    A fast FIFO (First In First Out) memory buffer capable of storing data at rates of 100 megabytes per second. The invention includes a data packer which concatenates small bit data words into large bit data words, a memory array having individual data storage addresses adapted to store the large bit data words, a data unpacker into which large bit data words from the array can be read and reconstructed into small bit data words, and a controller to control and keep track of the individual data storage addresses in the memory array into which data from the packer is being written and data to the unpacker is being read.

    Sandia National Laboratories has tested and evaluated a new infrasound sensor, the MB3a, manufactured by Seismo Wave. These infrasound sensors measure pressure output by a methodology developed by researchers at the French Alternative Energies and Atomic Energy Commission (CEA) and the technology was recently licensed to Seismo Wave for production and sales. The purpose of the infrasound sensor evaluation was to determine a measured sensitivity, transfer function, power, self-noise, dynamic range, seismic sensitivity, and self- calibration ability. The MB3a infrasound sensors are being evaluated for potential use in the International Monitoring System (IMS) of the Comprehensive Nuclear Test-Ban-Treaty Organization (CTBTO).

    Hyperglycemia is a critical risk factor for development and progression of breast cancer. We have recently reported that high glucose induces phosphorylation of histone H3 at Ser 10 as well as de-phosphorylation of GSK-3? at Ser 9 in MDA-MB-231 cells. Here, we elucidate the mechanism underlying hyperglycemia-induced proliferation in MDA-MB-231 breast cancer cells. We provide evidence that hyperglycemia led to increased DNA methylation and DNMT1 expression in MDA-MB-231 cells. High glucose condition led to significant increase in the expression of PCNA, cyclin D1 and decrease in the expression of PTPN 12, p21 and PTEN. It also induced hypermethylation of DNA at the promoter region of PTPN 12, whereas hypomethylation at Vimentin and Snail. Silencing of GSK-3? by siRNA prevented histone H3 phosphorylation and reduced DNMT1 expression. We show that chromatin obtained after immunoprecipitation with phospho-histone H3 was hypermethylated under high glucose condition, which indicates a cross-talk between DNA methylation and histone H3 phosphorylation. ChIP-qPCR analysis revealed up-regulation of DNMT1 and metastatic genes viz. Vimentin, Snail and MMP-7 by phospho-histone H3, which were down-regulated upon GSK-3? silencing. To the best of our knowledge, this is the first report which shows that interplay between GSK-3? activation, histone H3 phosphorylation and DNA methylation directs proliferation of breast cancer cells. - Highlights: High glucose induces phosphorylation of histone H3 and dephosphorylation of GSK-3?. Moreover, hyperglycemia also leads to increased DNA methylation in MDA-MB-231 cells. Inhibition of GSK-3? prevented histone H3 phosphorylation and reduced DNMT1 levels. Interplay exists between GSK-3?, histone H3 phosphorylation and DNA methylation.

    The results of a static test of total ionizing dose (TID) effects on an ISSI 4Mb PSRAM memory are reported in this work. The irradiation was performed at the IEAvs Laboratory of Ionizing Radiation with 1.17 and 1.32 MeV gamma-rays from a {sup 60}Co source at a dose rate of 2.5 krad/h up to an accumulated dose of 215.7 krad. The TID threshold for bit flip found in this experiment was 52.5 krad. From a sampling of 4096 memory addresses it was estimated a bit flip rate of approximately 50% at an accumulated dose of 215.7 krad.

    Well established theory can be used to combine single-phenomenology hypothesis tests into a multi-phenomenology event screening hypothesis test (Fisher's and Tippett's tests). Commonly used standard error in Ms:mb event screening hypothesis test is not fully consistent with physical basis. Improved standard error - Better agreement with physical basis, and correctly partitions error to include Model Error as a component of variance, correctly reduces station noise variance through network averaging. For 2009 DPRK test - Commonly used standard error 'rejects' H0 even with better scaling slope ({beta} = 1, Selby et al.), improved standard error 'fails to rejects' H0.

  19. A 5.5-Mb high-resolution integrated map of distal 11q13

    The distal part of 11q13, which contains several genes relevant to human diseases, has been poorly mapped as part of genome-wide mapping efforts. In the prospect of drawing a fine-scale integrated map of the area containing KRN1 and OMP, we have established a framework of markers by hybridization to DNA of somatic cell hybrids and by fluorescence in situ hybridization (FISH) on metaphase chromosomes. The probes studied were used to isolate 27 YACs and 16 cosmids that could be organized in three contigs covering approximately 6 Mb. These contigs were separated by two gaps that are likely to contain sequences underrepresented in YAC libraries. They were then integrated based on long-range restriction mapping and DNA-fiber FISH into a high-resolution physical map, which covers a 5.5-Mb region and includes 36 anonymous markers and 10 genes. This map will be used to search for genes within the 2/3 of this region where none have been localized as yet. It will also lay the ground for the characterization of an amplicon surrounding GARP in breast cancer and for the search of disease genes within this region. 42 refs., 2 figs., 2 tabs.

