    The Raft River Geothermal Site has been evaluated over the past eight years by the United States Geological Survey and the Idaho National Engineering Laboratory as a moderate-temperature geothermal resource. The geoscience data gathered in the drilling and testing of seven geothermal wells suggest that the Raft River thermal reservoir is: (a) produced from fractures found at the contact metamorphic zone, apparently the base of detached normal faulting from the Bridge and Horse Well Fault zones of the Jim Sage Mountains; (b) anisotropic, with the major axis of hydraulic conductivity coincident to the Bridge Fault Zone; (c) hydraulically connected to the shallow thermal fluid of the Crook and BLM wells based upon both geochemistry and pressure response; (d) controlled by a mixture of diluted meteoric water recharging from the northwest and a saline sodium chloride water entering from the southwest. Although the hydrogeologic environment of the Raft River geothermal area is very complex and unique, it is typical of many Basin and Range systems.

    The Raft River Geothermal Site is located in Southern Idaho's Raft River Valley, southwest of Malta, Idaho, in Cassia County. EG and G idaho, Inc., is the DOE's prime contractor for development of the Raft River geothermal field. Contract work has been progressing for several years towards creating a fully integrated utilization of geothermal water. Developmental progress has resulted in the drilling of seven major DOE wells. Four are producing geothermal water from reservoir temperatures measured to approximately 149 C (approximately 300 F). Closed-in well head pressures range from 69 to 102 kPa (100 to 175 psi). Two wells are scheduled for geothermal cold 60 C (140 F) water reinjection. The prime development effort is for a power plant designed to generate electricity using the heat from the geothermal hot water. The plant is designated as the ''5 MW(e) Raft River Research and Development Plant'' project. General site management assigned to EG and G has resulted in planning and development of many parts of the 5 MW program. Support and development activities have included: (1) engineering design, procurement, and construction support; (2) fluid supply and injection facilities, their study, and control; (3) development and installation of transfer piping systems for geothermal water collection and disposal by injection; and (4) heat exchanger fouling tests.

    Three conservative tracer tests have been conducted through the Bridge Fault fracture zone at the Raft River Geothermal (RRG) site. All three tests were conducted between injection well RRG-5 and production wells RRG-1 (790 m distance) and RRG-4 (740 m distance). The injection well is used during the summer months to provide pressure support to the production wells. The first test was conducted in 2008 using 136 kg of fluorescein tracer. Two additional tracers were injected in 2010. The first 2010 tracer injected was 100 kg fluorescein disodium hydrate salt on June, 21. The second tracer (100 kg 2,6-naphthalene disulfonic acid sodium salt) was injected one month later on July 21. Sampling of the two productions wells is still being performed to obtain the tail end of the second 2010 tracer test. Tracer concentrations were measured using HPLC with a fluorescence detector. Results for the 2008 test, suggest 80% tracer recover at the two production wells. Of the tracer recovered, 85% of tracer mass was recovered in well RRG-4 indicating a greater flow pathway connection between injection well and RRG-4 than RRG-1. Fluorescein tracer results appear to be similar between the 2008 and 2010 tests for well RRG-4 with peak concentrations arriving approximately 20 days after injection despite the differences between the injection rates for the two tests (~950 gpm to 475 gpm) between the 2008 and 2010. The two 2010 tracer tests will be compared to determine if the results support the hypothesis that rock contraction along the flow pathway due to the 55 oC cooler water injection alters the flow through the ~140 oC reservoir.

    We discovered a previously unobserved mechanism by which air bubbles detach from vibrating walls in glasses containing water. Chaotic oscillation and subsequent water jets appeared when a wall vibrated at greater than a critical level. Wave forms were developed at water-air interface of the bubble by the wall vibration, and water jets were formed when sufficiently grown wave-curvatures were collapsing. Droplets were pinched off from the tip of jets and fell to the surface of the glass. When the solid-air interface at the bubble-wall attachment point was completely covered with water, the bubble detached from the wall. The water jets were mainly generated by subharmonic waves and were generated most vigorously when the wall vibrated at the volume resonant frequency of the bubble. Bubbles of specific size can be removed by adjusting the frequency of the wall's vibration.

The influence of these different aged microstructures on thermomechanical fatigue (TMF) under either continuously cycled (CC) and creep-fatigue (CF) was investigated.     The origin of low-angle normal faults or detachment faults mantling metamorphic core complexes in the southwestern United States remains controversial. If [sigma][sub 1] is vertical during extension, the formation of, or even slip along, such low-angle normal faults is mechanically implausible. No records exist of earthquakes on low-angle normal faults in areas currently undergoing continental extension, except from an area of actively forming core complexes in the Solomon Sea, Papua New Guinea. In light of such geophysical and mechanical arguments, W.R. Buck and B. Wernicke and G.J. Axen proposed models in which detachment faults originate as high-angle normal faults, but rotate to low angles and become inactive as extension proceeds. These models are inconsistent with critical field relations in several core complexes. The Rawhide fault, an areally extensive detachment fault in western Arizona, propagated at close to its present subhorizontal orientation late in the Tertiary extension of the region. Neither the Wernicke and Axen nor Buck models predict such behavior; in fact, both models preclude the operation of low-angle normal faults. The authors recommend that alternative explanations or modifications of existing models are needed to explain the evidence that detachment faults form and operate with gentle dips.

