    In this April 21, 2008 Berkeley Lab event, a dynamic panel of Berkeley Lab scientists highlight breast cancer research advances related to susceptibility, early detection, prevention, and therapy - a biological systems approach to tackling the disease from the molecular and cellular levels, to tissues and organs, and ultimately the whole individual. Joe Gray, Berkeley Lab Life Sciences Division Director, explores how chromosomal abnormalities contribute to cancer and respond to gene-targeted therapies. Mina Bissell, former Life Sciences Division Director, approaches the challenge of breast cancer from the breast's three dimensional tissue microenvironment and how the intracellular ''conversation'' triggers malignancies. Mary Helen Barcellos-Hoff, Deputy Director, Life Sciences Division, identifies what exposure to ionizing radiation can tell us about how normal tissues suppress carcinogenesis. The panel is moderated by Susan M. Love, breast cancer research pioneer, author, President and Medical Director of the Dr. Susan Love Research Foundation.

    This past year we have made substantial progress in modeling the contribution of homeostatic regulation to low-dose radiation effects and carcinogenesis. We have worked to refine and apply our multistage carcinogenesis models to explicitly incorporate cell cycle states, simple and complex damage, checkpoint delay, slow and fast repair, differentiation, and apoptosis to study the effects of low-dose ionizing radiation in mouse intestinal crypts, as well as in other tissues. We have one paper accepted for publication in ''Advances in Space Research'', and another manuscript in preparation describing this work. I also wrote a chapter describing our combined cell-cycle and multistage carcinogenesis model that will be published in a book on stochastic carcinogenesis models edited by Wei-Yuan Tan. In addition, we organized and held a workshop on ''Biologically Based Modeling of Human Health Effects of Low dose Ionizing Radiation'', July 28-29, 2005 at Fred Hutchinson Cancer Research Center in Seattle, Washington. We had over 20 participants, including Mary Helen Barcellos-Hoff as keynote speaker, talks by most of the low-dose modelers in the DOE low-dose program, experimentalists including Les Redpath (and Mary Helen), Noelle Metting from DOE, and Tony Brooks. It appears that homeostatic regulation may be central to understanding low-dose radiation phenomena. The primary effects of ionizing radiation (IR) are cell killing, delayed cell cycling, and induction of mutations. However, homeostatic regulation causes cells that are killed or damaged by IR to eventually be replaced. Cells with an initiating mutation may have a replacement advantage, leading to clonal expansion of these initiated cells. Thus we have focused particularly on modeling effects that disturb homeostatic regulation as early steps in the carcinogenic process. There are two primary considerations that support our focus on homeostatic regulation. First, a number of epidemiologic studies using multistage carcinogenesis models that incorporate the ''initiation, promotion, and malignant conversion'' paradigm of carcinogenesis are indicating that promotion of initiated cells is the most important cellular mechanism driving the shape of the age specific hazard for many types of cancer. Second, we have realized that many of the genes that are modified in early stages of the carcinogenic process contribute to one or more of four general cellular pathways that confer a promotional advantage to cells when these pathways are disrupted.

    The 22 slides in this presentation treat the subject under the following headings: MaRIE XFEL Performance Parameters, Input Electron Beam Parameters, Undulator Design, Genesis Simulations, Risks, and Summary It is concluded that time-dependent Genesis simulations show the MaRIE XFEL can deliver the number of photons within the required bandwidth, provided a number of assumptions are met; the highest risks are associated with the electron beam driving the XFEL undulator; and risks associated with the undulator and/or distributed seeding technique may be evaluated or retired by performing early validation experiments.

    Ash from the massive 18 May 1980 eruption of Mount St. Helens readily gave off large amounts of carbonyl sulfide and carbon disulfide gases at room temperature. These findings suggest that the sulfur that enhances the Junge sulfate layer in the stratosphere after volcanic eruptions could be carried directly to the upper atmosphere as carbonyl sulfide and carbon disulfide adsorbed on ash particles from major volcanic eruptions.

Site Description - The Marys River Fir site is part of the "Synthesis of Remote Sensing and Field Observations to Model and Understand Disturbance and Climate Effects on the Carbon Balance of Oregon and Northern California (ORCA)". Located in the western region of Oregon the Marys River site represents the western extent of the climate gradient that spans eastward into the semi-arid basin of central Oregon. The sites that make up the eastern extent of the ORCA climate gradient is the Metolius site network (US-Me1, US-ME2, US-ME4, US-Me5) all of which are part of the TERRA PNW project at Oregon State University.

