    ETS, Inc., a pollution consulting firm with headquarters in Roanoke, Virginia, has developed a dry, limestone-based flue gas desulfurization (FGD) system. This SO{sub 2} removal system, called Limestone Emission Control (LEC), can be designed for installation on either new or existing coal-fired boilers. In the LEC process, the SO{sub 2} in the flue gas reacts with wetted granular limestone that is contained in a moving bed. A surface layer of principally calcium sulfate (CaSO{sub 4}) is formed on the limestone. Periodic removal of this surface layer by mechanical agitation allows high utilization of the limestone granules. A nominal 5,000 acfm LEC pilot plant has been designed, fabricated and installed on the slipstream of a 70,000 pph stoker boiler providing steam to Ohio University`s Athens, Ohio campus. A total of over 90 experimental trials have been performed using the pilot-scale moving-bed LEC dry scrubber as a part of this research project with run times ranging up to a high of 125 hours. SO{sub 2} removal efficiencies as high as 99.9% were achievable for all experimental conditions studied during which sufficient humidification was added to the LEC bed. The LEC process and conventional limestone scrubbing have been compared on an equatable basis using flue gas conditions that would be expected at the outlet of the electrostatic precipitator (ESP) of a 500 MW coal-fired power plant. The LEC was found to have a definite economic advantage in both direct capital costs and operating costs. Based on the success and findings of the present project, the next step in LEC process development will be a full-scale commercial demonstration unit.

    A combined license (COL) application is required by the Nuclear Regulatory Commission (NRC) for all proposed nuclear plants. The information requirements for a COL application are set forth in 10 CFR 52.79, Contents of Applications; Technical Information in Final Safety Analysis Report. An applicant for a modular high temperature gas-cooled reactor (HTGR) must develop and submit for NRC review and approval a COL application which conforms to these requirements. The technical information necessary to allow NRC staff to evaluate a COL application and resolve all safety issues related to a proposed nuclear plant is detailed and comprehensive. To this, Regulatory Guide (RG) 1.206, Combined License Applications for Nuclear Power Plants (LWR Edition), was developed to assist light water reactor (LWR) applicants in incorporating and effectively formatting required information for COL application review (Ref. 1). However, the guidance prescribed in RG 1.206 presumes a LWR design proposal consistent with the systems and functions associated with large LWR power plants currently operating under NRC license.

A nominal 5,000 acfm LEC pilot plant has been designed, fabricated and installed on the slipstream of a 70,000 pph stoker boiler providing steam to Ohio University`s Athens, Ohio campus. The pilot plant was normally operated on the slipstream of the Ohio Univ. boiler plant flue gas, but also had the capability of operating at higher inlet SO{sub 2} concentrations (typically equivalent to 3-1/2% sulfur coal) than those normally available from the flue gas slipstream. This was accomplished by injecting SO{sub 2} gas into the slipstream inlet. The pilot plant was instrumented to provide around-the-clock operation and was fully outfitted with temperature, SO{sub 2}, gas flow and pressure drop monitors.

Black-Right-Pointing-Pointer This is the first report of DFSP with a novel COL1A1 breakpoint in exon 5. -- Abstract: The detection of fusion transcripts of the collagen type 1{alpha}1 (COL1A1) and platelet-derived growth factor-BB (PDGFB) genes by genetic analysis has recognized as a reliable and valuable molecular tool for the diagnosis of dermatofibrosarcoma protuberans (DFSP). To detect the COL1A1-PDGFB fusion, almost previous reports performed reverse transcription polymerase chain reaction (RT-PCR) using multiplex forward primers from COL1A1. However, it has possible technical difficulties with respect to the handling of multiple primers and reagents in the procedure. The objective of this study is to establish a rapid, easy, and efficient one-step method of PCR using only a single primer pair to detect the fusion transcripts of the COL1A1 and PDGFB in DFSP. To validate new method, we compared the results of RT-PCR in five patients of DFSP between the previous method using multiplex primers and our established one-step RT-PCR using a single primer pair. In all cases of DFSP, the COL1A1-PDGFB fusion was detected by both previous method and newly established one-step PCR. Importantly, we detected a novel COL1A1 breakpoint in exon 5. The newly developed method is valuable to rapidly identify COL1A1-PDGFB fusion transcripts in DFSP.

    The research described in this product was performed in part in the Environmental Molecular Sciences Laboratory, a national scientific user facility sponsored by the Department of Energy's Office of Biological and Environmental Research and located at Pacific Northwest National Laboratory. The (TiO?)n clusters and their anions for n ) 1-4 have been studied with coupled cluster theory [CCSD(T)] and density functional theory (DFT). For n > 1, numerous conformations are located for both the neutral and anionic clusters, and their relative energies are calculated at both the DFT and CCSD(T) levels. The CCSD(T) energies are extrapolated to the complete basis set limit for the monomer and dimer and calculated up to the triple-? level for the trimer and tetramer. The adiabatic and vertical electron detachment energies of the anionic clusters to the ground and first excited states of the neutral clusters are calculated at both levels and compared with the experimental results. The comparison allows for the definitive assignment of the groundstate structures of the anionic clusters. Anions of the dimer and tetramer are found to have very closely lying conformations within 2 kcal/mol at the CCSD(T) level, whereas that of the trimer does not. In addition, accurate clustering energies and heats of formation are calculated for the neutral clusters and compared with the available experimental data. Estimates of the titanium-oxygen bond energies show that they are stronger than the group VIB transition metal-oxygen bonds except for tungsten. The atomization energies of these clusters display much stronger basis set dependence than the clustering energies. This allows the calculation of more accurate heats of formation for larger clusters on the basis of calculated clustering energies.