  10. Creep of CMSX-4 superalloy single crystals: Effects of rafting at high temperature

    The creep performance of (001)-orientated CMSX-4 superalloy single crystals at temperatures beyond 1000 C is analyzed. Rafting of the {gamma}{prime} structure occurs rapidly, e.g., for the 1150 C/100 MPa tests rafting is completed within the first 10 h. At this stage and for a considerable time thereafter the creep strain rate decreases with increasing strain, implying a creep hardening effect which is absent at lower temperatures when the kinetics of rafting is less rapid. Once a critical strain {epsilon}* of (0.7 {+-} 0.3)% is reached, the creep strain increases dramatically and failure occurs within a few tens of hours. It is demonstrated that methods of interpretation which, assume a proportionality between the creep strain rate and creep strain, are unable to account for creep hardening which occurs as a consequence of rafting. A modification is proposed which accounts for the blocking of the glide/climb of {l{underscore}brace}111{r{underscore}brace}{l{underscore}angle}1{bar 1}0{r{underscore}angle} creep dislocations which occurs as the number of vertical {gamma} channels is reduced and cellular dislocation networks become stabilized. Consequently, failure must be taken to be associated with creep cavitation, which occurs predominantly around casting porosity. It is emphasized that more work is required to quantify the interaction between the various creep damage mechanisms.

    It has been suggested that cholesterol may modulate amyloid-{beta} (A{beta}) formation, a causative factor of Alzheimer's disease (AD), by regulating distribution of the three key proteins in the pathogenesis of AD ({beta}-amyloid precursor protein (APP), {beta}-secretase (BACE1) and/or presenilin 1 (PS1)) within lipid rafts. In this work we tested whether cholesterol accumulation upon NPC1 dysfunction, which causes Niemann Pick type C disease (NPC), causes increased partitioning of APP into lipid rafts leading to increased CTF/A{beta} formation in these cholesterol-rich membrane microdomains. To test this we used CHO NPC1{sup -/-} cells (NPC cells) and parental CHOwt cells. By sucrose density gradient centrifugation we observed a shift in fl-APP/CTF compartmentalization into lipid raft fractions upon cholesterol accumulation in NPC vs. wt cells. Furthermore, {gamma}-secretase inhibitor treatment significantly increased fl-APP/CTF distribution in raft fractions in NPC vs. wt cells, suggesting that upon cholesterol accumulation in NPC1-null cells increased formation of APP-CTF and its increased processing towards A{beta} occurs in lipid rafts. Our results support that cholesterol overload, such as in NPC disease, leads to increased partitioning of APP/CTF into lipid rafts resulting in increased amyloidogenic processing of APP in these cholesterol-rich membranes. This work adds to the mechanism of the cholesterol-effect on APP processing and the pathogenesis of Alzheimer's disease and supports the role of lipid rafts in these processes.

    Tunneling nanotubes (TnTs) are long, non-adherent, actin-based cellular extensions that act as conduits for transport of cellular cargo between connected cells. The mechanisms of nanotube formation and the effects of the tumor microenvironment and cellular signals on TnT formation are unknown. In the present study, we explored exosomes as potential mediators of TnT formation in mesothelioma and the potential relationship of lipid rafts to TnT formation. Mesothelioma cells co-cultured with exogenous mesothelioma-derived exosomes formed more TnTs than cells cultured without exosomes within 2448 h; and this effect was most prominent in media conditions (low-serum, hyperglycemic medium) that support TnT formation (1.31.9-fold difference). Fluorescence and electron microscopy confirmed the purity of isolated exosomes and revealed that they localized predominantly at the base of and within TnTs, in addition to the extracellular environment. Time-lapse microscopic imaging demonstrated uptake of tumor exosomes by TnTs, which facilitated intercellular transfer of these exosomes between connected cells. Mesothelioma cells connected via TnTs were also significantly enriched for lipid rafts at nearly a 2-fold higher number compared with cells not connected by TnTs. Our findings provide supportive evidence of exosomes as potential chemotactic stimuli for TnT formation, and also lipid raft formation as a potential biomarker for TnT-forming cells. - Highlights: Exosomes derived from malignant cells can stimulate an increased rate in the formation of tunneling nanotubes. Tunneling nanotubes can serve as conduits for intercellular transfer of these exosomes. Most notably, exosomes derived from benign mesothelial cells had no effect on nanotube formation. Cells forming nanotubes were enriched in lipid rafts at a greater number compared with cells not forming nanotubes.   20. Folding associated with extensional faulting: Sheep Range detachment, southern Nevada

