    The Midwest Clean Energy Application Center (CEAC) was one of eight regional centers that promoted and assisted in transforming the market for combined heat and power (CHP), waste heat to power (WHP), and district energy (DE) technologies and concepts throughout the United States between October 1, 2009 and December 31, 2013. The key services the CEACs provided included: Market Opportunity Analyses – Supporting analyses of CHP market opportunities in diverse markets including industrial, federal, institutional, and commercial sectors. Education and Outreach – Providing information on the energy and non-energy benefits and applications of CHP to state and local policy makers, regulators, energy end-users, trade associations and others. Information was shared on the Midwest CEAC website: www.midwestcleanergy.org. Technical Assistance – Providing technical assistance to end-users and stakeholders to help them consider CHP, waste heat to power, and/or district energy with CHP in their facility and to help them through the project development process from initial CHP screening to installation. The Midwest CEAC provided services to the Midwest Region that included the states of Illinois, Indiana, Iowa, Kansas, Michigan, Minnesota, Missouri, Nebraska, North Dakota, Ohio, South Dakota, and Wisconsin.

    The Midwest Superconductivity Consortium, MISCON, in the fourth year of operations further strengthened its mission to advance the science and understanding of high T{sub c} superconductivity. The goals of the organization and the individual projects continue to reflect the current needs for new knowledge in the field and the unique capabilities of the institutions involved. Group efforts and cooperative laboratory interactions to achieve the greatest possible synergy under the Consortium continue to be emphasized. Industrial affiliations coupled with technology transfer initiatives were expanded. Activities of the participants during the past year achieved an interactive and high level of performance. The number of notable achievements in the field contributed by Consortium investigators increased. The programmatic research continues to focus upon key materials-related problems in two areas. The first area has a focus upon {open_quotes}Synthesis and Processing{close_quotes} while the second is centered around {open_quotes}Limiting Features in Transport Properties of High T{sub c} Materials{close_quotes}.

    This final report summarizes the Phase I research conducted by the Midwest regional Carbon Sequestration Partnership (MRCSP). The Phase I effort began in October 2003 and the project period ended on September 31, 2005. The MRCSP is a public/private partnership led by Battelle with the mission of identifying the technical, economic, and social issues associated with implementation of carbon sequestration technologies in its seven state geographic region (Indiana, Kentucky, Maryland, Michigan, Ohio, Pennsylvania, and West Virginia) and identifying viable pathways for their deployment. It is one of seven partnerships that together span most of the U.S. and parts of Canada that comprise the U.S. Department of Energy's (DOE's) Regional Carbon Sequestration Program led by DOE's national Energy Technology Laboratory (NETL). The MRCSP Phase I research was carried out under DOE Cooperative Agreement No. DE-FC26-03NT41981. The total value of Phase I was $3,513,513 of which the DOE share was $2,410,967 or 68.62%. The remainder of the cost share was provided in varying amounts by the rest of the 38 members of MRCSP's Phase I project. The next largest cost sharing participant to DOE in Phase I was the Ohio Coal Development Office within the Ohio Air Quality Development Authority (OCDO). OCDO's contribution was $100,000 and was contributed under Grant Agreement No. CDO/D-02-17. In this report, the MRCSP's research shows that the seven state MRCSP region is a major contributor to the U. S. economy and also to total emissions of CO2, the most significant of the greenhouse gases thought to contribute to global climate change. But, the research has also shown that the region has substantial resources for sequestering carbon, both in deep geological reservoirs (geological sequestration) and through improved agricultural and land management practices (terrestrial sequestration).     U.S. colleges and universities are in a position to make substantial solar project investments both on and off campus. To facilitate these investments, the Midwest Renewable Energy Association is working with university stakeholders at four universities to create solar investment proposals for consideration by university governance boards. This campus stakeholder engagement is led by student solar deployment teams and builds off of past program success with the U.S. Department of Energy Solar Decathlon and similar student-led projects. This effort showcases the potential of university solar photovoltaic (PV) investments, advances favorable board policies to govern PV investments, and provides a roadmap for universities across the country to deploy solar—advancing their sustainability goals.

    Distributed generation consists of the use of integrated or stand-alone natural gas or liquid fueled power generation equipment. These distributed resources (DR) may provide power in the 5-kW to 50-MW range and can provide power directly to the customer, thereby potentially deferring additional transmission and distribution systems upgrades by the utility and improving power quality and reliability for the customer. Compact, technically advanced units in a variety of technologies presently available include diesel engines, lean burning gas fired internal combustion engine generator sets, cogeneration packages, small microturbines, and fuel cells. This class of resources has the potential to provide a cost effective, reliable, addition to a utility`s generating resource mix and is once again gaining favor in era characterized both by uncertainty and opportunity. Strategic deployment of these resources can also eliminate or delay expensive central plant capacity additions. Installations may be tailored to meet customer technical and financial requirements with projects being developed by utilities on customer sites as well as by Independent Power Producers and others. Economic constraints as well as key environmental and operating issues must be understood and clarified for those seeking to capitalize on this approach. As the first phase of a project with the objective of developing a best practices approach to implementing DR, EPRI`s project Application Experiences with Distributed Resources in the Midwest examined these issues. This report contains the results of an in-depth technical survey, which was given to distributed resource sites throughout the Midwest. Aspects of DR projects involving electrical interconnection, siting and permitting, operations and maintenance and various operating practices are examined.

