  4. Characterization of protein redox dynamics induced during light-to-dark transitions and nutrient limitation in cyanobacteria

    SciTech Connect (OSTI)

Ansong, Charles; Sadler, Natalie C.; Hill, Eric A.; Lewis, Michael P.; Zink, Erika M.; Smith, Richard D.; Beliaev, Alex S.; Konopka, Allan; Wright, Aaron T.

    2014-07-03

Protein redox chemistry constitutes a major void in knowledge pertaining to photoautotrophic system regulation and signaling processes. We have employed a chemical biology approach to analyze redox sensitive proteins in live Synechococcus sp. PCC 7002 cells in both light and dark periods, and to understand how cellular redox balance is disrupted during nutrient perturbation. The present work identified several novel putative redox-sensitive proteins that are involved in the generation of reductant, macromolecule synthesis, and carbon flux through central metabolic pathways, and may be involved in cell signaling and response mechanisms. Furthermore, our research suggests that dynamic redox changes in response to specific nutrient limitations contribute to the regulatory changes driven by a shift from light to dark. Taken together, these results contribute to the high-level understanding of post-translational mechanisms regulating flux distributions and therefore present potential metabolic engineering targets for redirecting carbon towards biofuel precursors.

  5. Function allocation for humans and automation in the context of team dynamics

    SciTech Connect (OSTI)

Jeffrey C. Joe; John O'Hara; Jacques Hugo; Johanna Oxstrand

    2015-07-01

Within Human Factors Engineering, a decision-making process called function allocation (FA) is used during the design life cycle of complex systems to distribute the system functions, often identified through a functional requirements analysis, to all human and automated machine agents (or teammates) involved in controlling the system. Most FA methods make allocation decisions primarily by comparing the capabilities of humans and automation, but then also by considering secondary factors such as cost, regulations, and the health and safety of workers. The primary analysis of the strengths and weaknesses of humans and machines, however, is almost always considered in terms of individual human or machine capabilities. Yet, FA is fundamentally about teamwork in that the goal of the FA decision-making process is to determine what are the optimal allocations of functions among agents. Given this framing of FA, and the increasing use of and sophistication of automation, there are two related social psychological issues that current FA methods need to address more thoroughly. First, many principles for effective human teamwork are not considered as central decision points or in the iterative hypothesis and testing phase in most FA methods, when it is clear that social factors have numerous positive and negative effects on individual and team capabilities. Second, social psychological factors affecting team performance and can be difficult to translate to automated agents, and most FA methods currently do not account for this effect. The implications for these issues are discussed.

  6. Nutrient dynamics and nitrogen trace gas flux during ecosystem development in montane rain forest

    SciTech Connect (OSTI)

    Riley, R.H.; Vitousek, P.M.

    1995-01-01

Patterns of nitrogen trace gas emissions, soil nitrogen flux, and nutrient availability were evaluated at five sites that form a chronosequence in Hawaiian montane rain forest. The estimated age of basaltic parent material from which soils developed at the Kilauea site was 200 yr, 6000 yr at the Puu Makaala site, 185000 yr at the Kohala site, 1.65 x 10{sup 6} yr at the Molokai site, and 4.5 x 10{sup 6} yr at the Kauai site. Peak net N mineralization and nitrification values were found in soils from the 185000-yr-old Kohala site. Nitrogen content of foliage and leaf litter was highest in the intermediate age sites (Puu Makaala and Kohala) and N and P retranslocation was lowest at the Puu Makaala site. Soil cores fertilized with nitrogen had significantly higher rates of root ingrowth than control cores at the two youngest sites (200 and 6000 yr old) but not in older sites (185000 and 4.5 x 10{sup 6}-yr-old sites) and total fine root growth into control cores was greatest at the Kohala site. The highest N{sub 2}O emissions were found at the 185000-yr-old Kohala site, while the highest combined flux of N{sub 2}O + NO was observed at the 4.5 x 10{sup 6}-yr-old Kauai site. While overall N{sub 2}O emission rates were correlated with rates of N transformations, soil water content appeared to influence the magnitude of emissions of N{sub 2}O and the ratios of emissions of NO vs. N{sub 2}O. N{sub 2}O emissions occurred when water-filled pore space (WFPS) values were >40%, with highest emissions in at least two sites observed at WFPS values of 75%. Among sites, high N{sub 2}O emissions were associated with high soil N transformation rates. Large NO fluxes were observed only at the Kauai site when WFPS values were <60%. 50 refs., 8 figs., 4 tabs.