  20. Heavy reflector experiments in the IPEN/MB-01 reactor: Stainless steel, carbon steel and nickel

    New experiments devoted to the measurements of physical parameters of a light water core surrounded by a heavy reflector were performed in the IPEN/MB-01 research reactor facility. These experiments comprise three sets of heavy reflector (SS-304, Carbon Steel, and Nickel) in a form of laminates around 3 mm thick. Each set was introduced individually in the west face of the core of the IPEN/MB-01 reactor. The aim here is to provide high quality experimental data for the interpretation and validation of the SS-304 heavy reflector calculation methods. The experiments of Carbon Steel, which is composed mainly of iron, and Nickel were performed to provide a consistent and an interpretative check for the SS-304 reflector experiment. The experimental results comprise critical control bank positions, temperatures and reactivities as a function of the number of the plates. Particularly to the case of Nickel, the experimental data are unique of its kind. The theoretical analysis was performed by MCNP-5 with the nuclear data library ENDF/B-VII.0. It was shown that this nuclear data library has a very good performance up to thirteen plates and overestimates the reactivity for higher number of plates independently of the type of the reflector.

    Researches that aim to improve the performance of neutron transport codes and quality of nuclear cross section databases are very important to increase the accuracy of simulations and the quality of the analysis and prediction of phenomena in the nuclear field. In this context, relevant experimental data such as reactivity worth measurements are needed. Control rods may be made of several neutron absorbing materials that are used to adjust the reactivity of the core. For the reactor operation, these experimental data are also extremely important: with them it is possible to estimate the reactivity worth by the movement of the control rod, understand the reactor response at each rod position and to operate the reactor safely. This work presents a temperature correction approach for the control rod calibration problem. It is shown the control rod calibration data of the IPEN/MB-01 reactor, the integral and differential reactivity curves and a theoretical analysis, performed by the MCNP-5 reactor physics code, developed and maintained by Los Alamos National Laboratory, using the ENDF/B-VII.0 nuclear data library.

    This is a complementary work about the behavior analysis of the neutron lifetimes that was developed in the IPEN/MB-01 nuclear reactor facility. The macroscopic neutron noise technique was experimentally employed using pulse mode detectors for two stages of control rods insertion, where a total of twenty levels of subcriticality have been carried out. It was also considered that the neutron reflector density was treated as an additional group of delayed neutrons, being a sophisticated approach in the two-region kinetic theoretical model.

    Connexins (Cx), gap junction (GJ) proteins, are regarded as tumor suppressors, and Cx43 expression is often down regulated in breast tumors. We assessed the effect of Cx43 over-expression in 2D and 3D cultures of two breast adenocarcinoma cell lines: MCF-7 and MDA-MB-231. While Cx43 over-expression decreased proliferation of 2D and 3D cultures of MCF-7 by 56% and 80% respectively, MDA-MB-231 growth was not altered in 2D cultures, but exhibited 35% reduction in 3D cultures. C-terminus truncated Cx43 did not alter proliferation. Untransfected MCF-7 cells formed spherical aggregates in 3D cultures, and MDA-MB-231 cells formed stellar aggregates. However, MCF-7 cells over-expressing Cx43 formed smaller sized clusters and Cx43 expressing MDA-MB-231 cells lost their stellar morphology. Extravasation ability of both MCF-7 and MDA-MB-231 cells was reduced by 60% and 30% respectively. On the other hand, silencing Cx43 in MCF10A cells, nonneoplastic human mammary cell line, increased proliferation in both 2D and 3D cultures, and disrupted acinar morphology. Although Cx43 over-expression did not affect total levels of ?-catenin, ?-catenin and ZO-2, it decreased nuclear levels of ?-catenin in 2D and 3D cultures of MCF-7 cells, and in 3D cultures of MDA-MB-231 cells. Cx43 associated at the membrane with ?-catenin, ?-catenin and ZO-2 in 2D and 3D cultures of MCF-7 cells, and only in 3D conditions in MDA-MB-231 cells. This study suggests that Cx43 exerts tumor suppressive effects in a context-dependent manner where GJ assembly with ?-catenin, ?-catenin and ZO-2 may be implicated in reducing growth rate, invasiveness, and, malignant phenotype of 2D and 3D cultures of MCF-7 cells, and 3D cultures of MDA-MB-231 cells, by sequestering ?-catenin away from nucleus. - Highlights: Cx43 over-expressing MCF-7 and MDA-MB-231 were grown in 2D and 3D cultures. Proliferation and growth morphology were affected in a context dependent manner. Extravasation ability of both MCF-7 and MDA-MB-231 cells was reduced. Cx43-mediated gap junction complex assembly correlated with observed changes. We propose that membranous Cx43 sequesters ?-catenin away from the nucleus.

    Photodiode arrays are formed with close diode-to-diode spacing and minimized cross-talk between diodes in the array by isolating the diodes from one another with trenches that are formed between the photodiodes in the array. The photodiodes are formed of spaced regions in a base layer, each spaced region having an impurity type opposite to that of the base layer to define a p-n junction between the spaced regions and the base layer. The base layer meets a substrate at a boundary, with the substrate being much more heavily doped than the base layer with the same impurity type. The trenches extend through the base layer and preferably into the substrate. Minority carriers generated by absorption of light photons in the base layer can only migrate to an adjacent photodiode through the substrate. The lifetime and the corresponding diffusion length of the minority carriers in the substrate is very short so that all minority carriers recombine in the substrate before reaching an adjacent photodiode.

