    2010-10-22

Research highlights: {yields} Strong ADAM15 expression is found in normal melanocytes. {yields} ADAM15 expression is significantly downregulated in patients with melanoma metastasis. {yields} TGF-{beta} can downregulate ADAM15 expression in melanoma cells. {yields} Overexpression of ADAM15 in melanoma cells inhibits migration, proliferation and invasion of melanoma cells. {yields} Conclusion: ADAM15 represents an tumor suppressor protein in melanoma. -- Abstract: In a mouse melanoma metastasis model it has been recently shown that ADAM15 overexpression in melanoma cells significantly reduced the number of metastatic nodules on the lung. Unfortunately, the expression of ADAM15 in human melanoma tissue has not been determined so far. In our study, we characterized the expression of ADAM15 in tissue micro-arrays of patients with primary melanoma with melanoma metastasis. ADAM15 was expressed in melanocytes and endothelial cells of benign nevi and melanoma tissue. Importantly, ADAM15 was significantly downregulated in melanoma metastasis compared to primary melanoma. We further demonstrate that IFN-{gamma} and TGF-{beta} downregulate ADAM15 protein levels in melanoma cells. To investigate the role of ADAM15 in melanoma progression, we overexpressed ADAM15 in melanoma cells. Importantly, overexpression of ADAM15 in melanoma cells reduced the migration, invasion and the anchorage dependent and independent cell growth of melanoma cells. In summary, the downregulation of ADAM15 plays an important role in melanoma progression and ADAM15 act as a tumorsuppressor in melanoma.

  16. MiR-153 inhibits migration and invasion of human non-small-cell lung cancer by targeting ADAM19

    SciTech Connect (OSTI)

Shan, Nianxi; Shen, Liangfang; Wang, Jun; He, Dan; Duan, Chaojun

    2015-01-02

Highlights: • Decreased miR-153 and up-regulated ADAM19 are correlated with NSCLC pathology. • MiR-153 inhibits the proliferation and migration and invasion of NSCLC cells in vitro. • ADAM19 is a direct target of miR-153. • ADAM19 is involved in miR-153-suppressed migration and invasion of NSCLC cells. - Abstract: MiR-153 was reported to be dysregulated in some human cancers. However, the function and mechanism of miR-153 in lung cancer cells remains unknown. In this study, we investigated the role of miR-153 in human non-small-cell lung cancer (NSCLC). Using qRT-PCR, we demonstrated that miR-153 was significantly decreased in clinical NSCLC tissues and cell lines, and downregulation of miR-153 was significantly correlated with lymph node status. We further found that ectopic expression of miR-153 significantly inhibited the proliferation and migration and invasion of NSCLC cells in vitro, suggesting that miR-153 may be a novel tumor suppressor in NSCLC. Further integrated analysis revealed that ADAM19 is as a direct and functional target of miR-153. Luciferase reporter assay demonstrated that miR-153 directly targeted 3′UTR of ADAM19, and correlation analysis revealed an inverse correlation between miR-153 and ADAM19 mRNA levels in clinical NSCLC tissues. Knockdown of ADAM19 inhibited migration and invasion of NSCLC cells which was similar with effects of overexpression of miR-153, while overexpression of ADAM19 attenuated the function of miR-153 in NSCLC cells. Taken together, our results highlight the significance of miR-153 and ADAM19 in the development and progression of NSCLC.

ADAMS/WT is an wind-turbine-specific shell for the general-purpose mechanical system simulation package ADAMS5. It was developed under the guidance of the National Renewable Energy Laboratory to give engineers and analysts in the wind turbine community access to the analytical power of ADAMS, without having to become expert in its particular technology. The 1.4 version of ADAMS/WT is the most recent upgrade to the package, incorporating the most up-to-date version of the AeroDyn aerodynamic forcing subroutines from the University of Utah. It is also the first version to be made available on the Windows/NT platform. In version 1.4, ADAMS/WT has been significantly improved throughout and runs much faster. Automatic generation of standardized output has been added. The documentation has been extensively augmented with more detailed descriptions, more figures and more examples. ADAMS/WT remains the most powerful analytical tool available for horizontal-axis wind turbine development. 10 figs.

