    SciTech Connect (OSTI)

    Lee, S.; Barnes, O.

    2014-11-17

The hydraulic results demonstrate that pump head pressure of 20 psi recirculates about 5.6 liters/min flowrate through the existing 0.131-inch orifice when a valve connected to NT-41 is closed. In case of the valve open to NT-41, the solution flowrates to HB-Line tanks, NT-21 and NT-41, are found to be about 0.5 lpm and 5.2 lpm, respectively. The modeling calculations for the mixing operations of miscible fluids contained in the HB-Line tank NT-21 were performed by taking a three-dimensional Computational Fluid Dynamics (CFD) approach. The CFD modeling results were benchmarked against the literature results and the previous SRNL test results to validate the model. Final performance calculations were performed for the nominal case by using the validated model to quantify the mixing time for the HB-Line tank. The results demonstrate that when a pump recirculates a solution volume of 5.7 liters every minute out of the 72-liter tank contents containing two acid solutions of 2.7 M and 0 M concentrations (i.e., water), a minimum mixing time of 1.5 hours is adequate for the tank contents to get the tank contents adequately mixed. In addition, the sensitivity results for the tank contents of 8 M existing solution and 1.5 M incoming species show that the mixing time takes about 2 hours to get the solutions mixed.

Spores of the anaerobic bacterium Clostridium novyi-NT are able to germinate in and destroy hypoxic regions of tumors in experimental animals. Future progress in this area will benefit from a better understanding of the germination and outgrowth processes that are essential for the tumorilytic properties of these spores. Towards this end, we have used both transmission electron microscopy and atomic force microscopy to determine the structure of dormant as well as germinating spores. We found that the spores are surrounded by an amorphous layer intertwined with honeycomb parasporal layers. Moreover, the spore coat layers had apparently self-assembled and this assembly was likely to be governed by crystal growth principles. During germination and outgrowth, the honeycomb layers as well as the underlying spore coat and undercoat layers sequentially dissolved until the vegetative cell was released. In addition to their implications for understanding the biology of C. novyi-NT, these studies document the presence of proteinaceous growth spirals in a biological organism.

  15. Landfill reduction experience in The Netherlands

    SciTech Connect (OSTI)

    Scharff, Heijo

    2014-11-15

Highlights: • ‘Zero waste’ initiatives never consider risks, side effects or experience of achieved low levels of landfill. • This paper provides insight into what works and what not. • Where strong gradients in regulations and tax occur between countries, waste will find its way to landfills across borders. • Strong landfill reduction can create a fierce competition over the remaining waste to be landfilled resulting in losses. • At some point a public organisation should take responsibility for the operation of a ‘safety net’ in waste management. - Abstract: Modern waste legislation aims at resource efficiency and landfill reduction. This paper analyses more than 20 years of landfill reduction in the Netherlands. The combination of landfill regulations, landfill tax and landfill bans resulted in the desired landfill reduction, but also had negative effects. A fierce competition developed over the remaining waste to be landfilled. In 2013 the Dutch landfill industry generated €40 million of annual revenue, had €58 million annual costs and therefore incurred an annual loss of €18 million. It is not an attractive option to prematurely end business. There is a risk that Dutch landfill operators will not be able to fulfil the financial obligations for closure and aftercare. Contrary to the polluter pays principle the burden may end up with society. EU regulations prohibiting export of waste for disposal are in place. Strong differentials in landfill tax rate between nations have nevertheless resulted in transboundary shipment of waste and in non-compliance with the self-sufficiency and proximity principles. During the transformation from a disposal society to a recycling society, it is important to carefully plan required capacity and to guide the reorganisation of the landfill sector. At some point, it is no longer profitable to provide landfill services. It may be necessary for public organisations or the state to take responsibility for the continued operation of a ‘safety net’ in waste management. Regulations have created a financial incentive to pass on the burden of monitoring and controlling the impact of waste to future generations. To prevent this, it is necessary to revise regulations on aftercare and create incentives to actively stabilise landfills.

