    This SAND report summarizes the activities and outcomes of the Network and Ensemble Enabled Entity Extraction in Informal Text (NEEEEIT) LDRD project, which addressed improving the accuracy of conditional random fields for named entity recognition through the use of ensemble methods. Conditional random fields (CRFs) are powerful, flexible probabilistic graphical models often used in supervised machine learning prediction tasks associated with sequence data. Specifically, they are currently the best known option for named entity recognition (NER) in text. NER is the process of labeling words in sentences with semantic identifiers such as %E2%80%9Cperson%E2%80%9D, %E2%80%9Cdate%E2%80%9D, or %E2%80%9Corganization%E2%80%9D. Ensembles are a powerful statistical inference meta-method that can make most supervised machine learning methods more accurate, faster, or both. Ensemble methods are normally best suited to %E2%80%9Cunstable%E2%80%9D classification methods with high variance error. CRFs applied to NER are very stable classifiers, and as such, would initially seem to be resistant to the benefits of ensembles. The NEEEEIT project nonetheless worked out how to generalize ensemble methods to CRFs, demonstrated that accuracy can indeed be improved by proper use of ensemble techniques, and generated a new CRF code, %E2%80%9CpyCrust%E2%80%9D and a surrounding application environment, %E2%80%9CNEEEEIT%E2%80%9D, which implement those improvements. The summary practical advice that results from this work, then, is: When making use of CRFs for label prediction tasks in machine learning, use the pyCrust CRF base classifier with NEEEEIT's bagging ensemble implementation. (If those codes are not available, then de-stablize your CRF code via every means available, and generate the bagged training sets by hand.) If you have ample pre-processing computational time, do %E2%80%9Cforward feature selection%E2%80%9D to find and remove counter-productive feature classes. Conversely, if

    2004-2011 Final Report for AFCI University Fellowship Program. The goal of this effort was to be supportive of university students and university programs - particularly those students and programs that will help to strengthen the development of nuclear-related fields. The program also supported the stability of the nuclear infrastructure and developed research partnerships that are helping to enlarge the national nuclear science technology base. In this fellowship program, the U.S. Department of Energy sought master's degree students in nuclear, mechanical, or chemical engineering, engineering/applied physics, physics, chemistry, radiochemistry, or fields of science and engineering applicable to the AFCI/Gen IV/GNEP missions in order to meet future U.S. nuclear program needs. The fellowship program identified candidates and selected full time students of high-caliber who were taking nuclear courses as part of their degree programs. The DOE Academic Program Managers encouraged fellows to pursue summer internships at national laboratories and supported the students with appropriate information so that both the fellows and the nation's nuclear energy objectives were successful.

Also, the foundation, with its university affiliates, conducts ecological and radioecological research on the Idaho National Environmental Research Park. This research benefits major DOE-ID programs including waste management, environmental restoration, spent nuclear fuels, and land management issues. Major accomplishments during CY1995 can be divided into five categories: environmental surveillance program, environmental education, environmental services and support, ecological risk assessment, and research benefitting the DOE-ID mission.

    This document is one in a series of topical reports compiled by the Pacific Northwest National Laboratory to summarize technical information on selected topics important to the performance of a probabilistic seismic hazard analysis (PSHA) of the Hanford Site. The purpose of this report is to summarize available data and analyses relevant to the Yakima Fold Belt (YFB) that may bear on the question of whether or not the YFB behaves as a single seismotectonic province in which activity along one fold structure is representative of behavior along all other fold structures. This topic has met with a fairly high level of contention in the expert community and has the potential to result in significant impacts on an evaluation of seismic hazard at the Hanford Site. This report defines the relevant alternative conceptual models relevant to this technical issue and the arguments and data that support those models. It provides a brief description of the technical issue and principal uncertainties; a general overview on the nature of the technical issue, along with alternative conceptual models, supporting arguments and information, and uncertainties; and finally, it suggests some possible approaches for reducing uncertainties regarding this issue.