open, the operator is exposed to the potential risk of serious and minor cuts and abrasions when using and handling the saw. There is also potential for damage to the blades if the saw is not stored properly. Without guarding on the lower part of the blades, these can be damaged if the saw is dropped or rested on the cutting blades. Based upon the industrial hygiene sampling conducted for the other four saws demonstrated at FIU, noise levels, nuisance dust, and airborne fiberglass may be a problem when using this technology for the cutting of fiberglass-reinforced plywood crates. No industrial hygiene sampling was conducted while the Adamant saw was in use. Engineering controls should be used to eliminate these problems whenever possible. Where this is not possible, administrative controls, training, and proper personal protective equipment (PPE) should be used. Respirators should be used if engineering controls do not sufficiently control the dust or fiberglass generated. Respirators should be equipped with an organic vapor and acid gas cartridge with High Efficiency Particulate Air (HEPA) filter, since during the demonstration, the workers complained of an odd smell, which may have been the breakdown of the fiberglass.

A new neutron reflectometer SuperADAM has recently been built and commissioned at the Institut Laue-Langevin, Grenoble, France. It replaces the previous neutron reflectometer ADAM. The new instrument uses a solid state polarizer/wavelength filter providing a highly polarized (up to 98.6%) monochromatic neutron flux of 8 Multiplication-Sign 10{sup 4} n cm{sup -2} s{sup -1} with monochromatization {Delta}{lambda}/{lambda}= 0.7% and angular divergence {Delta}{alpha}= 0.2 mrad. The instrument includes both single and position sensitive detectors. The position sensitive detector allows simultaneous measurement of specular reflection and off-specular scattering. Polarization analysis for both specular reflection and off-specular scattering is achieved using either mirror analyzers or a {sup 3}He spin filter cell. High efficiency detectors, low background, and high flux provides a dynamic range of up to seven decades in reflectivity. Detailed specifications and the instrument capabilities are illustrated with examples of recently collected data in the fields of thin film magnetism and thin polymer films.

We obtained Spitzer Space Telescope Multiband Imaging Photometer for Spitzer (MIPS) 24 {mu}m and 70 {mu}m observations of 182 nearby, Hipparcos F- and G-type common proper motion single and binary systems in the nearest OB association, Scorpius-Centaurus. We also obtained Magellan/MIKE R {approx} 50,000 visual spectra at 3500-10500 A for 181 candidate ScoCen stars in single and binary systems. Combining our MIPS observations with those of other ScoCen stars in the literature, we estimate 24 {mu}m F+G-type disk fractions of 9/27 (33% {+-} 11%), 21/67 (31% {+-} 7%), and 25/71 (35% {+-} 7%) for Upper Scorpius ({approx}10 Myr), Upper Centaurus Lupus ({approx}15 Myr), and Lower Centaurus Crux ({approx}17 Myr), respectively. We confirm previous IRAS and MIPS excess detections and present new discoveries of 41 protoplanetary and debris disk systems, with fractional infrared luminosities ranging from L{sub IR}/L{sub *} = 10{sup -5} to 10{sup -2} and grain temperatures ranging from T{sub gr} = 40-300 K. We searched for an increase in 24 {mu}m excess at an age of 15-20 Myr, consistent with the onset of debris production predicted by coagulation N-body simulations of outer planetary systems. We found such an increase around 1.5 M{sub sun} stars but discovered a decrease in the 24 {mu}m excess around 1.0 M{sub sun} stars. We additionally discovered that the 24 {mu}m excess around 1.0 M{sub sun} stars is larger than predicted by self-stirred models. Finally, we found a weak anti-correlation between fractional infrared luminosity (L{sub IR}/L{sub *}) and chromospheric activity (R'{sub HK}), that may be the result of differences in stellar properties, such as mass, luminosity, and/or winds.

  12. Evaluation of ADAM/1 model for advanced coal-extraction concepts

    SciTech Connect (OSTI)

    Deshpande, G. K.; Gangal, M. D.

    1982-01-15

The Advanced Coal Extraction Project is sponsored by the Department of Energy at the Jet Propulsion Laboratory to define and develop advanced underground coal extraction systems which: (1) are suitable for significant remaining resources after the year 2000, and (2) promise a significant improvement in production cost and miner safety, with no degradation in miner health, environmental quality and resource recovery. System requirements in the five performance areas have been defined by Goldsmith and Lavin (1980). Several existing computer programs for estimating life-cycle cost of mining systems have been evaluated. A commercially available program ADAM/1 was found to be satisfactory in relation to the needs of the Advanced Coal Extraction Project. Two test cases were run to confirm the ability of the program to handle non-conventional mining equipment and procedures. The results were satisfactory. The model, therefore, is recommended to the project team for evaluation of their conceptual designs. Since the model is commercially available, data preparation instructions are not reproduced in this document; instead the reader is referred to the original documents for this information.