    A network of twelve monitoring sites around Mount St. Helens was established to evaluate possible short-lived radioactivity in the fallen ash. Seven sites were located near major population centers of Washington and Oregon, and five sites were located within 80 km of the volcano. Each site monitored the radioactivity present by the use of thermoluminescent dosimeters which recorded the total exposure to radioactivity over the exposure period. Eruptions occurring on July 22, August 7, and October 16 to 18, 1980 were monitored. No statistically significant quantities of measurable radon daughters were observed.

    Analytical and numerical solutions to the hydrodynamic equations of motion, constrained by physical properties of juvenile ejecta in the Mount St. Helens blast deposit, were used to investigate magmatic conditions required to produce the initial devastating blast phase of the eruption of May 18, 1980. Evidence that the blast was magmatic includes equivalence in volume of juvenile blast ejecta to preeruption inflation of the cone, substantial vesicularity of this ejecta, and continued vesiculation of large juvenile clasts after eruption. Observed or inferred ejecta velocities of 100 to 250 m/s are shown to require 0.2 to 0.7 wt% water vapor preexisting in magma unloaded by a landslide 200 to 900 m thick. These conditions imply total magmatic water contents of 0.7 to 1.7 wt%, respectively. Such low required water content suggests that volcanic blasts may be regarded as a normal consequence of magma intrusion into an unstable edifice.

    This report describes the 4-yr follow-up of 712 loggers exposed over an extended period to varying levels of fresh volcanic ash from the 1980 eruptions of Mt. St. Helens. Concerns related to the irritant effect the ash might have on the airways and also to its fibrogenic potential if exposures were intense and continued over many years. Our subjects were divided into 3 groups: high, low, and no exposure. Baseline testing was begun in June 1980, 1 month after the major eruption, and follow-up testing continued on an annual basis through 1984; 88% of the loggers have been tested at least 3 times. Analysis of lung function data showed that a significant, exposure-related decline in FEV1 occurred during the first year after the eruption. The decline was short-lived, however, and by 1984 the differences between exposure groups were no longer significant. Self-reported symptoms of cough, phlegm, and wheeze showed a similar pattern. No ash-related changes were seen in chest roentgenograms taken in 1980 and in 1984. Our findings are consistent with the hypothesis that the inhaled ash caused mucus hypersecretion and/or airway inflammation that reversed when the exposure levels decreased. The ash levels to which the loggers were exposed were low compared with permissible occupational levels for nuisance dusts, but generally higher than the total suspended particulate levels permissible in ambient air.

    The purpose of the Matter-Radiation Interactions in Extremes (MaRIE) facility at Los Alamos National Laboratory is to investigate the performance limits of materials in extreme environments. The MaRIE facility will utilize a 12 GeV linac to drive an X-ray Free-Electron Laser (FEL). Most of the same linac will also be used to perform electron radiography. The main linac is driven by two shorter linacs; one short linac optimized for X-FEL pulses and one for electron radiography. The RF systems have historically been the one of the largest single component costs of a linac. We will describe the details of the different types of RF systems required by each part of the linacs. Starting with the High Power RF system, we will present our methodology for the choice of RF system peak power and pulselength with respect to klystron parameters, modulator parameters, performance requirements and relative costs. We will also present an overview of the Low Level RF systems that are proposed for MaRIE and briefly describe their use with some proposed control schemes.

    Western Area Power Administration (Western) and the Platte River Power Authority (Platte River) propose to upgrade portions of the existing electric transmission and substation system that serves the Town of Estes Park, Colorado. The existing transmission lines between the Estes Power Plant Switchyard and the Marys Lake Substation include a 115,000 volt (115-kV) line and 69,000 volt (69-kV) line. Approximately one mile is a double-circuit 115/69-kV line on steel lattice structures, and approximately two miles consists of separate single-circuit 115-kV and a 69-kV lines, constructed on wood H-Frame structures. Both lines were constructed in 1951 by the US Bureau of Reclamation. The existing transmission lines are on rights-of-way (ROW) that vary from 75 feet to 120 feet and are owned by Western. There are 48 landowners adjacent to the existing ROW. All of the houses were built adjacent to the existing ROW after the transmission lines were constructed. Upgrading the existing 69-kV transmission line between the Marys Lake Substation and the Estes Power Plant Switchyard to 115-kV and expanding the Marys Lake Substation was identified as the most effective way in which to improve electric service to Estes Park. The primary purpose and need of the proposed project is to improve the reliability of electric service to the Town of Estes Park. Lack of reliability has been a historical concern, and reliability will always be less than desired until physical improvements are made to the electrical facilities serving Estes Park.

