    Quercetin is a bioflavonoid that exhibits several biological functions in vitro and in vivo. Quercetin 3-O-methyl ether (Q3) is a natural product reported to have pharmaceutical activities, including antioxidative and anticancer activities. However, little is known about the mechanism by which it protects cells from oxidative stress. This study was designed to investigate the mechanisms by which Q3 protects against Cu{sup 2+}-induced cytotoxicity. Exposure to Cu{sup 2+} resulted in the death of mouse liver FL83B cells, characterized by apparent apoptotic features, including DNA fragmentation and increased nuclear condensation. Q3 markedly suppressed Cu{sup 2+}-induced apoptosis and mitochondrial dysfunction, characterized by reduced mitochondrial membrane potential, caspase-3 activation, and PARP cleavage, in Cu{sup 2+}-exposed cells. The involvement of PI3K, Akt, Erk, FOXO3A, and Mn-superoxide dismutase (MnSOD) was shown to be critical to the survival of Q3-treated FL83B cells. The liver of both larval and adult zebrafish showed severe damage after exposure to Cu{sup 2+} at a concentration of 5 μM. Hepatic damage induced by Cu{sup 2+} was reduced by cotreatment with Q3. Survival of Cu{sup 2+}-exposed larval zebrafish was significantly increased by cotreatment with 15 μM Q3. Our results indicated that Cu{sup 2+}-induced apoptosis in FL83B cells occurred via the generation of ROS, upregulation and phosphorylation of Erk, overexpression of 14-3-3, inactivation of Akt, and the downregulation of FOXO3A and MnSOD. Hence, these results also demonstrated that Q3 plays a protective role against oxidative damage in zebrafish liver and remarked the potential of Q3 to be used as an antioxidant for hepatocytes.     We investigate the sensitivity of simulated cloud properties and surface precipitation to assumptions regarding the size distributions of the precipitating hydrometeors in a one-moment bulk microphysics scheme. Three sensitivity experiments were applied to two composites of 15 convective and 15 frontal stratiform intense precipitation events observed in a coastal midlatitude region (Belgium), which were evaluated against satellite-retrieved cloud properties and radar-rain-gauge derived surface precipitation. It is found that the cloud optical thickness distribution was well captured by all experiments, although a significant underestimation of cloudiness occurred in the convective composite. The cloud-top-pressure distribution was improved most by more realistic snow size distributions (including a temperature-dependent intercept parameter and non-spherical snow for the calculation of the slope parameter), due to increased snow depositional growth at high altitudes. Surface precipitation was far less sensitive to whether graupel or hail was chosen as the rimed ice species, as compared to previous idealized experiments. This smaller difference in sensitivity could be explained by the stronger updraught velocities and higher freezing levels in the idealized experiments compared to typical coastal midlatitude environmental conditions.

    This study investigates the sensitivity of moist processes and surface precipitation during three extreme precipitation events over Belgium to the representation of rain, snow and hail size distributions in a bulk one-moment microphysics parameterisation scheme. Sensitivities included the use of empirically derived relations to calculate the slope parameter and diagnose the intercept parameter of the exponential snow and rain size distributions and sensitivities to the treatment of hail/graupel. A detailed evaluation of the experiments against various high temporal resolution and spatially distributed observational data was performed to understand how moist processes responded to the implemented size distribution modifications. Net vapor consumption by microphysical processes was found to be unaffected by snow or rain size distribution modifications, while it was reduced replacing formulations for hail by those typical for graupel, mainly due to intense sublimation of graupel. Cloud optical thickness was overestimated in all experiments and all cases, likely due to overestimated snow amounts. The overestimation slightly deteriorated by modifying the rain and snow size distributions due to increased snow depositional growth, while it was reduced by including graupel. The latter was mainly due to enhanced cloud water collection by graupel and reduced snow depositional growth. Radar reflectivity and cloud optical thickness could only be realistically represented by inclusion of graupel during a stratiform case, while hail was found indispensable to simulate the vertical reflectivity profile and the surface precipitation structure. Precipitation amount was not much altered by any of the modifications made and the general overestimation was only decreased slightly during a supercell convective case.