    The Sheep Range detachment is a major Miocene extensional fault system of the Great Basin. Its major faults have a scoop shape, with straight, N-S traces extending 15-30 km and then abruptly turning to strike E-W. Tertiary deformation involved simultaneous normal faulting, sedimentation, landsliding, and strike-slip faulting. Folds occur in two settings: landslide blocks and drag along major faults. Folds occur in landslide blocks and beneath them. Most folds within landslide blocks are tight anticlines, with limbs dipping 40-60 degrees. Brecciation of the folds and landslide blocks suggests brittle deformation. Near Quijinump Canyon in the Sheep Range, at least three landslide blocks (up to 500 by 1500 m) slid into a small Tertiary basin. Tertiary limestone beneath the Paleozoic blocks was isoclinally folded. Westward dips reveal drag folds along major normal faults, as regional dips are consistently to the east. The Chowderhead anticline is the largest drag fold, along an extensional fault that offsets Ordovician units 8 km. East-dipping Ordovician and Silurian rocks in the Desert Range form the hanging wall. East-dipping Cambrian and Ordovician units in the East Desert Range form the foot wall and east limb of the anticline. Caught along the fault plane, the anticline's west-dipping west limb contains mostly Cambrian units.

    Occludin is an essential integral transmembrane protein regulating tight junction (TJ) integrity in brain endothelial cells. Phosphorylation of occludin is associated with its localization to TJ sites and incorporation into intact TJ assembly. The present study is focused on the role of lipid rafts in polychlorinated biphenyl (PCB)-induced disruption of occludin and endothelial barrier function. Exposure of human brain endothelial cells to 2,2′,4,4′,5,5′-hexachlorobiphenyl (PCB153) induced dephosphorylation of threonine residues of occludin and displacement of occludin from detergent-resistant membrane (DRM)/lipid raft fractions within 1 h. Moreover, lipid rafts modulated the reduction of occludin level through activation of matrix metalloproteinase 2 (MMP-2) after 24 h PCB153 treatment. Inhibition of protein phosphatase 2A (PP2A) activity by okadaic acid or fostriecin markedly protected against PCB153-induced displacement of occludin and increased permeability of endothelial cells. The implication of lipid rafts and PP2A signaling in these processes was further defined by co-immunoprecipitation of occludin with PP2A and caveolin-1, a marker protein of lipid rafts. Indeed, a significant MMP-2 activity was observed in lipid rafts and was increased by exposure to PCB153. The pretreatment of MMP-2 inhibitors protected against PCB153-induced loss of occludin and disruption of lipid raft structure prevented the increase of endothelial permeability. Overall, these results indicate that lipid raft-associated processes, such as PP2A and MMP-2 activation, participate in PCB153-induced disruption of occludin function in brain endothelial barrier.     Angiotensin II (ANG-II) receptors (AGTRs) contribute to renal physiology and pathophysiology, but the underlying mechanisms that regulate AGTR function in glomerular mesangium are poorly understood. Here, we show that AGTR1 is the functional AGTR subtype expressed in neonatal pig glomerular mesangial cells (GMCs). Cyclodextrin (CDX)-mediated cholesterol depletion attenuated cell surface AGTR1 protein expression and ANG-II-induced intracellular Ca{sup 2+} ([Ca{sup 2+}]{sub i}) elevation in the cells. The COOH-terminus of porcine AGTR1 contains a caveolin (CAV)-binding motif. However, neonatal GMCs express CAV-1, but not CAV-2 and CAV-3. Colocalization and in situ proximity ligation assay detected an association between endogenous AGTR1 and CAV-1 in the cells. A synthetic peptide corresponding to the CAV-1 scaffolding domain (CSD) sequence also reduced ANG-II-induced [Ca{sup 2+}]{sub i} elevation in the cells. Real-time imaging of cell growth revealed that ANG-II stimulates neonatal GMC proliferation. ANG-II-induced GMC growth was attenuated by EMD 66684, an AGTR1 antagonist; BAPTA, a [Ca{sup 2+}]{sub i} chelator; KN-93, a Ca{sup 2+}/calmodulin-dependent protein kinase II inhibitor; CDX; and a CSD peptide, but not PD 123319, a selective AGTR2 antagonist. Collectively, our data demonstrate [Ca{sup 2+}]{sub i}-dependent proliferative effect of ANG-II and highlight a critical role for lipid raft microdomains in AGTR1-mediated signal transduction in neonatal GMCs. - Highlights: AGTR1 is the functional AGTR subtype expressed in neonatal mesangial cells. Endogenous AGTR1 associates with CAV-1 in neonatal mesangial cells. Lipid raft disruption attenuates cell surface AGTR1 protein expression.     We show that the selective localization of cholesterol-rich domains and associated ganglioside receptors prefer to occur in the monolayer across continuous monolayer-bilayer junctions (MBJs) in supported lipid membranes. For the MBJs, glass substrates were patterned with poly(dimethylsiloxane) (PDMS) oligomers by thermally-assisted contact printing, leaving behind 3 nm-thick PDMS patterns. The hydrophobicity of the transferred PDMS patterns was precisely tuned by the stamping temperature. Lipid monolayers were formed on the PDMS patterned surface while lipid bilayers were on the bare glass surface. Due to the continuity of the lipid membranes over the MBJs, essentially free diffusion of lipids was allowed betweenmore » the monolayer on the PDMS surface and the upper leaflet of the bilayer on the glass substrate. The preferential localization of sphingomyelin, ganglioside GM1 and cholesterol in the monolayer region enabled to develop raft microdomains through coarsening of nanorafts. Furthermore, our methodology provides a simple and effective scheme of non-disruptive manipulation of the chemical landscape associated with lipid phase separations, which leads to more sophisticated applications in biosensors and as cell culture substrates.« less

