    This report presents the preliminary findings from the first phase of the Environmental Survey of the US Department of Energy (DOE) Fermi National Accelerator Laboratory (Fermilab), conducted September 14 through 25, 1987. The Survey is being conducted by an interdisciplinary team of environmental specialists led and managed by the Office of Environment, Safety and Health's Office of Environmental Audit. Individual participants for the Survey team are being supplied by a private contractor. The objective of the Survey is to identify environmental problems and areas of environmental risk associated with Fermilab. The Survey covers all environmental media and all areas of environmental regulation. It is being performed in accordance with the DOE Environmental Survey Manual. This phase of the Survey involves the review of existing site environmental data, observations of the operations performed at Fermilab, and interviews with site personnel. 110 refs., 26 figs., 41 tabs.

    IL-15 interacts with a heterotrimeric receptor that consists of the {beta} and {gamma} subunits of the IL-2 receptor (IL-2R) as well as a specific, high-affinity IL-15-binding subunit, which is designated IL-15R{alpha}. Since both the {beta} and the {gamma} subunits of the IL-2R are required for signaling by either IL-2 or IL-15, it is not surprising that these cytokines share many activities in vitro. However, the differential expression of these cytokines and the {alpha} chains of their receptors within various tissues and cell types suggests that IL-2 and IL-15 may perform at least partially distinct physiological functions. The production of IL-15 by macrophages, and possibly other cell types, in response to environmental stimuli and infectious agents suggests that IL-15 may play a role in protective immune responses, allograft rejection, and the pathogenesis of autoimmune diseases. 56 refs.

We report that the IL-7R{alpha} ectodomain uses glycosylation to modulate its binding constants to IL-7, unlike the other receptors in the {gamma}{sub c} family. IL-7 binds glycosylated IL-7R{alpha} 300-fold more tightly than unglycosylated IL-7R{alpha}, and the enhanced affinity is attributed primarily to an accelerated on rate. Structural comparison of IL-7 in complex to both forms of IL-7R{alpha} reveals that glycosylation does not participate directly in the binding interface. The SCID mutations of IL-7R{alpha} locate outside the binding interface with IL-7, suggesting that the expressed mutations cause protein folding defects in IL-7R{alpha}. The IL-7/IL-7R{alpha} structures provide a window into the molecular recognition events of the IL-7 signaling cascade and provide sites to target for designing new therapeutics to treat IL-7-related diseases.

    Successional Restoration is the method being used to restore the prairie at Fermilab on the former agricultural fields. This involves an initial planting, using aggressive species that have wide ecological tolerances which will grow well on abandoned agricultural fields. Collectively, these species are designated as the prairie matrix. The species used for this prairie matrix compete with and eventually eliminate most weedy species. They also provide an adequate fuel load capable of sustaining a fire within a few years after a site has been initially planted. Associated changes in the biological and physical structure of the soil help prepare the way for the successful introduction of plants of the later successional species. Only after the species of the prairie matrix are well established, is the species diversity increased by introducing species with narrower ecological tolerances. These species are thus characteristic of the later successional stages.

Here we show that micromolar concentrations of curcumin inhibited DNA synthesis by mouse CD4{sup +} T-lymphocytes, as well as interleukin-2 (IL-2) and CD25 ({alpha} chain of the high affinity IL-2 receptor) expression in response to antibody-mediated cross-linking of CD3 and CD28. Curcumin acted downstream of protein kinase C activation and intracellular Ca{sup 2+} release to inhibit I{kappa}B phosphorylation, which is required for nuclear translocation of the transcription factor NF{kappa}B. In addition, IL-2-dependent DNA synthesis by mouse CTLL-2 cells, but not constitutive CD25 expression, was impaired in the presence of curcumin, which demonstrated an inhibitory effect on IL-2 receptor (IL-2R) signaling. IL-2-induced phosphorylation of STAT5A and JAK3, but not JAK1, was diminished in the presence of curcumin, indicating inhibition of critical proximal events in IL-2R signaling. In line with the inhibitory action of curcumin on IL-2R signaling, pretreatment of CD4{sup +}CD25{sup +} regulatory T-cells with curcumin downregulated suppressor function, as well as forkhead box p3 (Foxp3) expression. We conclude that curcumin inhibits IL-2 signaling by reducing available IL-2 and high affinity IL-2R, as well as interfering with IL-2R signaling.

    This chapter summarizes the laboratory activities performed by PNNLs Vadose Zone Characterization Project in support of the Tank Farm Vadose Zone Program, led by CH2M HILL Hanford Group, Inc. The results of these studies are contained in numerous reports (Lindenmeier et al. 2002; Serne et al. 2002a, 2002b, 2002c, 2002d, 2002e; Lindenmeier et al. 2003; Serne et al. 2004a, 2004b; Brown et al. 2005, 2006a, 2007; Serne et al. 2007) and have generated much of the data reported in Chapter 22 (Geochemistry-Contaminant Movement), Appendix G (Geochemistry-Contaminant Movement), and Cantrell et al. (2007, SST WMA Geochemistry Data Package in preparation). Sediment samples and characterization results from PNNLs Vadose Zone Characterization Project are also shared with other science and technology (S&T) research projects, such as those summarized in Chapter 12 (Associated Science Activities).