    The Organization of Midwest ISO States (OMS) launched the Midwest Demand Resource Initiative (MWDRI) in 2007 to identify barriers to deploying demand response (DR) resources in the Midwest Independent System Operator (MISO) region and develop policies to overcome them. The MWDRI stakeholders decided that a useful initial activity would be to develop more detailed information on existing retail DR programs and dynamic pricing tariffs, program rules, and utility operating practices. This additional detail could then be used to assess any"seams issues" affecting coordination and integration of retail DR resources with MISO's wholesale markets. Working with state regulatory agencies, we conducted a detailed survey of existing DR programs, dynamic pricing tariffs, and their features in MISO states. Utilities were asked to provide information on advance notice requirements to customers, operational triggers used to call events (e.g. system emergencies, market conditions, local emergencies), use of these DR resources to meet planning reserves requirements, DR resource availability (e.g., seasonal, annual), participant incentive structures, and monitoring and verification (M&V) protocols. This report describes the results of this comprehensive survey and discusses policy implications for integrating legacy retail DR programs and dynamic pricing tariffs into organized wholesale markets. Survey responses from 37 MISO members and 4 non-members provided information on 141 DR programs and dynamic pricing tariffs with a peak load reduction potential of 4,727 MW of retail DR resource. Major findings of this study area:- About 72percent of available DR is from interruptible rate tariffs offered to large commercial and industrial customers, while direct load control (DLC) programs account for ~;;18percent.   13. Knockdown of dual specificity phosphatase 4 enhances the chemosensitivity of MCF-7 and MCF-7/ADR breast cancer cells to doxorubicin

    Background: Breast cancer is the major cause of cancer-related deaths in females world-wide. Doxorubicin-based therapy has limited efficacy in breast cancer due to drug resistance, which has been shown to be associated with the epithelial-to-mesenchymal transition (EMT). However, the molecular mechanisms linking the EMT and drug resistance in breast cancer cells remain unclear. Dual specificity phosphatase 4 (DUSP4), a member of the dual specificity phosphatase family, is associated with cellular proliferation and differentiation; however, its role in breast cancer progression is controversial. Methods: We used cell viability assays, Western blotting and immunofluorescent staining, combined with siRNA interference, to evaluate chemoresistance and the EMT in MCF-7 and adriamycin-resistant MCF-7/ADR breast cancer cells, and investigate the underlying mechanisms. Results: Knockdown of DUSP4 significantly increased the chemosensitivity of MCF-7 and MCF-7/ADR breast cancer cells to doxorubicin, and MCF-7/ADR cells which expressed high levels of DUSP4 had a mesenchymal phenotype. Furthermore, knockdown of DUSP4 reversed the EMT in MCF-7/ADR cells, as demonstrated by upregulation of epithelial biomarkers and downregulation of mesenchymal biomarkers, and also increased the chemosensitivity of MCF-7/ADR cells to doxorubicin. Conclusions: DUSP4 might represent a potential drug target for inhibiting drug resistance and regulating the process of the EMT during the treatment of breast cancer. - Highlights: We used different technologies to prove our conclusion. DUSP4 knockdown increased doxorubicin chemosensitivity in breast cancer cells. DUSP4 is a potential target for combating drug resistance in breast cancer. DUSP4 is a potential target for regulating the EMT in breast cancer.

    The mission of the MFRC Research and Development Program, is to provide technological advances in forensic science for the benefit of our regional partners as well as the forensic community at large. Key areas of forensic science need are identified through our interactions with our Midwest partners and our R&D advisory group, as well as through our participation in national meetings in forensic science. Under the sponsorship of the National Institute of Justice, the MFRC solicits proposals for the development of practical and useful technology, instrumentation, and methodology that address needs in areas related to forensic science and its application to operational crime laboratories. The MFRC facilitates proposal development by working to establish partnerships between researchers and our regional partners. The MFRC administers a peer-review of the proposals and then funds the selected projects at a cost of approximately $55,000 each, with a 12-month period of performance. The process for selection of these projects includes the following steps: (1) drafting of a call for proposals by MFRC staff, (2) review of the draft call by members of the R&D advisory committee, (3) review and approval of the call by NIJ, (4) issuance of the call to ISU, Ames Laboratory, regional partners, and research organizations, (5) receipt of proposals, (6) review of proposals by R&D advisory committee, (7) ranking and selection by MFRC staff using advisory committee reviews, with concurrence by NIJ, (8) notification of proposers, (9) receipt and review of progress reports by MFRC, (10) receipt and review of final reports by MFRC, R&D advisory committee, and NIJ. The decision to fund any specific project is based upon a peer-reviewed call-for-proposal system administered by the MFRC.     Land use change is a complex response to changing environmental and socioeconomic systems. Historical drivers of land use change include changes in the natural resource availability of a region, changes in economic conditions for production of certain products and changing policies. Most recently, introduction of policy incentives for biofuel production have influenced land use change in the US Midwest, leading to concerns that bioenergy production systems may compete with food production and land conservation. Here we explore how land use may be impacted by future climate mitigation measures by nesting a high resolution agricultural model (EPIC Environmental Policy Indicator Climate) for the US Midwest within a global integrated assessment model (GCAM Global Change Assessment Model). This approach is designed to provide greater spatial resolution and detailed agricultural practice information by focusing on the climate mitigation potential of agriculture and land use in a specific region, while retaining the global economic context necessary to understand the far ranging effects of climate mitigation targets. We find that until the simulated carbon prices are very high, the US Midwest has a comparative advantage in producing traditional food and feed crops over bioenergy crops. Overall, the model responds to multiple pressures by adopting a mix of future responses. We also find that the GCAM model is capable of simulations at multiple spatial scales and agricultural technology resolution, which provides the capability to examine regional response to global policy and economic conditions in the context of climate mitigation.