    A critical issue for optimal water management in proton-exchange-membrane fuel cells at lower temperatures is the removal of liquid water from the cell. This pathway is intimately linked with the phenomena of liquid-water droplet removal from surface of the gas-diffusion layer and into the flow channel. Thus, a good understanding of liquid-water transport and droplet growth and detachment from the gas-diffusion layer is critical. In this study, liquid-water droplet growth and detachment on the gas-diffusion layer surfaces are investigated experimentally to improve the understating of water transport through and removal from gas-diffusion layers. An experiment using a sliding-angle measurement is designed and used to quantify and directly measure the adhesion force for liquid-water droplets, and to understand the droplets? growth and detachment from the gas-diffusion layers.

    DDT is a highly lipophilic molecule known to deplete membrane rafts of their phosphoglycolipid and cholesterol contents. However, we have recently shown that DDT can also alter the thyroid homeostasis by inhibiting TSH receptor (TSHr) internalization. The present study was undertaken to verify whether DDT goitrogenic effects are due to the insecticide acting directly on TSHr or via alteration of the membrane rafts hosting the receptor itself. Our results demonstrate that, in CHO-TSHr transfected cells, TSHr is activated in the presence of TSH, while it is inhibited following DDT exposure. DDT can also reduce the endocytic vesicular traffic, alter the extension of multi-branched microvilli along their plasma membranes and induce TSHr shedding in vesicular forms. To verify whether TSHr displacement might depend on DDT altering the raft constitution of CHO-TSHr cell membranes the extent of TSHr and lipid raft co-localization was examined by confocal microscopy. Evidence shows that receptor/raft co-localization increased significantly upon exposure to TSH, while receptors and lipid rafts become dislodged on opposite cell poles in DDT-exposed CHO-TSHr cells. As a control, under similar culturing conditions, diphenylethylene, which is known to be a lipophilic substance that is structurally related to DDT, did not affect the extent of TSHr and lipid raft co-localization in CHO-TSHr cells treated with TSH. These findings corroborate and extend our view that, in CHO cells, the DDT disrupting action on TSHr is primarily due to the insecticide acting on membranes to deplete their raft cholesterol content, and that the resulting inhibition on TSHr internalization is due to receptor dislodgement from altered raft microdomains of the plasma membrane. - Highlights: >DDT is a pesticide with a severe environmental impact >Epidemiologic correlation exists between exposition to DDT and thyroid dysfunction >DDT is a lipophilic molecule that has been shown to inhibit TSH receptor

    This paper presents a critical assessment of the theory of photo-detachment diagnostic method used to probe the negative ion density and electronegativity α = n{sub -}/n{sub e}. In this method, a laser pulse is used to photo-detach all negative ions located within the electropositive channel (laser spot region). The negative ion density is estimated based on the assumption that the increase of the current collected by an electrostatic probe biased positively to the plasma is a result of only the creation of photo-detached electrons. In parallel, the background electron density and temperature are considered as constants during this diagnostics. While the numerical experiments performed here show that the background electron density and temperature increase due to the formation of an electrostatic potential barrier around the electropositive channel. The time scale of potential barrier rise is about 2 ns, which is comparable to the time required to completely photo-detach the negative ions in the electropositive channel (∼3 ns). We find that neglecting the effect of the potential barrier on the background plasma leads to an erroneous determination of the negative ion density. Moreover, the background electron velocity distribution function within the electropositive channel is not Maxwellian. This is due to the acceleration of these electrons through the electrostatic potential barrier. In this work, the validity of the photo-detachment diagnostic assumptions is questioned and our results illustrate the weakness of these assumptions.

  1. Spatially resolved spectroscopy of detached recombining plasmas in the University of Manchester Linear System divertor simulator

    The University of Manchester Linear System (ULS) [M. G. Rusbridge et al., Plasma Phys. Controlled Fusion 42, 588 (2000)] is an experiment in which a steady-state plasma stream is confined along a longitudinal magnetic field. The plasma passes through a tapered orifice into a separate gas target chamber (GTC) where it interacts with neutral gas at pressures of up to 15 mTorr. The upstream plasma beam is 6-14 mm in diameter with electron temperatures between 2 and 15 eV, and densities in the range of 10{sup 17}-10{sup 19} m{sup -3}. The primary aim of this study is to investigate physical processes relevant to gas target divertors in tokamaks. Upstream parameters, in terms of electron temperature and density, can be varied in the ULS enabling the investigation of electron-ion recombination (EIR) or molecular-activated recombination (MAR) processes. Detached recombining plasmas have been studied in the GTC using an axially scanning spectroscopic system with a spatial resolution of less than 5 mm. We have investigated two distinct plasma regimes having the same electron temperature upstream, but upstream densities that differ by an order of magnitude. Vibrationally excited molecules, which are a necessary prerequisite for MAR, have been detected in both these cases. In the higher density case, which ultimately detaches via EIR processes, these excited molecules are present upstream of the EIR region.

