Aspergillus niger is a filamentous ascomycete fungus that is ubiquitous in the environment and has been implicated in opportunistic infections of humans. In addition to its role as an opportunistic human pathogen, A. niger is economically important as a fermentation organism used for the production of citric acid. Industrial citric acid production by A. niger represents one of the most efficient, highest yield bioprocesses in use currently by industry. The genome size of A. niger is estimated to be between 35.5 and 38.5 megabases (Mb) divided among eight chromosomes/linkage groups that vary in size from 3.5 - 6.6 Mb. Currently, there are three independent A. niger genome projects, an indication of the economic importance of this organism. The rich amount of data resulting from these multiple A. niger genome sequences will be used for basic and applied research programs applicable to fermentation process development, morphology and pathogenicity.

Enzymes can be used to catalyze a myriad of chemical reactions and are a cornerstone in the biotechnology industry. Enzymes have a wide range of uses, ranging from medicine with the production of pharmaceuticals to energy were they are applied to biofuel production. It has been observed in Nigeria, that there is a correlation between the standard of living as measured by per capita GNP, and per capita energy consumption. As energy use per capita is tied to the importance of population increase it tends to drop during economic slow down. The per capita energy usage is put at 0.2 kw compared to 10 kw for USA and 4 kw for Europe respectively. Furthermore, analysis shows with the increase in population per year and a 2--5% growth in per capita GNP, require an increase of 5--8% in energy supply per year. The Country derives almost all its energy need from fossil fuels (petroleum, gas and coal), hydropower (the only renewable energy used for generating electricity at present) Wood, Animal, Human power and Wind. The Atmospheric Radiation Measurement (ARM) Program is the largest global change research program supported by the U.S. Department of Energy. The primary goal of the ARM Program is to improve the treatment of cloud and radiation physics in global climate models in order to improve the climate simulation capabilities of these models. To achieve this goal, ARM scientists and researchers around the world use continuous data obtained through the ARM Climate Research Facility. The ARM Mobile Facility (AMF) operates at non-permanent sites selected by the ARM Program. Sometimes these sites can become permanent ARM sites, as was the case with Graciosa Island in the Azores. It is now known as the Eastern North Atlantic permanent site. In January 2006 the AMF deployed to Niamey, Niger, West Africa, at the Niger Meteorological Office at Niamey International Airport. This deployment was timed to coincide with the field phases and Special Observing Periods of the African Monsoon Multidisciplinary Analysis (AMMA). The ARM Program participated in this international effort as a field campaign called "Radiative Divergence using AMF, GERB and AMMA Stations (RADAGAST).The primary purpose of the Niger deployment was to combine an extended series of measurements from the AMF with those from the Geostationary Earth Radiation Budget (GERB) Instrument on the Meteosat operational geostationary satellite in order to provide the first well-sampled, direct estimates of the divergence of solar and thermal radiation across the atmosphere. A large collection of data plots based on data streams from specific instruments used at Niamey are available via a link from ARM's Niamey, Niger site information page. Other data can be found at the related websites mentioned above and in the ARM Archive. Users will be requested to create a password, but the plots and data files are free for viewing and downloading. The ARM Archive physically resides at the Oak Ridge National Laboratory.

The filamentous fungus Aspergillus niger exhibits great diversity in its phenotype. It is found globally, both as marine and terrestrial strains, produces both organic acids and hydrolytic enzymes in high amounts, and some isolates exhibit pathogenicity. Although the genome of an industrial enzyme-producing A. niger strain (CBS 513.88) has already been sequenced, the versatility and diversity of this species compels additional exploration. We therefore undertook whole genome sequencing of the acidogenic A. niger wild type strain (ATCC 1015), and produced a genome sequence of very high quality. Only 15 gaps are present in the sequence and half the telomeric regions have been elucidated. Moreover, sequence information from ATCC 1015 was utilized to improve the genome sequence of CBS 513.88. Chromosome-level comparisons uncovered several genome rearrangements, deletions, a clear case of strain-specific horizontal gene transfer, and identification of 0.8 megabase of novel sequence. Single nucleotide polymorphisms per kilobase (SNPs/kb) between the two strains were found to be exceptionally high (average: 7.8, maximum: 160 SNPs/kb). High variation within the species was confirmed with exo-metabolite profiling and phylogenetics. Detailed lists of alleles were generated, and genotypic differences were observed to accumulate in metabolic pathways essential to acid production and protein synthesis. A transcriptome analysis revealed up-regulation of the electron transport chain, specifically the alternative oxidative pathway in ATCC 1015, while CBS 513.88 showed significant up-regulation of genes relevant to glucoamylase A production, such as tRNA-synthases and protein transporters. Our results and datasets from this integrative systems biology analysis resulted in a snapshot of fungal evolution and will support further optimization of cell factories based on filamentous fungi.[Supplemental materials (10 figures, three text documents and 16 tables) have been made available. Mapping N-linked Glycosylation Sites in the Secretome and Whole Cells of Aspergillus niger Using Hydrazide Chemistry and Mass Spectrometry

Protein glycosylation is known to play an essential role in both cellular functions and the secretory pathways; however, little information is available on the dynamics of glycosylated N-linked glycosites of fungi. Herein we present the first extensive mapping of glycosylated N-linked glycosites in industrial strain Aspergillus niger by applying an optimized solid phase enrichment of glycopeptide protocol using hydrazide modified magnetic beads. The enrichment protocol was initially optimized using mouse plasma and A. niger secretome samples, which was then applied to profile N-linked glycosites from both the secretome and whole cell lysates of A. niger. A total of 847 unique N-linked glycosites and 330 N-linked glycoproteins were confidently identified by LC-MS/MS. Based on gene ontology analysis, the identified N-linked glycoproteins in the whole cell lysate were primarily localized in the plasma membrane, endoplasmic reticulum, golgi apparatus, lysosome, and storage vacuoles. The identified N-linked glycoproteins are involved in a wide range of biological processes including gene regulation and signal transduction, protein folding and assembly, protein modification and carbohydrate metabolism. The extensive coverage of glycosylated N-linked glycosites along with identification of partial N-linked glycosylation in those enzymes involving in different biochemical pathways provide useful information for functional studies of N-linked glycosylation and their biotechnological applications in A. niger.