    CARB

    2005-11-01

The Yakama Nation is the largest tribe in the Pacific Northwest. The Yakama Nation Housing Authority (YNHA) is working to rehabilitate single family homes (two to four bedrooms) in its Adams View project using public and tax credit financing. It is in need of a major rehabilitation as a result of wear and tear after many years of use and overcrowding. The scope for the current CARB ''gut rehab'' project is 25 of the 40 homes in the Adams View development, but the proposed strategies could be replicated for the remaining 15 homes as well as for several other similar developments of the YNHA. On a larger scale, the system rehabilitation strategy developed for the Adam's View project should be replicable for most of the more than 4,300 housing units that were constructed in the Northwest (Washington, Oregon, and Idaho) under the US Department of Housing and Urban Development (HUD) Housing Act of 1937.

The paper develops an approach to the issues relating to sea level change that will assist the non-scientist and the applied scientist in making the most effective use of our existing and developing knowledge. The human perception of ''sea level'' and how that changes as societies change and develop are discussed. After some practical perspectives on the relationships between societies and sea levels are developed, an approach to developing the best available local prediction of sea level changes is outlined, and finally present knowledge and uncertainties about the future course of events that will influence ''sea level'' as defined in the practical sense is discussed.

    Grasso, Albert P.

    1986-01-01

A liquid level detector for low pressure boilers. A boiler tank, from which apor, such as steam, normally exits via a main vent, is provided with a vertical side tube connected to the tank at the desired low liquid level. When the liquid level falls to the level of the side tube vapor escapes therethrough causing heating of a temperature sensitive device located in the side tube, which, for example, may activate a liquid supply means for adding liquid to the boiler tank. High liquid level in the boiler tank blocks entry of vapor into the side tube, allowing the temperature sensitive device to cool, for example, to ambient temperature.

An apparatus for switch wear leveling includes a switching module that controls switching for two or more pairs of switches in a switching power converter. The switching module controls switches based on a duty cycle control technique and closes and opens each switch in a switching sequence. The pairs of switches connect to a positive and negative terminal of a DC voltage source. For a first switching sequence a first switch of a pair of switches has a higher switching power loss than a second switch of the pair of switches. The apparatus includes a switch rotation module that changes the switching sequence of the two or more pairs of switches from the first switching sequence to a second switching sequence. The second switch of a pair of switches has a higher switching power loss than the first switch of the pair of switches during the second switching sequence.

A growing number of low and middle income nations (LMCs) have adopted some sort of system for environmental impact assessment (EIA). However, generally many of these EIA systems are characterised by a low performance in terms of timely information dissemination, monitoring and enforcement after licencing. Donor actors (such as the World Bank) have attempted to contribute to a higher performance of EIA systems in LMCs by intervening at two levels: the project level (e.g. by providing scoping advice or EIS quality review) and the system level (e.g. by advising on EIA legislation or by capacity building). The aims of these interventions are environmental protection in concrete cases and enforcing the institutionalisation of environmental protection, respectively. Learning by actors involved is an important condition for realising these aims. A relatively underexplored form of learning concerns learning at EIA system-level via project level donor interventions. This 'indirect' learning potentially results in system changes that better fit the specific context(s) and hence contribute to higher performances. Our exploratory research in Ghana and the Maldives shows that thus far, 'indirect' learning only occurs incidentally and that donors play a modest role in promoting it. Barriers to indirect learning are related to the institutional context rather than to individual characteristics. Moreover, 'indirect' learning seems to flourish best in large projects where donors achieved a position of influence that they can use to evoke reflection upon system malfunctions. In order to enhance learning at all levels donors should thereby present the outcomes of the intervention elaborately (i.e. discuss the outcomes with a large audience), include practical suggestions about post-EIS activities such as monitoring procedures and enforcement options and stimulate the use of their advisory reports to generate organisational memory and ensure a better information dissemination.

    DOE Patents [OSTI]

Tokarz, Richard D. (West Richland, WA)

    1983-01-01

A length of metal sheathed metal oxide cable is perforated to permit liquid access to the insulation about a pair of conductors spaced close to one another. Changes in resistance across the conductors will be a function of liquid level, since the wetted insulation will have greater electrical conductivity than that of the dry insulation above the liquid elevation.