  7. Social impacts of earthquakes caused by gas extraction in the Province of Groningen, The Netherlands

    SciTech Connect (OSTI)

Voort, Nick van der Vanclay, Frank

    2015-01-15

Gas extraction from the Groningen gasfield in the northern Netherlands has led to localised earthquakes which are projected to become more severe. The social impacts experienced by local residents include: damage to property; declining house prices; concerns about the chance of dykes breaking; feelings of anxiety and insecurity; health issues; and anger. These social and emotional impacts are exacerbated by the increasing distrust Groningen people have towards the national government and the gas company, NAM, a partnership between Shell and ExxonMobil. The earthquakes have reopened discussions about the distribution of benefits from gas production and the extent to which benefits are retained locally. Mitigation of the impacts is attempted, but the lack of trust decreases the effectiveness of the mitigation measures. The extent of this experience of previously-unforeseen, unanticipated impacts suggests that a new social and environmental impact assessment needs to be undertaken, and a new Social Impact Management Plan (SIMP) and Impacts and Benefits Agreement (IBA) developed, so that the project can regain its legitimacy and social licence to operate. In addition to conventional gas, this paper has wider relevance for unconventional gas developments, for example shale gas extraction by hydraulic fracturing methods (fracking). - Highlights: • Gas production in Groningen has caused over 1000 earthquakes. • The induced seismicity has caused many social impacts. • Impacts include building damage, reduced house prices, fear and health issues. • Mitigation measures attempted to date are inadequate. • Distrust towards the national government and operator hinders mitigation efforts. • Gas production in Groningen has lost its social licence to operate.

The Towers Display Program provides a convenient means of graphically depicting current wind speed and direction from a network of meteorological monitoring stations. The program was designed primarily for emergency response applications and, therefore, plots observed wind directions as a transport direction, i.e., the direction toward which the wind would transport a release of an atmospheric contaminant. Tabular summaries of wind speed and direction as well as temperature, relative humidity, and atmospheric turbulence measured atmore » each monitoring station can be displayed. The current implementation of the product at SRS displays data from eight Weather INformation and Display (WIND) System meteorological towers at SRS, meteorological stations established jointly by SRS/WSRC and the Augusta/Richmond County Emergency Management Agency in Augusta, GA, and National Weather Service stations in Augusta, GA. Wind speed and direction are plotted in a Beaufort scale format at the location of the station on a geographic map of the area. A GUI provides for easy specification of a desired date and time for the data to be displayed.« less

  18. Energy conservation and electricity sector liberalization: Case-studies on the development of cogeneration, wind energy and demand-side management in the Netherlands, Denmark, Germany and the United Kingdom

    SciTech Connect (OSTI)

    Slingerland, S.

    1998-07-01

In this paper, the development of cogeneration, wind energy and demand-side management in the Netherlands, Denmark, Germany and the United Kingdom are compared. It is discussed to what extent these developments are determined by the liberalization process. Three key liberalization variables are identified: unbundling, privatization and introduction of competition. The analysis suggests that unbundling prior to introduction of full competition in generation is particularly successful in stimulating industrial cogeneration; simultaneous introduction of competition and unbundling mainly stimulates non-cogeneration gas-based capacity; and introduction of competition in itself is likely to impede the development of district-heating cogeneration. Furthermore, it is argued that development of wind energy and demand-side management are primarily dependent on the kind of support system set up by policy makers rather than on the liberalization process. Negative impacts of introduction of competition on integrated resource planning and commercial energy services could nevertheless be expected.

  8. Trials with a 100% pellet burden in blast furnace No. 6 at Hoogovens IJmuiden

    SciTech Connect (OSTI)

    Schoone, E.; Toxopeus, H.; Vos, D.

    1995-12-01

The burden consists of 50% high basicity sinter and 50% home made olivine pellets. Two coke oven plants produce the required coke, about 340 kg/t (680 lb/NT). The average pulverized coal injection rate is 150 kg/t (300 lb/NT). To anticipate the aging coke oven plant No. 2 the coal injection capacity will e increased by 50% in 1996, by the installation of a third coal grinding line. In the Netherlands environmental issues have a high impact on further developments. In particular the environmental regulations require a significant decrease of dust, SO{sub 2} and dioxins emitted by the sinter plant. The appropriate measures must be concluded in the second part of this decade. To avoid costly conventional solutions Hoogovens has been testing since April, 1994 the Emission Optimized Sintering (EOS). In case of failure of EOS, the situation of a (partially) closed sinter plant was tested. Purchased pellets replaced sinter, leading to a 100% pellet and an 80% pellet/20% sinter trial. The trials were executed in the first half of 1994 at blast furnace No. 6, equipped with a PW-bell less top. Results are described.

  2. Pseudocapacitive Lithium-Ion Storage in Oriented Anatase TiO2 Nanotube Arrays

    SciTech Connect (OSTI)

    Zhu, K.; Wang, Q.; Kim, J. H.; Pesaran, A. A.; Frank, A. J.