Given the instability of crude oil prices and the disruptions in crude oil supply chains, this article offers a complementing investment proposal through diversification of Nigeria's energy source and dependence. Therefore, the following issues were examined and reported: A comparative survey of coal and hydrocarbon reserve bases in Nigeria was undertaken and presented. An excursion into the economic, environmental, and technological justifications for the proposed diversification and roll-back to coal-based resource was also undertaken and presented. The technology available for coal beneficiation for environmental pollution control was reviewed and reported. The Fischer-Tropsch synthesis and its advances into Sasol's slurry phase distillate process were reviewed. Specifically, the adoption of Sasol's advanced synthol process and the slurry phase distillate process were recommended as ways of processing the products of coal gasification. The article concludes by discussing all the above-mentioned issues with regard to value addition as a means of wealth creation and investment.

Slump dominated upper slope reservoir facies, Intra Qua Iboe (Pliocene), Edop Field, offshore Nigeria

An integration of sedimentologic and 3D seismic data provides a basis for unraveling complex depositional processes and sand distribution of the Intra Qua Iboe (IQI) reservoir (Pliocene), Edop Field, offshore Nigeria. Nearly 3,000 feet of conventional core was examined in interpreting slump/slide/debris flow, bottom current, turbidity current, pelagic/hemipelagic, wave and tide dominated facies. The IQI was deposited on an upper slope in close proximity to the shelf edge. Through time, as the shelf edge migrated seaward, deposition began with a turbidite channel dominated slope system (IQI 1 and 2) and progressed through a slump/debris flow dominated slope system (IQI 3, the principal reservoir) to a tide and wave dominated, collapsed shelf-edge deltaic system (IQI 4). Using seismic time slices and corresponding depositional facies in the core, a sandy {open_quotes}fairway{open_quotes} has been delineated in the IQI 3. Because of differences in stacking patterns of sandy and muddy slump intervals, seismic facies show: (1) both sheet-like and mounded external forms (geometries), and (2) parallel/continuous as well as chaotic/hummocky internal reflections. In wireline logs, slump facies exhibits blocky, coarsening-up, fining-up, and serrated motifs. In the absence of conventional core, slump facies may be misinterpreted and even miscorrelated because seismic facies and log motifs of slumps and debris flows tend to mimic properties of turbidite fan deposits. The slump dominated reservoir facies is composed of unconsolidated fine-grained sand. Thickness of individual units varies from 1 to 34 feet, but amalgamated intervals reach a thickness of up to 70 feet and apparently form connected sand bodies. Porosity commonly ranges from 20 to 35%. Horizontal permeability commonly ranges from 1,000 to 3,000 md.

An integration of 3-D seismic and sedimentological information provides a basis for recognizing and mapping individual flow units within the Intra Qua Iboe (IQI) reservoir (Pliocene), Edop Field, offshore Nigeria. Core examination show the following depositional facies: A-Sandy slump/mass flow, B-Muddy slump/mass flow, C. Bottom current reworking. D-Non-channelized turbidity currents, E. Channelized (coalesced) turbidity currents. F-Channelized (isolated) turbidity currents, G-Pelagic/hemipelagic, H-Levee, I-Reworked slope, J-Wave dominated, and K-Tide dominated facies. With the exception of facies J and K, all these facies are of deep-water affinity. The IQI was deposited on an upper slope environment in close proximity to the shelf edge. Through time, as the shelf edge migrated scaward, deposition began with a channel dominated deep-water system (IQI 1 and 2) and progressed through a slump/debris flow dominated deep-water system (IQI 3, the principle reservoir) to a tide and wave dominated shallow-water system (IQI 4). Compositional and textural similarities between the deep-water facies result in similar log motifs. Furthermore, these depositional facies are not readily apparent as distinct seismic facies. Deep-water facies A, D, E, and F are reservoir facies, whereas facies B, C, G, H, and I are non-reservoir facies. However, Facies G is useful as a seismically mappable event throughout the study area. Mapping of these non-reservoir events provides the framework for understanding gross reservoir architecture. This study has resulted in seven defined reservoir units within the IQI, which serves as the architectural framework for ongoing reservoir characterization.

Material flows of mobile phones and accessories in Nigeria: Environmental implications and sound end-of-life management options

Presently, Nigeria is one of the fastest growing Telecom markets in the world. The country's teledensity increased from a mere 0.4 in 1999 to 10 in 2005 following the liberalization of the Telecom sector in 2001. More than 25 million new digital mobile lines have been connected by June 2006. Large quantities of mobile phones and accessories including secondhand and remanufactured products are being imported to meet the pent-up demand. This improvement in mobile telecom services resulted in the preference of mobile telecom services to fixed lines. Consequently, the contribution of fixed lines decreased from about 95% in year 2000 to less than 10% in March 2005. This phenomenal progress in information technology has resulted in the generation of large quantities of electronic waste (e-waste) in the country. Abandoned fixed line telephone sets estimated at 120,000 units are either disposed or stockpiled. Increasing quantities of waste mobile phones estimated at 8 million units by 2007, and accessories will be generated. With no material recovery facility for e-waste and/or appropriate solid waste management infrastructure in place, these waste materials end up in open dumps and unlined landfills. These practices create the potential for the release of toxic metals and halocarbons from batteries, printed wiring boards, liquid crystal display and plastic housing units. This paper presents an overview of the developments in the Nigerian Telecom sector, the material in-flow of mobile phones, and the implications of the management practices for wastes from the Telecom sector in the country.