AHR knockdown inhibits IGF-2 stimulated increases in MCF-7 proliferation. -- Abstract: Insulin like growth factor (IGF)-1 and IGF-2 stimulate normal growth, development and breast cancer cell proliferation. Cyclin D1 (CCND1) promotes cell cycle by inhibiting retinoblastoma protein (RB1). The aryl hydrocarbon receptor (AHR) is a major xenobiotic receptor that also regulates cell cycle. The purpose of this study was to investigate whether IGF-2 promotes MCF-7 breast cancer proliferation by inducing AHR. Western blot and quantitative real time PCR (Q-PCR) analysis revealed that IGF-2 induced an approximately 2-fold increase (P < .001) in the expression of AHR and CCND1. Chromatin immunoprecipitation (ChIP), followed by Q-PCR indicated that IGF-2 promoted (P < .001) a 7-fold increase in AHR binding on the CCND1 promoter. AHR knockdown significantly (P < .001) inhibited IGF-2 stimulated increases in CCND1 mRNA and protein. AHR knockdown cells were less (P < .001) responsive to the proliferative effects of IGF-2 than control cells. Collectively, our findings have revealed a new regulatory mechanism by which IGF-2 induction of AHR promotes the expression of CCND1 and the proliferation of MCF-7 cells. This previously uncharacterized pathway could be important for the proliferation of IGF responsive cancer cells that also express AHR.

Their desire for continuous improvement, willingness to seek outside assistance, and ambition to be leaders in their field are common themes. Problem solving to overcome challenges was accepted as part of doing business. It was concluded that crossing the gap from code-based building to high performance based building was a natural evolution for these leading builders.

Black-Right-Pointing-Pointer There is no direct molecular interplay of mediators of estrogen and epigenetic signaling. -- Abstract: Estrogen signaling and epigenetic modifications, in particular DNA methylation, are involved in regulation of gene expression in breast cancers. Here we investigated a potential regulatory cross-talk between these two pathways by identifying their common target genes and exploring underlying molecular mechanisms in human MCF-7 breast cancer cells. Gene expression profiling revealed that the expression of approximately 140 genes was influenced by both 17{beta}-estradiol (E2) and a demethylating agent 5-aza-2 Prime -deoxycytidine (DAC). Gene ontology (GO) analysis suggests that these genes are involved in intracellular signaling cascades, regulation of cell proliferation and apoptosis. Based on previously reported association with breast cancer, estrogen signaling and/or DNA methylation, CpG island prediction and GO analysis, we selected six genes (BTG3, FHL2, PMAIP1, BTG2, CDKN1A and TGFB2) for further analysis. Tamoxifen reverses the effect of E2 on the expression of all selected genes, suggesting that they are direct targets of estrogen receptor. Furthermore, DAC treatment reactivates the expression of all selected genes in a dose-dependent manner.     In March 2005, the Midwest Independent System Operator (MISO) will begin operating the first-ever wholesale market for electricity in the central and upper Midwestern portion of the United States. Region-wide, centralized, security-constrained, bid-based dispatch will replace the current system of decentralized dispatch by individual utilities and control areas. This report focuses on how the operation of generators may change under centralized dispatch. We analyze a stylized example of these changes by comparing a base case dispatch based on a ''snapshot'' taken from MISO's state estimator for an actual, historical dispatch (4 p.m., July 7, 2003) to a hypothetical, centralized dispatch that seeks to minimize the total system cost of production, using estimated cost data collected by the EIA. Based on these changes in dispatch, we calculate locational marginal prices, which in turn reveals the location of congestion within MISO's footprint, as well as the distribution of congestion revenues. We also consider two sensitivity scenarios that examine (1) the effect of changes in MISO membership (2003 vs. 2004 membership lists), and (2) different load and electrical data, based on a snapshot from a different date and time (1 p.m., Feb. 18, 2004). Although our analysis offers important insights into how the MISO market could operate when it opens, we do not address the question of the total benefits or costs of creating a wholesale market in the Midwest.