MCU silencing potentiated caspase-independent cell death initiated by ionomycin. MCU silencing promoted ionomycin-mediated cell death without changes in bulk Ca{sup 2+}. -- Abstract: The mitochondrial calcium uniporter (MCU) transports free ionic Ca{sup 2+} into the mitochondrial matrix. We assessed MCU expression in clinical breast cancer samples using microarray analysis and the consequences of MCU silencing in a breast cancer cell line. Our results indicate that estrogen receptor negative and basal-like breast cancers are characterized by elevated levels of MCU. Silencing of MCU expression in the basal-like MDA-MB-231 breast cancer cell line produced no change in proliferation or cell viability. However, distinct consequences of MCU silencing were seen on cell death pathways. Caspase-dependent cell death initiated by the Bcl-2 inhibitor ABT-263 was not altered by MCU silencing; whereas caspase-independent cell death induced by the calcium ionophore ionomycin was potentiated by MCU silencing. Measurement of cytosolic Ca{sup 2+} levels showed that the promotion of ionomycin-induced cell death by MCU silencing occurs independently of changes in bulk cytosolic Ca{sup 2+} levels. This study demonstrates that MCU overexpression is a feature of some breast cancers and that MCU overexpression may offer a survival advantage against some cell death pathways. MCU inhibitors may be a strategy to increase the effectiveness of therapies that act through the induction of caspase-independent cell death pathways in estrogen receptor negative and basal-like breast cancers.

    Prolactin receptor (PRL-R) activation regulates cell differentiation, proliferation, cell survival and motility of breast cells. Prolactin (PRL) and PRL-R over-expression are strongly implicated in breast cancer, particularly contributing to tumor growth and invasion in the more aggressive estrogen-receptor negative (ER?) disease. PRL-R antagonists have been suggested as potential therapeutic agents; however, mechanisms through which PRL-R antagonists exert their actions are not well-understood. Zinc (Zn) is a regulatory factor for over 10% of the proteome, regulating critical cell processes such as proliferation, cell signaling, transcription, apoptosis and autophagy. PRL-R signaling regulates Zn metabolism in breast cells. Herein we determined effects of PRL-R attenuation on cellular Zn metabolism and cell function in a model of ER-, PRL-R over-expressing breast cancer cells (MDA-MB-453). PRL-R attenuation post-transcriptionally increased ZnT2 abundance and redistributed intracellular Zn pools into lysosomes and mitochondria. ZnT2-mediated lysosomal Zn sequestration was associated with reduced matrix metalloproteinase 2 (MMP-2) activity and decreased invasion. ZnT2-mediated Zn accumulation in mitochondria was associated with increased mitochondrial oxidation. Our results suggest that PRL-R antagonism in PRL-R over-expressing breast cancer cells may reduce invasion through the redistribution of intracellular Zn pools critical for cellular function. - Highlights: PRL-R attenuation increased ZnT2 expression. PRL-R attenuation increased lysosomal and mitochondrial Zn accumulation. PRL-R attenuation decreased MMP-2 and invasion. PRL-R antagonists may modulate lysosomal and mitochondrial Zn pools.

    The rapid growth in computational capabilities has provided great opportunities for direct numerical simulations (DNS) of turbulent combustion, a type of simulations without any turbulence model. With the help of terascale high performance supercomputing (HPC) resources, we are now able to provide fundamental insight into turbulence-chemistry interaction in simple laboratory-scale turbulent flames with detailed chemistry using three-dimensional (3D) DNS. However, the actual domain size of 3D-DNS is still limited within {approx} O(10 cm{sup 3}) due to its tremendously high grid resolution required to resolve the smallest turbulent length scale as well as flame structures. Moreover, 3D-DNS will require more computing powers to investigate next-generation engines, of which operating conditions will be characterized by higher pressures, lower temperatures, and higher levels of dilution. In this talk, I will discuss the capabilities and limits of DNS of turbulent combustion and present some results of ignition/extinction characteristics of a highly diluted hydrogen flame counter-flowing against heated air. The results of our recent 3D-DNS of a spatially-developing turbulent lifted hydrogen jet flame in heated coflow will also be presented. The 3D-DNS was performed at a jet Reynolds number of 11,000 with {approx} 1 billion grid points, which required 3.5 million CPU hours on Cray XT3/XT4 at Oak Ridge National Laboratories.

