    Quercetin is a bioflavonoid that exhibits several biological functions in vitro and in vivo. Quercetin 3-O-methyl ether (Q3) is a natural product reported to have pharmaceutical activities, including antioxidative and anticancer activities. However, little is known about the mechanism by which it protects cells from oxidative stress. This study was designed to investigate the mechanisms by which Q3 protects against Cu{sup 2+}-induced cytotoxicity. Exposure to Cu{sup 2+} resulted in the death of mouse liver FL83B cells, characterized by apparent apoptotic features, including DNA fragmentation and increased nuclear condensation. Q3 markedly suppressed Cu{sup 2+}-induced apoptosis and mitochondrial dysfunction, characterized by reduced mitochondrial membrane potential, caspase-3 activation, and PARP cleavage, in Cu{sup 2+}-exposed cells. The involvement of PI3K, Akt, Erk, FOXO3A, and Mn-superoxide dismutase (MnSOD) was shown to be critical to the survival of Q3-treated FL83B cells. The liver of both larval and adult zebrafish showed severe damage after exposure to Cu{sup 2+} at a concentration of 5 μM. Hepatic damage induced by Cu{sup 2+} was reduced by cotreatment with Q3. Survival of Cu{sup 2+}-exposed larval zebrafish was significantly increased by cotreatment with 15 μM Q3. Our results indicated that Cu{sup 2+}-induced apoptosis in FL83B cells occurred via the generation of ROS, upregulation and phosphorylation of Erk, overexpression of 14-3-3, inactivation of Akt, and the downregulation of FOXO3A and MnSOD. Hence, these results also demonstrated that Q3 plays a protective role against oxidative damage in zebrafish liver and remarked the potential of Q3 to be used as an antioxidant for hepatocytes.     We investigate the sensitivity of simulated cloud properties and surface precipitation to assumptions regarding the size distributions of the precipitating hydrometeors in a one-moment bulk microphysics scheme. Three sensitivity experiments were applied to two composites of 15 convective and 15 frontal stratiform intense precipitation events observed in a coastal midlatitude region (Belgium), which were evaluated against satellite-retrieved cloud properties and radar-rain-gauge derived surface precipitation. It is found that the cloud optical thickness distribution was well captured by all experiments, although a significant underestimation of cloudiness occurred in the convective composite. The cloud-top-pressure distribution was improved most by more realistic snow size distributions (including a temperature-dependent intercept parameter and non-spherical snow for the calculation of the slope parameter), due to increased snow depositional growth at high altitudes. Surface precipitation was far less sensitive to whether graupel or hail was chosen as the rimed ice species, as compared to previous idealized experiments. This smaller difference in sensitivity could be explained by the stronger updraught velocities and higher freezing levels in the idealized experiments compared to typical coastal midlatitude environmental conditions.

    This study investigates the sensitivity of moist processes and surface precipitation during three extreme precipitation events over Belgium to the representation of rain, snow and hail size distributions in a bulk one-moment microphysics parameterisation scheme. Sensitivities included the use of empirically derived relations to calculate the slope parameter and diagnose the intercept parameter of the exponential snow and rain size distributions and sensitivities to the treatment of hail/graupel. A detailed evaluation of the experiments against various high temporal resolution and spatially distributed observational data was performed to understand how moist processes responded to the implemented size distribution modifications. Net vapor consumption by microphysical processes was found to be unaffected by snow or rain size distribution modifications, while it was reduced replacing formulations for hail by those typical for graupel, mainly due to intense sublimation of graupel. Cloud optical thickness was overestimated in all experiments and all cases, likely due to overestimated snow amounts. The overestimation slightly deteriorated by modifying the rain and snow size distributions due to increased snow depositional growth, while it was reduced by including graupel. The latter was mainly due to enhanced cloud water collection by graupel and reduced snow depositional growth. Radar reflectivity and cloud optical thickness could only be realistically represented by inclusion of graupel during a stratiform case, while hail was found indispensable to simulate the vertical reflectivity profile and the surface precipitation structure. Precipitation amount was not much altered by any of the modifications made and the general overestimation was only decreased slightly during a supercell convective case.