    Connexins (Cx), gap junction (GJ) proteins, are regarded as tumor suppressors, and Cx43 expression is often down regulated in breast tumors. We assessed the effect of Cx43 over-expression in 2D and 3D cultures of two breast adenocarcinoma cell lines: MCF-7 and MDA-MB-231. While Cx43 over-expression decreased proliferation of 2D and 3D cultures of MCF-7 by 56% and 80% respectively, MDA-MB-231 growth was not altered in 2D cultures, but exhibited 35% reduction in 3D cultures. C-terminus truncated Cx43 did not alter proliferation. Untransfected MCF-7 cells formed spherical aggregates in 3D cultures, and MDA-MB-231 cells formed stellar aggregates. However, MCF-7 cells over-expressing Cx43 formed smaller sized clusters and Cx43 expressing MDA-MB-231 cells lost their stellar morphology. Extravasation ability of both MCF-7 and MDA-MB-231 cells was reduced by 60% and 30% respectively. On the other hand, silencing Cx43 in MCF10A cells, nonneoplastic human mammary cell line, increased proliferation in both 2D and 3D cultures, and disrupted acinar morphology. Although Cx43 over-expression did not affect total levels of ?-catenin, ?-catenin and ZO-2, it decreased nuclear levels of ?-catenin in 2D and 3D cultures of MCF-7 cells, and in 3D cultures of MDA-MB-231 cells. Cx43 associated at the membrane with ?-catenin, ?-catenin and ZO-2 in 2D and 3D cultures of MCF-7 cells, and only in 3D conditions in MDA-MB-231 cells. This study suggests that Cx43 exerts tumor suppressive effects in a context-dependent manner where GJ assembly with ?-catenin, ?-catenin and ZO-2 may be implicated in reducing growth rate, invasiveness, and, malignant phenotype of 2D and 3D cultures of MCF-7 cells, and 3D cultures of MDA-MB-231 cells, by sequestering ?-catenin away from nucleus. - Highlights: Cx43 over-expressing MCF-7 and MDA-MB-231 were grown in 2D and 3D cultures. Depletion of FLII causes inhibition of proliferation of MCF-7 cells. - Abstract: The coordinated activities of multiple protein complexes are essential to the remodeling of chromatin structure and for the recruitment of RNA polymerase II (Pol II) to the promoter in order to facilitate the initiation of transcription in nuclear receptor-mediated gene expression. Flightless I (Drosophila) homolog (FLII), a nuclear receptor coactivator, is associated with the SWI/SNF-chromatin remodeling complex during estrogen receptor (ER)?-mediated transcription. However, the function of FLII in estrogen-induced chromatin opening has not been fully explored. Here, we show that FLII plays a critical role in establishing active histone modification marks and generating the open chromatin structure of ER? target genes. We observed that the enhancer regions of ER? target genes are heavily occupied by FLII, and histone H3K4me3 and Pol II binding induced by estrogen are decreased in FLII-depleted MCF-7 cells. Furthermore, formaldehyde-assisted isolation of regulatory elements (FAIRE)-quantitative polymerase chain reaction (qPCR) experiments showed that depletion of FLII resulted in reduced chromatin accessibility of multiple ER? target genes. These data suggest FLII as a key regulator of ER?-mediated transcription through its role in regulating chromatin accessibility for the binding of RNA Polymerase II and possibly other transcriptional coactivators.

    Perillyl alcohol (POH) is a dietary monoterpene with potential applications in chemoprevention and chemotherapy. Although clinical trials are under way, POH's physiological and pharmacological properties are still unclear. In the present study, the effect of POH on polycyclic aromatic hydrocarbon (PAH)-induced genotoxicity, and the related expression were examined in MCF-7 cells. Exposure to environmental toxicant increases the risk of cancer. Many of these compounds are pro-carcinogens and are biotransformed into their ultimate genotoxic structures by xenobiotic metabolizing enzymes. CYP1A1 and 1B1 are enzymes that catalyze the biotransformation of dimethylbenz[a]anthracene (DMBA). Our data revealed that 0.5 {mu}M of POH was effective in blocking DMBA-DNA binding. Ethoxyresorufin-O-deethylase (EROD) assay indicated that the administration of POH inhibited the DMBA-induced enzyme activity in MCF-7 cells. Enzyme kinetic analysis revealed that POH inhibited CYP1B1 but not CYP1A1 activity. Quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) assay also demonstrated that the monoterpene reduced CYP1B1 mRNA abundance induced by DMBA. The present study illustrated that POH might inhibit and downregulate CYP1B1, which could protect against PAH-induced carcinogenesis.

    In a traditional Black–Scholes market we develop a verification theorem for a general class of investment and consumption problems where the standard dynamic programming principle does not hold. The theorem is an extension of the standard Hamilton–Jacobi–Bellman equation in the form of a system of non-linear differential equations. We derive the optimal investment and consumption strategy for a mean-variance investor without pre-commitment endowed with labor income. In the case of constant risk aversion it turns out that the optimal amount of money to invest in stocks is independent of wealth. The optimal consumption strategy is given as a deterministic bang-bang strategy. In order to have a more realistic model we allow the risk aversion to be time and state dependent. Of special interest is the case were the risk aversion is inversely proportional to present wealth plus the financial value of future labor income net of consumption. Using the verification theorem we give a detailed analysis of this problem. It turns out that the optimal amount of money to invest in stocks is given by a linear function of wealth plus the financial value of future labor income net of consumption. The optimal consumption strategy is again given as a deterministic bang-bang strategy. We also calculate, for a general time and state dependent risk aversion function, the optimal investment and consumption strategy for a mean-standard deviation investor without pre-commitment. In that case, it turns out that it is optimal to take no risk at all.

    In 2009, the Illinois State Geological Survey (ISGS), in collaboration with the Midwest Geological Sequestration Consortium (MGSC), created a regional technology training center to disseminate carbon capture and sequestration (CCS) technology gained through leadership and participation in regional carbon sequestration projects. This technology training center was titled and branded as the Sequestration Training and Education Program (STEP). Over the last six years STEP has provided local, regional, national, and international education and training opportunities for engineers, geologists, service providers, regulators, executives, K-12 students, K-12 educators, undergraduate students, graduate students, university and community college faculty members, and participants of community programs and functions, community organizations, and others. The goal for STEP educational programs has been on knowledge sharing and capacity building to stimulate economic recovery and development by training personnel for commercial CCS projects. STEP has worked with local, national and international professional organizations and regional experts to leverage existing training opportunities and provide stand-alone training. This report gives detailed information on STEP activities during the grant period (2009-2015).