    The etiology of radiation carcinogenesis has been described in terms of aberrant changes that span several levels of biological organization. Growth factors regulate many important cellular and tissue functions including apoptosis, differentiation and proliferation. A variety of genetic and epigenetic changes of growth factors have been shown to contribute to cancer initiation and progression. It is known that cellular and tissue damage to ionizing radiation is in part initiated by the production of reactive oxygen species, which can activate cytokine signaling, and the DNA damage response pathways, most notably the ATM signaling pathway. Recently the transforming growth factor ? (TGF?) pathway has been shown to regulate or directly interact with the ATM pathway in the response to radiation. The relevance of this interaction with the ATM pathway is not known although p53 becomes phosphorylated and DNA damage responses are involved. However, growth factor interactions with DNA damage responses have not been elucidated particularly at low doses and further characterization of their relationship to cancer processes is warranted. Our goal will be to use a systems biology approach to mathematically and experimentally describe the low dose responses and cross-talk between the ATM and TGF? pathways initiated by low and high LET radiation. We will characterize ATM and TGF? signaling in epithelial and fibroblast cells using 2D models and ultimately extending to 3D organotypic cell culture models to begin to elucidate possible differences that may occur for different cell types and/or inter-cellular communication. We will investigate the roles of the Smad and Activating transcription factor 2 (ATF2) proteins as the potential major contributors to cross- talk between the TGF? and ATM pathways, and links to cell cycle control and/or the DNA damage response, and potential differences in their responses at low and high doses. We have developed various experimental approaches to apply to these problems using confocal microscopy and flow cytometry to detail changes at low dose/dose-rate in order to understand individual cell responses, and will establish our mathematical models based on the experimental findings resulting from changes in DNA repair, apoptosis and proliferation.

    The overall objective of this project was to demonstrate, through a series of real-world applications of existing technology, the benefits to the University of Nevada, Reno and the community, of various sustainability efforts. The project was very successful and has stimulated the Campus to take on more projects after seeing the successes of those initial ones funded through this project. The three areas of this work could broadly be described as energy efficiency, renewable energy and recycling. Under the first project, the campus did several projects replacing or changing heating and cooling systems, using state funding. The DOE funding initially funded the replacement of lights in one campus parking garage with LED lights. Subsequently, the campus facilities group recognized how effective this was and leveraged funds to do the other two garages. Similarly with the renewable energy project, once the first system was installed and working well, the campus committed funds to more than double that system. Lastly, the recycling efforts expanded the use and awareness on campus and led the campus to begin using a single-stream recycling program once it became available in this area, hopefully leading to more participation by the campus community. Thus, overall the project areas each did what they were intended to do, which was to demonstrate the usefulness of these sustainability programs and thus encourage the campus to do more. All this great work helps the campus’ goals overall, but without additional effort would not reach beyond the campus. This was the objective of the education and outreach effort. The combination of events, websites, and videos enabled us to reach many key decision makers and at the same time provide a long-term presence on the web that we can use to further educate people.     The objective of the visit to Hanford Site was to: demonstrate equipment, technology, and methods for calculating Pu production, measuring integrated reactor power, and storing and safeguarding PuO{sub 2}; demonstrate the shutdown of Hanford production reactors; and foster openness and transparency of Hanford operations. The first day`s visit was an introduction to Hanford and a review of the history of the reactors. The second day consisted of discussions on the production reactors, reprocessing operations, and PuO{sub 2} storage. The group divided on the third day to tour facilities. Group A toured the N reactor, K-West reactor, K-West Basins, B reactor, and participated in a demonstration and discussion of reactor modeling computer codes. Group B toured the Hanford Pu Storage Facility, 200-East Area, N-cell (oxide loadout station), the Automated Storage Facility, and the Nondestructive Assay Measurement System. Group discussions were held during the last day of the visit, which included scheduling of a US visit to Russia.

    Cell-cell communication in bacteria involves the production, release, and subsequent detection of chemical signaling molecules called autoinducers. This process, called quorum sensing, allows bacteria to regulate gene expression on a population-wide scale. Processes controlled by quorum sensing are usually ones that are unproductive when undertaken by an individual bacterium but become effective when undertaken by the group. For example, quorum sensing controls bioluminescence, secretion of virulence factors, biofilm formation, sporulation, and the exchange of DNA. Thus, quorum sensing is a mechanism that allows bacteria to function as multi-cellular organisms. Bacteria make, detect, and integrate information from multiple autoinducers, some of which are used exclusively for intra-species communication while others enable communication between species. Research is now focused on the development of therapies that interfere with quorum sensing to control bacterial virulence.

    A low-noise optical amplifier solves crosstalk problems in optical amplifiers by using an optical cavity oriented off-axis (e.g. perpendicular) to the direction of a signal amplified by the gain medium of the optical amplifier. Several devices are used to suppress parasitic lasing of these types of structures. The parasitic lasing causes the gain of these structures to be practically unusable. The lasing cavity is operated above threshold and the gain of the laser is clamped to overcome the losses of the cavity. Any increase in pumping causes the lasing power to increase. The clamping action of the gain greatly reduces crosstalk due to gain saturation for the amplified signal beam. It also reduces other nonlinearities associated with the gain medium such as four-wave mixing induced crosstalk. This clamping action can occur for a bandwidth defined by the speed of the laser cavity. The lasing field also reduces the response time of the gain medium. By having the lasing field off-axis, no special coatings are needed. Other advantages are that the lasing field is easily separated from the amplified signal and the carrier grating fluctuations induced by four-wave mixing are decreased. Two related methods reduce the amplified spontaneous emission power without sacrificing the gain of the optical amplifier.