    In order to reduce the current excesses of plutonium (both weapon grade and reactor grade) and other transuranium elements, a concept of inert matrix fuel (IMF) has been proposed for an uranium free transmutation of fissile actinides which excludes continuous uranium-plutonium conversion in thermal reactors and advanced systems. Magnesium oxide (MgO) is a promising candidate for inert matrix (IM) materials due to its high melting point (2827 C), high thermal conductivity (13 W/K {center_dot} m at 1000 C), good neutronic properties, and irradiation stability However, MgO reacts with water and hydrates easily, which prevents it from being used in light water reactors (LWRs) as an IM. To improve the hydration resistance of MgO-based inert matrix materials, Medvedev and coworkers have recently investigated the introduction of a secondary phase that acts as a hydration barrier. An MgO-ZrO{sub 2} composite was specifically studied and the results showed that the composite exhibited improved hydration resistance than pure MgO. However, ZrO{sub 2} is insoluble in most acids except HF, which is undesirable for fuel reprocessing. Moreover, the thermal conductivity of ZrO{sub 2} is low and typically less than 3 W {center_dot} m{sup -1} {center_dot} K{sup -1} at 1000 C. In search for an alternative composite strategy, Nd{sub 2}Zr{sub 2}O{sub 7}, an oxide compound with pyrochlore structure, has been proposed recently as a corrosion resistant phase, and MgO-Nd{sub 2}Zr{sub 2}O{sub 7} composites have been investigated as potential IM materials. An adequate thermal conductivity of 6 W {center_dot} m{sup -} 1 {center_dot} K{sup -1} at 1000 C for the MgO-Nd{sub 2}Zr{sub 2}O{sub 7} composite with 90 vol% MgO was recently calculated and reported. Other simulations proposed that the MgO-pyrochlore composites could exhibit higher radiation stability than previously reported.     The proposed straw separation system developed in the research project harvests the large internode sections of the straw which has the greater potential as a feedstock for lignocellulosic ethanol production while leaving the chaff and nodes in the field. This strategy ensures sustainable agriculture by preventing the depletion of soil minerals, and it restores organic matter to the soil in amounts and particle sizes that accommodate farmers’ needs to keep tillage and fertilizer costs low. A ton of these nutrient-rich plant tissues contains as much as $10.55 worth of fertilizer (economic and energy benefits), in terms of nitrogen, phosphorus, potassium, and other nutrients provided to the soil when incorporated by tillage instead of being burned. Biomass conversion to fermentable sugars for the purpose of producing fuels, chemicals, and other industrial products is well understood. Most bioenergy strategies rely on low-cost fermentable sugars for sustainability and economic viability in the marketplace. Exploitation of the “whole crop”—specifically, wheat straw or other plant material currently regarded as residue or waste—is a practical approach for obtaining a reliable and low-cost source of sugars. However, industrial-scale production of sugars from wheat straw, while technically feasible, is plagued by obstacles related to capital costs, energy consumption, waste streams, production logistics, and the quality of the biomass feedstock. Currently available separation options with combine harvesters are not able to achieve sufficient separation of the straw/stover and chaff streams to realize the full potential of selective harvest. Since ethanol yield is a function of feedstock structural carbohydrate content, biomass anatomical fractions of higher product yield can have a significant beneficial impact on minimum ethanol selling price. To address this advanced biomass separation computation engineering models were developed to more effectively and efficiently engineer high-fidelity and high throughput separation systems for biomass components. INL and Iowa State University developed a computational modeling strategy for simulating multi-phase flow with an integrated solver using various computational fluid dynamics (CFD) codes. ISU set up a classic multi-phase test problem to be solved by the various CFD codes. The benchmark case was based on experimental data for bubble gas holdup and bed expansion for a gas/solid fluidized bed. Preliminary fluidization experiments identified some unexpected fluidization behavior, where rather than the bed uniformly fluidizing, a “blow out” would occur where a hole would open up in the bed through which the air would preferentially flow, resulting in erratic fluidization. To improve understanding of this phenomena and aid in building a design tool, improved computational tools were developed. The virtual engineering techniques developed were tested and utilized to design a separation baffle in a CNH combine. A computational engineering approach involving modeling, analysis, and simulation was used in the form of virtual engineering to design a baffle separator capable of accomplishing the high-fidelity residue separation established by the performance targets. Through the use of the virtual engineering model, baffle designs were simulated to (1) determine the effect of the baffle on the airflow of the combine cleaning system, and (2) predict the effectiveness of the baffle in separating the residue streams. A baffle design was selected based on the virtual engineering modeling, built into the INL selective harvest test combine. The result of the baffle changes improved the crop separation capability of the combine, enabling downstream improvement in composition and theoretical ethanol yield. In addition, the positive results from the application of the virtual engineering tools to the CNH combine design resulted in further application of these tools to other INL areas of research. INL and the University of Idaho identified, characterized, and modified a key plant biosynthetic lignin gene, cinnamoyl CoA reductase (CCR), to assess its influence on the efficiency with which the resulting biomaterials interact with and are processed by engineering systems. The characterization of CCR genes and resulting antisense gene constructs enabled the modification and assessment of the impact of cell wall biosynthetic gene in wheat. This allows the model systems needed to study the structural and functional post-harvest properties of straw to be created and assessed.     Highlights: Black-Right-Pointing-Pointer Expression of Id3 but not Id1 is induced by Wnt3a stimulation in C2C12 cells. Black-Right-Pointing-Pointer Wnt3a induces Id3 expression via canonical Wnt/{beta}-catenin pathway. Black-Right-Pointing-Pointer Wnt3a-induced Id3 expression does not depend on BMP signaling activation. Black-Right-Pointing-Pointer Induction of Id3 expression is critical determinant in Wnt3a-induced cell proliferation and differentiation. -- Abstract: Canonical Wnt signaling plays important roles in regulating cell proliferation and differentiation. In this study, we report that inhibitor of differentiation (Id)3 is a Wnt-inducible gene in mouse C2C12 myoblasts. Wnt3a induced Id3 expression in a {beta}-catenin-dependent manner. Bone morphogenetic protein (BMP) also potently induced Id3 expression. However, Wnt-induced Id3 expression occurred independent of the BMP/Smad pathway. Functional studies showed that Id3 depletion in C2C12 cells impaired Wnt3a-induced cell proliferation and alkaline phosphatase activity, an early marker of osteoblast cells. Id3 depletion elevated myogenin induction during myogenic differentiation and partially impaired Wnt3a suppressed myogenin expression in C2C12 cells. These results suggest that Id3 is an important Wnt/{beta}-catenin induced gene in myoblast cell fate determination.

