    Research highlights: {yields} Strong ADAM15 expression is found in normal melanocytes. {yields} ADAM15 expression is significantly downregulated in patients with melanoma metastasis. {yields} TGF-{beta} can downregulate ADAM15 expression in melanoma cells. {yields} Overexpression of ADAM15 in melanoma cells inhibits migration, proliferation and invasion of melanoma cells. {yields} Conclusion: ADAM15 represents an tumor suppressor protein in melanoma. -- Abstract: In a mouse melanoma metastasis model it has been recently shown that ADAM15 overexpression in melanoma cells significantly reduced the number of metastatic nodules on the lung. Unfortunately, the expression of ADAM15 in human melanoma tissue has not been determined so far. In our study, we characterized the expression of ADAM15 in tissue micro-arrays of patients with primary melanoma with melanoma metastasis. ADAM15 was expressed in melanocytes and endothelial cells of benign nevi and melanoma tissue. Importantly, ADAM15 was significantly downregulated in melanoma metastasis compared to primary melanoma. We further demonstrate that IFN-{gamma} and TGF-{beta} downregulate ADAM15 protein levels in melanoma cells. To investigate the role of ADAM15 in melanoma progression, we overexpressed ADAM15 in melanoma cells. Importantly, overexpression of ADAM15 in melanoma cells reduced the migration, invasion and the anchorage dependent and independent cell growth of melanoma cells. In summary, the downregulation of ADAM15 plays an important role in melanoma progression and ADAM15 act as a tumorsuppressor in melanoma.

    The Advanced Coal Extraction Project is sponsored by the Department of Energy at the Jet Propulsion Laboratory to define and develop advanced underground coal extraction systems which: (1) are suitable for significant remaining resources after the year 2000, and (2) promise a significant improvement in production cost and miner safety, with no degradation in miner health, environmental quality and resource recovery. System requirements in the five performance areas have been defined by Goldsmith and Lavin (1980). Several existing computer programs for estimating life-cycle cost of mining systems have been evaluated. A commercially available program ADAM/1 was found to be satisfactory in relation to the needs of the Advanced Coal Extraction Project. Two test cases were run to confirm the ability of the program to handle non-conventional mining equipment and procedures. The results were satisfactory. The model, therefore, is recommended to the project team for evaluation of their conceptual designs. Since the model is commercially available, data preparation instructions are not reproduced in this document; instead the reader is referred to the original documents for this information.

    The Yakama Nation is the largest tribe in the Pacific Northwest. The Yakama Nation Housing Authority (YNHA) is working to rehabilitate single family homes (two to four bedrooms) in its Adams View project using public and tax credit financing. It is in need of a major rehabilitation as a result of wear and tear after many years of use and overcrowding. The scope for the current CARB ''gut rehab'' project is 25 of the 40 homes in the Adams View development, but the proposed strategies could be replicated for the remaining 15 homes as well as for several other similar developments of the YNHA. On a larger scale, the system rehabilitation strategy developed for the Adam's View project should be replicable for most of the more than 4,300 housing units that were constructed in the Northwest (Washington, Oregon, and Idaho) under the US Department of Housing and Urban Development (HUD) Housing Act of 1937.