    The Renewables and Energy Efficiency Planning (REEP) program was developed at the US Army Construction Engineering Research Laboratories (USACERL). This program allows for the analysis of 78 energy and water conservation opportunities at 239 major DOD installations. REEP uses a series of algorithms in conjunction with installation specific data to estimate the energy and water conservation potential for entire installations. The program provides the energy, financial, pollution, and social benefits of conservation initiatives. The open architecture of the program allows for simple modification of energy and water conservation variables, and installation database values to allow for individualized analysis. The program is essentially a high-level screening tool that can be used to help identify and focus preliminary conservation studies. The REEP program requires an IBM PC or compatible with a 80386 or 80486 microprocessor. It also requires approximately 4 megabytes of disk space and at least 8 megabytes of RAM. The system was developed for a Windows environment and requires Microsoft Windows 3.1{trademark} or higher to run properly.

    A data mining system uncovers patterns, associations, anomalies and other statistically significant structures in data. Data files are read and displayed. Objects in the data files are identified. Relevant features for the objects are extracted. Patterns among the objects are recognized based upon the features. Data from the Faint Images of the Radio Sky at Twenty Centimeters (FIRST) sky survey was used to search for bent doubles. This test was conducted on data from the Very Large Array in New Mexico which seeks to locate a special type of quasar (radio-emitting stellar object) called bent doubles. The FIRST survey has generated more than 32,000 images of the sky to date. Each image is 7.1 megabytes, yielding more than 100 gigabytes of image data in the entire data set.

    Cosmological N-body simulations on parallel computers produce large datasets - about five hundred Megabytes at a single output time, or tens of Gigabytes over the course of a simulation. These large datasets require further analysis before they can be compared to astronomical observations. We have implemented two methods for performing halo finding, a key part of the knowledge discovery process, on parallel machines. One of these is a parallel implementation of the friends of friends (FOF) algorithm, widely used in the field of N-body cosmology. The new isodensity (ID) method has been developed to overcome some of the shortcomings of FOR Both have been implemented on a variety of computer systems, and successfully used to extract halos from simulations with up to 256{sup 3} (or about 16.8 million) particles, which axe among the largest N-body cosmology simulations in existence.

    Detailed results of the Hanford Environmental Dose Reconstruction project (HEDR) iodine-131 release reconstruction are presented in this volume. Included are daily data on B, D, and F Plant, reactor operations from the P-Department Daily Reports (General Electric Company 1947). Tables of B and T Plant material processed from the three principal sources on separations plant operations: The Jaech report (Jaech undated), the 200 Area Report (Acken and Bird 1945; Bird and Donihee 1945), and the Metal History Reports (General Electric Company 1946). A transcription of the Jaech report is also provided because it is computer-generated and is not readily readable in its original format. The iodine-131 release data are from the STRM model. Cut-by-cut release estimates are provided, along with daily, monthly, and yearly summations. These summations are based on the hourly release estimates. The hourly data are contained in a 28 megabyte electronic file. Interested individuals may request a copy.

  4. Walking the sustainability assessment talk - Progressing the practice of environmental impact assessment (EIA)

    Internationally there is a growing demand for environmental impact assessment (EIA) to move away from its traditional focus towards delivering more sustainable outcomes. South Africa is an example of a country where the EIA system seems to have embraced the concept of sustainability. In this paper we test the existing objectives for EIA in South Africa against sustainability principles and then critique the effectiveness of EIA practice in delivering these objectives. The outcome of the research suggests that notwithstanding a strong and explicit sustainability mandate through policy and legislation, the effectiveness of EIA practice falls far short of what is mandated. This shows that further legislative reform is not required to improve effectiveness but rather a focus on changing the behaviour of individual professionals. We conclude by inviting further debate on what exactly practitioners can do to give effect to sustainability in EIA practice.

    To fully evaluate energy and emission impacts of advanced vehicle technologies and new transportation fuels, the fuel cycle from wells to wheels and the vehicle cycle through material recovery and vehicle disposal need to be considered. Sponsored by the U.S. Department of Energy's Office of Energy Efficiency and Renewable Energy (EERE), Argonne has developed a full life-cycle model called GREET (Greenhouse gases, Regulated Emissions, and Energy use in Transportation). It allows researchers and analysts to evaluate various vehicle and fuel combinations on a full fuel-cycle/vehicle-cycle basis. The first version of GREET was released in 1996. Since then, Argonne has continued to update and expand the model. The most recent GREET versions are the GREET 1 2012 version for fuel-cycle analysis and GREET 2.7 version for vehicle-cycle analysis.

    Plug loads represent 30percent of total electricity use in residential buildings. Significant energy savings would result from an accurate understanding of which miscellaneous electric devices are using energy, at what time, and in what quantity. Commercially available plug load monitoring and control solutions replace or limit the attached device's native controls - forcing the user to adapt to a separate set of controls associated with the monitoring and control hardware. A better solution is integration of these capabilities at the power supply level. In this paper, we demonstrate a method achieving this integration. Our solution allows unobtrusive power monitoring and control while retaining native device control features. Further, our prototype enables intelligent behaviors by allowing devices to respond to the state of one another automatically. The CPS enables energy savings while demonstrating an added level of functionality to the user. If CPS technology became widespread in devices, a combination of automated and human interactive solutions would enable high levels of energy savings in buildings.

