  11. HYDRAULICS AND MIXING EVALUATIONS FOR NT-21/41 TANKS

    SciTech Connect (OSTI)

    Lee, S.; Barnes, O.

    2014-11-17

The hydraulic results demonstrate that pump head pressure of 20 psi recirculates about 5.6 liters/min flowrate through the existing 0.131-inch orifice when a valve connected to NT-41 is closed. In case of the valve open to NT-41, the solution flowrates to HB-Line tanks, NT-21 and NT-41, are found to be about 0.5 lpm and 5.2 lpm, respectively. The modeling calculations for the mixing operations of miscible fluids contained in the HB-Line tank NT-21 were performed by taking a three-dimensional Computational Fluid Dynamics (CFD) approach. The CFD modeling results were benchmarked against the literature results and the previous SRNL test results to validate the model. Final performance calculations were performed for the nominal case by using the validated model to quantify the mixing time for the HB-Line tank. The results demonstrate that when a pump recirculates a solution volume of 5.7 liters every minute out of the 72-liter tank contents containing two acid solutions of 2.7 M and 0 M concentrations (i.e., water), a minimum mixing time of 1.5 hours is adequate for the tank contents to get the tank contents adequately mixed. In addition, the sensitivity results for the tank contents of 8 M existing solution and 1.5 M incoming species show that the mixing time takes about 2 hours to get the solutions mixed.

Spores of the anaerobic bacterium Clostridium novyi-NT are able to germinate in and destroy hypoxic regions of tumors in experimental animals. Future progress in this area will benefit from a better understanding of the germination and outgrowth processes that are essential for the tumorilytic properties of these spores. Towards this end, we have used both transmission electron microscopy and atomic force microscopy to determine the structure of dormant as well as germinating spores. We found that the spores are surrounded by an amorphous layer intertwined with honeycomb parasporal layers. Moreover, the spore coat layers had apparently self-assembled and this assembly was likely to be governed by crystal growth principles. During germination and outgrowth, the honeycomb layers as well as the underlying spore coat and undercoat layers sequentially dissolved until the vegetative cell was released. In addition to their implications for understanding the biology of C. novyi-NT, these studies document the presence of proteinaceous growth spirals in a biological organism.

  14. Landfill reduction experience in The Netherlands

    SciTech Connect (OSTI)

    Scharff, Heijo

    2014-11-15

Highlights: • ‘Zero waste’ initiatives never consider risks, side effects or experience of achieved low levels of landfill. • This paper provides insight into what works and what not. • Where strong gradients in regulations and tax occur between countries, waste will find its way to landfills across borders. • Strong landfill reduction can create a fierce competition over the remaining waste to be landfilled resulting in losses. • At some point a public organisation should take responsibility for the operation of a ‘safety net’ in waste management. - Abstract: Modern waste legislation aims at resource efficiency and landfill reduction. This paper analyses more than 20 years of landfill reduction in the Netherlands. The combination of landfill regulations, landfill tax and landfill bans resulted in the desired landfill reduction, but also had negative effects. A fierce competition developed over the remaining waste to be landfilled. In 2013 the Dutch landfill industry generated €40 million of annual revenue, had €58 million annual costs and therefore incurred an annual loss of €18 million. It is not an attractive option to prematurely end business. There is a risk that Dutch landfill operators will not be able to fulfil the financial obligations for closure and aftercare. Contrary to the polluter pays principle the burden may end up with society. EU regulations prohibiting export of waste for disposal are in place. Strong differentials in landfill tax rate between nations have nevertheless resulted in transboundary shipment of waste and in non-compliance with the self-sufficiency and proximity principles. During the transformation from a disposal society to a recycling society, it is important to carefully plan required capacity and to guide the reorganisation of the landfill sector. At some point, it is no longer profitable to provide landfill services. It may be necessary for public organisations or the state to take responsibility for the continued operation of a ‘safety net’ in waste management. Regulations have created a financial incentive to pass on the burden of monitoring and controlling the impact of waste to future generations. To prevent this, it is necessary to revise regulations on aftercare and create incentives to actively stabilise landfills.