    Florida International University's (FIU) Hemispheric Center for Environmental Technology (HCET) evaluated five saws for their effectiveness in cutting up specially prepared fiberglass-reinforced plywood crates. These crates were built as surrogates for crates that presently hold radioactive contaminated glove boxes at the Department of Energy's (DOE) Los Alamos facility. The Adamant circular saw was assessed on August 14, 2001. During the FIU test of efficacy, a team from the Operating Engineers National Hazmat Program (OENHP) evaluated the occupational safety and health issues associated with this technology. The Adamant was only used during a limited ''test'' on a regular plywood crate due to safety considerations of the tool for this application. The Adamant circular saw, a counter-rotating twin-cutter, constructed with blades that work differently than conventional cutting wheels with twin blades, each rotating in opposite directions. It is used to cut wood and metals. Each blade is approximately 8 3/4 inches in diameter with a maximum cutting depth of 2 1/2 inches. The machine has two rotation speeds: 1,900 and 2,900 rotations per minute (rpm). The saw is operated with an interlocked, guarded trigger switch located at the end of the saw opposite the cutting blades. To operate the saw, the safety interlock must be depressed prior to powering the saw with the trigger control. The saw is supported by a handle at the front of the saw near the cutting blades. The top part of the blades is guarded near the handle, with approximately three-fourths of the face of the blades exposed. The Adamant circular saw is an innovative technology used to cut metals and wood. Its safety features include: interlocking switch for powering the saw, overload indicator and shutoff, and an electronic brake that stops the engine immediately when the start button is released. The top part of the blades is guarded near the motor. With approximately three-fourths of the face of the blades open, the operator is exposed to the potential risk of serious and minor cuts and abrasions when using and handling the saw. There is also potential for damage to the blades if the saw is not stored properly. Without guarding on the lower part of the blades, these can be damaged if the saw is dropped or rested on the cutting blades. Based upon the industrial hygiene sampling conducted for the other four saws demonstrated at FIU, noise levels, nuisance dust, and airborne fiberglass may be a problem when using this technology for the cutting of fiberglass-reinforced plywood crates. No industrial hygiene sampling was conducted while the Adamant saw was in use. Engineering controls should be used to eliminate these problems whenever possible. Where this is not possible, administrative controls, training, and proper personal protective equipment (PPE) should be used. Respirators should be used if engineering controls do not sufficiently control the dust or fiberglass generated. Respirators should be equipped with an organic vapor and acid gas cartridge with High Efficiency Particulate Air (HEPA) filter, since during the demonstration, the workers complained of an odd smell, which may have been the breakdown of the fiberglass.

    Objectives: Project RC-1649, Advanced Chemical Measurement of Smoke from DoD-prescribed Burns was undertaken to use advanced instrumental techniques to study in detail the particulate and vapor-phase chemical composition of the smoke that results from prescribed fires used as a land management tool on DoD bases, particularly bases in the southeastern U.S. The statement of need (SON) called for (1) improving characterization of fuel consumption and (2) improving characterization of air emissions under both flaming and smoldering conditions with respect to volatile organic compounds, heavy metals, and reactive gases. The measurements and fuels were from several bases throughout the southeast (Camp Lejeune, Ft. Benning, and Ft. Jackson) and were carried out in collaboration and conjunction with projects 1647 (models) and 1648 (particulates, SW bases). Technical Approach: We used an approach that featured developing techniques for measuring biomass burning emission species in both the laboratory and field and developing infrared (IR) spectroscopy in particular. Using IR spectroscopy and other methods, we developed emission factors (EF, g of effluent per kg of fuel burned) for dozens of chemical species for several common southeastern fuel types. The major measurement campaigns were laboratory studies at the Missoula Fire Sciences Laboratory (FSL) as well as field campaigns at Camp Lejeune, NC, Ft. Jackson, SC, and in conjunction with 1648 at Vandenberg AFB, and Ft. Huachuca. Comparisons and fusions of laboratory and field data were also carried out, using laboratory fuels from the same bases. Results: The project enabled new technologies and furthered basic science, mostly in the area of infrared spectroscopy, a broadband method well suited to biomass burn studies. Advances in hardware, software and supporting reference data realized a nearly 20x improvement in sensitivity and now provide quantitative IR spectra for potential detection of ~60 new species and actual field quantification of several new species such as nitrous acid, glycolaldehyde, ?-/?-pinene and D-limonene. The new reference data also permit calculation of the global warming potential (GWP) of the greenhouse gases by enabling 1) detection of their ambient concentrations, and 2) quantifying their ability to absorb IR radiation.