    Protoapigenone is a unique flavonoid and enriched in many ferns, showing potent antitumor activity against a broad spectrum of human cancer cell lines. RY10-4, a modified version of protoapigenone, manifested better anti-proliferation activity in human breast cancer cell line MCF-7. The cytotoxicity of RY10-4 against MCF-7 cells is exhibited in both time- and concentration-dependent manners. Here we investigated a novel effect of RY10-4 mediated autophagy in autophagy defect MCF-7 cells. Employing immunofluorescence assay for microtubule-associated protein light-chain 3 (LC3), monodansylcadaverine staining, Western blotting analyses for LC3 and p62 as well as ultrastructural analysis by transmission electron microscopy, we showed that RY10-4 induced autophagy in MCF-7 cells but protoapigenone did not. Meanwhile, inhibition of autophagy by pharmacological and genetic approaches significantly increased the viability of RY10-4 treated cells, suggesting that the autophagy induced by RY10-4 played as a promotion mechanism for cell death. Further studies revealed that RY10-4 suppressed the activation of mTOR and p70S6K via the Akt/mTOR pathway. Our results provided new insights for the mechanism of RY10-4 induced cell death and the cause of RY10-4 showing better antitumor activity than protoapigenone, and supported further evidences for RY10-4 as a lead to design a promising antitumor agent. - Highlights: We showed that RY10-4 induced autophagy in MCF-7 cells but protoapigenone did not. Autophagy induced by RY10-4 played as a promotion mechanism for cell death. RY10-4 induced autophagy in MCF-7 cell through the Akt/mTOR pathway.     Collaborative efforts of Midwest Consortium have been put forth to add value to distiller's grains by further processing them into fermentable sugars, ethanol, and a protein rich co-product consistent with a pathway to a biorenewables industry (Schell et al, 2008). These studies were recently published in the enclosed special edition (Volume 99, Issue 12) of Bioresource Technology journal. Part of them have demonstrated the utilization of distillers grains as additional feedstock for increased ethanol production in the current dry grind process (Kim et al., 2008a, b; Dien et al.,2008, Ladisch et al., 2008a, b). Results showed that both liquid hot water (LHW) pretreatment and ammonia fiber expansion (AFEX) were effective for enhancing digestibility of distiller's grains. Enzymatic digestion of distiller's grains resulted in more than 90% glucose yield under standard assay conditions, although the yield tends to drop as the concentration of dry solids increases. Simulated process mass balances estimated that hydrolysis and fermentation of distillers grains can increase the ethanol yield by 14% in the current dry milling process (Kim et al., 2008c). Resulting co-products from the modified process are richer in protein and oil contents than conventional distiller's grains, as determined both experimentally and computationally. Other research topics in the special edition include water solubilization of DDGS by transesterification reaction with phosphite esters (Oshel el al., 2008) to improve reactivity of the DDGS to enzymes, hydrolysis of soluble oligomers derived from DDGS using functionalized mesoporous solid catalysts (Bootsma et al., 2008), and ABE (acetone, butanol, ethanol) production from DDGS by solventogenic Clostridia (Ezeji and Blaschek, 2008).     The aim was to analyze the mechanism of leptin-induced activity of telomerase in MCF-7 breast cancer cells. We found that leptin activated telomerase in a dose-dependent manner; leptin upregulated the expression of Human Telomerase Reverse Transcriptase (hTERT) at mRNA and protein levels; blockade of signal transducer and activator of transcription 3 (STAT3) phosphorylation significantly counteracted leptin-induced hTERT transcription and protein expression; chromatin immunoprecipitation analysis showed that leptin enhanced the binding of STAT3 to the hTERT promoter. This study uncovers a new mechanism of the proliferative effect of leptin on breast cancer cells and provides a new explanation of obesity-related breast cancer.

    This report provides estimates on energy consumption in the manufacturing sector of the U.S. economy based on data from the Manufacturing Energy Consumption Survey. The sample used in this report represented about 250,000 of the largest manufacturing establishments which account for approximately 98 percent of U.S. economic output from manufacturing, and an expected similar proportion of manufacturing energy use. The amount of energy use was collected for all operations of each establishment surveyed. Highlights of the report include profiles for the four major energy-consuming industries (petroleum refining, chemical, paper, and primary metal industries), and an analysis of the effects of changes in the natural gas and electricity markets on the manufacturing sector.     Household Vehicles Energy Consumption 1994 reports on the results of the 1994 Residential Transportation Energy Consumption Survey (RTECS). The RTECS is a national sample survey that has been conducted every 3 years since 1985. For the 1994 survey, more than 3,000 households that own or use some 6,000 vehicles provided information to describe vehicle stock, vehicle-miles traveled, energy end-use consumption, and energy expenditures for personal vehicles. The survey results represent the characteristics of the 84.9 million households that used or had access to vehicles in 1994 nationwide. (An additional 12 million households neither owned or had access to vehicles during the survey year.) To be included in then RTECS survey, vehicles must be either owned or used by household members on a regular basis for personal transportation, or owned by a company rather than a household, but kept at home, regularly available for the use of household members. Most vehicles included in the RTECS are classified as {open_quotes}light-duty vehicles{close_quotes} (weighing less than 8,500 pounds). However, the RTECS also includes a very small number of {open_quotes}other{close_quotes} vehicles, such as motor homes and larger trucks that are available for personal use.