Biodiesel as an Alternative Energy Resource in Southwest Nigeria

The Nigerian state faces unique issues that may provide an opportunity for rural economic growth. One of such is that major urban areas in the southwest of the country are beginning to have population increase and hence air quality problems that will require actions to reduce sources of pollution. One major pollution source is from exhaust emissions from cars and trucks. The use of alternative fuel sources such as biodiesel can make a significant reduction in certain exhaust emissions thus reducing pollution and improving air quality. The opportunity for economic growth in a single product economy like ours could lie in the processing of soybean oil and other suitable feedstocks produced within the country into biodiesel. The new fuel can be used by vehicles traversing the country thus reduce air pollution and providing another market for agricultural feedstocks while creating a value added market for animal fats and spent oils from industrial facilities. The benefits of biodiesel go far beyond the clean burning nature of the product. Bio diesel is a renewable resource helping to reduce the dependence of the economy on limited resources and imports, create a market for farmers and reduce the amount of waste oil, fat and grease being dumped into landfills and sewers.

This document describes the contents of a digital database containing maximum potential aboveground biomass, land use, and estimated biomass and carbon data for 1980. The biomass data and carbon estimates are associated with woody vegetation in Tropical Africa. These data were collected to reduce the uncertainty associated with estimating historical releases of carbon from land use change. Tropical Africa is defined here as encompassing 22.7 x 10{sup 6} km{sup 2} of the earth's land surface and is comprised of countries that are located in tropical Africa (Angola, Botswana, Burundi, Cameroon, Cape Verde, Central African Republic, Chad, Congo, Benin, Equatorial Guinea, Ethiopia, Djibouti, Gabon, Gambia, Ghana, Guinea, Ivory Coast, Kenya, Liberia, Madagascar, Malawi, Mali, Mauritania, Mozambique, Namibia, Niger, Nigeria, Guinea-Bissau, Zimbabwe (Rhodesia), Rwanda, Senegal, Sierra Leone, Somalia, Sudan, Tanzania, Togo, Uganda, Burkina Faso (Upper Volta), Zaire, and Zambia). The database was developed using the GRID module in the ARC/INFO{trademark} geographic information system. Source data were obtained from the Food and Agriculture Organization (FAO), the U.S. National Geophysical Data Center, and a limited number of biomass-carbon density case studies. These data were used to derive the maximum potential and actual (ca. 1980) aboveground biomass values at regional and country levels. The land-use data provided were derived from a vegetation map originally produced for the FAO by the International Institute of Vegetation Mapping, Toulouse, France.

Zing-Monkin area, located in the northern part of Adamawa Massif, is underlain by extensive exposures of moderately radioactive granodiorites, anatectic migmatites, equigranular granites, porphyritic granites and highly radioactive fine-grained granites with minor pegmatites. Selected major and trace element petrochemical investigations of the rocks show that a progression from granodiorite through migmatite to granites is characterised by depletion of MgO, CaO, Fe{sub 2}O{sub 3,} Sr, Ba, and Zr, and enrichment of SiO{sub 2} and Rb. This trend is associated with uranium enrichment and shows a chemical gradation from the more primitive granodiorite to the more evolved granites. Electron microprobe analysis shows that the uranium is content in uranothorite and in accessories, such as monazite, titanite, apatite, epidote and zircon. Based on petrochemical and mineralogical data, the more differentiated granitoids (e.g., fine-grained granite) bordering the Benue Trough are the immediate source of the uranium prospect in Bima Sandstone within the Trough. Uranium was derived from the granitoids by weathering and erosion. Transportation and subsequent interaction with organic matter within the Bima Sandstone led to precipitation of insoluble secondary uranium minerals in the Benue Trough.

Recent years have witnessed a growing recognition of the link between emissions of carbon dioxide (CO{sub 2}) and changes in the global climate. of all anthropogenic activities, energy production and use generate the single largest portion of these greenhouse gases. Although developing countries currently account for a small share of global carbon emissions, their contribution is increasing rapidly. Due to the rapid expansion of energy demand in these nations, the developing world`s share in global modern energy use rose from 16 to 27 percent between 1970 and 1990. If the growth rates observed over the past 20 years persist, energy demand in developing nations will surpass that in the countries of the Organization for Economic Cooperation and Development (OECD) early in the 21st century. The study seeks to examine the forces that galvanize the growth of energy use and carbon emissions, to assess the likely future levels of energy and CO{sub 2} in selected developing nations and to identify opportunities for restraining this growth. The purpose of this report is to provide the quantitative information needed to develop effective policy options, not to identify the options themselves. A combined study was carried out for the countries of the Gulf Cooperation Council (Bahrain, Kuwait, Oman, Qatar, Saudi Arabia and the United Arab Emirates).

A simulation methodology for predicting the time-course of enzymatic digestions is described. The model is based solely on the enzyme's subsite architecture and concomitant binding energies. This allows subsite binding energies to be used to predict the evolution of the relative amounts of different products during the digestion of arbitrary mixtures of oligomeric or polymeric substrates. The methodology has been specifically demonstrated by studying the fragmentation of a population of oligogalacturonides of varying degrees of polymerization, when digested by endo-polygalacturonase II (endo-PG II) from Aspergillus niger.

The following seven individuals were fully funded (complete airfare, accommodation, registration, meals) to participate in the Conference –Yerima Ladan (Head, ASECNA Forecast Office, Niamey Airport); Dr. Ousmane Manga Adamou (University of Niamey); Abdou Adam Abdoul-Aziz Abebe (Forecasater, ASECNA Forecast Office, Niamey Airport); Hassane Abdou (Forecaster, ASECNA Forecast Office, Niamey Airport); Saley Diori (Forecaster, ASECNA Forecast Office, Niamey Airport); Alhassane Diallo (Meteorological Engineer, Burkina Faso Weather Service). Four rigs refurbished for West Africa drilling