  14. Use of Treated Municipal Wastewater as Power Plant Cooling System Makeup Water: Tertiary Treatment versus Expanded Chemical Regimen for Recirculating Water Quality Management

    Treated municipal wastewater is a common, widely available alternative source of cooling water for thermoelectric power plants across the U.S. However, the biodegradable organic matter, ammonia-nitrogen, carbonate and phosphates in the treated wastewater pose challenges with respect to enhanced biofouling, corrosion, and scaling, respectively. The overall objective of this study was to evaluate the benefits and life cycle costs of implementing tertiary treatment of secondary treated municipal wastewater prior to use in recirculating cooling systems. The study comprised bench- and pilot-scale experimental studies with three different tertiary treated municipal wastewaters, and life cycle costing and environmental analyses of various tertiary treatment schemes. Sustainability factors and metrics for reuse of treated wastewater in power plant cooling systems were also evaluated. The three tertiary treated wastewaters studied were: secondary treated municipal wastewater subjected to acid addition for pH control (MWW_pH); secondary treated municipal wastewater subjected to nitrification and sand filtration (MWW_NF); and secondary treated municipal wastewater subjected nitrification, sand filtration, and GAC adsorption (MWW_NFG). Tertiary treatment was determined to be essential to achieve appropriate corrosion, scaling, and biofouling control for use of secondary treated municipal wastewater in power plant cooling systems. The ability to control scaling, in particular, was found to be significantly enhanced with tertiary treated wastewater compared to secondary treated wastewater. MWW_pH treated water (adjustment to pH 7.8) was effective in reducing scale formation, but increased corrosion and the amount of biocide required to achieve appropriate biofouling control. Corrosion could be adequately controlled with tolytriazole addition (4-5 ppm TTA), however, which was the case for all of the tertiary treated waters. For MWW_NF treated water, the removal of ammonia by nitrification helped to reduce the corrosivity and biocide demand. Also, the lower pH and alkalinity resulting from nitrification reduced the scaling to an acceptable level, without the addition of anti-scalant chemicals. Additional GAC adsorption treatment, MWW_NFG, yielded no net benefit. Removal of organic matter resulted in pitting corrosion in copper and cupronickel alloys. Negligible improvement was observed in scaling control and biofouling control. For all of the tertiary treatments, biofouling control was achievable, and most effectively with pre-formed monochloramine (2-3 ppm) in comparison with NaOCl and ClO2. Life cycle cost (LCC) analyses were performed for the tertiary treatment systems studied experimentally and for several other treatment options. A public domain conceptual costing tool (LC3 model) was developed for this purpose. MWW_SF (lime softening and sand filtration) and MWW_NF were the most cost-effective treatment options among the tertiary treatment alternatives considered because of the higher effluent quality with moderate infrastructure costs and the relatively low doses of conditioning chemicals required. Life cycle inventory (LCI) analysis along with integration of external costs of emissions with direct costs was performed to evaluate relative emissions to the environment and external costs associated with construction and operation of tertiary treatment alternatives. Integrated LCI and LCC analysis indicated that three-tiered treatment alternatives such as MWW_NSF and MWW_NFG, with regular chemical addition for treatment and conditioning and/or regeneration, tend to increase the impact costs and in turn the overall costs of tertiary treatment. River water supply and MWW_F alternatives with a single step of tertiary treatment were associated with lower impact costs, but the contribution of impact costs to overall annual costs was higher than all other treatment alternatives. MWW_NF and MWW_SF alternatives exhibited moderate external impact costs with moderate infrastructure and chemical conditioner dosing, which makes them (especially MWW_NF) better treatment alternatives from the environmental sustainability perspective since they exhibited minimal contribution to environmental damage from emissions.

    The structural integrity of reinforced concrete structures in blast events is important for critical facilities. This paper presents experimental data generated for calibrating detailed finite element models that predict the performance of reinforced concrete wall panels with a wide range of construction details under blast loading. The test specimens were 1.2 m square wall panels constructed using Normal Weight Concrete (NWC) or Fiber Reinforced Concrete (FRC). FRC consists of macro-synthetic fibers dispersed in NWC. Five types of panels were tested: NWC panels with steel bar reinforcement (Type A); FRC panels without additional reinforcement (Type B); FRC panels with steel bar reinforcement (Type C); NWC panels with glass fiber reinforced polymer (GFRP) bar reinforcement (Type D); and NWC panels reinforced with steel bar reinforcement and external bidirectional GFRP overlays on both faces (Type E). An additional three Type C panels were used as control specimens (CON). Each panel type was constructed with three thicknesses: 152 mm, 254 mm, and 356 mm. The panels were instrumented with strain gauges, and accelerometers; in addition, pressure sensors and high speed videos were employed during the blast events. Panel types C and E had the best performance, whereas panel type B did not perform well. Preliminary dynamic simulations show crack patterns similar to the experimental results.