    1999-05-20

The Towers Display Program provides a convenient means of graphically depicting current wind speed and direction from a network of meteorological monitoring stations. The program was designed primarily for emergency response applications and, therefore, plots observed wind directions as a transport direction, i.e., the direction toward which the wind would transport a release of an atmospheric contaminant. Tabular summaries of wind speed and direction as well as temperature, relative humidity, and atmospheric turbulence measured atmore » each monitoring station can be displayed. The current implementation of the product at SRS displays data from eight Weather INformation and Display (WIND) System meteorological towers at SRS, meteorological stations established jointly by SRS/WSRC and the Augusta/Richmond County Emergency Management Agency in Augusta, GA, and National Weather Service stations in Augusta, GA. Wind speed and direction are plotted in a Beaufort scale format at the location of the station on a geographic map of the area. A GUI provides for easy specification of a desired date and time for the data to be displayed.« less

  17. Energy conservation and electricity sector liberalization: Case-studies on the development of cogeneration, wind energy and demand-side management in the Netherlands, Denmark, Germany and the United Kingdom

    SciTech Connect (OSTI)

    Slingerland, S.

    1998-07-01

In this paper, the development of cogeneration, wind energy and demand-side management in the Netherlands, Denmark, Germany and the United Kingdom are compared. It is discussed to what extent these developments are determined by the liberalization process. Three key liberalization variables are identified: unbundling, privatization and introduction of competition. The analysis suggests that unbundling prior to introduction of full competition in generation is particularly successful in stimulating industrial cogeneration; simultaneous introduction of competition and unbundling mainly stimulates non-cogeneration gas-based capacity; and introduction of competition in itself is likely to impede the development of district-heating cogeneration. Furthermore, it is argued that development of wind energy and demand-side management are primarily dependent on the kind of support system set up by policy makers rather than on the liberalization process. Negative impacts of introduction of competition on integrated resource planning and commercial energy services could nevertheless be expected.

  7. Trials with a 100% pellet burden in blast furnace No. 6 at Hoogovens IJmuiden

    SciTech Connect (OSTI)

    Schoone, E.; Toxopeus, H.; Vos, D.

    1995-12-01

The burden consists of 50% high basicity sinter and 50% home made olivine pellets. Two coke oven plants produce the required coke, about 340 kg/t (680 lb/NT). The average pulverized coal injection rate is 150 kg/t (300 lb/NT). To anticipate the aging coke oven plant No. 2 the coal injection capacity will e increased by 50% in 1996, by the installation of a third coal grinding line. In the Netherlands environmental issues have a high impact on further developments. In particular the environmental regulations require a significant decrease of dust, SO{sub 2} and dioxins emitted by the sinter plant. The appropriate measures must be concluded in the second part of this decade. To avoid costly conventional solutions Hoogovens has been testing since April, 1994 the Emission Optimized Sintering (EOS). In case of failure of EOS, the situation of a (partially) closed sinter plant was tested. Purchased pellets replaced sinter, leading to a 100% pellet and an 80% pellet/20% sinter trial. The trials were executed in the first half of 1994 at blast furnace No. 6, equipped with a PW-bell less top. Results are described.

  19. Pseudocapacitive Lithium-Ion Storage in Oriented Anatase TiO2 Nanotube Arrays

    SciTech Connect (OSTI)

    Zhu, K.; Wang, Q.; Kim, J. H.; Pesaran, A. A.; Frank, A. J.

    2012-06-07

We report on the synthesis and electrochemical properties of oriented anatase TiO{sub 2} nanotube (NT) arrays as electrodes for Li-ion batteries. The TiO{sub 2} NT electrodes displayed both pseudocapacitive Li{sup +} storage associated with the NT surface and the Li{sup +} storage within the bulk material. The relative contribution of the pseudocapacitive and bulk storages depends strongly on the scan rate. While the charges are stored primarily in the bulk at low scan rates (<< 1 mV/s), the surface storage dominates the total storage capacity at higher scan rates (>1 mV/s). The storage capacity of the NT electrodes as a function of charge/discharge rates showed no dependence on the NT film thickness, suggesting that the Li{sup +} insertion/extraction processes occur homogeneously across the entire length of NT arrays. These results indicated that the electron conduction along the NT walls and the ion conduction within the electrolyte do not cause significant hindering of the charge/discharge kinetics for NT electrode architectures. As a result of the surface pseudocapacitive storage, the reversible Li{sup +} storage capacities for TiO{sub 2} NT electrodes were higher than the theoretical storage capacity for bulk anatase TiO{sub 2} materials.

