 may be useful in designing approaches to enhance biofuel production in plants. During the tenure of this project, we determined the molecular mechanisms by which LEC1 acts as a transcription factor in embryos. We also identified genes directly regulated by LEC1 and showed that many of these genes are involved in maturation processes. This information has been useful in dissecting the gene regulatory networks controlling embryo development. Finally, LEC1 is a novel isoform of a transcription factor that is conserved among eukaryotes, and LEC1 is active primarily in seeds. Therefore, we determined that the LEC1-type transcription factors first appeared in lycophytes during land plant evolution. Together, this study provides basic information that has implications for biofuel production.

    We extract the statistical uncertainties for the pion-nucleon ($\\pi N$) low energy constants (LECs) up to fourth order $\\mathcal{O}(Q^4)$ in the chiral expansion of the nuclear effective Lagrangian. The LECs are optimized with respect to experimental scattering data. For comparison, we also present an uncertainty quantification that is based solely on \\pin{} scattering phase shifts. Statistical errors on the LECs are critical in order to estimate the subsequent uncertainties in \\textit{ab initio} modeling of light and medium mass nuclei which exploit chiral effective field theory. As an example of the this, we present the first complete predictions with uncertainty quantification of peripheral phase shifts of elastic proton-neutron scattering.

    We present procedures based on Bayesian statistics for effective field theory (EFT) parameter estimation from data. The extraction of low-energy constants (LECs) is guided by theoretical expectations that supplement such information in a quantifiable way through the specification of Bayesian priors. A prior for natural-sized LECs reduces the possibility of overfitting, and leads to a consistent accounting of different sources of uncertainty. A set of diagnostic tools are developed that analyze the fit and ensure that the priors do not bias the EFT parameter estimation. The procedures are illustrated using representative model problems and the extraction of LECs for the nucleon mass expansion in SU(2) chiral perturbation theory from synthetic lattice data.

    The licensing framework established by the U.S. Nuclear Regulatory Commission under Title 10 of the Code of Federal Regulations (10 CFR) Part 52, “Licenses, Certifications, and Approvals for Nuclear Power Plants,” provides requirements for standard design certifications (DCs) and combined license (COL) applications. The intent of this process is the early reso- lution of safety issues at the DC application stage. Subsequent COL applications may incorporate a DC by reference. Thus, the COL review will not reconsider safety issues resolved during the DC process. However, a COL application that incorporates a DC by reference must demonstrate that relevant site-specific de- sign parameters are within the bounds postulated by the DC, and any departures from the DC need to be justified. This paper provides an overview of several seismic analysis issues encountered during a review of recent DC applications under the 10 CFR Part 52 process, in which the authors have participated as part of the safety review effort.

    An appraisal of the potential performance of different Low Energy Cooling (LEC) systems in nonresidential buildings in California is being conducted using computer simulation. The paper presents results from the first phase of the study, which addressed the systems that can be modeled, with the DOE-2.1E simulation program. The following LEC technologies were simulated as variants of a conventional variable-air-volume system with vapor compression cooling and mixing ventilation in the occupied spaces: Air-side indirect and indirect/direct evaporative pre-cooling. Cool beams. Displacement ventilation. Results are presented for four populous climates, represented by Oakland, Sacramento, Pasadena and San Diego. The greatest energy savings are obtained from a combination of displacement ventilation and air-side indirect/direct evaporative pre-cooling. Cool beam systems have the lowest peak demand but do not reduce energy consumption significantly because the reduction in fan energy is offse t by a reduction in air-side free cooling. Overall, the results indicate significant opportunities for LEC technologies to reduce energy consumption and demand in nonresidential new construction and retrofit.