    A powerful method to aid in understanding the performance of parallel applications uses log or trace files containing time-stamped events and states (pairs of events). These trace files can be very large, often hundreds or even thousands of megabytes. Because of the cost of accessing and displaying such files, other methods are often used that reduce the size of the tracefiles at the cost of sacrificing detail or other information. This paper describes a hierarchical trace file format that provides for display of an arbitrary time window in a time independent of the total size of the file andmore » roughly proportional to the number of events within the time window. This format eliminates the need to sacrifice data to achieve a smaller trace file size (since storage is inexpensive, it is necessary only to make efficient use of bandwidth to that storage). The format can be used to organize a trace file or to create a separate file of annotations that may be used with conventional trace files. We present an analysis of the time to access all of the events relevant to an interval of time and we describe experiments demonstrating the performance of this file format.« less

    Highlights: The function of the mitochondria fatty acid synthesis pathway is partially unknown. Overexpression of the pathway causes transcriptional activation through PPARs. Knock down of the pathway attenuates that activation. The last enzyme in the pathway regulates its own transcription. Products of the mtFASII pathway are able to drive nuclear transcription. -- Abstract: Mammalian cells contain two fatty acid synthesis pathways, the cytosolic FASI pathway, and the mitochondrial FASII pathway. The selection behind the conservation of the mitochondrial pathway is not completely understood, given the presence of the cytosolic FAS pathway. In this study, we show through heterologous gene reporter systems and PCR-based arrays that overexpression of MECR, the last step in the mtFASII pathway, causes modulation of gene expression through the PPAR pathway. Electromobility shift assays (EMSAs) demonstrate that overexpression of MECR causes increased binding of PPARs to DNA, while cell fractionation and imaging studies show that MECR remains localized to the mitochondria. Interestingly, knock down of the mtFASII pathway lessens the effect of MECR on this transcriptional modulation. Our data are most consistent with MECR-mediated transcriptional activation through products of the mtFASII pathway, although we cannot rule out MECR acting as a coactivator. Further investigation into the physiological relevance of this communication will be necessary to better understand some of the phenotypic consequences of deficits in this pathway observed in animal models and human disease.

  5. A New On-the-Fly Sampling Method for Incoherent Inelastic Thermal Neutron Scattering Data in MCNP6

    At thermal energies, the scattering of neutrons in a system is complicated by the comparable velocities of the neutron and target, resulting in competing upscattering and downscattering events. The neutron wavelength is also similar in size to the target's interatomic spacing making the scattering process a quantum mechanical problem. Because of the complicated nature of scattering at low energies, the thermal data files in ACE format used in continuous-energy Monte Carlo codes are quite large { on the order of megabytes for a single temperature and material. In this paper, a new storage and sampling method is introduced that is orders of magnitude less in size and is used to sample scattering parameters at any temperature on-the-fly. In addition to the reduction in storage, the need to pre-generate thermal scattering data tables at fine temperatures has been eliminated. This is advantageous for multiphysics simulations which may involve temperatures not known in advance. A new module was written for MCNP6 that bypasses the current S(?,?) table lookup in favor of the new format. The new on-the-fly sampling method was tested for graphite for two benchmark problems at ten temperatures: 1) an eigenvalue test with a fuel compact of uranium oxycarbide fuel homogenized into a graphite matrix, 2) a surface current test with a \\broomstick" problem with a monoenergetic point source. The largest eigenvalue difference was 152pcm for T= 1200K. For the temperatures and incident energies chosen for the broomstick problem, the secondary neutron spectrum showed good agreement with the traditional S(?,?) sampling method. These preliminary results show that sampling thermal scattering data on-the-fly is a viable option to eliminate both the storage burden of keeping thermal data at discrete temperatures and the need to know temperatures before simulation runtime.

  6. Energy use and domestic hot water consumption - Phase 1. Final report

    New York State Energy Research & Development Authority has sponsored a project to develop comprehensive operating data on combined domestic hot water (DHW) and heating systems to be used in system design and specifications. The new, more precise DHW flow data (broken down on a per capita basis) result in a better foundation for sizing decisions. Thirty New York City multifamily buildings with combined steam heating and DHW plants were instrumented to monitor apartment, outdoor, boiler and DHW temperatures, and burner on-off times. In nine buildings, which had been upgraded, additional data collected were: stack temperature, DHW flow in 15-minute increments, oil and boiler make-up water flows, and DHW temperature before and after the mixing valve and on the return line. The data set collected for 14 months amounts to a data base of over 110 megabytes. This report presents DHW demand patterns, seasonal variations, weekday vs. weekend consumption, consumption vs. occupancy levels, coincidence of 15-, 60-, 120-, and 180- minute demand periods, and average vs. peak demand levels, as well as figures for energy to generate DHW. The format of guidelines presented in this report differs from those currently in use. The {open_quotes}Low - Medium - High{close_quotes} (LMH) users set of guidelines provide a range, rather than a specific, singular gallonage standard. By providing LMH tables and guidelines for their application the design engineer or contractor can then use these as a tool from which to select the appropriate level that matches the site being served. Values are presented for both New York sizing guidelines (developed from this study), as well as a set of {open_quote}National{close_quote} figures (derived from a compilation of studies conducted in other cities). The LMH approach and {open_quote}National{close_quote} guidelines were adopted for inclusion in the 1995 ASHRAE Handbook revision.