In April and May 1990, Shell Petroleum Development Co. of Nigeria Ltd. awarded Noble Drilling West Africa Inc. four separate contracts to drill oil and gas wells in the inland waterways of Nigeria. The contracted rigs included a shallow water jack up, the NN-1, and three posted barges, the Gene Rosser, the Chuck Syring, and the Lewis Dugger. The jack up was built in 1978, and the three posted barges are 1980s vintage. Three of the rigs have been idle for a number of years. The Shell Nigeria contracts required major modifications to the rigs before putting them into international service. Noble replaced or refurbished all major pieces of equipment in the drilling, power, and service systems on the rigs. Rig crews serviced all other equipment. A significant amount of general service piping and electrical wiring was replaced. Each rig also required additional motor control centers to support the new drilling and mud processing equipment. Alfa-Laval waste-heat water desalination plants and new sewage treatment units were installed on all four rigs. Because of the tidal variances and high silt conditions expected in the African waterways, all engine cooling systems were converted from heat exchangers to radiators. Rotary tables were made common on all rigs at 37 1/2 in. Noble had all traveling equipment completely inspected and modified as necessary. Strict attention was paid to certification and documentation of all equipment. Safety upgrades conformed to both Shell and Noble standards. Fire and gas detection systems were installed throughout each rig. Water and foam deluge systems were installed in the wellhead areas, and new foam systems and monitors were installed on the helldecks.

Africa's natural gas: potentialities and letdowns

Although Africa has experienced 10 times less hydrocarbon exploration than Western Europe, its proved gas reserves already amount to 220-223 trillion CF or 7% of world reserves, while Europe holds 6% or 167 TCF. Yet Africa marketed only 1.3 TCF in 1982 against Europe's 6.5 TCF. Because of the lack of domestic demand for gas, Africa flares up to 21% of its gas output. Algeria is the continent's primary gas consumer, with Egypt, Libya, and Nigeria trying to expand local gas markets. The vast majority of marketed African gas goes to Europe, either as gas sent through the Trans-Med pipeline or as LNG via tanker.

Oil and gas developments in central and southern Africa in 1987

Significant rightholding changes took place in central and southern Africa during 1987. Angola, Benin, Congo, Gabon, Ghana, Guinea, Guinea Bissau, Mauritania, Seychelles, Somali Republic, Tanzania, Zaire, and Zambia announced awards or acreage open for bidding. Decreases in exploratory rightholdings occurred in Cameroon, Congo, Cote d'Ivoire, Equatorial Guinea, Gabon, Kenya, Namibia, South Africa, and Tanzania. More wells and greater footage were drilled in 1987 than in 1986. Total wells increased by 18% as 254 wells were completed compared to 217 in 1986. Footage drilled during the year increased by 46% as about 1.9 million ft were drilled compared to about 1.3 million ft in 1986. The success rate for exploration wells in 1987 improved slightly to 36% compared to 34% in 1986. Significant discoveries were made in Nigeria, Angola, Congo, and Gabon. Seismic acquisition in 1987 was the major geophysical activity during the year. Total oil production in 1987 was 773 million bbl (about 2.1 million b/d), a decrease of 7%. The decrease is mostly due to a 14% drop in Nigerian production, which comprises 60% of total regional production. The production share of OPEC countries (Nigeria and Gabon) versus non-OPEC countries of 67% remained unchanged from 1986. 24 figs., 5 tabs.

Resin-assisted Enrichment of N-terminal Peptides for Characterizing Proteolytic Processing

Proteolytic processing is a ubiquitous, irreversible posttranslational modification that plays an important role in cellular regulation in all living organisms. Herein we report a resin-assisted positive selection method for specifically enriching protein N-terminal peptides to facilitate the characterization of proteolytic processing events by liquid chromatography-tandem mass spectrometry. In this approach, proteins are initially reduced and alkylated and their lysine residues are converted to homoarginines. Then, protein N-termini are selectively converted to reactive thiol groups. We demonstrate that these sequential reactions were achieved with nearly quantitative efficiencies. Thiol-containing N-terminal peptides are then captured (>98% efficiency) by a thiol-affinity resin, a significant improvement over the traditional avidin/biotin enrichment. Application to cell lysates of Aspergillus niger, a filamentous fungus of interest for biomass degradation, enabled the identification of 1672 unique protein N-termini and proteolytic cleavage sites from 690 unique proteins.

Solar electricity for Africa: The case of Kenya

This paper presents results of two recent World Bank efforts made in Kenya, Niger, and Cameroon to study the impact of two different renewable projects, one a Micro-Lights program involving about 500 lanterns and the second a survey of 410 households using solar electricity systems. The Micro-Lights program showed that users have distinct preferences in the style of the lamps, that they are willing to spend cash, and that they demand good quality. They may be initially satisfied, but rapidly want more from their purchases. The photoelectric system survey touched less than 1% of such households, and looked at user education, system size, satisfaction, expectations, age of system, appliances, and expectations.

Challenges in Whole-Genome Annotation of Pyrosequenced Eukaryotic Genomes

Pyrosequencing technologies such as 454/Roche and Solexa/Illumina vastly lower the cost of nucleotide sequencing compared to the traditional Sanger method, and thus promise to greatly expand the number of sequenced eukaryotic genomes. However, the new technologies also bring new challenges such as shorter reads and new kinds and higher rates of sequencing errors, which complicate genome assembly and gene prediction. At JGI we are deploying 454 technology for the sequencing and assembly of ever-larger eukaryotic genomes. Here we describe our first whole-genome annotation of a purely 454-sequenced fungal genome that is larger than a yeast (>30 Mbp). The pezizomycotine (filamentous ascomycote) Aspergillus carbonarius belongs to the Aspergillus section Nigri species complex, members of which are significant as platforms for bioenergy and bioindustrial technology, as members of soil microbial communities and players in the global carbon cycle, and as agricultural toxigens. Application of a modified version of the standard JGI Annotation Pipeline has so far predicted ~;;10k genes. ~;;12percent of these preliminary annotations suffer a potential frameshift error, which is somewhat higher than the ~;;9percent rate in the Sanger-sequenced and conventionally assembled and annotated genome of fellow Aspergillus section Nigri member A. niger. Also,>90percent of A. niger genes have potential homologs in the A. carbonarius preliminary annotation. Weconclude, and with further annotation and comparative analysis expect to confirm, that 454 sequencing strategies provide a promising substrate for annotation of modestly sized eukaryotic genomes. We will also present results of annotation of a number of other pyrosequenced fungal genomes of bioenergy interest.