    2014-10-21

Plant and microbial toxins are considered bioterrorism threat agents because of their extreme toxicity and/or ease of availability. Additionally, some of these toxins are increasingly responsible for accidental food poisonings. The current study utilized an ELISA-based protein antibody microarray for the multiplexed detection of ten biothreat toxins, botulinum neurotoxins (BoNT) A, B, C, D, E, F, ricin, shiga toxins 1 and 2 (Stx), and staphylococcus enterotoxin B (SEB), in buffer and complex biological matrices. The multiplexed assay displayed a sensitivity of 1.3 pg/mL (BoNT/A, BoNT/B, SEB, Stx-1 and Stx-2), 3.3 pg/mL (BoNT/C, BoNT/E, BoNT/F) and 8.2 pg/mL (BoNT/D, ricin). All assays demonstrated high accuracy (75-120 percent recovery) and reproducibility (most coefficients of variation < 20%). Quantification curves for the ten toxins were also evaluated in clinical samples (serum, plasma, nasal fluid, saliva, stool, and urine) and environmental samples (apple juice, milk and baby food) with overall minimal matrix effects. The multiplex assays were highly specific, with little crossreactivity observed between the selected toxin antibodies. The results demonstrate a multiplex microarray that improves current immunoassay sensitivity for biological warfare agents in buffer, clinical, and environmental samples.

The interaction between carbon and BN nanotubes (NT) and transition metal Co and Ni supports was studied using electronic structure calculations. Several configurations of interfaces were considered, and the most stable ones were used for electronic structure analysis. All NT/Co interfaces were found to be more energetically favorable than NT/Ni, and conductive carbon nanotubes demonstrate slightly stronger bonding than semiconducting ones. The presence of contact-induced spin polarization was established for all nanocomposites. It was found that the contact-induced polarization of BNNT leads to the appearance of local conductivity in the vicinity of the interface while the rest of the nanotube lattice remains to be insulating.

A linear transformation method for generic multinuclide decay calculations is presented together with its properties and implications. The method takes advantage of the linear form of the decay solution N(t) = F(t)N{sub 0}, where N(t) is a column vector that represents the numbers of atoms of the radioactive nuclides in the decay chain, N{sub 0} is the initial value vector of N(t), and F(t) is a lower triangular matrix whose time-dependent elements are independent of the initial values of the system.

  5. Magnetic field fluctuations observed in the heliosheath and interstellar magnetic field by Voyager 1 at 115.7-124.9 AU during 2011-2013

    Burlaga, L. F.; Ness, N. F.; Florinski, V.; Heerikhuisen, J. E-mail: nfnudel@yahoo.com

    2014-09-10

We discuss microscale fluctuations of the hour averages of the magnetic field B observed on a scale of one day by Voyager 1 (V1) from 2011.0 to 2012.3143 (when it was within the distant heliosheath, where the average magnetic field strength (B) = 0.17 nT) and during the interval from 2012.6503 to 2013.5855 (when it was within the interstellar plasma with (B) = 0.47 nT). In both regions, the fluctuations were primarily compressive fluctuations, varying along the average B (?T direction in RTN coordinates). In the heliosheath, the average of the daily standard deviations (SDs) of the compressive and transverse components of B were (SD{sub c}) = 0.010 nT and (SD{sub t}) ? 0.005 nT (which is the limit of the measurement). In the distant heliosheath (SD{sub c})/(B) = 0.06, and the distributions of SD were skewed and highly kurtotic. The interstellar magnetic field (ISMF) strength was B = 0.48 nT, but the fluctuations were below the limit of measurement: (SD{sub c}) = 0.004 nT and (abs(SD{sub t})) = 0.004 nT. The distributions of these interstellar SDs have skewness and kurtosis consistent with a Gaussian noise distribution. We also discuss the fluctuations of 48 s averages of B on a scale of 1 day during a 30 day interval when V1 was observing the ISMF. For the fluctuations in all three components of B, SD = 0.010 nT, which gives an upper limit on the fluctuations of the ISMF on the scales observed by V1. This SD rules out the possibility that there is significant power in electromagnetic fluctuations generated by pickup ion ring instabilities at these scales, which strongly constrains models of the IBEX ribbon.