    Organophosphorus (OP) nerve agents such as (S)-sarin are among the most highly toxic compounds that have been synthesized. Engineering enzymes that catalyze the hydrolysis of nerve agents ( bioscavengers ) is an emerging prophylactic approach to diminishing their toxic effects. Although its native function is not known, diisopropyl fluorophosphatase (DFPase) from Loligo vulgaris catalyzes the hydrolysis of OP compounds. Here, we investigate the mechanisms of diisopropylfluorophosphate (DFP) and (S)-sarin hydrolysis by DFPase with quantum mechanical/molecular mechanical (QM/MM) umbrella sampling simulations. We find that the mechanism for hydrolysis of DFP involves nucleophilic attack by Asp229 on phosphorus to form a pentavalent intermediate. P F bond dissociation then yields a phosphoacyl enzyme intermediate in the rate-limiting step. The simulations suggest that a water molecule, coordinated to the catalytic Ca2+, donates a proton to Asp121 and then attacks the tetrahedral phosphoacyl intermediate to liberate the diisopropylphosphate product. In contrast, the calculated free energy barrier for hydrolysis of (S)-sarin by the same mechanism is highly unfavorable, primarily due to the instability of the pentavalent phosphoenzyme species. Instead, simulations suggest that hydrolysis of (S)-sarin proceeds by a mechanism in which Asp229 could activate an intervening water molecule for nucleophilic attack on the substrate. These findings may lead to improved strategies for engineering DFPase and related six-bladed -propeller folds for more efficient degradation of OP compounds.

    The overall goal of the project was to define the cellular processes that underlie embryo development in plants at a mechanistic level. Our studies focused on a critical transcriptional regulator, Arabidopsis LEAFY COTYLEDON (LEC1), that is necessary and sufficient to induce processes required for embryo development. Because LEC1 regulates lipid accumulation during the maturation phase of embryo development, information about LEC1 may be useful in designing approaches to enhance biofuel production in plants. During the tenure of this project, we determined the molecular mechanisms by which LEC1 acts as a transcription factor in embryos. We also identified genes directly regulated by LEC1 and showed that many of these genes are involved in maturation processes. This information has been useful in dissecting the gene regulatory networks controlling embryo development. Finally, LEC1 is a novel isoform of a transcription factor that is conserved among eukaryotes, and LEC1 is active primarily in seeds. Therefore, we determined that the LEC1-type transcription factors first appeared in lycophytes during land plant evolution. Together, this study provides basic information that has implications for biofuel production.

    Lymphatic vessels play an important role in tissue fluid homeostasis, intestinal fat absorption and immunosurveillance. Furthermore, they are involved in pathologic conditions, such as tumor cell metastasis and chronic inflammation. In comparison to blood vessels, the molecular phenotype of lymphatic vessels is less well characterized. Performing comparative gene expression analysis we have recently found that coxsackie- and adenovirus receptor (CAR) is significantly more highly expressed in cultured human, skin-derived lymphatic endothelial cells (LECs), as compared to blood vascular endothelial cells. Here, we have confirmed these results at the protein level, using Western blot and FACS analysis. Immunofluorescence performed on human skin confirmed that CAR is expressed at detectable levels in lymphatic vessels, but not in blood vessels. To address the functional significance of CAR expression, we modulated CAR expression levels in cultured LECs in vitro by siRNA- and vector-based transfection approaches. Functional assays performed with the transfected cells revealed that CAR is involved in distinct cellular processes in LECs, such as cell adhesion, migration, tube formation and the control of vascular permeability. In contrast, no effect of CAR on LEC proliferation was observed. Overall, our data suggest that CAR stabilizes LEC-LEC interactions in the skin and may contribute to lymphatic vessel integrity.

The LECs are optimized with respect to experimental scattering data. For comparison, we also present an uncertainty quantification that is based solely on \\pin{} scattering phase shifts. Statistical errors on the LECs are critical in order to estimate the subsequent uncertainties in \\textit{ab initio} modeling of light and medium mass nuclei which exploit chiral effective field theory. As an example of the this, we present the first complete predictions with uncertainty quantification of peripheral phase shifts of elastic proton-neutron scattering.