    2012-06-07

We report on the synthesis and electrochemical properties of oriented anatase TiO{sub 2} nanotube (NT) arrays as electrodes for Li-ion batteries. The TiO{sub 2} NT electrodes displayed both pseudocapacitive Li{sup +} storage associated with the NT surface and the Li{sup +} storage within the bulk material. The relative contribution of the pseudocapacitive and bulk storages depends strongly on the scan rate. While the charges are stored primarily in the bulk at low scan rates (<< 1 mV/s), the surface storage dominates the total storage capacity at higher scan rates (>1 mV/s). The storage capacity of the NT electrodes as a function of charge/discharge rates showed no dependence on the NT film thickness, suggesting that the Li{sup +} insertion/extraction processes occur homogeneously across the entire length of NT arrays. These results indicated that the electron conduction along the NT walls and the ion conduction within the electrolyte do not cause significant hindering of the charge/discharge kinetics for NT electrode architectures. As a result of the surface pseudocapacitive storage, the reversible Li{sup +} storage capacities for TiO{sub 2} NT electrodes were higher than the theoretical storage capacity for bulk anatase TiO{sub 2} materials.

  5. Development of an ELISA microarray assay for the sensitive and simultaneous detection of ten biodefense toxins.

    2014-10-21

Plant and microbial toxins are considered bioterrorism threat agents because of their extreme toxicity and/or ease of availability. Additionally, some of these toxins are increasingly responsible for accidental food poisonings. The current study utilized an ELISA-based protein antibody microarray for the multiplexed detection of ten biothreat toxins, botulinum neurotoxins (BoNT) A, B, C, D, E, F, ricin, shiga toxins 1 and 2 (Stx), and staphylococcus enterotoxin B (SEB), in buffer and complex biological matrices. The multiplexed assay displayed a sensitivity of 1.3 pg/mL (BoNT/A, BoNT/B, SEB, Stx-1 and Stx-2), 3.3 pg/mL (BoNT/C, BoNT/E, BoNT/F) and 8.2 pg/mL (BoNT/D, ricin). All assays demonstrated high accuracy (75-120 percent recovery) and reproducibility (most coefficients of variation < 20%). Quantification curves for the ten toxins were also evaluated in clinical samples (serum, plasma, nasal fluid, saliva, stool, and urine) and environmental samples (apple juice, milk and baby food) with overall minimal matrix effects. The multiplex assays were highly specific, with little crossreactivity observed between the selected toxin antibodies. The results demonstrate a multiplex microarray that improves current immunoassay sensitivity for biological warfare agents in buffer, clinical, and environmental samples.

In this process, Na2CO3 reacts with CO2 and water to form sodium bicarbonate at the temperature of the flue gas

  8. Atomic-scale studies on the effect of boundary coherency on stability in twinned Cu

    SciTech Connect (OSTI)

Niu, Rongmei; Han, Ke Su, Yi-Feng; Salters, Vincent J.

    2014-01-06

The stored energy and hardness of nanotwinned (NT) Cu are related to interaction between dislocations and (111)-twin boundaries (TBs) studied at atomic scales by high-angle annular dark-field scanning transmission electron microscope. Lack of mobile dislocations at coherent TBs (CTBs) provides as-deposited NT Cu a rare combination of stability and hardness. The introduction of numerous incoherent TBs (ITBs) reduces both the stability and hardness. While storing more energy in their ITBs than in the CTBs, deformed NT Cu also exhibits high dislocation density and TB mobility and therefore has increased the driving force for recovery, coarsening, and recrystallization.

  9. Linear Transformation Method for Multinuclide Decay Calculation

    SciTech Connect (OSTI)

    Ding Yuan

    2010-12-29

A linear transformation method for generic multinuclide decay calculations is presented together with its properties and implications. The method takes advantage of the linear form of the decay solution N(t) = F(t)N{sub 0}, where N(t) is a column vector that represents the numbers of atoms of the radioactive nuclides in the decay chain, N{sub 0} is the initial value vector of N(t), and F(t) is a lower triangular matrix whose time-dependent elements are independent of the initial values of the system.

Kuzubov, Alexander A.; Kovaleva, Evgenia A. Avramov, Paul; Kuklin, Artem V.; Mikhaleva, Natalya S.; Tomilin, Felix N.; Sakai, Seiji; Entani, Shiro; Matsumoto, Yoshihiro; Naramoto, Hiroshi

    2014-08-28

The interaction between carbon and BN nanotubes (NT) and transition metal Co and Ni supports was studied using electronic structure calculations. Several configurations of interfaces were considered, and the most stable ones were used for electronic structure analysis. All NT/Co interfaces were found to be more energetically favorable than NT/Ni, and conductive carbon nanotubes demonstrate slightly stronger bonding than semiconducting ones. The presence of contact-induced spin polarization was established for all nanocomposites. It was found that the contact-induced polarization of BNNT leads to the appearance of local conductivity in the vicinity of the interface while the rest of the nanotube lattice remains to be insulating.