  12. Diurnal activity of soil surface arthropods in agroecosystems: design for an inexpensive time-sorting pitfall trap

    SciTech Connect (OSTI)

    Blumberg, A.Y.; Crossley, D.A. Jr.

    1986-01-01

The design for an inexpensive time-sorting pitfall trap is presented. The basis of the mechanism is a rotary stepping solenoid powered by lantern batteries. Traps were utilized to sample soil surface arthropods at two hour intervals for five 24 hr periods in 1983. One trap each was placed in conventional tillage (CT) and no-tillage (NT) agroecosystems. Soil arthropod surface activity was greatest in CT on 9 July during the dawn and dusk periods but the data did not indicate other dominant trends. Activity in NT was greatest during dusk on 27 June, but again no other dominant trends were evident. When CT and NT are combined over the sample dates, surface soil arthropod activity peaked during dusk, with a smaller activity peak at dawn. 10 refs., 6 figs.

Botulinum neurotoxins A and B (BoNT/A and B) are neuromuscular blocking agents which inhibit neurotransmission by cleaving the intra-cellular presynaptic SNARE proteins SNAP-25 and VAMP2, localized respectively in plasma membrane and synaptic vesicles. These neurotoxins are both dangerous pathogens and powerful therapeutic agents with numerous clinical and cosmetic applications. Consequently there is a need for in vitro assays of their biological activity to screen for potential inhibitors and to replace the widely used in vivo mouse assay. Surface plasmon resonance (SPR) was used to measure membrane vesicle capture by antibodies against SNAP-25 and VAMP2. Substrate cleavage by BoNTs modified capture providing a method to assay toxin activity. Firstly using synaptic vesicles as a substrate, a comparison of the EC{sub 50}s for BoNT/B obtained by SPR, ELISA or flow cytometry indicated similar sensitivity although SPR assays were more rapid. Sonication of brain or neuronal cultures generated plasma membrane fragments with accessible intra-cellular epitopes adapted to measurement of BoNT/A activity. SPR responses were proportional to antigen concentration permitting detection of as little as 4 pM SNAP-25 in crude lysates. BoNT/A activity was assayed using monoclonal antibodies that specifically recognize a SNAP-25 epitope generated by the proteolytic action of the toxin. Incubation of intact primary cultured neurons with BoNT/A yielded an EC{sub 50} of 0.5 pM. The SPR biosensor method was sensitive enough to monitor BoNT/A and B activity in cells cultured in a 96-well format providing an alternative to experimental animals for toxicological assays.

A fluorescence sandwich immunoassay using high affinity antibodies and quantum dot (QD) reporters has been developed for detection of botulinum toxin serotype A (BoNT/A). For the development of the assay, a nontoxic recombinant fragment of the holotoxin (BoNT/A-HC-fragment) has been used as a structurally valid simulant for the full toxin molecule. The antibodies used, AR4 and RAZ1, bind to nonoverlapping epitopes present on both the full toxin and on the recombinant fragment. In one format, the immunoassay is carried out in a 96-well plate with detection in a standard plate reader. Detection down to 31 pM of the BoNT/Hc-fragment was demonstrated with a total incubation time of 3 hours, using AR4 as the capture antibody and QD-coupled RAZ1 as the reporter. In a second format, the AR4 capture antibody was coupled to Sepharose beads, and the immunochemical reactions were carried out in microcentrifuge tubes with an incubation time of 1 hour. These beads were subsequently captured and concentrated in a rotating rod renewable surface flow cell as part of a sequential injection fluidic system. This flow cell was equipped with a fiber optic system for fluorescence measurements. In PBS buffer solution matrix, the BoNT/A-HC-fragment was detected to concentrations as low as 5 pM using the fluidic measurement approach.