    A detachable current and voltage sensor provides an isolated and convenient device to measure current passing through a conductor such as an AC branch circuit wire, as well as providing an indication of an electrostatic potential on the wire, which can be used to indicate the phase of the voltage on the wire, and optionally a magnitude of the voltage. The device includes a housing formed from two portions that mechanically close around the wire and that contain the current and voltage sensors. The current sensor is a ferrite cylinder formed from at least three portions that form the cylinder when the sensor is closed around the wire with a hall effect sensor disposed in a gap between two of the ferrite portions along the circumference to measure current. A capacitive plate or wire is disposed adjacent to, or within, the ferrite cylinder to provide the indication of the voltage.

    In this study, self-detached nanoporous titanium-based metal oxide was synthesized for the first time by ultrafast anodization in a fluoride-free electrolyte containing 10% HNO{sub 3}. The nanoporous oxide has through-holes with diameters ranging from 10 to 60 nm. The as-formed oxides are amorphous, and were transformed to crystalline structures by annealing. The performance of a dye sensitized solar cell using nanoporpous Ti–10Zr oxide (TZ10) was further studied. It was found that the TZ10 film could increase both the short-circuit current and the open-circuit photovoltage of the solar cell. The overall efficiency of the solar cell was 6.99%, an increase of 20.7% as compared to that using a pure TiO{sub 2} (P25) film. - Graphical abstract: The nanoporous Ti–xZr(x=10, 30) oxide layers are fabricated by anodizing in a dilute nitric acid solvent. The power conversion efficiency of the DSSC by a covering of a Ti–10Zr thin film is increased by 20.7%, with an η of 7.69% , a short circuit current of 12.4 mA/cm{sup 2}, a open circuit voltage of 0.833 V, and a fill factor of 0.679. - Highlights: • Self-detached nanoporous titanium-based metal (TiZr) oxide was synthesized. • The TiZr oxides have through-hole nanopores with diameters ranging from 10 to 60 nm. • The nanoporous Ti–10Zr oxide can improve the power conversion efficiency of a DSSC.

  4. Numerical simulations used for a validity check on the laser induced photo-detachment diagnostic method in electronegative plasmas

    Laser photo-detachment is used as a method to measure or determine the negative ion density and temperature in electronegative plasmas. In essence, the method consists of producing an electropositive channel (negative ion free region) via pulsed laser photo-detachment within an electronegative plasma bulk. Electrostatic probes placed in this channel measure the change in the electron density. A second pulse might be used to track the negative ion recovery. From this, the negative ion density and temperature can be determined. We study the formation and relaxation of the electropositive channel via a two-dimensional Particle-In-Cell/Mote Carlo collision model. The simulation is mainly carried out in a Hydrogen plasma with an electronegativity of α = 1, with a parametric study for α up to 20. The temporal and spatial evolution of the plasma potential and the electron densities shows the formation of a double layer (DL) confining the photo-detached electrons within the electropositive channel. This DL evolves into two fronts that move in the opposite directions inside and outside of the laser spot region. As a consequence, within the laser spot region, the background and photo-detached electron energy distribution function relaxes/thermalizes via collisionless effects such as Fermi acceleration and Landau damping. Moreover, the simulations show that collisional effects and the DL electric field strength might play a non-negligible role in the negative ion recovery within the laser spot region, leading to a two-temperature negative ion distribution. The latter result might have important effects in the determination of the negative ion density and temperature from laser photo detachment diagnostic.

    Populations of Yellowstone cutthroat trout Oncorhynchus clarkii bouvierii have declined throughout their native range as a result of habitat fragmentation, overharvest, and introductions of nonnative trout that have hybridized with or displaced native populations. The degree to which these factors have impacted the current genetic population structure of Yellowstone cutthroat trout populations is of primary interest for their conservation. In this study, we examined the genetic diversity and genetic population structure of Yellowstone cutthroat trout in Idaho and Nevada with data from six polymorphic microsatellite loci. A total of 1,392 samples were analyzed from 45 sample locations throughout 11 major river drainages. We found that levels of genetic diversity and genetic differentiation varied extensively. The Salt River drainage, which is representative of the least impacted migration corridors in Idaho, had the highest levels of genetic diversity and low levels of genetic differentiation. High levels of genetic differentiation were observed at similar or smaller geographic scales in the Portneuf River, Raft River, and Teton River drainages, which are more altered by anthropogenic disturbances. Results suggested that Yellowstone cutthroat trout are naturally structured at the major river drainage level but that habitat fragmentation has altered this structuring. Connectivity should be restored via habitat restoration whenever possible to minimize losses in genetic diversity and to preserve historical processes of gene flow, life history variation, and metapopulation dynamics. However, alternative strategies for management and conservation should also be considered in areas where there is a strong likelihood of nonnative invasions or extensive habitat fragmentation that cannot be easily ameliorated.

