Following an Internal Medicine Residency at Case Western Reserve University Hospitals of Cleveland and a Hematology-Oncology Fellowship at the University of North Carolina at Chapel Hill, Dr. Agre returned to Johns Hopkins as a postdoctoral fellow in cell biology. Intracellular route and biological activity of exogenously delivered Rep proteins from the adeno-associated virus type 2

The two large Rep proteins, Rep78 and Rep68, from the adeno-associated virus type 2 (AAV-2) are required for AAV-2 DNA replication, site-specific integration, and for the regulation of viral gene expression. The study of their activities is dependent on the ability to deliver these proteins to the cells in a time and dose-dependent manner. We evaluated the ability of a protein transduction domain (PTD) derived from the human immunodeficiency virus 1 (HIV-1) TAT protein to drive the cellular internalization of exogenously delivered PTD-fused Rep68 proteins. This analysis unexpectedly revealed that recombinant Rep68 alone, in the absence of any PTD, could be endocytosed by the cells. Rep68 as the chimeric TAT-Rep68 proteins were internalized through endocytosis in clathrin-coated vesicles and retained in late endosomes/lysosomes with no detectable nuclear localization. In the presence of adenovirus, the Rep proteins could translocate into the nucleus where they displayed a biological activity. These findings support recent reports on the mechanism of entry of TAT-fused proteins and also revealed a new property of Rep68.

RepPop: A Database for Repetitive Elements in Populus Trichocarpa

The populus was selected as the first tree with the genome to be sequenced, mainly due to its small genome size, the wide deployment worldwide (30+ species), and its short juvenile period. Its rich content of cellulose, which is one of the most important source for biofuel. A female clone of P. trichocarpa was chosen to be sequenced. The current assembly of Populus genome is release 1.0, whose small insert end-sequence coverage is 7.5X, and it was released in June 2004. It consists of 22,012 sequences (including the 19 chromosomes) and the total length is 485,510,911 bps. The data was downloaded from the offical site of the Populus trichocarpa genome sequencing project. The latest version of the genome can be found at the Poplar Genome Project at JGI Eukaryotic Genomics. Duplication regions introduce significant difficulties into the correct assemblying of sequence contigs. We identified all the repetitive elements in the populus genome. We further assign each of them as different classes of repetitive elements, including DNA transposons, RNA retrotransposons, Miniature Inverted-repeat Transposable Elements (MITE), Simple Sequence Repeats (SSR), and Segmental Duplications (SD), etc. We organized the annotations into this easily browsable, searchable, and blastable database, RepPop, for the whole community.[From website for RepPop at http://csbl.bmb.uga.edu/~ffzhou/RepPop/

In this work we consider the Peter principle and anti-Peter principle as the discrete logistic and discrete inverse logistic equation. Especially we discuss imprecisely estimated (by hierarchical control mechanism) carrying capacity, i.e. boundary (in)competence level of a hierarchy member. It implies that Peter principle holds two sub-principles. In the first one objective boundary competence level is increased for estimation error. In the second one objective boundary competence level is decreased for estimation error. Similarly, anti-Peter principle holds two sub-principles too. All this implies that paradoxical situations that follow from Peter and anti-Peter principle can be simply removed by decrease of the error of hierarchical (social) control. Also we discuss cases by Peter principle when error of the boundary competence level by estimation grows up. (Then, in fact, there is no estimation error but stimulation of the boundary level by control mechanism.) By first Peter sub-principle it implies anarchy in the social structure or, correspondingly, cancer in the biology and medicine, schizophrenia in the psychology and inflation in the economy. By second Peter sub-principle it implies a totalitary social structure (dictature or caste regime) or multiplex sclerosis and other autoimmune diseases in biology and medicine, servile mentality or low value complex in psychology and depression by hyperactive political influences in economy. Finally, monotonus changes of the stimulated part of boundary level cause corresponding phase transitions discussed on the example of the introspection in the psychology.

CO{sub 2} Injectivity, Storage Capacity, Plume Size, and Reservoir and Seal Integrity of the Ordovician St. Peter Sandstone and the Cambrian Potosi Formation in the Illnois Basin  

SciTech Connect

The Cambro-Ordovician strata of the Illinois and Michigan Basins underlie most of the states of Illinois, Indiana, Kentucky, and Michigan. This interval also extends through much of the Midwest of the United States and, for some areas, may be the only available target for geological sequestration of CO{sub 2}. We evaluated the Cambro-Ordovician strata above the basal Mt. Simon Sandstone reservoir for sequestration potential. The two targets were the Cambrian carbonate intervals in the Knox and the Ordovician St. Peter Sandstone. The evaluation of these two formations was accomplished using wireline data, core data, pressure data, and seismic data from the USDOE-funded Illinois Basin ?? Decatur Project being conducted by the Midwest Geological Sequestration Consortium in Macon County, Illinois. Interpretations were completed using log analysis software, a reservoir flow simulator, and a finite element solver that determines rock stress and strain changes resulting from the pressure increase associated with CO{sub 2} injection. Results of this research suggest that both the St. Peter Sandstone and the Potosi Dolomite (a formation of the Knox) reservoirs may be capable of storing up to 2 million tonnes of CO{sub 2} per year for a 20-year period. Reservoir simulation results for the St. Peter indicate good injectivity and a relatively small CO{sub 2} plume. While a single St. Peter well is not likely to achieve the targeted injection rate of 2 million tonnes/year, results of this study indicate that development with three or four appropriately spaced wells may be sufficient. Reservoir simulation of the Potosi suggest that much of the CO{sub 2} flows into and through relatively thin, high permeability intervals, resulting in a large plume diameter compared with the St. Peter.