  13. New Fabrication Method Improves the Efficiency and Economics of Solar Cells (Fact Sheet)

Synthetic fabrication strategy optimizes the illumination geometry and transport properties of dye-sensitized solar cells. Using oriented titanium oxide (TiO{sub 2}) nanotube (NT) arrays has shown promise for dye-sensitized solar cells (DSSCs). High solar conversion efficiency requires that the incident light enters the cell from the photoelectrode side. However, for NT-based DSSCs, the light normally enters the cell through the counter electrode because a nontransparent titanium foil is typically used as the substrate for forming the aligned NTs and for making electrical contact with them. It has been synthetically challenging to prepare transparent TiO{sub 2} NT electrodes by directly anodizing Ti metal films on transparent conducting oxide (TCO) substrates because it is difficult to control the synthetic conditions. National Renewable Energy Laboratory (NREL) researchers have developed a general synthetic strategy for fabricating transparent TiO{sub 2} NT films on TCO substrates. With the aid of a conducting Nb-doped TiO{sub 2} (NTO) layer between the Ti film and TCO substrate, the Ti film can be anodized completely without degrading the TCO. The NTO layer protects the TCO from degradation through a self-terminating mechanism by arresting the electric field-assisted dissolution process at the NT-NTO interface. NREL researchers found that the illumination direction and wavelength of the light incident on the DSSCs strongly influenced the incident photon-to-current conversion efficiency, light-harvesting, and charge-collection properties, which, in turn, affect the photocurrent density, photovoltage, and solar energy conversion efficiency. Researchers also examined the effects of NT film thickness on the properties and performance of DSSCs and found that illuminating the cell from the photoelectrode side substantially increased the conversion efficiency compared with illuminating it from the counter-electrode side. This method solves a key challenge in fabricating NT-based DSSCs and determines an optimal illumination direction to use in these cells. The synthetic fabrication strategy will improve the economics and conversion efficiency of DSSCs.

The seven antigenically distinct serotypes (A to G) of botulinum neurotoxin (BoNT) are responsible for the deadly disease botulism. BoNT serotype A (BoNT/A) exerts its lethal action by cleaving the SNARE protein SNAP-25, leading to inhibition of neurotransmitter release, flaccid paralysis and autonomic dysfunction. BoNTs are dichain proteins: the heavy chain is responsible for neurospecific binding, internalization and translocation, and the light chain is responsible for substrate cleavage. Because of their extreme toxicity and prior history of weaponization, the BoNTs are considered to be potential bioterrorism agents. No post-symptomatic therapeutic interventions are available for BoNT intoxication other than critical care; therefore it is imperative to develop specific antidotes against this neurotoxin. To this end, a cyclic peptide inhibitor (CPI-1) was synthesized and found to inhibit BoNT/A light chain (Balc) with high affinity. When tested in a cell-free Frster resonance excitation transfer (FRET) assay, CPI-1 was found to have a Ki of 13.9 nM using full-length Balc448 and 42.1 nM using a truncated crystallizable form of light chain (Balc424). Co-crystallization of CPI-1 with Balc424 revealed that in the Balc-CPI-1 complex, the inhibitor adopts a helical conformation, occupies a high percentage of the active site cavity and interacts in an amphipathic manner with critical active site residues. The data suggest that CPI-1 prevents SNAP-25 from accessing the Balc active site by blocking both the substrate binding path at the surface and the Zn2+ binding region involved in catalysis. This is in contrast to linear peptide inhibitors described to date which block only the latter

  15. Removing Structural Disorder from Oriented TiO2 Nanotube Arrays: Reducing the Dimensionality of Transport and Recombination in Dye-Sensitized Solar Cells