    A methodology is presented to calculate the total combined cost (TCC) of water sourcing, water treatment and condenser fouling in the recirculating cooling systems of thermoelectric power plants. The methodology is employed to evaluate the economic viability of using treated municipal wastewater (MWW) to replace the use of freshwater as makeup water to power plant cooling systems. Cost analyses are presented for a reference power plant and five different tertiary treatment scenarios to reduce the scaling tendencies of MWW. Results indicate that a 550 MW sub-critical coal fired power plant with a makeup water requirement of 29.3 ML/day has a TCC of $3.0 - 3.2 million/yr associated with the use of treated MWW for cooling. (All costs USD 2009). This translates to a freshwater conservation cost of $0.29/kL, which is considerably lower than that of dry air cooling technology, $1.5/kL, as well as the 2020 conservation cost target set by the U.S. Department of Energy, $0.74/kL. Results also show that if the available price of freshwater exceeds that of secondary-treated MWW by more than $0.13-0.14/kL, it can be economically advantageous to purchase secondary MWW and treat it for utilization in the recirculating cooling system of a thermoelectric power plant.

    The Nuclear Material Disposition Project group asked the Savannah River National Lab (SRNL) Research and Development Engineering (R&DE) department to help procure, test, and deploy a remote crawler to recover the 2014 Inspection Crawler (IC) that tipped over in the H-Canyon Air Exhaust Tunnel. R&DE wrote a Procurement Specification for a Recovery Crawler (RC) and SRNS Procurement Department awarded the contract to Power Equipment Manufacturing Inc. (PEM). The PEM RC was based on their standard sewer inspection crawler with custom arms and forks added to the front. The arms and forks would be used to upright the 2014 Inspection Crawler. PEM delivered the RC and associated cable reel, 2014 Inspection Crawler mockup, and manuals in late April 2015. R&DE and the team tested the crawler in May of 2015 and made modifications based on test results and Savannah River Site (SRS) requirements. R&DE delivered the RC to H-Area at the end of May. The team deployed the RC on June 9, 10, and 11, 2015 in the H-Canyon Air Exhaust Tunnel. The RC struggled with some obstacles in the tunnel, but eventually made it to the IC. The team spent approximately five hours working to upright the IC and eventually got it on its wheels. The IC travelled approximately 20 feet and struggled to drive over debris on the air tunnel floor. Unfortunately the IC tripped over trying to pass this obstacle. The team decided to leave the IC in this location and inspect the tunnel with the RC. The RC passed the IC and inspected the tunnel as it travelled toward H-Canyon. The team turned the RC around when it was about 20 feet from the H-Canyon crossover tunnel. From that point, the team drove the RC past the manway towards the new sand filter and stopped approximately 20 feet from the new sand filter. The team removed the RC from the tunnel, decontaminated the RC, and stored it the manway building, 294-2H. The RC deployment confirmed the IC was not in a condition to perform useful tunnel inspections and would require significant maintenance to become inspection ready. The RC traveled approximately 660 feet in the tunnel and viewed the tunnel and ceiling wall surfaces that were not blocked by existing ducts. This deployment also documented the tunnel obstacles for future inspections. Overall, the RC deployment was a success.

    The disclosure provides a waste heat recovery system and method in which pressure in a Rankine cycle (RC) system of the WHR system is regulated by diverting working fluid from entering an inlet of an energy conversion device of the RC system. In the system, an inlet of a controllable bypass valve is fluidly coupled to a working fluid path upstream of an energy conversion device of the RC system, and an outlet of the bypass valve is fluidly coupled to the working fluid path upstream of the condenser of the RC system such that working fluid passing through the bypass valve bypasses the energy conversion device and increases the pressure in a condenser. A controller determines the temperature and pressure of the working fluid and controls the bypass valve to regulate pressure in the condenser.