    An integrated model is being developed to advance our understanding of the interactions between human activities, terrestrial system and water cycle, and how system interactions will be affected by a changing climate at the regional scale. As a first step towards that goal, a global integrated assessment model including a waterdemand model is coupled offline with a land surface hydrology routing water resources management model. A spatial and temporal disaggregation approach is developed to project the annual regional water demand simulations into a daily time step and subbasin representation. The model demonstrated reasonable ability to represent the historical flow regulation and water supply over the Midwest (Missouri, Upper Mississippi and Ohio). Implications for the future flow regulation, water supply and supply deficit are investigated using a climate change projection with the B1 emission scenario which affects both natural flow and water demand. Over the Midwest, changes in flow regulation are mostly driven by the change in natural flow due to the limited storage capacity over the Ohio and Upper Mississippi river basins. The changes in flow and demand have a combined effect on the Missouri Summer regulated flow. The supply deficit tends to be driven by the change in flow over the region. Spatial analysis demonstrates the relationship between the supply deficit and the change in demand over urban areas not along a main river or with limited storage, and over areas upstream of groundwater dependent fields with therefore overestimated demand.

    Various components of refining energy were determined experimentally and compared with those calculated on the basis of the dimensions of morphological elements of wood. The experimentally determined fiberization energy of spruce was 6 to 60 times larger than the calculated value and that of birch 3 to 15 times larger. The energy consumed in reducing the Canadian standard freeness of isolated fibers from 500 to 150 ml was found to be approximately 1/3 of the total fiber development energy for both spruce and birch TMP. Chip size affected the refining energy consumption; the total energy dropped by approximately 30% when chip size was reduced from 16 mm to 3 mm in the case of spruce and approximately 40% for birch. 6 refs.

    This presents information about household end-use consumption of energy and expenditures for that energy. These data were collected in the 1993 Residential Energy Consumption Survey; more than 7,000 households were surveyed for information on their housing units, energy consumption and expenditures, stock of energy-consuming appliances, and energy-related behavior. The information represents all households nationwide (97 million). Key findings: National residential energy consumption was 10.0 quadrillion Btu in 1993, a 9% increase over 1990. Weather has a significant effect on energy consumption. Consumption of electricity for appliances is increasing. Houses that use electricity for space heating have lower overall energy expenditures than households that heat with other fuels. RECS collected data for the 4 most populous states: CA, FL, NY, TX.

    Though electric power is a major operating cost of oil production, few producers have systematically evaluated their power consumption for ways to be more efficient. There is significant money to be saved by doing so, and now is a good time to make an evaluation because new power options are at hand. They range from small turbo generators that can run on casing head gas and power one or two lift pumps, to rebuilt major turbines and ram-jet powered generators that can be set in a multi-well field and deliver power at bargain prices. Power industry deregulation is also underway. Opportunities for more advantageous power contracts from competitive sources are not far off. This two-part series covers power efficiency and power options. This article reviews steps you can take to evaluate the efficiency of your power use and go about improving it. Part 2 will discuss opportunities for use of distributed power and changes you can expect from decentralized power.

    Today, emphasis is placed on reducing power consumption. Computers are large consumers; therefore it is important to know the total consumption of computing systems. Since their optimal functioning requires quite strict environmental conditions, without much variation in temperature and humidity, reducing energy consumption cannot be made without monitoring environmental parameters. Thus, the present work uses a multifunctional electric meter UPT 210 for power consumption monitoring. Two applications were developed: software which carries meter readings provided by electronic and programming facilitates remote device and a device for temperature monitoring and control. Following temperature variations that occur both in the cooling system, as well as the ambient, can reduce energy consumption. For this purpose, some air conditioning units or some computers are stopped in different time slots. These intervals were set so that the economy is high, but the work's Datacenter is not disturbed.

A novel class of hybrid compound synthesized by introducing an ITC moiety on PTER backbone was evaluated for its anti-cancer efficacy in hormone-dependent breast cancer cell line (MCF-7) in vitro and Ehrlich ascitic tumor bearing mice model in vivo. The novel hybrid molecule showed significant in vitro anti-cancer activity (IC{sub 50}=25±0.38) when compared to reference compound PTER (IC{sub 50}=65±0.42). The conjugate molecule induced both S and G2/M phase cell cycle arrest as indicated by flow cytometry analysis. In addition, the conjugate induced cell death was characterized by changes in cell morphology, DNA fragmentation, activation of caspase-9, release of cytochrome-c into cytosol and increased Bax: Bcl-2 ratio. The conjugate also suppressed the phosphorylation of Akt and ERK. The conjugate induced cell death was significantly increased in presence of A6730 (a potent Akt1/2 kinase inhibitor) and PD98059 (a specific ERK inhibitor). Moreover, the conjugated PTER inhibited tumor growth in Ehrlich ascitic cell induced tumor bearing mice as observed by reduction in tumor volume compared to untreated animals. Collectively, the pro-apoptotic effect of conjugate is mediated through the activation of caspases, and is correlated with the blockade of the Akt and ERK signaling pathways in MCF-7 cells. - Highlights: • Conjugate was prepared by appending isothiocyanate moiety on pterostilbene backbone. • Conjugate showed anticancer effects at comparatively lower dose than pterostilbene. • Conjugate caused blockage of the Akt and ERK signaling pathways in MCF-7 cells. • Conjugate significantly reduced solid tumor volume as compared to pterostilbene.