  10. Magnetic field fluctuations observed in the heliosheath and interstellar magnetic field by Voyager 1 at 115.7-124.9 AU during 2011-2013

We discuss microscale fluctuations of the hour averages of the magnetic field B observed on a scale of one day by Voyager 1 (V1) from 2011.0 to 2012.3143 (when it was within the distant heliosheath, where the average magnetic field strength (B) = 0.17 nT) and during the interval from 2012.6503 to 2013.5855 (when it was within the interstellar plasma with (B) = 0.47 nT). In both regions, the fluctuations were primarily compressive fluctuations, varying along the average B (?T direction in RTN coordinates). In the heliosheath, the average of the daily standard deviations (SDs) of the compressive and transverse components of B were (SD{sub c}) = 0.010 nT and (SD{sub t}) ? 0.005 nT (which is the limit of the measurement). In the distant heliosheath (SD{sub c})/(B) = 0.06, and the distributions of SD were skewed and highly kurtotic. The interstellar magnetic field (ISMF) strength was B = 0.48 nT, but the fluctuations were below the limit of measurement: (SD{sub c}) = 0.004 nT and (abs(SD{sub t})) = 0.004 nT. The distributions of these interstellar SDs have skewness and kurtosis consistent with a Gaussian noise distribution. We also discuss the fluctuations of 48 s averages of B on a scale of 1 day during a 30 day interval when V1 was observing the ISMF. For the fluctuations in all three components of B, SD = 0.010 nT, which gives an upper limit on the fluctuations of the ISMF on the scales observed by V1. This SD rules out the possibility that there is significant power in electromagnetic fluctuations generated by pickup ion ring instabilities at these scales, which strongly constrains models of the IBEX ribbon.

  14. Polyneutron Chain Reactions

    SciTech Connect (OSTI)

    John C. Fisher

    2000-11-12

Although helium atoms do not form molecules, a sufficiently large number will bind into a stable liquid droplet. A comparable situation is expected for neutrons, with a sufficiently large number binding into a stable droplet of neutron matter. Such polyneutron droplets can be viewed as isotopes of an element with nuclear charge Z=0, tentatively denoted neutrium, symbol Nt. Because of the relatively weak binding of neutrons compared with that of a mix of neutrons and protons, the minimum number of neutrons required for stability of a droplet is fairly large. Early estimates of {approx}60 may be reduced to a dozen or so by the BCS pairing interaction. The Nt entries with N{>=}12 are new to the table of isotopes. Because all of them are beta-unstable, none is expected to persist as a free particle. Yet, some may occasionally be produced by means to be described below, and it is of interest to examine their decay chains and their interactions with charged nuclei to ascertain how their presence might be revealed. Although these reactions are interesting, they cannot be taken seriously without identifying a source for the initial Nt isotope that begins the chain. Here, we consider possible interactions between {sup 16}O and {sup A}Nt. Although there is no strong interaction between them, we can expect a very weak residual attraction that can form a loosely bound {sup 16}O {sup A}Nt nuclear molecule. This is not a compound nucleus in the usual sense because, considered as fluids, the {sup 16}O and {sup A}Nt droplets are immiscible. For a droplet with fewer than about 60 neutrons, beta decay of {sup A}Nt is prevented by the buildup of Coulomb energy associated with transforming {sup A}Nt into {sup A}H in close proximity to {sup 16}O. Thus, it is possible that {sup 16}O {sup A}Nt molecules can persist indefinitely and that a few of them may be present in ordinary water as supermassive oxygen nuclei. Because the binding of these molecules is weak, the {sup A}Nt component can tunnel to an adjacent nucleus, and if the adjacent nucleus is {sup 18}O, a chain reaction can begin. The circumstances under which it can develop to produce macroscopic consequences depend on the mix of reactants and upon the appropriate removal of poisons and addition of fresh reactants to the reaction volume. With the proper conditions, there can be generation of sensible excess energy, helium, and other reaction products associated with the various cold fusion reactions.