    The American Academy of Microbiology convened a colloquium November 6-7, 2003, in Key Largo, Florida, to discuss the application of genomic methods to the study of pathogenesis. Professionals in the fields of genomics, bacteriology, virology, eukaryotic microbiology, medicine, clinical diagnostics, bioinformatics, and forensics contributed their expertise to discussions on the recent advancements in the field and the outlook for future research. A number of recommendations were made for ensuring that progress in the field continues. The availability of genome sequences from pathogenic bacteria, viruses, and fungi and other eukaryotes has opened new horizons for the field of pathogenesis. The genomes of over 100 bacterial pathogens have been fully sequenced, and scientists are busy investigating the mechanisms that regulate the diversity of bacterial pathogens and their myriad abilities to evade host defenses. Close to 1,200 viral genomes have been sequenced, and virologists are now beginning to examine the genomes of those viruses that cause undetected, cryptic infections. These virus-host interactions can serve as a reservoir of viral genes that later emerge in novel pathogens with the potential to infect humans, economically important animals, or crops. A number of eukaryotic microbes, including several pathogenic fungi, have also been sequenced, revealing unimagined diversity among these important pathogens. Certain themes have emerged from analyses of pathogen genome sequences, and the possibility exists that a sequence-based common thread may be found linking pathogens from different taxa. The results of genome sequencing efforts have also informed the study of pathogenesis, helping to identify the ways in which pathogens bring about disease. The advances of the past several years have been great, and we are closer than ever to a comprehensive understanding of pathogenesis, but much work lies ahead. If the science is to move forward, the genome sequences of many more organisms are needed. The sequences of many hosts, pathogens, their nonpathogenic relatives, commensals, as well as a diverse array of microorganisms, are all needed to complete the picture of pathogenesis and provide a phylogenetic framework for understanding the phenomenon. Moreover, improvements are needed in the two most important tools of genomics: annotation methodologies and sequence databases.

Aspects of remote sensing use covered include the significance of lineaments and their effects on ground-water systems, waste-site use and geotechnical characterization, the estimation of reservoir submerging losses using CIR aerial photographs, and satellite-based investigation of the significance of surficial deposits for surface mining operations. Other topics presented include the location of potential ground subsidence and collapse features in soluble carbonate rock, optical Fourier analysis of surface features of interest in geotechnical engineering, geotechnical applications of U.S. Government remote sensing programs, updating the data base for a Geographic Information System, the joint NASA/Geosat Test Case Project, the selection of remote data telemetry methods for geotechnical applications, the standardization of remote sensing data collection and transmission, and a comparison of airborne Goodyear electronic mapping system/SAR with satelliteborne Seasat/SAR radar imagery.

    Aspects of solar measurements, solar collectors, selective coatings, thermal storage, phase change storage, and heat exchangers are discussed. The analysis and testing of flat-plate solar collectors are addressed. The development and uses of plastic collectors, a solar water heating system, solar energy collecting oil barrels, a glass collector panel, and a two-phase thermosyphon system are considered. Studies of stratification in thermal storage, of packed bed and fluidized bed systems, and of thermal storage in solar towers, in wall passive systems, and in reversible chemical reactions are reported. Phase change storage by direct contact processes and in residential solar space heating and cooling is examined, as are new materials and surface characteristics for solar heat storage. The use of R-11 and Freon-113 in heat exchange is discussed.

    The U.S. Bureau of Mines diskette contains analysis and related source data for 2,357 natural gas samples collected from miscellaneous states, which include the following states: Alabama, Arkansas (except Arkoma Basin), Florida, Georgia, Illinois, Indiana, Iowa, Kentucky, Louisiana, Maryland, Michigan, Mississippi, Missouri, New Jersey, New York, North Carolina, Ohio, Pennsylvania, Tennessee, Virginia, and West Virginia. All samples were obtained and analyzed as part of the Bureau's investigations of occurrences of helium in natural gases of countries with free market economies. The survey has been conducted since 1917. The analysis contained on the diskette contain the full range of component analysis data. Five files are on the diskette: READ.ME, MISC.TXT, MISC.DBF, USHEANAL.DBF, and BASINCDE.TXT.

    There are several definitions of symbiosis, but in this book it involves an association where one organism (the symbiont) lives within or on the body of another organism (the host), regardless of the actual effect on the host. Some symbioses are mutualistic, some parasitic, and some involve commensalism, in which one partner derives some benefit without either harming or benefiting the other. This is the second volume in this exciting and rapidly advancing topic by these editors. The first volume was published in 2003 and during the intervening three years additional data have been produced that make this book a useful addition to your library.     We present a computational framework for integrating a state-of-the-art numerical weather prediction (NWP) model in stochastic unit commitment/economic dispatch formulations that account for wind power uncertainty. We first enhance the NWP model with an ensemble-based uncertainty quantification strategy implemented in a distributed-memory parallel computing architecture. We discuss computational issues arising in the implementation of the framework and validate the model using real wind-speed data obtained from a set of meteorological stations. We build a simulated power system to demonstrate the developments.