Well performance graph simplifies field calculations

Graphic Methods are widely employed in order to understand overall well behavior using only surface parameters. The authors propose a new graphic method, used successfully by Agip for oil and gas wells in Italy, Libya, Nigeria and Tunisia. The well performance graph helps solve many production problems, including estimation of: inflow performance relationship; causes of rate decline throughout well life; and production rate and bottomhole flowing pressure for various pressures upstream of the surface choke, and vice-versa. This method differs from others by using flow behavior through the choke for both critical and subcritical conditions. Equations describing flow through the formation, string and surface choke are also used. Results are quite reliable when these theoretical equations are calibrated with field data, either from the well concerned or from nearby wells producing the same fluid. This article describes the technique as it applies to oil wells. The methodology for gas wells is similar.

TEWS98 was a four week residential program which focused on providing a meaningful science education experience while developing critical thinking skills. Thirty three students, three teachers, four college interns and the camp administrator lived and worked together while developing solutions to several worldwide problems. These problems are called the Discovery Topics and they are: (1) design the worlds perfect house; (2) how many people can the world hold; and (3) predict the hot stocks for the year 2030. The participants, both students and staff came from different countries all over the world The following countries were represented: The United Kingdom, Sierra Leone (West Africa), Ireland, USA Nigeria, West Indies and Barbados.

Generation of transgenic wheat (Triticum aestivum L.) accumulating heterologous endo-xylanase or ferulic acid esterase in the endosperm

Endo-xylanase (from Bacillus subtilis) or ferulic acid esterase (from Aspergillus niger) were expressed in wheat under the control of the endosperm specific 1DX5 glutenin promoter. Constructs both with and without the endoplasmic reticulum retention signal KDEL were used. Transgenic plants were recovered in all four cases but no qualitative differences could be observed whether KDEL was added or not. Endo-xylanase activity in transgenic grains was increased between two and three fold relative to wild type. The grains were shriveled and had a 25-33% decrease in mass. Extensive analysis of the cell walls showed a 10-15% increase in arabinose to xylose ratio, a 50% increase in the proportion of water extractable arabinoxylan, and a shift in the MW of the water extractable arabinoxylan from being mainly larger than 85 kD to being between 2 kD and 85 kD. Ferulic acid esterase expressing grains were also shriveled and the seed weight was decreased by 20-50%. No ferulic acid esterase activity could be detected in wild type grains whereas ferulic acid esterase activity was detected in transgenic lines. The grain cell walls had 15-40% increase in water unextractable arabinoxylan and a decrease in monomeric ferulic acid between 13 and 34%. In all the plants the observed changes are consistent with a plant response that serves to minimize the effect of the heterologously expressed enzymes by increasing arabinoxylan biosynthesis and cross-linking.

This study draws examples from a mapping project of climate change vulnerability in Niger, and discusses the lessons...Geographical Information Systems have not been as successful in the non-governmental humanitarian sector as GIS professionals had hoped. If geospatial tools are to become more widely used and valuable within the humanitarian sector, it is perhaps time for a new approach to be taken. A debate is currently underway about the potential role of Google Earth in emergency relief operations. This research extends the scope of this discussion into the wider development sector, and asks whether Google Earth has the potential to become a useful tool in various aspects of development NGO work. Information management tools need to have wide support within an organisation to be successful, and it seems that many are very interested in what can be done using compromise geospatial/geovisualisation tools such as Google Earth. However there is also scepticism about its usefulness. This research suggests practical applications and recommendations for good use so that actors within the development sector can take the research further....

Sequencing the Black Aspergilli species complex

The ~15 members of the Aspergillus section Nigri species complex (the "Black Aspergilli") are significant as platforms for bioenergy and bioindustrial technology, as members of soil microbial communities and players in the global carbon cycle, and as food processing and spoilage agents and agricultural toxigens. Despite their utility and ubiquity, the morphological and metabolic distinctiveness of the complex's members, and thus their taxonomy, is poorly defined. We are using short read pyrosequencing technology (Roche/454 and Illumina/Solexa) to rapidly scale up genomic and transcriptomic analysis of this species complex. To date we predict 11197 genes in Aspergillus niger, 11624 genes in A. carbonarius, and 10845 genes in A. aculeatus. A. aculeatus is our most recent genome, and was assembled primarily from 454-sequenced reads and annotated with the aid of >2 million 454 ESTs and >300 million Solexa ESTs. To most effectively deploy these very large numbers of ESTs we developed 2 novel methods for clustering the ESTs into assemblies. We have also developed a pipeline to propose orthologies and paralogies among genes in the species complex. In the near future we will apply these methods to additional species of Black Aspergilli that are currently in our sequencing pipeline.

Characterization of Vertical Velocity and Drop Size Distribution Parameters in Widespread Precipitation at ARM Facilities

Extended, high-resolution measurements of vertical air motion and median volume drop diameter D0 in widespread precipitation from three diverse Atmospheric Radiation Measurement Program (ARM) locations [Lamont, Oklahoma, Southern Great Plains site (SGP); Niamey, Niger; and Black Forest, Germany] are presented. The analysis indicates a weak (0-10 cm{sup -1}) downward air motion beneath the melting layer for all three regions, a magnitude that is to within the typical uncertainty of the retrieval methods. On average, the hourly estimated standard deviation of the vertical air motion is 0.25 m s{sup -1} with no pronounced vertical structure. Profiles of D0 vary according to region and rainfall rate. The standard deviation of 1-min-averaged D0 profiles for isolated rainfall rate intervals is 0.3-0.4 mm. Additional insights into the form of the raindrop size distribution are provided using available dual-frequency Doppler velocity observations at SGP. The analysis suggests that gamma functions better explain paired velocity observations and radar retrievals for the Oklahoma dataset. This study will be useful in assessing uncertainties introduced in the measurement of precipitation parameters from ground-based and spaceborne remote sensors that are due to small-scale variability.