    The American Academy of Microbiology convened a colloquium November 6-7, 2003, in Key Largo, Florida, to discuss the application of genomic methods to the study of pathogenesis. Professionals in the fields of genomics, bacteriology, virology, eukaryotic microbiology, medicine, clinical diagnostics, bioinformatics, and forensics contributed their expertise to discussions on the recent advancements in the field and the outlook for future research. A number of recommendations were made for ensuring that progress in the field continues. The availability of genome sequences from pathogenic bacteria, viruses, and fungi and other eukaryotes has opened new horizons for the field of pathogenesis. The genomes of over 100 bacterial pathogens have been fully sequenced, and scientists are busy investigating the mechanisms that regulate the diversity of bacterial pathogens and their myriad abilities to evade host defenses. Close to 1,200 viral genomes have been sequenced, and virologists are now beginning to examine the genomes of those viruses that cause undetected, cryptic infections. These virus-host interactions can serve as a reservoir of viral genes that later emerge in novel pathogens with the potential to infect humans, economically important animals, or crops. A number of eukaryotic microbes, including several pathogenic fungi, have also been sequenced, revealing unimagined diversity among these important pathogens. Certain themes have emerged from analyses of pathogen genome sequences, and the possibility exists that a sequence-based common thread may be found linking pathogens from different taxa. The results of genome sequencing efforts have also informed the study of pathogenesis, helping to identify the ways in which pathogens bring about disease. The advances of the past several years have been great, and we are closer than ever to a comprehensive understanding of pathogenesis, but much work lies ahead. If the science is to move forward, the genome sequences of many more organisms are needed. The sequences of many hosts, pathogens, their nonpathogenic relatives, commensals, as well as a diverse array of microorganisms, are all needed to complete the picture of pathogenesis and provide a phylogenetic framework for understanding the phenomenon. Moreover, improvements are needed in the two most important tools of genomics: annotation methodologies and sequence databases.

    The 7-acre Whitehouse Waste Oil Pits site was used by Allied Petroleum Products (Allied) to dispose of acidic waste oil sludges from its oil reclamation process in Whitehouse, Duval County, Florida. A cypress swamp system and residential area are immediately adjacent to the site. The acid sludge produced in the first step and clay used to decolorize the oil were dumped into the unlined pits at the site. A 1985 ROD addressed source control as a containment remedy consisting of a slurry wall construction, soil cap, and a ground water recovery and treatment system; however, EPA has re-evaluated the 1985 ROD selection and determined that the containment remedy failed to meet the requirements of SARA. As a result, the ROD Amendment focuses on an alternative for treating Whitehouse wastes by eliminating direct contact risk associated with pit soil/sludge wastes and preventing contaminated ground water in the surficial aquifer from migrating laterally. The primary contaminants of concern that affect the soil, sediment, surface water, and ground water are VOCs, including benzene, toluene, and xylenes; organics, including PCBs and phenols; and metals, including arsenic, chromium, and lead. The amended remedial action for the site are included.

    There are several definitions of symbiosis, but in this book it involves an association where one organism (the symbiont) lives within or on the body of another organism (the host), regardless of the actual effect on the host. Some symbioses are mutualistic, some parasitic, and some involve commensalism, in which one partner derives some benefit without either harming or benefiting the other. This is the second volume in this exciting and rapidly advancing topic by these editors. The first volume was published in 2003 and during the intervening three years additional data have been produced that make this book a useful addition to your library.     The U.S. Bureau of Mines diskette contains analysis and related source data for 2,357 natural gas samples collected from miscellaneous states, which include the following states: Alabama, Arkansas (except Arkoma Basin), Florida, Georgia, Illinois, Indiana, Iowa, Kentucky, Louisiana, Maryland, Michigan, Mississippi, Missouri, New Jersey, New York, North Carolina, Ohio, Pennsylvania, Tennessee, Virginia, and West Virginia. All samples were obtained and analyzed as part of the Bureau's investigations of occurrences of helium in natural gases of countries with free market economies. The survey has been conducted since 1917. The analysis contained on the diskette contain the full range of component analysis data. Five files are on the diskette: READ.ME, MISC.TXT, MISC.DBF, USHEANAL.DBF, and BASINCDE.TXT.

    We present a computational framework for integrating a state-of-the-art numerical weather prediction (NWP) model in stochastic unit commitment/economic dispatch formulations that account for wind power uncertainty. We first enhance the NWP model with an ensemble-based uncertainty quantification strategy implemented in a distributed-memory parallel computing architecture. We discuss computational issues arising in the implementation of the framework and validate the model using real wind-speed data obtained from a set of meteorological stations. We build a simulated power system to demonstrate the developments.

    This poster describes the Fast Physics Testbed for the new FAst-physics System Testbed and Research (FASTER) project. The overall objective is to provide a convenient and comprehensive platform for fast turn-around model evaluation against ARM observations and to facilitate development of parameterizations for cloud-related fast processes represented in global climate models. The testbed features three major components: a single column model (SCM) testbed, an NWP-Testbed, and high-resolution modeling (HRM). The web-based SCM-Testbed features multiple SCMs from major climate modeling centers and aims to maximize the potential of SCM approach to enhance and accelerate the evaluation and improvement of fast physics parameterizations through continuous evaluation of existing and evolving models against historical as well as new/improved ARM and other complementary measurements. The NWP-Testbed aims to capitalize on the large pool of operational numerical weather prediction products. Continuous evaluations of NWP forecasts against observations at ARM sites are carried out to systematically identify the biases and skills of physical parameterizations under all weather conditions. The highresolution modeling (HRM) activities aim to simulate the fast processes at high resolution to aid in the understanding of the fast processes and their parameterizations. A four-tier HRM framework is established to augment the SCM- and NWP-Testbeds towards eventual improvement of the parameterizations.