    Exposure to persistent organic pollutants, such as polychlorinated biphenyls (PCBs), can lead to chronic inflammation and the development of vascular diseases. Because cell adhesion molecules (CAMs) of the cerebrovascular endothelium regulate infiltration of inflammatory cells into the brain, we have explored the molecular mechanisms by which ortho-substituted polychlorinated biphenyls (PCBs), such as PCB153, can upregulate CAMs in brain endothelial cells. Exposure to PCB153 increased expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1), as well as elevated adhesion of leukocytes to brain endothelial cells. These effects were impeded by inhibitors of EGFR, JAKs, or Src activity. In addition, pharmacological inhibition of NADPH oxidase or disruption of lipid rafts by cholesterol depleting agents blocked PCB153-induced phosphorylation of JAK and Src kinases and upregulation of CAMs. In contrast, silencing of caveolin-1 by siRNA interference did not affect upregulation of ICAM-1 and VCAM-1 in brain endothelial cells stimulated by PCB153. Results of the present study indicate that lipid raft-dependent NADPH oxidase/JAK/EGFR signaling mechanisms regulate the expression of CAMs in brain endothelial cells and adhesion of leukocytes to endothelial monolayers. Due to its role in leukocyte infiltration, induction of CAMs may contribute to PCB-induced cerebrovascular disorders and neurotoxic effects in the CNS.

    We have developed a high speed, miniature scanning probe microscope (MSPM) integrated with a Positioning Unit (PU) for accurately positioning the MSPM on a large substrate. This combination enables simultaneous, parallel operation of many units on a large sample for high throughput measurements. The size of the MSPM is 19 × 45 × 70 mm{sup 3}. It contains a one-dimensional flexure stage with counter-balanced actuation for vertical scanning with a bandwidth of 50 kHz and a z-travel range of more than 2 μm. This stage is mechanically decoupled from the rest of the MSPM by suspending it on specific dynamically determined points. The motion of the probe, which is mounted on top of the flexure stage is measured by a very compact optical beam deflection (OBD). Thermal noise spectrum measurements of short cantilevers show a bandwidth of 2 MHz and a noise of less than 15 fm/Hz{sup 1/2}. A fast approach and engagement of the probe to the substrate surface have been achieved by integrating a small stepper actuator and direct monitoring of the cantilever response to the approaching surface. The PU has the same width as the MSPM, 45 mm and can position the MSPM to a pre-chosen position within an area of 275×30 mm{sup 2} to within 100 nm accuracy within a few seconds. During scanning, the MSPM is detached from the PU which is essential to eliminate mechanical vibration and drift from the relatively low-resonance frequency and low-stiffness structure of the PU. Although the specific implementation of the MSPM we describe here has been developed as an atomic force microscope, the general architecture is applicable to any form of SPM. This high speed MSPM is now being used in a parallel SPM architecture for inspection and metrology of large samples such as semiconductor wafers and masks.

    Clustering an anion with one or more neutral molecules is a stabilizing process that enhances the oxidation potential of the complex relative to the free ion. Several hydrogen bond clusters (i.e., A HX, where A = H2PO4 and CF3CO2 and HX = MeOH, PhOH, and Me2NOH or Et2NOH) are examined by photoelectron spectroscopy and M06-2X and CCSD(T) computations. Remarkably, these species are experimentally found to have adiabatic detachment energies that are smaller than those for the free ion and reductions of 0.47 to 1.87 eV are predicted computationally. Hydrogen atom and proton transfers upon vertical photodetachment are two limiting extremes on the neutral surface in a continuum of mechanistic pathways that account for these results, and the whole gamut of possibilities are predicted to occur.

    We present the discovery of an extended ring of ultraviolet (UV) emission surrounding the asymptotic giant branch (AGB) star U Hya in archival observations performed by the Galaxy Evolution Explorer. This is the third discovery of extended UV emission from a carbon AGB star and the first from an AGB star with a detached shell. From imaging and photometric analysis of the FUV and NUV images, we determined that the UV ring has a radius of ∼110'', thus indicating that the emitting material is likely associated with the detached shell seen in the infrared. We find that scattering of the central point source of NUV and FUV emission by the dust shell is negligible. Moreover, we find that scattering of the interstellar radiation field by the dust shell can contribute at most ∼10% of the FUV flux. Morphological and photometric evidence suggests that shocks caused by the star's motion through space and, possibly, shock-excited H{sub 2} molecules are the most likely origins of the UV flux. In contrast to previous examples of extended UV emission from AGB stars, the extended UV emission from U Hya does not show a bow-shock-like structure, which is consistent with a lower space velocity and lower interstellar medium density. This suggests the detached dust shell is the source of the UV-emitting material and can be used to better understand the formation of detached shells.