Transcription factor binding sites showed distinct spatial distribution on genes. -- Abstract: Background: Breast cancer cells respond to estrogen in a concentration dependent fashion, resulting in proliferation or apoptosis. The mechanism of this concentration dependent differential outcome is not well understood yet. Methodology: Meta-analysis of the expression data of MCF7 cells treated with low (1 nM) or high (100 nM) dose of estradiol (E2) was performed. We identified genes differentially expressed at the low or the high dose, and examined the nature of regulatory elements in the vicinity of these genes. Specifically, we looked for the difference in the presence, abundance and spatial distribution of binding sites for estrogen receptor (ER) and selected transcription factors (TFs) in the genomic region up to 25 kb upstream and downstream from the transcription start site (TSS) of these genes. Results: It was observed that at high dose E2 induced the expression of stress responsive genes, while at low dose, genes involved in cell cycle were induced. We found that the occurrence of transcription factor binding regions (TFBRs) for certain factors such as Sp1 and SREBP1 were higher on regulatory regions of genes expressed at low dose. At high concentration of E2, genes with a higher frequency of Oct-1 binding regions were predominantly involved. In addition, there were differences in the spatial distribution pattern of the TFBRs in the genomic regions among the two sets of genes. The proper selection and maintenance of melting and holding equipment are clearly important factors in minimizing energy consumption in die-casting operations [2]. In addition to energy consumption, furnace selection also influences metal loss due to oxidation, metal quality, and maintenance requirements. Other important factors influencing energy consumption in a die-casting facility include geographic location, alloy(s) cast, starting form of alloy (solid or liquid), overall process flow, casting yield, scrap rate, cycle times, number of shifts per day, days of operation per month, type and size of die-casting form of alloy (solid or liquid), overall process flow, casting yield, scrap rate, cycle times, number of shifts per day, days of operation per month, type and size of die-casting machine, related equipment (robots, trim presses), and downstream processing (machining, plating, assembly, etc.). Each of these factors also may influence the casting quality and productivity of a die-casting enterprise. In a die-casting enterprise, decisions regarding these issues are made frequently and are based on a large number of factors. Therefore, it is not surprising that energy consumption can vary significantly from one die-casting enterprise to the next, and within a single enterprise as function of time.

The 1987 RECS, seventh in a series of national surveys of households and their energy suppliers, provides baseline information on household energy use in the United States. Data from the seven RECS and its companion survey, the Residential Transportation Energy Consumption Survey (RTECS), are made available to the public in published reports such as this one, and on public use data files. This report presents data for the four Census regions and nine Census divisions on the consumption of and expenditures for electricity, natural gas, fuel oil and kerosene (as a single category), and liquefied petroleum gas (LPG). Data are also presented on consumption of wood at the Census region level. The emphasis in this report is on graphic depiction of the data.     Cadmium (Cd) is a common environmental toxicant and an established carcinogen. Epidemiological studies implicate Cd with human breast cancer. Low micromolar concentrations of Cd promote proliferation of human breast cancer cells in vitro. The growth promotion of breast cancer cells is associated with the activation of MAPK/ERK pathway. This study explores the mechanism of Cd-induced activation of MAPK/ERK pathway. Specifically, the role of cell surface receptors ERα, EGFR, and Src kinase was evaluated in human breast cancer MCF-7 cells treated with 1–3 μM Cd. The activation of ERK was studied using a serum response element (SRE) luciferase reporter assay. Receptor phosphorylation was detected by Western blot analyses. Cd treatment increased both the SRE reporter activity and ERK1/2 phosphorylation in a concentration-dependent manner. Cd treatment had no effect on reactive oxygen species (ROS) generation. Also, blocking the entry of Cd into the cells with manganese did not diminish Cd-induced activation of MAPK/ERK. These results suggest that the effect of Cd was likely not caused by intracellular ROS generation, but through interaction with the membrane receptors. While Cd did not appear to activate either EGFR or Src kinase, their inhibition completely blocked the Cd-induced activation of ERK as well as cell proliferation. Similarly, silencing ERα with siRNA or use of ERα antagonist blocked the effects of Cd. Based on these results, it is concluded that not only ERα, but also basal activities of EGFR and Src kinase are essential for Cd-induced signal transduction and activation of MAPK/ERK pathway for breast cancer cell proliferation. - Highlights: • Low micromolar concentrations of Cd rapidly activate ERK1/2 in MCF-7 cells. • Signal transduction and resulting cell proliferation require EGFR, ERα, and Src. • These findings implicate Cd in promotion of breast cancer.