    1993-11-01

A Peruvian FCC unit's operations were improved by increasing the regenerator's catalyst level. This increase resulted in lower stack losses, an improved temperature profile, increased catalyst activity and a lower catalyst consumption rate. A more stable operation saved this Peruvian refiner over $131,000 per year in catalyst alone. These concepts and data may be suitable for your FCC unit as well.

  1. Levelized Power Generation Cost Codes

Energy Science and Technology Software Center (OSTI)

    1996-04-30

LPGC is a set of nine microcomputer programs for estimating power generation costs for large steam-electric power plants. These programs permit rapid evaluation using various sets of economic and technical ground rules. The levelized power generation costs calculated may be used to compare the relative economics of nuclear and coal-fired plants based on life-cycle costs. Cost calculations include capital investment cost, operation and maintenance cost, fuel cycle cost, decommissioning cost, and total levelized power generationmore » cost. These programs can be used for quick analyses of power generation costs using alternative economic parameters, such as interest rate, escalation rate, inflation rate, plant lead times, capacity factor, fuel prices, etc. The two major types of electric generating plants considered are pressurized water reactor (PWR) and pulverized coal-fired plants. Data are also provided for the Large Scale Prototype Breeder (LSPB) type liquid metal reactor.« less

  5. Load Leveling Battery System Costs

Energy Science and Technology Software Center (OSTI)

    1994-10-12

SYSPLAN evaluates capital investment in customer side of the meter load leveling battery systems. Such systems reduce the customer's monthly electrical demand charge by reducing the maximum power load supplied by the utility during the customer's peak demand. System equipment consists of a large array of batteries, a current converter, and balance of plant equipment and facilities required to support the battery and converter system. The system is installed on the customer's side of themore » meter and controlled and operated by the customer. Its economic feasibility depends largely on the customer's load profile. Load shape requirements, utility rate structures, and battery equipment cost and performance data serve as bases for determining whether a load leveling battery system is economically feasible for a particular installation. Life-cycle costs for system hardware include all costs associated with the purchase, installation, and operation of battery, converter, and balance of plant facilities and equipment. The SYSPLAN spreadsheet software is specifically designed to evaluate these costs and the reduced demand charge benefits; it completes a 20 year period life cycle cost analysis based on the battery system description and cost data. A built-in sensitivity analysis routine is also included for key battery cost parameters. The life cycle cost analysis spreadsheet is augmented by a system sizing routine to help users identify load leveling system size requirements for their facilities. The optional XSIZE system sizing spreadsheet which is included can be used to identify a range of battery system sizes that might be economically attractive. XSIZE output consisting of system operating requirements can then be passed by the temporary file SIZE to the main SYSPLAN spreadsheet.« less

  9. Liquid-level sensing device

    DOE Patents [OSTI]

    Goldfuss, G.T.

    1975-09-16

This invention relates to a device for sensing the level of a liquid while preventing the deposition and accumulation of materials on the exterior surfaces thereof. Two dissimilar metal wires are enclosed within an electrical insulating material, the wires being joined together at one end to form a thermocouple junction outside the insulating material. Heating means is disposed within the electrical insulating material and maintains the device at a temperature substantially greater than that of the environment surrounding the device, the heating means being electrically insulated from the two dissimilar thermocouple wires. In addition, a metal sheath surrounds and contacts both the electrical insulating material and the thermocouple junction. Electrical connections are provided for connecting the heating means with a power source and for connecting the thermocouple wires with a device for sensing the electrical potential across the thermocouple junction. 10. Fluorescent optical liquid level sensor

    DOE Patents [OSTI]

    Weiss, Jonathan D. (Albuquerque, NM)

    2001-01-01

A liquid level sensor comprising a transparent waveguide containing fluorescent material that is excited by light of a first wavelength and emits at a second, longer wavelength. The upper end of the waveguide is connected to a light source at the first wavelength through a beveled portion of the waveguide such that the input light is totally internally reflected within the waveguide above an air/liquid interface in a tank but is transmitted into the liquid below this interface. Light is emitted from the fluorescent material only in those portions of the waveguide that are above the air/liquid interface, to be collected at the upper end of the waveguide by a detector that is sensitive only to the second wavelength. As the interface moves down in the tank, the signal strength from the detector will increase.