Mercury concentrations in Maine sport fishes

To assess mercury contamination of fish in Maine, fish were collected from 120 randomly selected lakes. The collection goal for each lake was five fish of the single most common sport fish species within the size range commonly harvested by anglers. Skinless, boneless fillets of fish from each lake were composited, homogenized, and analyzed for total mercury. The two most abundant species, brook trout Salvelinus fontinalis and smallmouth bass Micropterus dolomieu, were also analyzed individually. The composite fish analyses indicate high concentrations of mercury, particularly in large and long-lived nonsalmonid species. Chain pickerel Esox niger, smallmouth bass, largemouth bass Micropterus salmoides, and white perch Morone americana had the highest average mercury concentrations, and brook trout and yellow perch Perca flavescens had the lowest. The mean species composite mercury concentration was positively correlated with a factor incorporating the average size and age of the fish. Lakes containing fish with high mercury concentrations were not clustered near known industrial or population centers but were commonest in the area within 150 km of the seacoast, reflecting the geographical distribution of species that contained higher mercury concentrations. Stocked and wild brook trout were not different in length or weight, but wild fish were older and had higher mercury concentrations. Fish populations maintained by frequent introductions of hatchery-produced fish and subject to high angler exploitation rates may consist of younger fish with lower exposure to environmental mercury and thus contain lower concentrations than wild populations.

World crude output overcomes Persian Gulf disruption

Several OPEC producers made good on their promises to replace 2.7 MMbpd of oil exports that vanished from the world market after Iraq took over Kuwait. Even more incredibly, they accomplished this while a breathtaking 1.2- MMbopd reduction in Soviet output took place during the course of 1991. After Abu Dhabi, Indonesia, Iran, Libya, Nigeria, Saudi Arabia and Venezuela turned the taps wide open, their combined output rose 2.95 MMbopd. Put together with a 282,000-bopd increase by Norway and contributions from smaller producers, this enabled world oil production to remain within 400,000 bopd of its 1990 level. The 60.5-MMbopd average was off by just 0.7%. This paper reports that improvement took place in five of eight regions. Largest increases were in Western Europe and Africa. Greatest reductions occurred in Eastern Europe and the Middle East. Fifteen nations produced 1 MMbopd or more last year, compared with 17 during 1990.

African oil plays

The vast continent of Africa hosts over eight sedimentary basins, covering approximately half its total area. Of these basins, only 82% have entered a mature exploration phase, 9% have had little or no exploration at all. Since oil was first discovered in Africa during the mid-1950s, old play concepts continue to bear fruit, for example in Egypt and Nigeria, while new play concepts promise to become more important, such as in Algeria, Angola, Chad, Egypt, Gabon, and Sudan. The most exciting developments of recent years in African oil exploration are: (1) the Gamba/Dentale play, onshore Gabon; (2) the Pinda play, offshore Angola; (3) the Lucula/Toca play, offshore Cabinda; (4) the Metlaoui play, offshore Libya/Tunisia; (5) the mid-Cretaceous sand play, Chad/Sudan; and (6) the TAG-I/F6 play, onshore Algeria. Examples of these plays are illustrated along with some of the more traditional oil plays. Where are the future oil plays likely to develop No doubt, the Saharan basins of Algeria and Libya will feature strongly, also the presalt of Equatorial West Africa, the Central African Rift System and, more speculatively, offshore Ethiopia and Namibia, and onshore Madagascar, Mozambique, and Tanzania.

Africa gaining importance in world LPG trade

Major LPG projects planned or under way in Africa will increase the importance of that region`s presence in world LPG trade. Supplies will nearly double between 1995 and 2005, at which time they will remain steady for at least 10 years. At the same time that exports are leveling, however, increasing domestic demand for PG is likely to reduce export-market participation by Algeria, Nigeria, Egypt, and Libya. The growth of Africa`s participation in world LPG supply is reflected in comparisons for the next 15--20 years. Total world supply of LPG in 1995 was about 165 million metric tons (tonnes), of which Africans share was 7.8 million tonnes. By 2000, world supply will grow to slightly more than 200 million tonnes, with Africa`s share expected to increase to 13.2 million tonnes (6.6%). And by 2005, world LPG supply will reach nearly 230 million tonnes; Africa`s overall supply volumes by that year will be nearly 16.2 million tonnes (7%). World LPG supply for export in 1995 was on order of 44 million tonnes with Africa supply about 4 million tonnes (9%). By 2005, world export volumes of LPG will reach nearly 70 million tonnes; Africa`s share will have grown by nearly 10 million tonnes (14.3%).

Attitude towards littering as a mediator of the relationship between personality attributes and responsible environmental behavior

Highlights: > Independently, altruism and locus of control contributed significantly toward attitude towards littering. > Altruism and locus of control jointly contributed significantly to attitude towards littering. > The results further show a significant joint influence of altruism and locus of control on REB. > The independent contributions reveal that altruism and locus of control contribute significantly to REB. > Attitude towards littering mediates the relationship between locus of control and REB. - Abstract: The study tested whether attitude towards littering mediates the relationship between personality attributes (altruism and locus of control) and responsible environmental behavior (REB) among some residents of Ibadan metropolis, Nigeria. Using multistage sampling technique, measures of each construct were administered to 1360 participants. Results reveal significant independent and joint influence of personality attributes on attitude towards littering and responsible environmental behavior, respectively. Attitude towards littering also mediates the relationship between personality characteristics and REB. These findings imply that individuals who possess certain desirable personality characteristics and who have unfavorable attitude towards littering have more tendencies to engage in pro-environmental behavior. Therefore, stakeholders who have waste management as their priority should incorporate this information when guidelines for public education and litter prevention programs are being developed. It is suggested that psychologists should be involved in designing of litter prevention strategies. This will ensure the inclusion of behavioral issues in such strategies. An integrated approach to litter prevention that combines empowerment, cognitive, social, and technical solutions is recommended as the most effective tool of tackling the litter problem among residents of Ibadan metropolis.