    Background: Organotin compounds (OTCs) are a large class of synthetic chemicals with widely varying properties. Due to their potential adverse health effects, their use has been restricted in many countries. Humans are exposed to OTCs mostly through fish consumption. Objectives: The aim of this study was to describe OTC exposure through fish consumption and to assess the associated potential health risks in a Finnish population. Methods: An extensive sampling of Finnish domestic fish was carried out in the Baltic Sea and freshwater areas in 2005-2007. In addition, samples of imported seafood were collected in 2008. The chemical analysis was performed in an accredited testing laboratory during 2005-2008. Average daily intake of the sum of dibutyltin (DBT), tributyltin (TBT), triphenyltin (TPhT) and dioctyltin (DOT) ({Sigma}OTCs) for the Finnish population was calculated on the basis of the measured concentrations and fish consumption rates. Results: The average daily intake of {Sigma}OTCs through fish consumption was 3.2 ng/kg bw day{sup -1}, which is 1.3% from the Tolerable Daily Intake (TDI) of 250 ng/kg bw day{sup -1} set by the European Food Safety Authority. In total, domestic wild fish accounted for 61% of the {Sigma}OTC intake, while the intake through domestic farmed fish was 4.0% and the intake through imported fish was 35%. The most important species were domestic perch and imported salmon and rainbow trout. Conclusions: The Finnish consumers are not likely to exceed the threshold level for adverse health effects due to OTC intake through fish consumption.

    This report presents data on the US consumption and expenditures for residential use of natural gas, electricity, fuel oil or kerosene, and liquefied petroleum gas (LPG) from April 1982 through March 1983. Data on the consumption of wood for this period are also presented. The consumption and expenditures data are based on actual household bills, obtained, with the permission of the household. from the companies supplying energy to the household. Data on wood consumption are based on respondent recall of the amount of wood burned during the winter and are subject to memory errors and other reporting errors described in the report. These data come from the 1982 Residential Energy Consumption Survey (RECS), the fifth in a series of comparable surveys beginning in 1978. The 1982 survey is the first survey to include, as part of its sample, a portion of the same households interviewed in the 1980 survey. A separate report is planned to report these longitudinal data. This summary gives the highlights of a comparison of the findings for the 5 years of RECS data. The data cover all types of housing units in the 50 states and the District of Columbia including single-family units, apartments, and mobile homes. For households with indirect energy costs, such as costs that are included in the rent or paid by third parties, the sonsumption and expenditures data are estimated and included in the figures reported here. The average household consumption of natural gas, electricity, fuel oil or kerosene, and LPG dropped in 1982 from the previous year, hitting a 5-year low since the first Residential Energy Consumption Survey (RECS) was conducted in 1978. The average consumption was 103 (+-3) million Btu per household in 1982, down from 114 (+-) million Btu in 1981. The weather was the main contributing factor. 8 figures, 46 tables.

    Lighting represents a substantial fraction of commercial electricity consumption. A wide range of initiatives in the Department of Energy`s (DOE) National Energy Strategy have focused on commercial lighting as a potential source of energy conservation. This report provides a statistical profile of commercial lighting, to examine the potential for lighting energy conservation in commercial buildings. The principal conclusion from this analysis is that energy use for lighting could be reduced by as much as a factor of four using currently available technology. The analysis is based primarily on the Energy Information Administration`s (EIA) 1986 Commercial Buildings Energy Consumption Survey (CBECS). The more recent 1989 survey had less detail on lighting, for budget reasons. While changes have occurred in the commercial building stock since 1986, the relationships identified by this analysis are expected to remain generally valid. In addition, the analytic approach developed here can be applied to the data that will be collected in the 1992 CBECS.

    The Commercial Buildings Energy Consumption Survey (CBECS) that is conducted by the Energy Information Administration (EIA) is the primary source of energy data for commercial buildings in the United States. The survey began in 1979 and has subsequently been conducted in 1983, 1986, and 1989. The next survey will cover energy consumption during the year 1992. The building characteristic data will be collected between August 1992 and early December 1992. Requests for energy consumption data are mailed to the energy suppliers in January 1993, with data due by March 1993. Before each survey is sent into the field, the data users' needs are thoroughly assessed. The purpose of this report is to document the findings of that user-needs assessment for the 1992 survey.

    Electrical energy consumption control apparatuses and electrical energy consumption control methods are described. According to one aspect, an electrical energy consumption control apparatus includes processing circuitry configured to receive a signal which is indicative of current of electrical energy which is consumed by a plurality of loads at a site, to compare the signal which is indicative of current of electrical energy which is consumed by the plurality of loads at the site with a desired substantially sinusoidal waveform of current of electrical energy which is received at the site from an electrical power system, and to use the comparison to control an amount of the electrical energy which is consumed by at least one of the loads of the site.

    This report is the first of a series of reports based on data from the 1987 RECS. The 1987 RECS is the seventh in the series of national surveys of households and their energy suppliers. These surveys provide baseline information on how households in the United States use energy. A cross section of housing types such as single-family detached homes, townhouses, large and small apartment buildings, condominiums, and mobile homes were included in the survey. Data from the RECS and a companion survey, the Residential Transportation Energy Consumption Survey (RTECS), are available to the public in published reports such as this one and on public use tapes. 10 figs., 69 tabs.

    This report, Household Energy Consumption and Expenditures 1990, is based upon data from the 1990 Residential Energy Consumption Survey (RECS). Focusing on energy end-use consumption and expenditures of households, the 1990 RECS is the eighth in a series conducted since 1978 by the Energy Information Administration (EIA). Over 5,000 households were surveyed, providing information on their housing units, housing characteristics, energy consumption and expenditures, stock of energy-consuming appliances, and energy-related behavior. The information provided represents the characteristics and energy consumption of 94 million households nationwide.