Science Conference Proceedings (OSTI)

Small- and Ultra-Small Angle Neutron Scattering (SANS and USANS) provide powerful tools for quantitative analysis of porous rocks, yielding bulk statistical information over a wide range of length scales. This study utilized (U)SANS to characterize shallowly buried quartz arenites from the St. Peter Sandstone. Backscattered electron imaging was also used to extend the data to larger scales. These samples contain significant volumes of large-scale porosity, modified by quartz overgrowths, and neutron scattering results show significant sub-micron porosity. While previous scattering data from sandstones suggest scattering is dominated by surface fractal behavior over many orders of magnitude, careful analysis of our data shows both fractal and pseudo-fractal behavior. The scattering curves are composed of subtle steps, modeled as polydispersed assemblages of pores with log-normal distributions. However, in some samples an additional surface-fractal overprint is present, while in others there is no such structure, and scattering can be explained by summation of non-fractal structures. Combined with our work on other rock-types, these data suggest that microporosity is more prevalent, and may play a much more important role than previously thought in fluid/rock interactions.

Permeabilities of 10.2 cm in length, 2.5 cm in diameter Kayenta (porosity, 20.7, +- 1.66%) and St. Peter (porosity, 13.6, +- 0.13%) sandstones to Magmamax No. 1 brine containing suspended solids were determined from 70 to 90/sup 0/C at 10.3-MPa confining pressure. Measurements were performed without filters, with one 10-..mu..m filter, and with two 10-..mu..m filters inserted upstream of the core sample. In all cases, there was a dramatic decrease in permeability within the first hour of flow or few hundred pore volumes of flow through the core. Experiments conducted without filters or with one filter yield permeabilities that represent both the rock and the 2- to 3-mm amorphous silica-iron layer on the top face of the core. The experimental results show that if the Salton Sea Geothermal Field (SSGF) were composed of porous, sedimentary formations similar to Kayenta sandstone, long-term injection of unmodified Magmamax brine would not be feasible. In the case of acidified brine, most of the permeability decline may result from the mobilization of calcite.

As of 2:00 PM EDT today (June 30), 3,592,559 customers are without power in the affected areas. This is a decrease from the 3,854,162 customers reported this morning in Situation Report #1. Utilities throughout the affected area are continuing to assess damage and restore power. Restoration is expected to take up to a week in some areas. Utilities have made request for outside workers to assist with restoration. Restoration estimates and efforts by electric utilities are reported below. Electricity Outages by State

The current outages now include the State of New Jersey. The outages are a decrease from the 3,592,559 customers reported in Situation Report #2 yesterday afternoon. Utilities throughout the affected area are continuing to assess damage. Major damage has occurred to components of the electric distribution network. Utilities are focusing initial restoration on

This report presents information on the preliminary process knowledge to be used in characterizing all Department of Energy (DOE)-owned Spent Nuclear Fuel (SNF) types that potentially exhibit a Resource Conservation and Recovery Act (RCRA) characteristic. This report also includes the process knowledge, analyses, and rationale used to preliminarily exclude certain SNF types from RCRA regulation under 40 CFR {section}261.4(a)(4), ``Identification and Listing of Hazardous Waste,`` as special nuclear and byproduct material. The evaluations and analyses detailed herein have been undertaken as a proactive approach. In the event that DOE-owned SNF is determined to be a RCRA solid waste, this report provides general direction for each site regarding further characterization efforts. The intent of this report is also to define the path forward to be taken for further evaluation of specific SNF types and a recommended position to be negotiated and established with regional and state regulators throughout the DOE Complex regarding the RCRA-related policy issues.

We point out the priority of our paper (Mahajan et al. 2001, Phys. Plasmas, 8, 1340) over (Aschwanden et al. 2007, Astrophys J., 659, 1673) in introducing the term "Formation and primary heating of the solar corona" working out explicit models (theory as well as simulation) for coronal structure formation and heating. On analyzing the Aschwanden et al. (2007) scenario of coronal heating process (shifted to the chromospheric heating) we stress, that for efficient loop formation, the primary upflows of plasma in chromosphere/transition region should be relatively cold and fast (as opposed to hot). It is during trapping and accumulation in closed field structures, that the flows thermalize (due to the dissipation of the short scale flow energy) leading to a bright and hot coronal structure. The formation and primary heating of a closed coronal structure (loop at the end) are simultaneous and a process like the "filling of the empty coronal loop by hot upflows" is purely speculative and totally unlikely.

The goal of these studies is to ensure long-term stable containment of air in the underground reservoirs used in conjunction with compressed air energy storage (CAES) plants. The specific objective is to develop stability criteria and engineering guidelines for designing CAES reservoirs in each of the three major reservoir types, including aquifers, salt cavities, and mined hard rock caverns. This document characterizes the geologic nature of porous media constituents native to the aquifer field test site near Pittsfield, Illinois. The geologic samples were subjected to geochemical evaluations to determine anticipated responses to cyclic air injection, heating and moisture - conditions typical of an operating CAES reservoir. This report documents the procedures used and results obtained from these analyses.

What cultural values motivated the various stakeholders in the Challenge: Google, professors, students, and businesses? Google Google was motivated by corporate values of providing hands-on learning for undergraduate and graduate students in business-related disciplines such as advertising, ecommerce, management information systems, and marketing. As Google obtains 95 % of its income from AdWords (J. Murphy, personal communication, 26 Feb, 2009), the Challenge was a way to create top-of-mind awareness among professors, students who would be future business employees, and businesses that have not used AdWords in the last six months preceding the Challenge period and would benefit from a test-drive of a three-week campaign with US$200 sponsored by Google.