    This poster describes the Fast Physics Testbed for the new FAst-physics System Testbed and Research (FASTER) project. The overall objective is to provide a convenient and comprehensive platform for fast turn-around model evaluation against ARM observations and to facilitate development of parameterizations for cloud-related fast processes represented in global climate models. The testbed features three major components: a single column model (SCM) testbed, an NWP-Testbed, and high-resolution modeling (HRM). The web-based SCM-Testbed features multiple SCMs from major climate modeling centers and aims to maximize the potential of SCM approach to enhance and accelerate the evaluation and improvement of fast physics parameterizations through continuous evaluation of existing and evolving models against historical as well as new/improved ARM and other complementary measurements. The NWP-Testbed aims to capitalize on the large pool of operational numerical weather prediction products. Continuous evaluations of NWP forecasts against observations at ARM sites are carried out to systematically identify the biases and skills of physical parameterizations under all weather conditions. The highresolution modeling (HRM) activities aim to simulate the fast processes at high resolution to aid in the understanding of the fast processes and their parameterizations. A four-tier HRM framework is established to augment the SCM- and NWP-Testbeds towards eventual improvement of the parameterizations.

    Uncertainties associated with solar forecasts present challenges to maintain grid reliability, especially at high solar penetrations. This study aims to quantify the errors associated with the day-ahead solar forecast parameters and the theoretical solar power output for a 51-kW solar power plant in a utility area in the state of Vermont, U.S. Forecasts were generated by three numerical weather prediction (NWP) models, including the Rapid Refresh, the High Resolution Rapid Refresh, and the North American Model, and a machine-learning ensemble model. A photovoltaic (PV) performance model was adopted to calculate theoretical solar power generation using the forecast parameters (e.g., irradiance, cell temperature, and wind speed). Errors of the power outputs were quantified using statistical moments and a suite of metrics, such as the normalized root mean squared error (NRMSE). In addition, the PV model's sensitivity to different forecast parameters was quantified and analyzed. Results showed that the ensemble model yielded forecasts in all parameters with the smallest NRMSE. The NRMSE of solar irradiance forecasts of the ensemble NWP model was reduced by 28.10% compared to the best of the three NWP models. Further, the sensitivity analysis indicated that the errors of the forecasted cell temperature attributed only approximately 0.12% to the NRMSE of the power output as opposed to 7.44% from the forecasted solar irradiance.

    We carry out a comprehensive investigation comparing the three-dimensional magnetic field restructuring, flare energy release, and the helioseismic response of two homologous flares, the 2011 September 6 X2.1 (FL1) and September 7 X1.8 (FL2) flares in NOAA AR 11283. In our analysis, (1) a twisted flux rope (FR) collapses onto the surface at a speed of 1.5 km s{sup –1} after a partial eruption in FL1. The FR then gradually grows to reach a higher altitude and collapses again at 3 km s{sup –1} after a fuller eruption in FL2. Also, FL2 shows a larger decrease of the flux-weighted centroid separation of opposite magnetic polarities and a greater change of the horizontal field on the surface. These imply a more violent coronal implosion with corresponding more intense surface signatures in FL2. (2) The FR is inclined northward and together with the ambient fields, it undergoes a southward turning after both events. This agrees with the asymmetric decay of the penumbra observed in the peripheral regions. (3) The amounts of free magnetic energy and nonthermal electron energy released during FL1 are comparable to those of FL2 within the uncertainties of the measurements. (4) No sunquake was detected in FL1; in contrast, FL2 produced two seismic emission sources S1 and S2 both lying in the penumbral regions. Interestingly, S1 and S2 are connected by magnetic loops, and the stronger source S2 has a weaker vertical magnetic field. We discuss these results in relation to the implosion process in the low corona and the sunquake generation.