    A combination 2/4-probe method was used to measure electrical resistances across a pure, monolithic CVD-SiC disc sample with contact resistance at the SiC/metallic electrode interfaces. By comparison of the almost simultaneous 2/4-probe measurements, the specific contact resistance (Rc) and its temperature dependence were determined for two types (sputtered gold and porous nickel) electrodes from room temperature (RT) to ?973 K. The Rc-values behaved similarly for each type of metallic electrode: Rc > ?1000 ? cm2 at RT, decreasing continuously to ?110 ? cm2 at 973 K. The temperature dependence of the inverse Rc indicated thermally activated electrical conduction across the SiC/metallic interface with an apparent activation energy of ?0.3 eV. For the flow channel insert application in a fusion reactor blanket, contact resistance potentially could reduce the transverse electrical conductivity by about 50%.

    We have probed a section (l {approx} 150, b {approx} -60) of the trailing tidal arm of the Sagittarius dwarf spheroidal galaxy by identifying a sample of Red Clump (RC) stream stars. RC stars are not generally found in the halo field, but are found in significant numbers in both the Sagittarius galaxy and its tidal streams, making them excellent probes of stream characteristics. Our target sample was selected using photometric data from the Sloan Digital Sky Survey, Data Release 6, which was constrained in color to match the Sagittarius RC stars. Spectroscopic observations of the target stars were conducted at Kitt Peak National Observatory using the WIYN telescope. The resulting spectroscopic sample is magnitude limited and contains both main-sequence disk stars and evolved RC stars. We have developed a method to systematically separate these two stellar classes using kinematic information and a Bayesian approach for surface gravity determination. The resulting RC sample allows us to determine an absolute stellar density of {rho} = 2.7 {+-} 0.5 RC stars kpc{sup -3} at this location in the stream. Future measurements of stellar densities for a variety of populations and at various locations along the streams will lead to a much improved understanding of the original nature of the Sagittarius galaxy and the physical processes controlling its disruption and subsequent stream generation.

    The objective of this project was to develop and evaluate an innovative membrane technology at process conditions that would be representative of Integrated Gasification Combined Cycle (IGCC) advanced power generation with pre-combustion capture of carbon dioxide (CO2). This research focused on hydrogen (H2)-selective zeolite membranes that could be utilized to separate conditioned syngas into H2-rich and CO2-rich components. Both experiments and process design and optimization calculations were performed to evaluate the concept of ultra-thin membranes made from zeolites nanosheets. In this work, efforts in the laboratory were made to tackle two fundamental challenges in application of zeolite membranes in harsh industrial environments, namely, membrane thickness and membrane stability. Conventional zeolite membranes have thicknesses in the micron range, limiting their performance. In this research, we developed a method for fabrication of ultimately thin zeolite membranes based on zeolite nanosheets. A range of layered zeolites (MWW, RWR, NSI structure types) suitable for hydrogen separation was successfully exfoliated to their constituent nanosheets. Further, membranes were made from one of these zeolites, MWW, to demonstrate the potential of this group of materials. Moreover, long-term steam stability of these zeolites (up to 6 months) was investigated in high concentrations of steam (35 mol% and 95 mole%), high pressure (10 barg), and high temperatures (350 °C and 600 °C) relevant to conditions of water-gas-shift and steam methane reforming reactions. It was found that certain nanosheets are stable, and that stability depends on the concentration of structural defects. Additionally, models that represent a water-gas-shift (WGS) membrane reactor equipped with the zeolite membrane were developed for systems studies. These studies had the aim of analyzing the effect of the membrane reactor integration into IGCC plants in terms of performance and economic aspects of the plants. Specifically, simulation and design optimization studies were performed using the developed stand-alone membrane reactor models to identify the membrane selectivity and permeance characteristics necessary to achieve desired targets of CO2 capture and H2 recovery, as well as guide the selection of the optimal reactor design that minimizes the membrane cost as a function of its surface area required. The isothermal membrane reactor model was also integrated into IGCC system models using both the MATLAB and Aspen software platforms and techno-economic analyses of the integrated plants have been carried out to evaluate the feasibility of replacing current technologies for pre-combustion capture by the proposed novel approach in terms of satisfying stream constraints and achieving the DOE target goal of 90% CO2 capture. The results of the performed analyses based on present value of annuity calculations showed break even costs for the membrane reactor within the feasible range for membrane fabrication. However, the predicted membrane performance used in these simulations exceeded the performance achieved experimentally. Therefore, further work is required to improve membrane performance.