  17. Intranuclear interactomic inhibition of NF-κB suppresses LPS-induced severe sepsis

    2015-08-28

Suppression of nuclear factor-κB (NF-κB) activation, which is best known as a major regulator of innate and adaptive immune responses, is a potent strategy for the treatment of endotoxic sepsis. To inhibit NF-κB functions, we designed the intra-nuclear transducible form of transcription modulation domain (TMD) of RelA (p65), called nt-p65-TMD, which can be delivered effectively into the nucleus without influencing the cell viability, and work as interactomic inhibitors via disruption of the endogenous p65-mediated transcription complex. nt-p65-TMD effectively inhibited the secretion of pro-inflammatory cytokines, including TNF-α, IL-1β, or IL-6 from BV2 microglia cells stimulated by lipopolysaccharide (LPS). nt-p65-TMD did not inhibit tyrosine phosphorylation of signaling mediators such as ZAP-70, p38, JNK, or ERK involved in T cell activation, but was capable of suppressing the transcriptional activity of NF-κB without the functional effect on that of NFAT upon T-cell receptor (TCR) stimulation. The transduced nt-p65-TMD in T cell did not affect the expression of CD69, however significantly inhibited the secretion of T cell-specific cytokines such as IL-2, IFN-γ, IL-4, IL-17A, or IL-10. Systemic administration of nt-p65-TMD showed a significant therapeutic effect on LPS-induced sepsis model by inhibiting pro-inflammatory cytokines secretion. Therefore, nt-p65-TMD can be a novel therapeutics for the treatment of various inflammatory diseases, including sepsis, where a transcription factor has a key role in pathogenesis, and further allows us to discover new functions of p65 under normal physiological condition without genetic alteration. - Highlights: • The nt-p65-TMD is intra-nuclear interactomic inhibitor of endogenous p65. • The nt-p65-TMD effectively inhibited the secretion of pro-inflammatory cytokines. • The excellent therapeutic potential of nt-p65-TMD was confirmed in sepsis model.

    SciTech Connect (OSTI)

    Blumberg, A.Y.; Crossley, D.A. Jr.

    1986-01-01

The design for an inexpensive time-sorting pitfall trap is presented. The basis of the mechanism is a rotary stepping solenoid powered by lantern batteries. Traps were utilized to sample soil surface arthropods at two hour intervals for five 24 hr periods in 1983. One trap each was placed in conventional tillage (CT) and no-tillage (NT) agroecosystems. Soil arthropod surface activity was greatest in CT on 9 July during the dawn and dusk periods but the data did not indicate other dominant trends. Activity in NT was greatest during dusk on 27 June, but again no other dominant trends were evident. When CT and NT are combined over the sample dates, surface soil arthropod activity peaked during dusk, with a smaller activity peak at dawn. 10 refs., 6 figs.

  20. A protein chip membrane-capture assay for botulinum neurotoxin activity

    SciTech Connect (OSTI)

Marconi, Severine; Ferracci, Geraldine; Berthomieu, Maelys; Kozaki, Shunji; Miquelis, Raymond; Boucraut, Jose; Seagar, Michael

    2008-12-15

Botulinum neurotoxins A and B (BoNT/A and B) are neuromuscular blocking agents which inhibit neurotransmission by cleaving the intra-cellular presynaptic SNARE proteins SNAP-25 and VAMP2, localized respectively in plasma membrane and synaptic vesicles. These neurotoxins are both dangerous pathogens and powerful therapeutic agents with numerous clinical and cosmetic applications. Consequently there is a need for in vitro assays of their biological activity to screen for potential inhibitors and to replace the widely used in vivo mouse assay. Surface plasmon resonance (SPR) was used to measure membrane vesicle capture by antibodies against SNAP-25 and VAMP2. Substrate cleavage by BoNTs modified capture providing a method to assay toxin activity. Firstly using synaptic vesicles as a substrate, a comparison of the EC{sub 50}s for BoNT/B obtained by SPR, ELISA or flow cytometry indicated similar sensitivity although SPR assays were more rapid. Sonication of brain or neuronal cultures generated plasma membrane fragments with accessible intra-cellular epitopes adapted to measurement of BoNT/A activity. SPR responses were proportional to antigen concentration permitting detection of as little as 4 pM SNAP-25 in crude lysates. BoNT/A activity was assayed using monoclonal antibodies that specifically recognize a SNAP-25 epitope generated by the proteolytic action of the toxin. Incubation of intact primary cultured neurons with BoNT/A yielded an EC{sub 50} of 0.5 pM. The SPR biosensor method was sensitive enough to monitor BoNT/A and B activity in cells cultured in a 96-well format providing an alternative to experimental animals for toxicological assays.
