    We report on the discovery, isolation, and use of a novel yellow fluorescent protein. Lucigen Yellow (LucY) binds one FAD molecule within its core, thus shielding it from water and maintaining its structure so that fluorescence is 10-fold higher than freely soluble FAD. LucY displays excitation and emission spectra characteristic of FAD, with 3 excitation peaks at 276nm, 377nm, and 460nm and a single emission peak at 530nm. These excitation and emission maxima provide the large Stokes shift beneficial to fluorescence experimentation. LucY belongs to the MurB family of UDP-N-acetylenolpyruvylglucosamine reductases. The high resolution crystal structure shows that in contrastmore » to other structurally resolved MurB enzymes, LucY does not contain a potentially quenching aromatic residue near the FAD isoalloxazine ring, which may explain its increased fluorescence over related proteins. Using E. coli as a system in which to develop LucY as a reporter, we show that it is amenable to circular permutation and use as a reporter of protein-protein interaction. Fragmentation between its distinct domains renders LucY non-fluorescent, but fluorescence can be partially restored by fusion of the fragments to interacting protein domains. Thus, LucY may find application in Protein-fragment Complementation Assays for evaluating protein-protein interactions.« less

    LUCI, the Laboratory for Underground CO{sub 2} Investigations, is an experimental facility being planned for the DUSEL underground laboratory in South Dakota, USA. It is designed to study vertical flow of CO{sub 2} in porous media over length scales representative of leakage scenarios in geologic carbon sequestration. The plan for LUCI is a set of three vertical column pressure vessels, each of which is {approx}500 m long and {approx}1 m in diameter. The vessels will be filled with brine and sand or sedimentary rock. Each vessel will have an inner column to simulate a well for deployment of down-hole logging tools. The experiments are configured to simulate CO{sub 2} leakage by releasing CO{sub 2} into the bottoms of the columns. The scale of the LUCI facility will permit measurements to study CO{sub 2} flow over pressure and temperature variations that span supercritical to subcritical gas conditions. It will enable observation or inference of a variety of relevant processes such as buoyancy-driven flow in porous media, Joule-Thomson cooling, thermal exchange, viscous fingering, residual trapping, and CO{sub 2} dissolution. Experiments are also planned for reactive flow of CO{sub 2} and acidified brines in caprock sediments and well cements, and for CO{sub 2}-enhanced methanogenesis in organic-rich shales. A comprehensive suite of geophysical logging instruments will be deployed to monitor experimental conditions as well as provide data to quantify vertical resolution of sensor technologies. The experimental observations from LUCI will generate fundamental new understanding of the processes governing CO{sub 2} trapping and vertical migration, and will provide valuable data to calibrate and validate large-scale model simulations.

    Number 13 Blast Furnace at Gary began injecting Pulverized Coal in March 1993. The injection level was increased over the next nine months until a level off 409 lbs/THM was achieved for the month of December 1993. Several major areas were critical in achieving this high level of Pulverized coal injection (PCI) including furnace conditions, lance position, tuyere blockage, operating philosophy, and outages. The paper discusses the modifications made to achieve this level of injection. This injection level decreased charged dry coke rate from 750 lbs/THM to about 625 lbs/THM, while eliminating 150 lbs/THM of oil and 20 lbs/THM of natural gas. Assuming a 1.3 replacement ratio for an oil/natural gas mixture, overall coke replacement for the coal is about 0.87 lbs coke/lbs coal. Gary Works anticipates levels of 500 lbs/THM are conceivable.

    No. 13 Blast Furnace at US Steel`s Gary Works is a 35 tuyere furnace with a 36.5 ft. hearth capable of producing over 9,000 tons of hot metal per day. The current casthouse design was placed in service following the second reline in the fall of 1979. This design anticipated daily production rates averaging 7,500 tons of hot metal per day and provided for removable troughs at two of the three tapholes. At the time, the troughs were rammed with a high alumina/silicon carbide granular ramming material that provided the operators with trough campaign lives between 60,000--70,000 tons of hot metal produced. As refractory technology progressed, low cement/low moisture castables were introduced to the trough systems on No. 13 Blast Furnace. The immediate success of the castables was tempered by emergence of a new unexpected problem. That problem was the thermal expansion of the castable. The paper describes the problems that resulted in the need to modify the trough design, the new design of the trough, and its improvement in iron trough campaign life and reliability.

    A new spectral restoration algorithm of reflection electron energy loss spectra is proposed. It is based on the maximum likelihood principle as implemented in the iterative Lucy-Richardson approach. Resolution is enhanced and point spread function recovered in a semi-blind way by forcing cyclically the zero loss to converge towards a Dirac peak. Synthetic phonon spectra of TiO{sub 2} are used as a test bed to discuss resolution enhancement, convergence benefit, stability towards noise, and apparatus function recovery. Attention is focused on the interplay between spectral restoration and quasi-elastic broadening due to free carriers. A resolution enhancement by a factor up to 6 on the elastic peak width can be obtained on experimental spectra of TiO{sub 2}(110) and helps revealing mixed phonon/plasmon excitations.

    Coal injection was initiated on No. 13 Blast Furnace in 1993 with 400 lb/THM achieved in 9 months. In early 1994, cold weather and coal preparation upsets led to the use of a second injectant, oil atomized by natural gas, to supplement the coal. Various combinations of coal and oil were investigated as total injection was increased to 450 lb/THM. Beginning in the last half of 1994, a continuing effort has been made to increase furnace production while maintaining this high co-injection level. Typical furnace production is now in excess of 10,000 THM/day compared with about 8500 THM/day in late 1993.

    Nuclear power facilities, both commercial and government operated, generate material called Dry Active Waste (DAW). DAW is a by-product of maintenance and operation of the power systems which contain radioactive materials. DAW can be any material contaminated with radioactive particles as long as it is not a fluid, typically: paper, cardboard, wood, plastics, cloth, and any other solid which is contaminated and determined to be dry. DAW is generated when any material is exposed to loose radioactive particles and subsequently becomes contaminated. In the United States, once a material is contaminated it must be treated as radioactive waste and disposed of in accordance with the requirements of Title 10 of the Code of Federal Regulations. Problems facing all commercial and non-commercial nuclear facilities are escalating costs of processing DAW and volumetric reduction of the DAW generated. Currently, approximately 85% of all DAW generated at a typical facility is comprised of anti-contamination clothing and protective barrier materials. Facilities that generate low-level radioactive waste need to dramatically reduce their waste volumes. This curtailment is required for several reasons: the number of radioactive waste repositories now accepting new waste is limited; the current cost of burial at an operating dump site is significant. Costs can be as high as $4,000 for a single 55 gallon drum; the cost of burial is constantly increasing; onsite storage of low-level radioactive waste is costly and results in a burial fee at plant decommissioning.

    We report a combined photoelectron velocity-map imaging spectroscopy and density functional theory investigation on lutetium monoxide anion. Transition between the X {sup 1}?{sup +} anion electronic ground state and the neutral X {sup 2}?{sup +} electronic ground state is observed. Vibrationally resolved spectra were obtained at four different photon energies, providing a wealth of spectroscopic information for the electronic ground states of the anionic lutetium monoxide and corresponding neutral species. Franck-Condon simulations of the ground-state transition are performed to assign vibrational structure in the spectra and to assist in identifying the observed spectral bands. The electronic ground state of LuO{sup ?} is found to have a vibrational frequency of 743 10 cm{sup ?1} and an equilibrium bond length of 1.841 . The electron affinity of LuO is measured to be 1.624 0.002 eV. The fundamental frequency of ground-state LuO is estimated to be 839 10 cm{sup ?1}.