  20. Plant Available Nutrients, Barrow, Alaska, Ver. 1

DOE Data Explorer [Office of Scientific and Technical Information (OSTI)]

Sloan, Victoria; Liebig, Jenny; Curtis, Bryan; Hahn, Melanie; Iversen, Colleen; Siegrist, Julie

    2014-02-19

This dataset consists of measurements of plant available nutrients made using Plant Root Simulator probes (Western Ag Innovations Inc.) during 2012 and 2013. In 2012, Ca, Mg, K, P, Fe, Mn, Cu, Zn, B, S, Pb, Al, Cd, NO3-N and NH4-N were measured during spring, summer and winter in the centers, edges and troughs of four polygons in each of four areas of contrasting moisture regime and polygon type. In 2013, probes were installed in centers, edges and troughs of four polygons in each of two areas (high-centered and low-centered polygons) at two-week intervals and at 3 soil depths to capture fine-scale season dynamics of NO3-N and NH4-N. PRS probes are ion exchange resin membranes held in plastic supports that are inserted into soil to measure ion supply in situ. The anion and cation exchange with the membrane is intended to mimic plant uptake and thus provide a relevant measure of soil nutrient bioavailability. Measurements are made per area of probe membrane and cannot be converted to concentrations or related to soil volume.

  19. Worker Safety and Security Teams Team Member Handbook

    SciTech Connect (OSTI)

    Sievers, Cindy S.

    2012-06-11

Worker Safety and Security Teams (WSSTs) are an effective way to promote safe workplaces. While WSSTs have a variety of structures and roles, they have one thing in common - employees and management collaborate to find ways to prevent accidents, injuries, and illnesses on the job. The benefits for all concerned are obvious in that employees have a safe place to work, employers save money on lost work time and workers compensation costs, and everyone returns home safe and healthy each day. A successful WSST will have the support and wholehearted participation of management and employees. LANL has a WSST at the institutional level (IWSST) and at all directorates and many divisions. The WSSTs are part of LANL's Voluntary Protection Program (VPP). The WSSTs meet at least monthly and follow an agenda covering topics such as safety shares, behavior based safety (BBS) observations, upcoming events or activities, issues, etc. A WSST can effectively influence safety programs and provide recommendations to managers, who have the resources and authority to implement changes in the workplace. WSSTs are effective because they combine the knowledge, expertise, perspective, enthusiasm, and effort of a variety of employees with diverse backgrounds. Those with experience in a specific job or work area know what the hazards or potential hazards are, and generally have ideas how to go about controlling them. Those who are less familiar with a job or area play a vital role too, by seeing what others may have overlooked or taken for granted. This booklet will cover the structure and operations of WSSTs, what needs to be done in order to be effective and successful, and how you can help, whether you're a WSST member or not.

  4. Distributed control of multi-robot teams: Cooperative baton passing task

    SciTech Connect (OSTI)

    Parker, L.E.

    1998-11-01

This research addresses the problem of achieving fault tolerant cooperation within small- to medium-sized teams of heterogeneous mobile robots. The author describes a novel behavior-based, fully distributed architecture, called ALLIANCE, that utilizes adaptive action selection to achieve fault tolerant cooperative control. The robots in this architecture possess a variety of high-level functions that they can perform during a mission, and must at all times select an appropriate action based on the requirements of the mission, the activities of other robots, the current environmental conditions, and their own internal states. Since such cooperative teams often work in dynamic and unpredictable environments, the software architecture allows the team members to respond robustly and reliably to unexpected environmental changes and modifications in the robot team that may occur due to mechanical failure, the learning of new skills, or the addition or removal of robots from the team by human intervention. After presenting ALLIANCE, they describe the implementation of this architecture on a team of physical mobile robots performing a cooperative baton passing task. These experiments illustrate the ability of ALLIANCE to achieve adaptive, fault-tolerant cooperative control amidst dynamic changes during the task.