    The National Radon Database (NRDB) was developed by the United States Environmental Protection Agency (USEPA) to distribute information in two recent radon surveys: the EPA/State Residential Radon Surveys and the National Residential Radon Survey. The National Residential Radon Surveys collected annual average radon measurements on all levels of approximately 5,700 homes nationwide. Information collected during survey includes a detailed questionnaire on house characteristics, as well as radon measurements. The radon survey data for Volume 6 is contained on two diskettes. The data diskettes are accompanied by comprehensive documentation on the design and implementation of the survey, the development and use of sampling weights, a summary of survey results, and information concerning the household questionnaire.

    Inhibition of the epidermal growth factor (EGF)-receptor (EGFR) has become a promising anticancer treatment strategy. In addition, application of retinoids yields encouraging results for cancer prevention and therapy. Many tumors express no or low amounts of retinoic acid receptor-{beta}2 (RAR-{beta}2) due to epigenetic silencing via DNA hypermethylation. RAR-{beta}2 is the main mediator of the antiproliferative effect of retinoids. RAR-{beta}2 re-expression causes reversal of transformation, cell cycle arrest, and restoration of retinoid sensitivity. RAR-{beta}2 is thus a tumor suppressor. Western blotting, colorimetric in vitro cell proliferation assays, and reverse transcription-polymerase chain reaction demonstrated that the EGFR inhibitor PD153035 not only blocked activation of EGFR and inhibited cell growth, but also stimulated RAR-{beta} expression in MDA-MB-468 breast and OVCAR-3 ovarian carcinoma cells. Upregulation of RAR-{beta} by PD153035 was confirmed by real-time reverse transcription-polymerase chain reaction. In contrast, expression of other retinoid receptors and of estrogen receptor-{alpha} was not affected. PD153035-mediated re-induction of RAR-{beta} was associated with demethylation of the RAR-{beta}2 gene promoter P2 as demonstrated by methylation-specific polymerase chain reaction. These novel results on the ErbB/retinoid receptor cross-talk may be useful for designing future anticancer combination regimens.

    In July of 2012, scientists leading two different research teams, working independently of each other, announced that they had almost certain proof of the long-sought Higgs boson. Though Cern did not call the discovery "official", many physicists conceded the evidence was now so compelling they had surely found the missing particle. The formal confirmation will come over the next few months of further investigation. The experiments are taking place at the Large Hadron Collider (LHC), and this third lecture in the DOE Science Speaker Series is given by one of those announcing scientists in July. He is Dr. Joseph Incandela, the current spokesperson for the Compact Muon Solenoid (CMS) Experiment at CERN. He was heavily involved in the search for the top quark at Fermi and is from the University of California, Santa Barbara. The title he gives his presentation is "Searching for the genetic code of our universe: Discovery at the LHC."

  1. A New Approach for Fingerprint Image Compression

    The FBI has been collecting fingerprint cards since 1924 and now has over 200 million of them. Digitized with 8 bits of grayscale resolution at 500 dots per inch, it means 2000 terabytes of information. Also, without any compression, transmitting a 10 Mb card over a 9600 baud connection will need 3 hours. Hence we need a compression and a compression as close to lossless as possible: all fingerprint details must be kept. A lossless compression usually do not give a better compression ratio than 2:1, which is not sufficient. Compressing these images with the JPEG standard leads to artefacts which appear even at low compression rates. Therefore the FBI has chosen in 1993 a scheme of compression based on a wavelet transform, followed by a scalar quantization and an entropy coding : the so-called WSQ. This scheme allows to achieve compression ratios of 20:1 without any perceptible loss of quality. The publication of the FBI specifies a decoder, which means that many parameters can be changed in the encoding process: the type of analysis/reconstruction filters, the way the bit allocation is made, the number of Huffman tables used for the entropy coding. The first encoder used 9/7 filters for the wavelet transform and did the bit allocation using a high-rate bit assumption. Since the transform is made into 64 subbands, quite a lot of bands receive only a few bits even at an archival quality compression rate of 0.75 bit/pixel. Thus, after a brief overview of the standard, we will discuss a new approach for the bit-allocation that seems to make more sense where theory is concerned. Then we will talk about some implementation aspects, particularly for the new entropy coder and the features that allow other applications than fingerprint image compression. Finally, we will compare the performances of the new encoder to those of the first encoder.