    Diabetic nephropathy is the most common cause of chronic kidney disease. We investigated the ability of intracellular galectin-1 (Gal-1), a prototype of endogenous lectin, to prevent renal fibrosis by regulating cell signaling under a high glucose (HG) condition. We demonstrated that overexpression of Gal-1 reduces type I collagen (COL1) expression and transcription in human renal epithelial cells under HG conditions and transforming growth factor-{beta}1 (TGF-{beta}1) stimulation. Matrix metalloproteinase 1 (MMP1) is stimulated by Gal-1. HG conditions and TGF-{beta}1 treatment augment expression and nuclear translocation of Gal-1. In contrast, targeted inhibition of Gal-1 expression reduces COL1 expression and increases MMP1 expression. The Smad3 signaling pathway is inhibited, whereas two mitogen-activated protein kinase (MAPK) pathways, p38 and extracellular signal-regulated kinase (ERK), are activated by Gal-1, indicating that Gal-1 regulates these signaling pathways in COL1 production. Using specific inhibitors of Smad3, ERK, and p38 MAPK, we showed that ERK MAPK activated by Gal-1 plays an inhibitory role in COL1 transcription and that activation of the p38 MAPK pathway by Gal-1 plays a negative role in MMP1 production. Taken together, two MAPK pathways are stimulated by increasing levels of Gal-1 in the HG condition, leading to suppression of COL1 expression and increase of MMP1 expression.

    Cadmium-resistant cells were developed in our laboratory with rat lung epithelial cells (LECs) by stepwise exposure of LECs to cadmium chloride from 1 {mu}M to 20 {mu}M after 20 passages. To investigate the Cd-resistant phenotype in a long-term perspective, cadmium-resistant cells adapted to 20 {mu}M cadmium (Cd{sup R}) were then cultured in the absence of cadmium for various passages [Cd{sup R}(-n)]. All these adapted cells were significantly protected from cadmium toxicity as compared to parental cadmium-sensitive LECs (Cd{sup S}). The cadmium-resistant phenotype of adapted cells was relatively stable in the absence of cadmium for as long as 40 passages. Basal mRNA level of metallothionein-1 (MT-1) was dramatically higher in Cd{sup R} than in Cd{sup R}(-), which may account for the higher Cd-resistance of Cd{sup R} than Cd{sup R}(-). MT-1 mRNA level decreased drastically in Cd{sup R} after cadmium removal, suggesting that the high basal level of MT-1 in Cd{sup R} may be only partially responsible for cadmium-resistance. Treatment of cells with high levels of cadmium resulted in decreased phosphorylation of c-Jun N-terminal kinase (JNK1/2) in adapted cells than in sensitive cells and this cadmium-induced JNK activity was blocked by JNK inhibitor II, SP600125. Ro318220, a strong activator of JNK, reverted cadmium-sensitive phenotype in adapted cells. Taken together, our results suggest that during cadmium adaptation, cells develop tolerance to cell death, generally due to perturbation of the JNK signaling pathway and the nonresponsiveness of JNK phosphorylation is critical for the Cd-tolerance in these cells.

Project successes, lessons learned, and suggestions for improvement are discussed. Objectives of the North Anna COL project included preparation and submittal of a COLA to the USNRC incorporating ESBWR technology for a third unit a the North Anna Power Station site, support for the NRC review process and mandatory hearing, obtaining NRC approval of the COLA and issuance of a COL, and development of a business case necessary to support a decision on building a new nuclear power plant at the North Anna site.

    Behavioral avoidance of copper (Cu), cobalt (Co), and a Cu and Co mixture in soft water differed greatly between rainbow trout (Oncorhynchus mykiss) and chinook salmon (O. tshawytscha). Chinook salmon avoided at least 0.7 {micro}g Cu/L, 24 {micro}g Co/L, and the mixture of 1.0 {micro}g Cu/L and 0.9 {micro}g Co/L, whereas rainbow trout avoided at least 1.6 {micro}g Cu/L, 180 {micro}g Co/L, and the mixture of 2.6 {micro}g Cu/L and 2.4 {micro}g Co/L. Chinook salmon were also more sensitive to the toxic effects of Cu in that they failed to avoid {ge}44 {micro}g Cu/L, whereas rainbow trout failed to avoid {ge}180 {micro}g Cu/L. Furthermore, following acclimation to 2 {micro}g Cu/L, rainbow trout avoided 4 {micro}g Cu/L and preferred clean water, but chinook salmon failed to avoid any Cu concentrations and did not prefer clean water. The failure to avoid high concentrations of metals by both species suggests that the sensory mechanism responsible for avoidance responses was impaired. Exposure to Cu concentrations that were not avoided could result in lethality from prolonged Cu exposure or in impairment of sensory-dependent behaviors that are essential for survival and reproduction.