    Wind Generation Feasibility Warm Springs Power and Water Enterprises (WSPWE) is a corporate entity owned by the Confederated Tribes of the Warm Springs Reservation, located in central Oregon. The organization is responsible for managing electrical power generation facilities on tribal lands and, as part of its charter, has the responsibility to evaluate and develop renewable energy resources for the Confederated Tribes of Warm Springs. WSPWE recently completed a multi-year-year wind resource assessment of tribal lands, beginning with the installation of wind monitoring towers on the Mutton Mountains site in 2003, and collection of on-site wind data is ongoing. The study identified the Mutton Mountain site on the northeastern edge of the reservation as a site with sufficient wind resources to support a commercial power project estimated to generate over 226,000 MWh per year. Initial estimates indicate that the first phase of the project would be approximately 79.5 MW of installed capacity. This Phase 2 study expands and builds on the previously conducted Phase 1 Wind Resource Assessment, dated June 30, 2007. In order to fully assess the economic benefits that may accrue to the Tribes through wind energy development at Mutton Mountain, a planning-level opinion of probable cost was performed to define the costs associated with key design and construction aspects of the proposed project. This report defines the Mutton Mountain project costs and economics in sufficient detail to allow the Tribes to either build the project themselves or contract with a developer under the most favorable terms possible for the Tribes.

The ceramic industry internationally is beginning to benefit from both the energy use and drying time reductions it achieves, while on the basis of further theoretical work carried out since 1993 other industries, including the bioenergy sector, should also soon begin to exploit its advantages. As global warming becomes a reality and oil reserves decline, superheated steam drying and gasification of biomass will contribute to the mitigation of those problems.

    This report serves to summarize the major activities completed as part of Virginia Electric and Power Company's North Anna construction and operating license demonstration project with DOE. Project successes, lessons learned, and suggestions for improvement are discussed. Objectives of the North Anna COL project included preparation and submittal of a COLA to the USNRC incorporating ESBWR technology for a third unit a the North Anna Power Station site, support for the NRC review process and mandatory hearing, obtaining NRC approval of the COLA and issuance of a COL, and development of a business case necessary to support a decision on building a new nuclear power plant at the North Anna site.

    Timely analysis of cyber-security information necessitates automated information extraction from unstructured text. While state-of-the-art extraction methods produce extremely accurate results, they require ample training data, which is generally unavailable for specialized applications, such as detecting security related entities; moreover, manual annotation of corpora is very costly and often not a viable solution. In response, we develop a very precise method to automatically label text from several data sources by leveraging related, domain-specific, structured data and provide public access to a corpus annotated with cyber-security entities. Next, we implement a Maximum Entropy Model trained with the average perceptron on a portion of our corpus (~750,000 words) and achieve near perfect precision, recall, and accuracy, with training times under 17 seconds.