    The discovery of a detached dust shell around the Wolf-Rayet (WR) star WR60-6 in the young stellar cluster VVV CL036 is reported. This shell is uncovered through the Spitzer-MIPS 24 μm image, where it appears brightest, and it is invisible at shorter wavelengths. Using new APEX observations and other data available from the literature, we have estimated some of the shell parameters: the inner and outer radii of 0.15 and 0.90 pc, respectively; the overall systemic velocity of the molecular {sup 12}CO(3 → 2) emission of –45.7 ± 2.3 km s{sup –1}; an expansion velocity of the gas of 16.3 ± 1 km s{sup –1}; the dust temperature and opacity of 122 ± 12 K and 1.04, respectively; and an age of 2.8 × 10{sup 4} yr. The WR star displays some cyclic variability. The mass computed for the WR60-6 nebula indicates that the material was probably ejected during its previous stages of evolution. In addition, we have identified a bright spot very close to the shell, which can be associated with the Midcourse Space Experiment source G312.13+00.20.

    We propose a novel method for determining the ages of low-mass, pre-main-sequence stellar systems using the apsidal motion of low-mass detached eclipsing binaries. The apsidal motion of a binary system with an eccentric orbit provides information regarding the interior structure constants of the individual stars. These constants are related to the normalized stellar interior density distribution and can be extracted from the predictions of stellar evolution models. We demonstrate that low-mass, pre-main-sequence stars undergoing radiative core contraction display rapidly changing interior structure constants (greater than 5% per 10 Myr) that, when combined with observational determinations of the interior structure constants (with 5%-10% precision), allow for a robust age estimate. This age estimate, unlike those based on surface quantities, is largely insensitive to the surface layer where effects of magnetic activity are likely to be most pronounced. On the main sequence, where age sensitivity is minimal, the interior structure constants provide a valuable test of the physics used in stellar structure models of low-mass stars. There are currently no known systems where this technique is applicable. Nevertheless, the emphasis on time domain astronomy with current missions, such as Kepler, and future missions, such as LSST, has the potential to discover systems where the proposed method will be observationally feasible.

The ductless mini-split heat pump, lighting and advanced air leakage control were the most cost effective measures. The future challenge will be to value engineer the performance levels indicated here in modeling using production based practices at a significantly lower cost.

  14. Measuring tides and binary parameters from gravitational wave data and eclipsing timings of detached white dwarf binaries

    The discovery of the most compact detached white dwarf (WD) binary SDSS J065133.33+284423.3 has been discussed in terms of probing the tidal effects in WDs. This system is also a verification source for the space-based gravitational wave (GW) detector, eLISA, or the evolved Laser Interferometer Space Antenna, which will observe short-period compact Galactic binaries with P {sub orb} ≲ 5 hr. We address the prospects of performing tidal studies using eLISA binaries by showing the fractional uncertainties in the orbital decay rate, f-dot , and the rate of that decay, f{sup ¨} expected from both the GW and electromagnetic (EM) data for some of the high-f binaries. We find that f-dot and f{sup ¨} can be measured using GW data only for the most massive WD binaries observed at high frequencies. From timing the eclipses for ∼10 yr, we find that f-dot can be known to ∼0.1% for J0651. We find that from GW data alone, measuring the effects of tides in binaries is (almost) impossible. We also investigate the improvement in the knowledge of the binary parameters by combining the GW amplitude and inclination with EM data with and without f-dot . In our previous work, we found that EM data on distance constrained the 2σ uncertainty in chirp mass to 15%-25% whereas adding f-dot reduces it to 0.11%. EM data on f-dot also constrain the 2σ uncertainty in distance to 35%-19%. EM data on primary mass constrain the secondary mass m {sub 2} to factors of two to ∼40% whereas adding f-dot reduces this to 25%. Finally, using single-line spectroscopic data constrains 2σ uncertainties in both the m {sub 2}, d to factors of two to ∼40%. Adding EM data on f-dot reduces these 2σ uncertainties to ≤25% and 6%-19%, respectively. Thus we find that EM measurements of f-dot and radial velocity are valuable in constraining eLISA binary parameters.

    The cross-field power loss due to radiation, plasma, and neutrals are measured for hydrogen discharges in a linear divertor simulator experiment. Radiation appears to be the dominant power loss channel; however, power loss due to heating of H{sub 2} neutrals is found to be quite significant, being only 2x weaker than radiation in the higher neutral pressure experiments. The H{sub 2} vibrational temperature T{sub vib} is found to be the most important channel for carrying neutral energy out of the plasma--more important than either kinetic temperature T{sub kin} or rotational temperature T{sub rot}. Power carried radially to the wall by plasma cross-field transport is found to be negligible when compared to neutral and radiation losses. These results demonstrate the importance of including of H{sub 2} neutrals in understanding power balance in detached tokamak divertors.