    Abstract.—The influence of a surgically implanted acoustic micro-transmitter and passive integrated transponder (PIT) tag on the growth and survival of hatchery-reared juvenile Chinook salmon was examined. Growth and survival were compared between treatment (implanted) and control fish within three fork length (FL) size groups (80-89, 90-99, and 100-109 mm). The acoustic micro-transmitter and PIT tag implanted in our study had a combined weight of 0.74 g. Weights of study fish ranged from 4.7 to 16.3 g for treatment fish and from 5.1 to 16.8 g for control fish. The burden for the combined acoustic and PIT tag experienced by implanted fish ranged from 8.8% to 15.7% for the 80-89 mm FL group, 6.0-10.9% for the 90-99 mm FL group, and 4.5-8.6% for the 100-109 mm FL group. Results indicated that growth and survival were size-dependent among implanted juvenile Chinook salmon. Significant differences in growth rate and survival were observed between treatment and control fish in the 80-89 mm FL group. Within this group, growth of fish smaller than 88.5 mm FL (tag burden > 10.0%) was negatively affected by the implantation or presence of an acoustic micro-transmitter and PIT tag. Survival of fish in the 90-99 mm FL group did not differ between treatment and control fish. However, survival of implanted fish within this size group that were smaller than 97.2 mm FL (tag burden > 7.4%) was negatively influenced. These results indicate that the burden of an acoustic micro-transmitter and PIT tag should be maintained at or below about 7.0% for studies that use hatchery-reared juvenile Chinook salmon.

    BA-PIRC has worked with several community-scale builders within the hot humid climate zone to improve performance of production, or community scale, housing. Tommy Williams Homes (Gainesville, FL), Lifestyle Homes (Melbourne, FL), and Habitat for Humanity (various locations, FL) have all been continuous partners of the Building America program and are the subjects of this report to document achievement of the Building America goal of 30% whole house energy savings packages adopted at the community scale. Key aspects of this research include determining how to evolve existing energy efficiency packages to produce replicable target savings, identifying what builders' technical assistance needs are for implementation and working with them to create sustainable quality assurance mechanisms, and documenting the commercial viability through neutral cost analysis and market acceptance. This report documents certain barriers builders overcame and the approaches they implemented in order to accomplish Building America (BA) Program goals that have not already been documented in previous reports.

    We investigated the Schottky barrier of Pt/4H-SiC contact as a function of 4H-SiC surface properties which effectively controlled by electronic cyclotron resonance hydrogen plasma pretreatment for different periods and annealing. Electrically homogeneous contacts are observed when the Fermi level (FL) is 'pinned (Bardeen limit)' and 'free-pinned (Schottky limit).' However, a partial pinning of FL leads to Gaussian distribution of inhomogeneous barrier height. These results could be correlated with changes in the magnitude and spatial distribution of surface state density after different pretreatments.

    Mercury iodide (HgI{sub 2}) nanocrystals (NCs) were successfully grown in a barium phosphate glass matrix synthesized by fusion. Growth control of HgI{sub 2} NCs was investigated by Atomic Force Microscopy (AFM), Optical Absorption (OA), Fluorescence (FL), and X-ray diffraction (XRD). AFM images reveal the formation of HgI{sub 2} nanocrystals in host glass matrix. HgI{sub 2} NCs growth was evidenced by an OA and FL band red-shift with increasing annealing time. XRD measurements revealed the β crystalline phase of the HgI{sub 2} nanocrystals.

    Measurements of wind fields in the earth's troposphere with daily global coverage is widely considered as a significant advance for forecasting and transport studies. For optimal use by NWP (Numerical Weather Prediction) models the horizontal and vertical resolutions should be approximately 100 km and 1 km, respectively. For boundary layer studies vertical resolution of a few hundred meters seems essential. Earth-orbiting Doppler lidar has a unique capability to measure global winds in the troposphere with the high vertical resolution required. The lidar approach depends on transmission of pulses with high spectral purity and backscattering from the atmospheric aerosol particles or layered clouds to provide a return signal. Recent field measurement campaigns using NASA research aircraft have resulted in collection of aerosol and cloud data which can be used to optimize the Doppler lidar instrument design and measurement strategy. 5 refs.