    Just as regulation is an ever-present and crucial factor in most of the day-to-day operating decisions of regulated natural gas transmission and distribution companies, it is also an important factor to consider in assessing the application of the antitrust laws to the conduct of these regulated entities. The legal guidelines are far from clear and are evolving. However, as regulated entities find themselves in an ever more competitive environment, it becomes necessary for their legal counsel to monitor closely the developments in the law concerning immunities from the antitrust laws by virtue of regulation imposed by government. The author hopes that this article will provide a useful starting point for that analysis.

Despite the ever-present desire for models of greater fidelity at the expense of analysis and computation resources, models of moderate to low fidelity are still required at many levels of decision-making. Problems can arise due to the issue of consistency among the family of models used for analysis. To address this and other problems, models of variable resolution have been suggested. However, such variable resolution architectures inherently carry their own set of issues which must be resolved in order to be useful. First, what are the structural requirements for a variable resolution model; and second, how are interactions between entities governed, especially when the entities have different resolutions? This paper addresses these issues and discusses key mechanisms needed to develop a variable resolution combat simulation that meets several core requirements for such models: seamless aggregation/disaggregation, appropriate interactions between entities of differing resolution, and control of the aggregation/disaggregation process.

Anomaly- and signature-based IDS technologies have developed methods to cover information technology-based networks activity and protocols effectively. However, these IDS technologies do not include the fine protocol granularity required to ensure network security inside an environment with weak protocols lacking authentication and encryption. By implementing a more specific and more intelligent packet inspection mechanism, tailored traffic flow analysis, and unique packet tampering detection, IDS technology developed specifically for SCADA environments can be deployed with confidence in detecting malicious activity.

    The DOE complex currently has 332 underground storage tanks (USTs) that have been used to process and store radioactive and chemical mixed waste generated from weapon materials production. Very little of the over 100 million gallons of high-level and low-level radioactive liquid waste has been treated and disposed of in final form. Two waste storage tank design types are prevalent across the DOE complex: single-shell wall and double-shell wall designs. They are made of stainless steel, concrete, and concrete with carbon steel liners, and their capacities vary from 5000 gallons (19 m{sup 3}) to 10{sup 6} gallons (3785 m{sup 3}). The tanks have an overburden layer of soil ranging from a few feet to tens of feet. Responding to the need for remediation of tank waste, driven by Federal Facility Compliance Agreements (FFCAs) at all participating sites, the Underground Storage Tank Integrated Demonstration (UST-ID) Program was created by the US DOE Office of Technology Development in February 1991. Its mission is to focus the development, testing, and evaluation of remediation technologies within a system architecture to characterize, retrieve, treat to concentrate, and dispose of radioactive waste stored in USTs at DOE facilities. The ultimate goal is to provide safe and cost-effective solutions that are acceptable to the public and the regulators. The UST-ID has focused on five DOE locations: the Hanford Site, which is the host site, in Richland, Washington; the Fernald Site in Fernald, Ohio; the Idaho National Engineering Laboratory near Idaho Falls, Idaho; the Oak Ridge Reservation in Oak Ridge, Tennessee, and the Savannah River Site in Savannah River, South Carolina.

    A nanofluidic channel fabricated in fused silica with an approximately 500 nm square cross section was used to isolate, detect and identify individual quantum dot conjugates. The channel enables the rapid detection of every fluorescent entity in solution. A laser of selected wavelength was used to excite multiple species of quantum dots and organic molecules, and the emission spectra were resolved without significant signal rejection. Quantum dots were then conjugated with organic molecules and detected to demonstrate efficient multicolor detection. PCH was used to analyze coincident detection and to characterize the degree of binding. The use of a small fluidic channel to detect quantum dots as fluorescent labels was shown to be an efficient technique for multiplexed single molecule studies. Detection of single molecule binding events has a variety of applications including high throughput immunoassays.