  11. Energy flow, nutrient cycling, and ecosystem resilience

    SciTech Connect (OSTI)

    DeAngelis, D.L.

    1980-08-01

The resilience, defined here as the speed with which a system returns to equilibrium state following a perturbation, is investigated for both food web energy models and nutrient cycling models. Previous simulation studies of food web energy models have shown that resilience increases as the flux of energy through the food web per unit amount of energy in the steady state web increases. Studies of nutrient cycling models have shown that resilience increases as the mean number of cycles that nutrient (or other mineral) atoms make before leaving the system decreases. In the present study these conclusions are verified analytically for general ecosystem models. The behavior of resilience in food web energy models and nutrient cycling models is a reflection of the time that a given unit, whether of energy or matter, spends in the steady state system. The shorter this residence time is, the more resilient the system is.

Final Report for the NA22 Simulations, Algorithm and Modeling (SAM) Ontology Assessment Team's efforts from FY09-FY11. The Ontology Assessment Team began in May 2009 and concluded in September 2011. During this two-year time frame, the Ontology Assessment team had two objectives: (1) Assessing the utility of knowledge representation and semantic technologies for addressing nuclear nonproliferation challenges; and (2) Developing ontological support tools that would provide a framework for integrating across the Simulation, Algorithm and Modeling (SAM) program. The SAM Program was going through a large assessment and strategic planning effort during this time and as a result, the relative importance of these two objectives changed, altering the focus of the Ontology Assessment Team. In the end, the team conducted an assessment of the state of art, created an annotated bibliography, and developed a series of ontological support tools, demonstrations and presentations. A total of more than 35 individuals from 12 different research institutions participated in the Ontology Assessment Team. These included subject matter experts in several nuclear nonproliferation-related domains as well as experts in semantic technologies. Despite the diverse backgrounds and perspectives, the Ontology Assessment team functioned very well together and aspects could serve as a model for future inter-laboratory collaborations and working groups. While the team encountered several challenges and learned many lessons along the way, the Ontology Assessment effort was ultimately a success that led to several multi-lab research projects and opened up a new area of scientific exploration within the Office of Nuclear Nonproliferation and Verification.

Red teams that address complex systems have rarely taken advantage of Modeling and Simulation (M&S) in a way that reproduces most or all of a red-blue team exchange within a computer. Chess programs, starting with IBM's Deep Blue, outperform humans in that red-blue interaction, so why shouldn't we think computers can outperform traditional red teams now or in the future? This and future position papers will explore possible ways to use M&S to augment or replace traditional red teams in some situations, the features Red Team M&S should possess, how one might connect live and simulated red teams, and existing tools in this domain.

    2004-01-01

Large government organizations such as the Department of Energy (DOE) are challenged with identifying and implementing best geospatial information management practices to ensure that operational needs are met and government objectives are achieved. Geographic Information System (GIS) professionals, complex wide within the Department, conduct spatial information management practices on a daily basis to complete a wide variety of science and engineering tasks. The DOE Office of the CIO recognized the wealth of geospatial information management knowledge within the DOE complex and formed the DOE GIS Core Team in 2001 as a result. The team is comprised of GIS experts-representing all major DOE labs, site facilities, and programs-who volunteer their time to address issues impacting the entire complex. These include the President's management agenda (with emphasis on the Geospatial One-Stop), homeland security, emergency response, site management, software and geospatial data licensing, and federal, national, and international standards governing the creation and dissemination of geospatial data. The strength of the DOE GIS Core Team is the wide diversity of GIS and scientific expertise represented on the team, which allows it to provide the DOE CIO's office with sound guidance on complex wide issues from a GIS practitioner's perspective. The Core Team's mission is 'to foster technical excellence and communication, to identify and advocate best business practices, and to provide sound recommendations on policy and standards.' As a first step toward identifying best practices the feam conducted a survey of all known GIS assets across the DOE complex. Assembling an On-Site Team Assembling an On-Site Team Assembling an on-site team is the second step in planning for a federal site solar project. A solar project team is important both to help get the work done and to make sure that all issues are considered. Even small oversights can be costly in terms of dollars and time, and can result in a failure to accomplish project goals. One of the most important features of the team should be its alignment with the project's goals. The project goals