Reinterpretation of depositional processes in a classic flysch sequence (Pennsylvania Jackfork Group), Ouachita Mountains, Arkansas and Oklahoma: Discussion

The contribution by Shanmugam and Moiola (1995) on the depositional processes of the Pennsylvanian Jackfork Group (Formation) in Arkansas and Oklahoma highlights a few important processes that are often overlooked. Their work on the fabric of some high-density flow deposits is interesting in light of the debate over the nature of these types of deposits (Lowe, 1982; Hiscott, 1994). However, we disagree with some of the observations and interpretations they use in making their argument for a new depositional model, and submit that (1) turbidity current deposits (turbidites) are a major lithofacies component in the DeGray Spillway cut is not difficult, and (4) because it is not necessary to preserve conventional dogma, a change in nomenclature is more appropriate than a change in depositional models. Finally, their call for application of their debris-flow model to the North Sea, Gulf of Mexico, offshore Nigeria, and elsewhere is disturbing because they would have the petroleum industry relinquish the idea of predictability in deep-water reservoirs.

International crude oil prices rose moderately in January 1995, under the combined influences of lower production and higher demand. Crude oil output both from the Organization of Petroleum Exporting Countries (OPEC) and from non-OPEC nations declined from December levels due to a variety of causes, including severe weather in the North Sea and a platform explosion offshore Nigeria. At the same time, strong demand, especially from Asia, revised buying patterns and tightened markets worldwide. In the United States, reformulated gasoline (RFG) remained the dominant market influence, with the official start of the program at the retail level on January 1. Repercussions from the {open_quotes}opting out{close_quotes} of counties in Pennsylvania, New York, and Maine in December continued to disrupt supply patterns and confuse markets. Gasoline prices spiked at mid-month, but ended near where they began, while distillate prices declined due to warm weather and high inventories. A sharp seasonal decline in motor gasoline volumes led total refiner sales of the major petroleum products down 5.9 percent from December levels. January market and sales activity for crude oil and the principal petroleum products is summarized in the following sections.

Organizational Politics, Turnover Intention and Organizational Commitment as Predictors of Employees ’ Efficiency and Effectiveness in Academia

This study examined the prediction of organizational politics, turnover intention and organizational commitment on employees ’ efficiency and effectiveness in academia. It made use of 200 randomly selected employees of Babcock University Ilishan, Ilishan – Remo, Ogun State, Nigeria. The age range of the respondents was between 25 and 53 years with mean age of 36.1 and standard deviation of 3.01. The expost-facto research design was employed. Three standard instruments: Perception of Organizational Politics Scale (POPS) by Kacmar and Carlson (1994); Organization Commitment Scale by Meyer and Allen (1991); Turnover Intention Scale by O Driscoll and Benhrs (1994) ; respondents ’ annual performance evaluation report were used in generating data for the study. Data Analysis involved the use of correlation and multiple regressions. The result indicated that the predictor variables combined and separately made significant contribution to the prediction of the criterion variable. Also, high and positive correlations were found between organizational politics and turnover intention, work efficiency and organizational commitment. On the basis of the findings, it was suggested that a happy employee is a better employee.

The common perception in exploration is that deep-water sands are predominantly a product of low- and high-density turbidity currents, and that submarine-fan models with channel/levee and lobe elements are the norm. The reality, however, is that deep-water systems are extremely complex and variable in terms of depositional processes and sand-body geometries. For example, the Bourna Sequence, composed of T{sub a}, T{sub b}, T{sub c}, T{sub d}, and T{sub e} divisions, is believed to be the product of a turbidity current. However, recent core and outcrop studies show that the complete and partial Bouma sequences also can be explained by processes other than turbidity currents, such as sandy debris flows (i.e., {open_quotes}T{sub a}{close_quotes}) and bottom-current reworking (i.e., {open_quotes}T{sub b}, T{sub c} and T{sub d}{close_quotes}). Massive sands are interpreted routinely as high-density turbidites, but the reality is that the term {open_quotes}high-density turbidity current{close_quotes} commonly refers to sandy debris flow in terms of flow theology and sediment-support mechanism. Deep-water sequences in the North Sea, Norwegian Sea, Offshore Gabon, Offshore Nigeria, Gulf of Mexico, and the Ouachita Mountains are generally considered to be turbidite-rich submarine fans. However, the reality is that these sequences are composed predominantly of sandy slumps and debris flows, not turbidites. Fan models are attractive to explorationists because of their predictable sheet-like geometries; however, these simplistic conceptual models are obsolete because they defy reality. Although the turbidite paradigm is alive and well for now in the minds of many sedimentologists and sequence stratigraphers, the turbidites themselves that form the foundation for fan models are becoming an endangered facies!

Identification and characterization of the polyketide synthase involved in ochratoxin A biosynthesis in Aspergillus carbonarius