  12. High level white noise generator

    DOE Patents [OSTI]

Borkowski, Casimer J.; Blalock, Theron V.

    1979-01-01

A wide band, stable, random noise source with a high and well-defined output power spectral density is provided which may be used for accurate calibration of Johnson Noise Power Thermometers (JNPT) and other applications requiring a stable, wide band, well-defined noise power spectral density. The noise source is based on the fact that the open-circuit thermal noise voltage of a feedback resistor, connecting the output to the input of a special inverting amplifier, is available at the amplifier output from an equivalent low output impedance caused by the feedback mechanism. The noise power spectral density level at the noise source output is equivalent to the density of the open-circuit thermal noise or a 100 ohm resistor at a temperature of approximately 64,000 Kelvins. The noise source has an output power spectral density that is flat to within 0.1% (0.0043 db) in the frequency range of from 1 KHz to 100 KHz which brackets typical passbands of the signal-processing channels of JNPT's. Two embodiments, one of higher accuracy that is suitable for use as a standards instrument and another that is particularly adapted for ambient temperature operation, are illustrated in this application.

  13. Optimizing High Level Waste Disposal

    SciTech Connect (OSTI)

    Dirk Gombert

    2005-09-01

If society is ever to reap the potential benefits of nuclear energy, technologists must close the fuel-cycle completely. A closed cycle equates to a continued supply of fuel and safe reactors, but also reliable and comprehensive closure of waste issues. High level waste (HLW) disposal in borosilicate glass (BSG) is based on 1970s era evaluations. This host matrix is very adaptable to sequestering a wide variety of radionuclides found in raffinates from spent fuel reprocessing. However, it is now known that the current system is far from optimal for disposal of the diverse HLW streams, and proven alternatives are available to reduce costs by billions of dollars. The basis for HLW disposal should be reassessed to consider extensive waste form and process technology research and development efforts, which have been conducted by the United States Department of Energy (USDOE), international agencies and the private sector. Matching the waste form to the waste chemistry and using currently available technology could increase the waste content in waste forms to 50% or more and double processing rates. Optimization of the HLW disposal system would accelerate HLW disposition and increase repository capacity. This does not necessarily require developing new waste forms, the emphasis should be on qualifying existing matrices to demonstrate protection equal to or better than the baseline glass performance. Also, this proposed effort does not necessarily require developing new technology concepts. The emphasis is on demonstrating existing technology that is clearly better (reliability, productivity, cost) than current technology, and justifying its use in future facilities or retrofitted facilities. Higher waste processing and disposal efficiency can be realized by performing the engineering analyses and trade-studies necessary to select the most efficient methods for processing the full spectrum of wastes across the nuclear complex. This paper will describe technologies being

The Low-Level Waste Technical Strategy document describes the mechanisms which the Low-Level Waste Program Office plans to implement to achieve its mission. The mission is to manage the receipt, immobilization, packaging, storage/disposal and RCRA closure (of the site) of the low-level Hanford waste (pretreated tank wastes) in an environmentally sound, safe and cost-effective manner. The primary objective of the TWRS Low-level waste Program office is to vitrify the LLW fraction of the tank waste and dispose of it onsite.

    Flanagan, G.U.; /Purdue U.

    2007-04-01

The CDF Run II Level-2 calorimeter trigger is implemented in hardware and is based on an algorithm used in Run I. This system insured good performance at low luminosity obtained during the Tevatron Run II. However, as the Tevatron instantaneous luminosity increases, the limitations of the current system due to the algorithm start to become clear. In this paper, we will present an upgrade of the Level-2 calorimeter trigger system at CDF. The upgrade is based on the Pulsar board, a general purpose VME board developed at CDF and used for upgrading both the Level-2 tracking and the Level-2 global decision crate. This paper will describe the design, hardware and software implementation, as well as the advantages of this approach over the existing system.

    1996-04-01

The purpose of this paper is to present the results of the testing of the recent improvements made to the two-phase level tracking model in RELAP5/MOD3.2. The level model was originally developed during the development of the TRAC-BWR computer code and was subsequently modified by the Pennsylvania State University (PSU). The modifications developed at PSU concern the way in which the two-phase level is moved from volume to volume as the thermal-hydraulic conditions in the system being simulated change during the course of a transients. The other components in the level tracking model remain as described in the original implementation of the model.