    The heterogeneous nature of typical sedimentary formations can play a major role in the propagation of the CO2 plume, eventually dampening the accumulation of mobile phase underneath the caprock. From core flooding experiments, it is also known that contrasts in capillary threshold pressure due to different pore size can affect the flow paths of the invading and displaced fluids and consequently influence the build- up of non-wetting phase (NWP) at interfaces between geological facies. The full characterization of the geologic variability at all relevant scales and the ability to make observations on the spatial and temporal distribution of the migration and trapping of supercritical CO2 is not feasible from a practical perspective. To provide insight into the impact of well-defined heterogeneous systems on the flow dynamics and trapping efficiency of supercritical CO2 under drainage and imbibition conditions, we present an experimental investigation at the meter scale conducted in synthetic sand reservoirs packed in a quasi-two-dimensional flow-cell. Two immiscible displacement experiments have been performed to observe the preferential entrapment of NWP in simple heterogeneous porous media. The experiments consisted of an injection, a fluid redistribution, and a forced imbibition stages conducted in an uncorrelated permeability field and a homogeneous base case scenario. We adopted x-ray attenuation analysis as a non-destructive technique that allows a precise measurement of phase saturations throughout the entire flow domain. By comparing a homogeneous and a heterogeneous scenario we have identified some important effects that can be attributed to capillary barriers, such as dampened plume advancement, higher non-wetting phase saturations, larger contact area between the injected and displaced phases, and a larger range of non-wetting phase saturations.

    The Wind Forecast Improvement Project (WFIP) is a public-private research program, the goals of which are to improve the accuracy of short-term (0-6 hr) wind power forecasts for the wind energy industry and then to quantify the economic savings that accrue from more efficient integration of wind energy into the electrical grid. WFIP was sponsored by the U.S. Department of Energy (DOE), with partners that include the National Oceanic and Atmospheric Administration (NOAA), private forecasting companies (WindLogics and AWS Truepower), DOE national laboratories, grid operators, and universities. WFIP employed two avenues for improving wind power forecasts: first, through the collection of special observations to be assimilated into forecast models to improve model initial conditions; and second, by upgrading NWP forecast models and ensembles. The new observations were collected during concurrent year-long field campaigns in two high wind energy resource areas of the U.S. (the upper Great Plains, and Texas), and included 12 wind profiling radars, 12 sodars, 184 instrumented tall towers and over 400 nacelle anemometers (provided by private industry), lidar, and several surface flux stations. Results demonstrate that a substantial improvement of up to 14% relative reduction in power root mean square error (RMSE) was achieved from the combination of improved NOAA numerical weather prediction (NWP) models and assimilation of the new observations. Data denial experiments run over select periods of time demonstrate that up to a 6% relative improvement came from the new observations. The use of ensemble forecasts produced even larger forecast improvements. Based on the success of WFIP, DOE is planning follow-on field programs.

    Resiliency and cyber security of modern critical infrastructures is becoming increasingly important with the growing number of threats in the cyber-environment. This paper proposes an extension to a previously developed fuzzy logic based anomaly detection network security cyber sensor via incorporating Type-2 Fuzzy Logic (T2 FL). In general, fuzzy logic provides a framework for system modeling in linguistic form capable of coping with imprecise and vague meanings of words. T2 FL is an extension of Type-1 FL which proved to be successful in modeling and minimizing the effects of various kinds of dynamic uncertainties. In this paper, T2 FL provides a basis for robust anomaly detection and cyber security state awareness. In addition, the proposed algorithm was specifically developed to comply with the constrained computational requirements of low-cost embedded network security cyber sensors. The performance of the system was evaluated on a set of network data recorded from an experimental cyber-security test-bed.

    Nimonic-80A is a kind of nickel-based superalloys which is used in high temperature components of land gas turbines. In this paper, the influence of four design factors: die temperature, strain rate, friction coefficient and geometry size of ring sample over the variation of internal diameters (VID) and forging load (FL) was studied. It was done by means of design methodology based on DOE-designated full factorial and FE simulations. FEM and experimental results showed that the variation of internal diameters and forging load had inverse proportion to the die temperature. Regression models were developed by using the response surface methodology (RSM) for VID and FL. Rate of the dynamic recrystallization varied depending on different amounts of die temperature. The results can be used in the semi-isothermal forging of complex part of the Nimonic-80A.

    Optimizing transverse particle distributions in the accumulator ring is one of most important factors to the future performance of the Spallation Neutron Source (SNS) [l]. This can only be achieved by optimizing the injection bumps that paint the beam in phase space. The process is complex due to the vague distribution inputs and the multiple optimization goals. Furthermore, the priority of the optimization criteria could change at different operational stages. We propose optimizing transverse phase space painting with fuzzy logic and present our initial studies toward that end. The focus of this paper is on how the problem can be solved with a Fuzzy Logic (FL) expert system through the creation of a set of rules that can be applied by the system. Various particle distributions, from computer simulations, are analyzed with FL and the results are compared and discussed. Finally, a run-time optimization control system is proposed.