    In this work the seismic behavior of masonry buildings with mixed structural system, consisting of perimeter masonry walls and internal r.c. frames, is studied by means of non linear static (pushover) analyses. Several aspects, like the distribution of seismic action between masonry and r.c. elements, the local and global behavior of the structure, the crisis of the connections and the attainment of the ultimate strength of the whole structure are examined. The influence of some parameters, such as the masonry compressive and tensile strength, on the structural behavior is investigated. The numerical analyses are also repeated on a building in which the r.c. internal frames are replaced with masonry walls.

  10. Effects of Contact Resistance on Electrical Conductivity Measurements of SiC-Based Materials

    A combination 2/4-probe method was used to measure electrical resistances across a pure, monolithic CVD-SiC disc sample with contact resistance at the SiC/metallic electrode interfaces. By comparison of the almost simultaneous 2/4-probe measurements, the specific contact resistance and its temperature dependence were determined for two types (sputtered gold and porous nickel) electrodes from RT to ~700C. The specific contact resistance values (Rc) behaved similarly for each type of metallic electrode: Rc >~1000 ?-cm2 at RT, decreasing continuously to ~1-10 ?-cm2 at 700C. The temperature dependence of the inverse Rc indicated thermally activated electrical conduction across the SiC/metallic interface with an apparent activation energy of ~0.3 eV. For the flow channel insert application in a fusion reactor blanket, contact resistance potentially could reduce the transverse electrical conductivity by ~1/2.

    Purpose: This study aims to correlate thymidylate synthase (TS) gene polymorphisms with the tumor response to preoperative 5-fluorouracil (5-FU)-based chemoradiation therapy (CRT) in patients with rectal cancer. Methods and Materials: Forty-four patients with rectal cancer treated with 5-FU-based preoperative CRT were prospectively enrolled in this study. Thymidylate synthase expression and TS gene polymorphisms were evaluated in tumor obtained before preoperative CRT and were correlated with the pathologic response, as assessed by histopathologic staging (pTNM) and tumor regression grade. Results: Patients exhibited 2R/3R and 3R/3R tandem repeat polymorphisms in the TS gene. With regard to TS expression in these genotypes, 2R/3RC and 3RC/3RC were defined as the low-expression group and 2R/3RG, 3RC/3RG, and 3RG/3RG as the high-expression group. There was no significant correlation between TS expression and tumor response. There was no significant difference in the tumor response between patients homozygous for 3R/3R and patients heterozygous for 2R/3R. However, 13 of 14 patients in the low-expression group with a G>C single-nucleotide polymorphism (SNP) (2R/3RC [n = 5] or 3RC/3RC [n = 9]) exhibited a significantly greater tumor downstaging rate, as compared with only 12 of 30 patients in the high-expression group without the SNP (2R/3RG [n = 10], 3RC/3RG [n = 9], or 3RG/3RG [n = 11]) (p = 0.001). The nodal downstaging rate was also significantly greater in this low-expression group, as compared with the high-expression group (12 of 14 vs. 14 of 30, p = 0.014). However, there was no significant difference in the tumor regression grade between these groups. Conclusions: This study suggests that SNPs within the TS enhancer region affect the tumor response to preoperative 5-FU-based CRT in rectal cancer.

    An anticoincidence device is described for a pair of adjacent channels of a multi-channel pulse height analyzer for preventing the lower channel from generating a count pulse in response to an input pulse when the input pulse has sufficient magnitude to reach the upper level channel. The anticoincidence circuit comprises a window amplifier, upper and lower level discriminators, and a biased-off amplifier. The output of the window amplifier is coupled to the inputs of the discriminators, the output of the upper level discriminator is connected to the resistance end of a series R-C network, the output of the lower level discriminator is coupled to the capacitance end of the R-C network, and the grid of the biased-off amplifier is coupled to the junction of the R-C network. In operation each discriminator produces a negative pulse output when the input pulse traverses its voltage setting. As a result of the connections to the R-C network, a trigger pulse will be sent to the biased-off amplifier when the incoming pulse level is sufficient to trigger only the lower level discriminator.