    In 2005, the US Department of Energy (DOE) launched the third generation of closure contracts, including the River Corridor Closure (RCC) Contract at Hanford. Over the past decade, significant progress has been made on cleaning up the river shore that bordes Hanford. However, the most important cleanup challenges lie ahead. In March 2005, DOE awarded the Hanford River Corridor Closure Contract to Washington Closure Hanford (WCH), a limited liability company owned by Washington Group International, Bechtel National and CH2M HILL. It is a single-purpose company whose goal is to safely and efficiently accelerate cleanup in the 544 km{sup 2} Hanford river corridor and reduce or eliminate future obligations to DOE for maintaining long-term stewardship over the site. The RCC Contract is a cost-plus-incentive-fee closure contract, which incentivizes the contractor to reduce cost and accelerate the schedule. At $1.9 billion and seven years, WCH has accelerated cleaning up Hanford's river corridor significantly compared to the $3.2 billion and 10 years originally estimated by the US Army Corps of Engineers. Predictable funding is one of the key features of the new contract, with funding set by contract at $183 million in fiscal year (FY) 2006 and peaking at $387 million in FY2012. Another feature of the contract allows for Washington Closure to perform up to 40% of the value of the contract and subcontract the balance. One of the major challenges in the next few years will be to identify and qualify sufficient subcontractors to meet the goal.

    The protection of worker health and safety is of paramount concern at the Savannah River Site. Since the site is one of the largest nuclear sites in the nation, radiation safety is a key element in the protection program. This report is a compendium of the results in 1988 of the programs at the Savannah River Plant and the Savannah River Laboratory to protect the radiological health of employees. By any measure, the radiation protection performance at this site in 1988 was the best since the beginning of operations. This accomplishment was made possible by the commitment and support at all levels of the organizations to reduce radiation exposures to ALARA (As Low As Reasonably Achievable). The report provides detailed information about the radiation doses received by departments and work groups within these organizations. It also includes exposure data for recent years to allow Plant and Laboratory units to track the effectiveness of their ALARA efforts. Many of the successful practices and methods that reduced radiation exposure are described. A new goal for personnel contamination cases has been established for 1989. Only through continual and innovative efforts to minimize exposures can the goals be met. The radiation protection goals for 1989 and previous years are included in the report. 27 figs., 58 tabs.

    The Hood River Passive Project was developed by Root Design Build of Hood River Oregon using the Passive House Planning Package (PHPP) to meet all of the requirements for certification under the European Passive House standards. The Passive House design approach has been gaining momentum among residential designers for custom homes and BEopt modeling indicates that these designs may actually exceed the goal of the U.S. Department of Energy's (DOE) Building America program to "reduce home energy use by 30%-50%" (compared to 2009 energy codes for new homes). This report documents the short term test results of the Shift House and compares the results of PHPP and BEopt modeling of the project. The design includes high R-Value assemblies, extremely tight construction, high performance doors and windows, solar thermal DHW, heat recovery ventilation, moveable external shutters and a high performance ductless mini-split heat pump. Cost analysis indicates that many of the measures implemented in this project did not meet the BA standard for cost neutrality. The ductless mini-split heat pump, lighting and advanced air leakage control were the most cost effective measures. The future challenge will be to value engineer the performance levels indicated here in modeling using production based practices at a significantly lower cost.

    The clip has a long slot with a round enlarged end, flat on one side. The badge has a headed member that is circular except for a flat side. The badge is connected to, or disconnected from, the clip by insertion of the headed member through the enlarged end of the slot in the clip. The badge is unlikely to be accidentally disconnected from the clip while being worn on a worker's clothing, because the weight of the badge keeps the headed member away from the enlarged end of the slot in the clip, and the headed member can pass through the enlarged end of the slot only at one relative angular position between badge and clip, namely, that angle at which the flat side of the headed member is coincident with the flat side of the enlarged end of the slot.     Bonneville Power Administration (BPA) is preparing an EA to assess potential environmental impacts of a proposal to rebuild its 24-mile long, 115 kilovolt Bonneville-Hood River transmission line. The existing line runs between the Bonneville Powerhouse at Bonneville Dam in Multnomah County, Oregon, and BPA's existing Hood River Substation in Hood River County, Oregon. The project would include replacing structures and conductor wires, improving access roads, and constructing new access roads or trails where needed.

USDA Forest Service, Savannah River, Aiken, SC. 28 pp. Abstract - This paper explains the rationale and results of a 1991 road erosion inventory conducted by members of the USDA Forest Service – Savannah River (FS-SR) and USDA Natural Resources Conservation Service (NRCS). The inventory provided information for the Department of Energy - Savannah River (DOE-SR) to justify the need for developing an erosion and sediment control program with appropriate funding, personnel, and equipment. Federally managed since the early 1950’s, the SRS is located on 198,344 acres (80,301 hectares) in the South Carolina counties of Aiken, Barnwell, and Allendale. Located along the eastern border of the Savannah River, the SRS is located within the Upper and Lower Coastal Plains of South Carolina.