    The negative regulator of DNA-binding/cell-differentiation Id2 is a small protein containing a central helix-loop-helix (HLH) motif and a C-terminal nuclear export signal (NES). Whereas the former is essential for Id2 dimerization and nuclear localization, the latter is responsible for the transport of Id2 from the nucleus to the cytoplasm. Whereas the isolated Id2 HLH motif is highly helical, large C-terminal Id2 fragments including the NES sequence are either unordered or aggregation-prone. To study the conformational properties of the isolated NES region, we synthesized the Id2 segment 103-124. The latter was insoluble in water and only temporarily soluble in water/alcohol mixtures, where it formed quickly precipitating {beta}-sheets. Introduction of a positively charged N-terminal tail prevented aggressive precipitation and led to aggregates consisting of long fibrils that bound thioflavin T. These results show an interesting structural aspect of the Id2 NES region, which might be of significance for both protein folding and function.

    Nine strains were identified to grow with gasoline as sole sulfur source. Two different genes were cloned from Gordonia terrae KGB1 and tested for the ability to support gasoline BDS. The first of these, fmoA, was cloned by screening a KGB1 gene library for the ability to convert indole to indigo (a sulfur-regulated capability in KGB1). The fmoA gene was overexpressed in a gasoline tolerant strain of Pseudomonas putida PpG1 and the recombinant strain was shown to convert thiophene to a dimer of thiophene sulfoxide at rates nearly two orders of magnitude higher than KGB1 could catalyze the reaction. Despite this high activity the recombinant PpG1 was unable to demonstrate any activity against gasoline either in shake flask or in bench-scale gasoline BDS bioreactor. A second gene (toeA) was cloned from KGB1 and shown to support growth of Rhodococcus erythropolis JB55 on gasoline. The toeA gene was also identified in another gasoline strain T. wratislaviensis EMT4, and was identified as a homolog of dszA from R. erythropolis IGTS8. Expression of this gene in JB55 supported conversion of DBTO2 (the natural substrate for DszA) to HPBS, but activity against gasoline was low and BDS results were inconsistent. It appeared that activity was directed against C2- and C3-thiophenes. Efforts to increase gene expression by plasmid manipulation, by addition of flavin reductase genes, or by expression in PpG1 were unsuccessful. The DszC protein (DBT monooxygenase) from IGTS8 has very little activity against the sulfur compounds in gasoline, but a mutant enzyme with a substitution of phenylalanine for valine at position 261 was shown to have an altered substrate range. This alteration resulted in increased activity against gasoline, with activity towards mainly C3- and C4-thiophenes and benzothiophene. A mutant library of dszB was constructed by RACHITT (W. C. Coco et al., DNA shuffling method for generating highly recombined genes and evolved enzymes. 2001. Nature Biotech. 19:354-359) method of in vitro recombination. Methods for analysis were developed and a preliminary analysis of the library was performed. A preliminary gasoline process design was constructed and process economics were determined based upon assumptions made from experimental results. The projected cost of gasoline BDS was determined to be competitive with current competing technologies.

    Highlights: •Id1 expression enables naïve T cell proliferation without anti-CD28 co-stimulation. •Id1 expression facilitates T cells survival when stimulated with anti-CD3. •Elevation of IL-2 production by Id1 contributes increased proliferation and survival. •Id1 potentiates NF-κB activation by anti-CD3 stimulation. -- Abstract: Although the role of E proteins in the thymocyte development is well documented, much less is known about their function in peripheral T cells. Here we demonstrated that CD4 promoter-driven transgenic expression of Id1, a naturally occurring dominant-negative inhibitor of E proteins, can substitute for the co-stimulatory signal delivered by CD28 to facilitate the proliferation and survival of naïve CD4{sup +} cells upon anti-CD3 stimulation. We next discovered that IL-2 production and NF-κB activity after anti-CD3 stimulation were significantly elevated in Id1-expressing cells, which may be, at least in part, responsible for the augmentation of their proliferation and survival. Taken together, results from this study suggest an important role of E and Id proteins in peripheral T cell activation. The ability of Id proteins to by-pass co-stimulatory signals to enable T cell activation has significant implications in regulating T cell immunity.

    The original PROP code developed by AeroVironment, Inc. and its various versions have been in use for wind turbine performance predictions for over ten years. Due to its simplicity, rapid execution times and relatively accurate predictions, it has become an industry standard in the US. The Europeans have similar blade-element/momentum methods in use for design. Over the years, PROP has continued to be improved (in its accuracy and capability), e.g., PROPSH, PROPPC, PROP93, and PropID. The latter version incorporates a unique inverse design capability that allows the user to specify the desired aerodynamic characteristics from which the corresponding blade geometry is determined. Through this approach, tedious efforts related to manually adjusting the chord, twist, pitch and rpm to achieve desired aerodynamic/performance characteristics can be avoided, thereby making it possible to perform more extensive trade studies in an effort to optimize performance. Past versions of PropID did not have supporting graphics software. The more current version to be discussed includes a Matlab-based graphical user interface (GUI) and additional features that will be discussed in this paper.