    2014-06-01

Probabilistic Risk Assessment (PRA) and Human Reliability Assessment (HRA) are important technical contributors to the United States (U.S.) Nuclear Regulatory Commission’s (NRC) risk-informed and performance based approach to regulating U.S. commercial nuclear activities. Furthermore, all currently operating commercial NPPs in the U.S. are required by federal regulation to be staffed with crews of operators. Yet, aspects of team performance are underspecified in most HRA methods that are widely used in the nuclear industry. There are a variety of "emergent" team cognition and teamwork errors (e.g., communication errors) that are 1) distinct from individual human errors, and 2) important to understand from a PRA perspective. The lack of robust models or quantification of team performance is an issue that affects the accuracy and validity of HRA methods and models, leading to significant uncertainty in estimating HEPs. This paper describes research that has the objective to model and quantify team dynamics and teamwork within NPP control room crews for risk informed applications, thereby improving the technical basis of HRA, which improves the risk-informed approach the NRC uses to regulate the U.S. commercial nuclear industry.

  15. Multiple soil nutrient competition between plants, microbes, and mineral surfaces: model development, parameterization, and example applications in several tropical forests

DOE Public Access Gateway for Energy & Science Beta (PAGES Beta)

    Zhu, Q.; Riley, W. J.; Tang, J.; Koven, C. D.

    2015-03-05

Soil is a complex system where biotic (e.g., plant roots, micro-organisms) and abiotic (e.g., mineral surfaces) consumers compete for resources necessary for life (e.g., nitrogen, phosphorus). This competition is ecologically significant, since it regulates the dynamics of soil nutrients and controls aboveground plant productivity. Here we develop, calibrate, and test a nutrient competition model that accounts for multiple soil nutrients interacting with multiple biotic and abiotic consumers. As applied here for tropical forests, the Nutrient COMpetition model (N-COM) includes three primary soil nutrients (NH4+, NO3?, and POx (representing the sum of PO43?, HPO42?, and H2PO4?)) and five potential competitors (plantmoreroots, decomposing microbes, nitrifiers, denitrifiers, and mineral surfaces). The competition is formulated with a quasi-steady-state chemical equilibrium approximation to account for substrate (multiple substrates share one consumer) and consumer (multiple consumers compete for one substrate) effects. N-COM successfully reproduced observed soil heterotrophic respiration, N2O emissions, free phosphorus, sorbed phosphorus, and free NH4+ at a tropical forest site (Tapajos). The overall model posterior uncertainty was moderately well constrained. Our sensitivity analysis revealed that soil nutrient competition was primarily regulated by consumer-substrate affinity rather than environmental factors such as soil temperature or soil moisture. Our results imply that the competitiveness (from most to least competitive) followed this order: (1) for NH4+, nitrifiers ~ decomposing microbes > plant roots, (2) for NO3?, denitrifiers ~ decomposing microbes > plant roots, (3) for POx, mineral surfaces > decomposing microbes ~ plant roots. Although smaller, plant relative competitiveness is of the same order of magnitude as microbes. We then applied the N-COM model to analyze field nitrogen and phosphorus perturbation experiments in two tropical forest sites (in Hawaii and Puerto Rico) not used in model development or calibration. Under soil inorganic nitrogen and phosphorus elevated conditions, the model accurately replicated the experimentally observed competition among different nutrient consumers. Although we used as many observations as we could obtain, more nutrient addition experiments in tropical systems would greatly benefit model testing and calibration. In summary, the N-COM model provides an ecologically consistent representation of nutrient competition appropriate for land BGC models integrated in Earth System Models.less