    This paper utilizes cyclodextrin-based host-guest chemistry in a microfluidic device to modulate the crystallization of membrane proteins and the process of concentration of membrane protein samples. Methyl-{beta}-cyclodextrin (MBCD) can efficiently capture a wide variety of detergents commonly used for the stabilization of membrane proteins by sequestering detergent monomers. Reaction Center (RC) from Blastochloris viridis was used here as a model system. In the process of concentrating membrane protein samples, MBCD was shown to break up free detergent micelles and prevent them from being concentrated. The addition of an optimal amount of MBCD to the RC sample captured loosely bound detergent from the protein-detergent complex and improved sample homogeneity, as characterized by dynamic light scattering. Using plug-based microfluidics, RC crystals were grown in the presence of MBCD, giving a different morphology and space group than crystals grown without MBCD. The crystal structure of RC crystallized in the presence of MBCD was consistent with the changes in packing and crystal contacts hypothesized for removal of loosely bound detergent. The incorporation of MBCD into a plug-based microfluidic crystallization method allows efficient use of limited membrane protein sample by reducing the amount of protein required and combining sparse matrix screening and optimization in one experiment. The use of MBCD for detergent capture can be expanded to develop cyclodextrin-derived molecules for fine-tuned detergent capture and thus modulate membrane protein crystallization in an even more controllable way.

    In this article we report a detailed study of resorcinol-formaldehyde (RF) aerogels prepared under different processing conditions, [resorcinol]/[catalyst] (R/C) ratios in the starting sol-gel solutions, using continuous flow hyperpolarized 129Xe NMR in combination with solid-state 13C and two-dimensional wide-line separation (2D-WISE) NMR techniques. The degree of polymerization and the mobility of the cross-linking functional groups in RF aerogels are examined and correlated with the R/C ratios. The origin of different adsorption regions is evaluated using both co-adsorption of chloroform and 2D EXSY 129Xe NMR. A hierarchical set of Xe exchange processes in RF aerogels is found using 2D EXSY 129Xe NMR. The exchange of Xe gas follows the sequence (from fastest to slowest): mesopores with free gas, gas in meso- and micro-pores, free gas with micropores, and, finally, among micropore sites. The volume-to-surface-area (Vg/S) ratios for aerogels are measured for the first time without the use of geometric models. The Vg/S parameter, which is related both to the geometry and the interconnectivity of the pore space, has been found to correlate strongly with the R/C ratio and exhibits an unusually large span: an increase in the R/C ratio from 50 to 500 results in about a 5-fold rise in Vg/S.

    A pulse processing system (10) for use in an X-ray spectrometer in which a ain channel pulse shaper (12) and a fast channel pulse shaper (13) each produce a substantially symmetrical triangular pulse (f, p) for each event detected by the spectrometer, with the pulse width of the pulses being substantially independent of the magnitude of the detected event and with the pulse width of the fast pulses (p) being substantially shorter than the pulse width of the main channel pulses (f). A pile-up rejector circuit (19) allows output pulses to be generated, with amplitudes linearly related to the magnitude of the detected events, whenever the peak of a main channel pulse (f) is not affected by a preceding or succeeding main channel pulse, while inhibiting output pulses wherein peak magnitudes of main channel pulses are affected by adjacent pulses. The substantially symmetrical triangular main channel pulses (f) are generated by the weighted addition (27-31) of successive RC integrations (24, 25, 26) of an RC differentiated step wave (23). The substantially symmetrical triangular fast channel pulses (p) are generated by the RC integration ( 43) of a bipolar pulse (o) in which the amplitude of the second half is 1/e that of the first half, with the RC time constant of integration being equal to one-half the width of the bipolar pulse.

    GE Lighting Solutions will develop precise and efficient manufacturing techniques for the remote phosphor platform of warm-white LED products. In volume, this will be demonstrated to drive significant materials, labor and capital productivity to achieve a maximum possible 53% reduction in overall cost. In addition, the typical total color variation for these white LEDs in production will be well within the ANSI bins and as low as a 4-step MacAdam ellipse centered on the black body curve. Achievement of both of these objectives will be demonstrated while meeting a performance target of > 75 lm/W for a warm-white LED and a reliability target of <30% lumen drop / <2-step MacAdam ellipse shift, estimated over 50,000 hrs.