    Organic semiconductors have the unique ability to conduct both ionic and electronic charge carriers in thin films, an emerging advantage in applications such as light-emitting devices, transistors, and electrochromic devices, among others. Evidence suggests that the profiles of ions and electrochemical doping in the polymer film during operation significantly impact the performance and stability of the device. However, few studies have directly characterized ion profiles within LECs. Here, we present profiles of ion distributions in LECs following application of voltage, via time-of-flight secondary ion mass spectrometry. Ion distributions were characterized with regard to film thickness, salt concentration, applied voltage, and relaxation over time. Results provide insight into the correlation between ion profiles and device performance, as well as potential approaches to tuning electrochemical doping processes in LECs.

    We study the parity-violating effects in $nd\\rightarrow$ $^3H\\gamma$ process using pionless effective field theory (EFT($//!/!/!/pi$)) at thermal energy. For the weak $NN$ interaction the parity-violating Lagrangian contains five independent low-energy coupling constants (LECs). One can fix the coupling constants by comparison of the calculated observables with a sufficient number of experimental data. So, we need five observables to obtain LECs. One of the five parity-violating observables is the photon circular polarization ($P_\\gamma$) in $nd\\rightarrow$ $^3H\\gamma$ process. We calculate $P_\\gamma$ at the leading order in terms of LECs. We compare our results with the previous $P_\\gamma$ calculation based on the model of Desplanques, Donoghue, and Holstein.

    The licensing framework established by the U.S. Nuclear Regulatory Commission under Title 10 of the Code of Federal Regulations (10 CFR) Part 52, Licenses, Certifications, and Approvals for Nuclear Power Plants, provides requirements for standard design certifications (DCs) and combined license (COL) applications. The intent of this process is the early reso- lution of safety issues at the DC application stage. Subsequent COL applications may incorporate a DC by reference. Thus, the COL review will not reconsider safety issues resolved during the DC process. However, a COL application that incorporates a DC by reference must demonstrate that relevant site-specific de- sign parameters are within the bounds postulated by the DC, and any departures from the DC need to be justified. This paper provides an overview of several seismic analysis issues encountered during a review of recent DC applications under the 10 CFR Part 52 process, in which the authors have participated as part of the safety review effort.

    The licensing framework established by the U.S. Nuclear Regulatory Commission under Title 10 of the Code of Federal Regulations (10 CFR) Part 52, Licenses, Certifications, and Approvals for Nuclear Power Plants, provides requirements for standard design certifications (DCs) and combined license (COL) applications. The intent of this process is the early reso- lution of safety issues at the DC application stage. Subsequent COL applications may incorporate a DC by reference. Thus, the COL review will not reconsider safety issues resolved during the DC process. However, a COL application that incorporates a DC by reference must demonstrate that relevant site-specific de- sign parameters are confined within the bounds postulated by the DC, and any departures from the DC need to be justified. This paper provides an overview of structural design chal- lenges encountered in recent DC applications under the 10 CFR Part 52 process, in which the authors have participated as part of the safety review effort.

    An appraisal of the potential performance of different Low Energy Cooling (LEC) systems in nonresidential buildings in California is being conducted using computer simulation. The paper presents results from the first phase of the study, which addressed the systems that can be modeled, with the DOE-2.1E simulation program. The following LEC technologies were simulated as variants of a conventional variable-air-volume system with vapor compression cooling and mixing ventilation in the occupied spaces: Air-side indirect and indirect/direct evaporative pre-cooling. Cool beams. Displacement ventilation. Results are presented for four populous climates, represented by Oakland, Sacramento, Pasadena and San Diego. The greatest energy savings are obtained from a combination of displacement ventilation and air-side indirect/direct evaporative pre-cooling. Cool beam systems have the lowest peak demand but do not reduce energy consumption significantly because the reduction in fan energy is offse t by a reduction in air-side free cooling. Overall, the results indicate significant opportunities for LEC technologies to reduce energy consumption and demand in nonresidential new construction and retrofit.

    Diabetic nephropathy is the most common cause of chronic kidney disease. We investigated the ability of intracellular galectin-1 (Gal-1), a prototype of endogenous lectin, to prevent renal fibrosis by regulating cell signaling under a high glucose (HG) condition. We demonstrated that overexpression of Gal-1 reduces type I collagen (COL1) expression and transcription in human renal epithelial cells under HG conditions and transforming growth factor-{beta}1 (TGF-{beta}1) stimulation. Matrix metalloproteinase 1 (MMP1) is stimulated by Gal-1. HG conditions and TGF-{beta}1 treatment augment expression and nuclear translocation of Gal-1. In contrast, targeted inhibition of Gal-1 expression reduces COL1 expression and increases MMP1 expression. The Smad3 signaling pathway is inhibited, whereas two mitogen-activated protein kinase (MAPK) pathways, p38 and extracellular signal-regulated kinase (ERK), are activated by Gal-1, indicating that Gal-1 regulates these signaling pathways in COL1 production. Using specific inhibitors of Smad3, ERK, and p38 MAPK, we showed that ERK MAPK activated by Gal-1 plays an inhibitory role in COL1 transcription and that activation of the p38 MAPK pathway by Gal-1 plays a negative role in MMP1 production. Taken together, two MAPK pathways are stimulated by increasing levels of Gal-1 in the HG condition, leading to suppression of COL1 expression and increase of MMP1 expression.