We report on the influence of morphological disorder, arising from bundling of nanotubes (NTs) and microcracks in films of oriented TiO{sub 2} NT arrays, on charge transport and recombination in dye-sensitized solar cells (DSSCs). Capillary stress created during evaporation of liquids from the mesopores of dense TiO{sub 2} NT arrays was of sufficient magnitude to induce bundling and microcrack formation. The average lateral deflection of the NTs in the bundles increased with the surface tension of the liquids and with the film thicknesses. The supercritical CO{sub 2} drying technique was used to produce bundle-free and crack-free NT films. Charge transport and recombination properties of sensitized films were studied by frequency-resolved modulated photocurrent/photovoltage spectroscopies. Transport became significantly faster with decreased clustering of the NTs, indicating that bundling creates additional pathways via intertube contacts. Removing such contacts alters the transport mechanism from a combination of one and three dimensions to the expected one dimension and shortens the electron-transport pathway. Reducing intertube contacts also resulted in a lower density of surface recombination centers by minimizing distortion-induced surface defects in bundled NTs. A causal connection between transport and recombination is observed. The dye coverage was greater in the more aligned NT arrays, suggesting that reducing intertube contacts increases the internal surface area of the films accessible to dye molecules. The solar conversion efficiency and photocurrent density were highest for DSSCs incorporating films with more aligned NT arrays owing to an enhanced light-harvesting efficiency. Removing structural disorder from other materials and devices consisting of nominally one-dimensional architectures (e.g., nanowire arrays) should produce similar effects.

We have investigated the structure and electronic properties of BN [1-4] and BC4N [5] nanotubes using density functional theory (DFT) simulations. The interaction between boron and neutron produce Li which has the potential to destabilize the structure as well as adversely affect the electronic properties. We have observed that addition of Li reduces the band gap of BNNT and BC4N nanotubes and makes them almost metallic. To prevent the reduction of band gap and to maintain structural integrity, we have proposed adding dopants in the nanotubes prior to irradiation with neutrons. We have successfully identified several dopants (F, O, Cl) which can retain the semiconducting and structural properties of BC4N nanostructures. In addition to BN NT, another promising nanostructure material for neutron detection is the BC4N nanotube (BC4N NT). BC4N nanotube brushes have been synthesized by the high temperature reaction of amorphous carbon nanotube (a-CNT) brushes with a mixture of boric acid and urea [5]. The most stable structure is predicted to be the one where BN3 and NB3 units are connected by a BN bond. The ground-state structures of BC4N nanotubes having different chirality; configurations (6,0) and (8,0) zigzag nanotubes, and (6,6) armchair nanotube have been reported to be semi conducting with various energy gaps. The (6,6) armchair BC4N nanotube shows the largest band gap (1.0 eV). The thermal stability of the BC4N nanotube is very high (up to 900 K); the presence of B and N makes this system more stable compared to the well-known carbon nanotube. Lithium produced due to interaction of boron and neutron modifies the structure and the electronic properties of the nanotube material. After losing its energy, Li can be absorbed in the vacant sites (e.g., N or B in BN NT or N, B or C in BC4N NT, or absorbed as an adatom in various parts of the nanotube system. So the nanostructure material which is initially suitable for direct conversion neutron detection can deteriorate after the accumulation of lithium. Our strategy in this study is to tailor the initial configuration such that the accumulation of lithium does not alter the properties. In this annual report we present the structure and electronic properties of pure BN NT and BC4N nanotubes, followed by an investigation on the effect of lithium (as adatoms and substitutional dopants in both BN NT and BC4N NT). Finally we tune the properties of (BNNT + Li) and (BC4N NT + Li) systems by adding appropriate dopants to negate the adverse effects of lithium.

The persistent photoconductivity (PPC) effect in nanotube arrays of barium titanate and TiO{sub 2} (BTO/TiO{sub 2}NT) was studied at room temperature under daylight illumination. The BTO/TiO{sub 2}NT heterostructures exhibited a giant PPC effect that was six orders of magnitude higher than the dark conductivity, followed by a slow relaxation for 3 h. The PPC in this material was explained by the existence of defects at the surfaces and the interfaces of the investigated heterostructures. The sample was prepared using a two-step synthesis: the anodization of a Ti-foil and a subsequent hydrothermal synthesis. The structural and electrical characteristics were studied by micro-Raman spectroscopy, field-emission-gun scanning electron microscopy, and impedance spectroscopy.

Carbon fibre reinforced cement based materials (CFRCs) offer the possibility of fabricating 'smart' electrically conductive materials. Rheology of the fresh mix is crucial to satisfactory moulding and fresh CFRC conforms to the Bingham model with slight structural breakdown. Both yield stress and plastic viscosity increase with increasing fibre length and volume concentration. Using a modified Viskomat NT, the concentration dependence of CFRC rheology up to 1.5% fibre volume is reported.