    The role of tumor stroma in regulation of breast cancer growth has been widely studied. However, the details on the type of heterocellular cross-talk between stromal and breast cancer cells (BCCs) are still poorly known. In the present study, in order to investigate the intercellular communication between human mesenchymal stromal cells (hMSCs) and breast cancer cells (BCCs, MDA-MB-231), we recruited cell-internalizing quantum dots (i-QD) generated by conjugation of cell-internalizing anti-mortalin antibody and quantum dots (QD). Co-culture of illuminated and color-coded hMSCs (QD655) and BCCs (QD585) revealed the intercellular transfer of QD655 signal from hMSCs to BCCs. The amount of QD double positive BCCs increased gradually within 48 h of co-culture. We found prominent intercellular transfer of QD655 in hanging drop co-culture system and it was non-existent when hMSCs and BBCs cells were co-cultured in trans-well system lacking imminent cellcell contact. Fluorescent and electron microscope analyses also supported that the direct cell-to-cell interactions may be required for the intercellular transfer of QD655 from hMSCs to BCCs. To the best of our knowledge, the study provides a first demonstration of transcellular crosstalk between stromal cells and BCCs that involve direct contact and may also include a transfer of mortalin, an anti-apoptotic and growth-promoting factor enriched in cancer cells.

One of them is noninvasive anticancer therapy photodynamic therapy (PDT), where singlet oxygen (SO), generated under the irradiation of light with appropriate wavelengths, kills cancer cells. In this study, authors report graphene oxide (GO) noncovalent functionalization with methylene blue (MB). MB molecules underwent adsorption on the surface of GO. Detailed characterization of the obtained material was carried out with UVvis spectroscopy, Raman spectroscopy, FT-IR spectroscopy, and confocal laser scanning microscopy. Furthermore, its performance in singlet oxygen generation (SOG) under irradiation of laser with excitation wavelengths of 785 nm was investigated. Interestingly, GO functionalized with MB (MBGO) showed enhanced efficiency in singlet oxygen generation compared to pristine MB. The efficiency in SOG was detected by photobleaching of 9,10-anthracenediyl-bis(methylene)dimalonic acid (ABMDMA). These results indicate the material is promising in PDT anticancer therapy and further in vitro and in vivo studies are required.

    The ability to develop type II collagen (CII)-induced arthritis (CIA) in mice is associated with the major histocompatibility I-A gene and with as yet poorly defined regulatory molecules of the major histocompatibility complex (MHC) class II antigen processing and presentation pathway. H2-M molecules are thought to be involved in the loading of antigenic peptides into the MHC class II binding cleft. We sequenced H2-Ma, H2-Mb1, and H2-Mb2 genes from CIA-susceptible and -resistant mouse strains and identified four different Ma and Mb2 alleles, and three different Mb1 alleles defined by polymorphic residues within the predicted peptide binding groove. Most CIA-resistant mouse strains share common Ma, Mb1, and Mb2 alleles. In contrast, H2-M alleles designated Ma-III, Ma-IV, Mb1-III, and Mb2-IV could be exclusively identified in the CIA-susceptible H2{sup r} and H2{sup q} haplotypes, suggesting that allelic H2-M molecules may modulate the composition of different CII peptides loaded onto MHC class II molecules, presumably presenting {open_quotes}arthritogenic{close_quotes} epitopes to T lymphocytes. 42 refs., 4 figs., 3 tabs.

    This document is one of two reports on the Integrated ISIS protocol. Required by the IAB/IESG in order for an Internet routing protocol to advance to Draft Standard Status. Integrated ISIS is an Interior Gateway Protocol and is designed to carry both IP and ISO CLNP routing information. Integrated ISIS is currently designated as a Proposed Standard. The protocol was first published in RFC 1195. Internet Draft was published subsequently to RFC 1195 and documents the current version of the protocol. This report documents experience with Integrated ISIS. This includes reports on interoperability testing, field experience and the current state of Integrated ISIS implementations. It also presents a summary of the Integrated ISIS Management Information Base (MIB), and a summary of the Integrated ISIS authentication mechanism.

  1. The interactions of azure B, a metabolite of methylene blue, with acetylcholinesterase and butyrylcholinesterase

    Methylene blue (MB) is reported to possess diverse pharmacological actions and is attracting increasing attention for the treatment of neurodegenerative disorders such as Alzheimer's disease. Among the pharmacological actions of MB, is the significant inhibition of acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). These activities may, at least in part, underlie MB's beneficial effects in Alzheimer's disease. MB is metabolized to yield N-demethylated products of which azure B, the monodemethyl metabolite, is the predominant species. Azure B has been shown to be pharmacologically active and also possesses a variety of biological actions. Azure B therefore may contribute to the pharmacological profile of MB. Based on these considerations, the present study investigates the possibility that azure B may, similar to MB, act as an inhibitor of human AChE and BuChE. The results document that azure B inhibits AChE and BuChE with IC{sub 50} values of 0.486 ?M and 1.99 ?M, respectively. The results further show that azure B inhibits AChE and BuChE reversibly, and that the modes of inhibition are most likely competitive. Although the AChE and BuChE inhibitory activities of azure B are twofold and fivefold, respectively, less potent than those recorded for MB [IC{sub 50}(AChE) = 0.214 ?M; IC{sub 50}(BuChE) = 0.389 ?M] under identical conditions, azure B may be a contributor to MB's in vivo activation of the cholinergic system and beneficial effects in Alzheimer's disease. - Highlights: Methylene blue (MB) is a known inhibitor of AChE and BuChE. Azure B, the major metabolite of MB, also is an inhibitor of AChE and BuChE. Azure B may be a contributor to MB's in vivo activation of the cholinergic system. Azure B may contribute to MB's potential in Alzheimer's disease therapy.