    St. Mary's Hospital, Pierre, South Dakota, with the assistance of the US Department of Energy, drilled a 2176 ft well into the Madison Aquifer ot secure 108/sup 0/F artesian flow water at 385 gpm (475 psig shut-in pressure). The objective was to provide heat for domestic hot water and to space heat 163,768 sq. ft. Cost savings for the first three years were significant and, with the exception of a shutdown to replace some corroded pipe, the system has operated reliably and continuously for the last four years.

    Environmental and breathing-zone samples were analyzed for polynuclear aromatic hydrocarbons (PAH), total particulates, and respirable free silica at the Airco Company (SIC-3624), Saint Marys, Pennsylvania in January, 1984. The evaluation was requested confidentially because of concern over exposures to soot, coal tar pitch volatiles, and sand in the car bottom and sagger bake operations. Forty-three employees were interviewed. Two of 19 total particulate samples exceeded the OSHA standard of 15 milligrams per cubic meter (mg/m3), 17.3 and 32.7 mg/m3. Benzene soluble fractions ranged from 0.5 to 5.0 mg/m3. The OSHA standard for benzene soluble fractions is 0.2 mg/m3. Two of seven samples of silica were above the limit of detection, 0.09 and 0.06 mg/m3. In bulk samples, the benzene soluble fractions ranged from 0.44 to 860 mg/gram and the PAH content from 0 to 26,124 micrograms per gram. Employees working in the bake areas reported a significant excess incidence of symptoms such as skin, nose and eye irritation, cough, sore or dry throat, chest tightness, and breathing difficulty. The authors conclude that a health hazard exists at the facility. Recommendations include enclosing vehicles used in moving electrodes, cleaning up spilled dust, and controlling fumes emitted from the sagger kilns.

    Long-period (100 to 260 s) Love and Rayleigh waves excited by the eruption of Mount St. Helens on May 18, 1980, and recorded by ID, SRO, and ASRO stations were analyzed to determine the mechanism of the eruption. The amplitude radiation patterns of both Rayleigh and Love waves are two lobed with nodal direction in E5/sup 0/S for Rayleigh waves and in N5/sup 0/E for Love waves. These radiation patterns preclude any double-couple mechanism. The radiation pattern, the initial phase, the relatively large amplitude ratio of Love to Rayleigh waves and the existence of clear nodes in the radiation patterns of fundamental mode and higher-mode Rayleigh waves suggest that the source is represented by an almost horizontal (less than 15/sup 0/ from the horizontal) single force pointed toward S5/sup 0/W. The surface wave spectra fall off very rapidly at periods shorter than 75 s suggesting a very slow source process. Although the details of the source time history could not be determined, a smooth bell-shaped time function: f/sub 0/s(t) = (1/2)f/sub 0/(1-cos( (t)/(tau) ..pi..)) for 0< or =t< or =2tau and f/sub 0/s(t) = 0 for t> or =2tau, with tau = 75 s is considered appropriate on the basis of comparison between synthetic and observed seismograms and of the shape of the source spectrum. The peak value of the force f/sub 0/ is about 10/sup 18/ dynes. The tailing end of the source time function could not be resolved, and some overshoot may be added. The magnitude and the time history of the force can be explained by a northward landslide followed by a lateral blast observed at the time of the eruption. Two distinct events about 110 apart can be identified on body wave and short-period surface wave records. The first event may correspond to the earthquake which triggered the landslide and the lateral blast. The second event appears to correspond to a second large earthquake and explosion which took place about 2 minutes after the first earthquake.

    This project uses numerical techniques to assess the structural integrity and capacity of the bridge foundations and, as a result, reduces the risk associated with reusing the same foundation for a new superstructure. Nondestructive test methods of different types were used in combination with the numerical modeling and analysis. The onsite tests included visual inspection, tomography, ground penetrating radar, drilling boreholes and coreholes, and the laboratory tests on recovered samples. The results were utilized to identify the current geometry of the structure with foundation, including the hidden geometry of the abutments and piers, and soil and foundation material properties. This data was used to build the numerical models and run computational analyses on a high performance computer cluster to assess the structural integrity of the bridge and foundations including the suitability of the foundation for reuse with a new superstructure and traffic that will increase the load on the foundations. Computational analysis is more cost-effective and gives an advantage of getting more detailed knowledge about the structural response. It also enables to go beyond non-destructive testing and find the failure conditions without destroying the structure under consideration.