Ochratoxin A (OTA) is a potent mycotoxin produced by Aspergillus and Penicillium species and is a common contaminant of a wide variety of food commodities, with Aspergillus carbonarius being the main producer of OTA contamination in grapes and wine. The molecular structure of OTA is composed of a dihydroisocoumarin ring linked to phenylalanine and, as shown in different producing fungal species, a polyketide synthase (PKS) is a component of the OTA biosynthetic pathway. Similar to observations in other filamentous ascomycetes, the genome sequence of A. carbonarius contains a large number of genes predicted to encode PKSs. In this work a pks gene identified within the putative OTA cluster of A. carbonarius, designated as AcOTApks, was inactivated and the resulting mutant strain was unable to produce OTA, confirming the role of AcOTApks in this biosynthetic pathway. AcOTApks protein is characteristic of the highly reduced (HR)-PKS family, and also contains a putative methyltransferase domain likely responsible for the addition of the methyl group to the OTA polyketide structure. AcOTApks is different from the ACpks protein that we previously described which showed an expression profile compatible with OTA production. We performed phylogenetic analyses of the ?-ketosynthase and acyl-transferase domains of the OTA PKSs which had been identified and characterized in different OTA producing fungal species. The phylogenetic results were similar for both the two domains analyzed and showed that OTA PKS of A. carbonarius, Aspergillus niger, and Aspergillus ochraceus clustered in a monophyletic group with 100% bootstrap support suggesting a common origin, while the other OTA PKSs analyzed were phylogenetically distant. A qRT-PCR assay monitored AcOTApks expression during fungal growth and concomitant production of OTA by A. carbonarius in synthetic grape medium. A clear correlation between the expression profile of AcOTApks and kinetics of OTA production was observed with AcOTApks which reached its maximum level of transcription before OTA accumulation in mycelium reached its highest level, confirming the fact that gene transcription always precedes phenotypic production.

Partitioned Waveform Inversion Applied to Eurasia and Northern Africa

This report summarizes the data analysis achieved during Heather Bedle's eleven-week Technical Scholar internship at Lawrence Livermore National Labs during the early summer 2006. The work completed during this internship resulted in constraints on the crustal and upper mantle S-velocity structure in Northern Africa, the Mediterranean, the Middle East, and Europe, through the fitting of regional waveform data. This data extends current raypath coverage and will be included in a joint inversion along with data from surface wave group velocity measurements, S and P teleseismic arrival time data, and receiver function data to create an improved velocity model of the upper mantle in this region. The tectonic structure of the North African/Mediterranean/Europe/Middle Eastern study region is extremely heterogeneous. This region consists of, among others, stable cratons and platforms such as the West Africa Craton, and Baltica in Northern Europe; oceanic subduction zones throughout the Mediterranean Sea where the African and Eurasian plate collide; regions of continental collision as the Arabian Plate moves northward into the Turkish Plate; and rifting in the Red Sea, separating the Arabian and Nubian shields. With such diverse tectonic structures, many of the waveforms were difficult to fit. This is not unexpected as the waveforms are fit using an averaged structure. In many cases the raypaths encounter several tectonic features, complicating the waveform, and making it hard for the software to converge on a 1D average structure. Overall, the quality of the waveform data was average, with roughly 30% of the waveforms being discarded due to excessive noise that interfered with the frequency ranges of interest. An inversion for the 3D S-velocity structure of this region was also performed following the methodology of Partitioned Waveform Inversion (Nolet, 1990; Van der Lee and Nolet, 1997). The addition of the newly fit waveforms drastically extends the range of the model. The model now extends as far east in Africa to cover Chad and Niger, and reaches south to cover Zambia. The model is also stretched eastward to cover the eastern half of India, and northward to cover the southern portion of Scandinavia.

Three Blind Men and the Elephant

Just like the blind men in the popular story of perceiving the elephant, the three major constituencies participating in the energy debate have greatly different perceptions of the problem. The constituency that is worried about climate change believes the energy problem is caused by profligate use of fossil fuel that has dramatically changed our atmosphere. The energy security group sees dangerous reliance on foreign sources of oil increasingly held by countries hostile to the US. The economic vitality group sees high energy prices and their effect on the economy and our life-style. Just like the blind men, each of the three constituencies perceives a different problem. And just as with the blind men, while each perspective is right as a piece of the elephant, it takes all the perspectives together to actually solve the problem. Environmentalists focus on solutions responding to the scientific consensus that greenhouse gases are creating rapid climate change. The tipping point has come: it is now a consensus position among scientists the global warming is being affected by anthropogenic activity to 90% certainty according to the last IPCC report. Although they still struggle with the prediction of how much global temperatures will rise if we do nothing--is it 5 deg or 10 under BAU? This group believes that we cannot afford to take a chance because we get only one chance. We can not afford to do this kind of experiment with the Earth. Any choice which decreases our CO{sub 2} footprint is favored, even if it means a decrease in standard of living. The energy security constituency sees the geo-politics of oil becoming increasingly dire. They look at oil money being used to fund anti-American activities of groups such as the Wahabis in Saudi Arabia, Hezbollah in Lebanon and the infamous Al Qaeda. They quip that the Iraq war is the first war where we are paying for both sides. They note Iran and the Shia throughout the Middle East seeing the possibility of controlling 2/3 of the world's oil. They see oil and gas being used by Russia to exert political power using the gas tap and Hugo Chavez in Venezuela clearly anti-American and now a virtual dictator who controls 15% of our oil imports. Conflicts in Nigeria over oil wealth and corruption affected our oil supply. Countries such as China are at best unwilling to join political action against countries such as Sudan that supply them oil, and at worst, selling them arms in order to cement their relationships with respect to importing oil. This security constituency favors ending our vulnerability by ending our ''addiction to foreign oil''. This group thinks that there is no domestic source of energy that is bad. They will be happy to see our corn turned into ethanol; our coal turned into liquid fuel for transportation. No matter that the price of tortillas doubles in Mexico, we expand corn farming at the expense of the environment, our tanks and pipes in gas stations corrode and leak, or we make liquid fuel from coal, thus increasing the carbon footprint by 30% per unit of energy. The economic vitality group sees increasing international demand for oil occurring simultaneously with a peaking supply of light sweet crude. They see an oil market where higher prices drive more production of oil which is heavier and more sour (supply follows demand). However, fast growth in world-wide demand increases even faster and prices will go up. For example, China adds 10,000 cars per month, and there is an uncanny correlation between the price of oil and the amount of oil imported by China. The security contingent also worries about reliability of supply as affected by pipeline leaks in Alaska or hurricanes or potential terrorism. This constituency thinks the problem is one of capacity and favors solutions that will increase oil production, reservoirs, pipelines and refineries. They believe that the energy system will be determined by the market and want solutions that favor investment in capacity.