  17. Computerized economic and statistical investigation of the Alabama liquid asphalt market for public entities

    This study outlines the development of an economic data base and techniques utilized in identifying noncompetitive practices in the sealed bid market for liquid asphalt products purchased by public entities in the State of Alabama. It describes the organization of data and methods for displaying salient characteristics of market conduct and performance. Likely areas of anticompetitive activity are identified from an examination of conditional factors influencing collusion in a market and of circumstantial evidence of collusive behavior of the vendors. Methods of detecting and analyzing suspicious behavior are indicated and applied to selected data. The conclusion reached was that collusion was present in the Alabama liquid asphalt market during 1971-1978. An antitrust action was initiated by the State. Damages were calculated from the data base using a GLM regression model. An out-of-court settlement was negotiated by the defendant vendors.

  19. A 12-MW-scale pilot study of in-duct scrubbing (IDS) using a rotary atomizer

    A low-cost, moderate-removal efficiency, flue gas desulfurization (FGD) technology was selected by the US Department of Energy for pilot demonstration in its Acid Rain Precursor Control Technology Initiative. The process, identified as In-Duct Scrubbing (IDS), applies rotary atomizer techniques developed for lime-based spray dryer FGD while utilizing existing flue gas ductwork and particulate collectors. IDS technology is anticipated to result in a dry desulfurization process with a moderate removal efficiency (50% or greater) for high-sulfur coal-fired boilers. The critical elements for successful application are: (1) adequate mixing of sorbent droplets with flue gas for efficient reaction contact, (2) sufficient residence time to produce a non-wetting product, and (3) appropriate ductwork cross-sectional area to prevent deposition of wet reaction products before particle drying is comple. The ductwork in many older plants, previously modified to meet 1970 Clean Air Act requirements for particulate control, usually meet these criteria. A 12 MW-scale IDS pilot plant was constructed at the Muskingum River Plant of the American Electric Power System. The pilot plant, which operates from a slipstrem attached to the air-preheater outlet duct from the Unit 5 boiler at the Muskingum River Plant (which burns about 4% sulfur coal), is equipped with three atomizer stations to test the IDS concept in vertical and horizontal configurations. In addition, the pilot plant is equipped to test the effect of injecting IDS off- product upstream of the atomizer, on SO{sub 2}and NO{sub x} removals.

    The mammalian BTG/Tob family is a group of proteins with anti-proliferative ability, and there are six members including BTG1, BTG2/PC3/Tis21, BTG3/ANA, BTG4/PC3B, Tob1/Tob and Tob2. Among them, Tob subfamily members, specifically Tob1/Tob and Tob2, have the most extensive C-terminal regions. As previously reported, overexpression of BTG/Tob proteins is associated with the inhibition of G1 to S-phase cell cycle progression and decreased cell proliferation in a variety of cell types. Tob subfamily proteins have similar anti-proliferative effects on cell cycle progression in cultured tumor cells. An important unresolved question is whether or not they have function in rapidly proliferating cells, such as embryonic stem cells (ESCs). Tob1 and Tob2 were expressed ubiquitously in mouse ESCs (mESCs), suggesting a possible role in early embryonic development and mESCs. To address the above question and explore the possible functions of the Tob subfamily in ESCs, we established ESCs from different genotypic knockout inner cell mass (ICM). We found that Tob1{sup −/−}, Tob2{sup −/−}, and Tob1/2 double knockout (DKO, Tob1{sup −/−} & Tob2{sup −/−}) ESCs grew faster than wild type (WT) ESCs without losing pluripotency, and we provide a possible mechanistic explanation for these observations: Tob1 and Tob2 inhibit the cell cycle via degradation of Id3 mRNA, which is a set of directly targeted genes of BMP4 signaling in mESCs that play critical roles in the maintenance of ESC properties. Together, our data suggest that BTG/Tob family protein Tob1 and Tob2 regulation cell proliferation does not compromise the basic properties of mESCs. - Highlights: • We established mouse Tob1/2 double knockout embryonic stem cells. • Tob1 and Tob2 inhibit the proliferation of ESCs without effect on pluripotency. • Tob1 and Tob2 involved in the degradation of Id3 in mESCs.