    Sublethally irradiated mice undergo thymic regeneration which follows a phenotypic pattern of events similar to that observed during normal fetal development. Thymic regeneration after irradiation is the product of a limited pool of intrathymic radioresistant stem cells undergoing simultaneous differentiation. We show that in this model of T cell development, thymic regeneration follows a pathway in which the IL-2R is transiently expressed on CD4-/CD8- cells. IL-2R expression occurred during the exponential growth period of thymic regeneration, and IL-2R blocking prevented this explosive growth. Flow cytometry analysis revealed that the IL-2R blockade affected primarily the development of the immature CD3-/CD4-/CD8- (triple negative) cells and their ability to generate CD3+/CD4+/CD8+ or CD3+/CD4+/CD8- and CD3+/CD4-/CD8+ thymocytes. Thus, our findings demonstrate that blocking of the IL-2R resulted in an arrest in proliferation and differentiation by intrathymic radioresistant stem cells, indicating that the IL-2/IL-2R pathway is necessary for the expansion of immature triple negative T cells.

    Human peripheral blood T cell expression of IL-2 receptors (IL-2R), detected by both immunocytofluorometry and /sup 125/I-IL-2 binding, was studied using lymphocytes stimulated with monoclonal anti-T3 antibodies (Leu-4, OKT3). Lymphocytes, isolated from healthy individuals, were prescreened and classified as Leu-4 responders or non-responders according to 72 h /sup 3/H-thymidine incorporation experiments. Leu-4 non-responder lymphocytes, though capable of normal IL-2R expression and IL-2 secretion when cultured with OKT3 (IgG2a), expressed little to no IL-2R nor secreted IL-2 when stimulated with Leu-4 (IgG1). In addition, the amount of IL-2 secreted by Leu-4 stimulated, Leu-4 responder cells, was one-third- to one-fifth of that detected when OKT3 was used as the stimulant. The addition of recombinant IL-2 (rIL-2) to a Leu-4 stimulated, Leu-4 non-responder lymphocyte culture, resulted in the expression of IL-2R and cellular proliferation, indicating that IL-2 upregulated its biologically relevant receptor. As expected, cyclosporin-A (CSA) inhibited the secretion of IL-2 and subsequent proliferation of Leu-4 stimulated, Leu-4 responder cells. Unexpectedly, however, the expression of IL-2R was also blocked. Exogenous rIL-2 partially reversed the effect of CSA on IL-2R expression and proliferation. The results indicate that IL-2 may provide an additional, required signal for optimal IL-2R expression.

    Carbon dioxide fluxes and stocks in terrestrial ecosystems are key measurements needed to constrain quantification of regional carbon sinks and sources and the mechanisms controlling them. This information is required to produce a sound carbon budget for North America. This project examines CO2 and energy fluxes from agricultural land and from restored tallgrass prairie to compare their carbon sequestration potentials. The study integrates eddy covariance measurements with biometric measurements of plant and soil carbon stocks for two systems in northeastern Illinois: 1) long-term cultivated land in corn-soybean rotation with conventional tillage, and 2) a 15 year-old restored prairie that represents a long-term application of CRP conversion of cultivated land to native vegetation. The study contributes to the North American Carbon Program (NACP) by providing information on the magnitude and distribution of carbon stocks and the processes that control carbon dynamics in cultivated and CRP-restored land in the Midwest.     A case-control association study was conducted in Caucasian patients with schizophrenia (DSM-III-R, n = 42) and unaffected controls (n = 47) matched for ethnicity and area of residence. Serum interleukin-2 receptor (IL-2R) concentrations, as well as a dinucleotide repeat polymorphism in the IL-2RP chain gene, were examined in both groups. No significant differences in IL-2R concentrations or in the distribution of the polymorphism were noted. This study does not support an association between schizophrenia and the IL-2RP gene locus, contrary to the suggestive evidence from linkage analysis in multicase families. 17 refs., 2 tabs.

    A standard laboratory module for automatically producing a solution of cominants from a soil sample. A sonication tip agitates a solution containing the soil sample in a beaker while a stepper motor rotates the sample. An aspirator tube, connected to a vacuum, draws the upper layer of solution from the beaker through a filter and into another beaker. This beaker can thereafter be removed for analysis of the solution. The standard laboratory module encloses an embedded controller providing process control, status feedback information and maintenance procedures for the equipment and operations within the standard laboratory module.

    The overall objective of the Workshop was to identify opportunities for accelerating the use of both natural gas (NG) and hydrogen (H{sub 2}) as motor fuels and in stationary power applications. Specific objectives of the Workshop were to: (1) Convene industry and other stakeholders to share current status/state-of-the-art of NG and H{sub 2} infrastructure; (2) Identify key challenges (including non-technical challenges, such as permitting, installation, codes, and standards) preventing or delaying the widespread deployment of NG and H{sub 2} infrastructure. Identify synergies between NG and H{sub 2} fuels; and (3) Identify and prioritize opportunities for addressing the challenges identified above, and determine roles and opportunities for both the government and industry stakeholders. Plenary speakers and panel discussions summarized the current status of the NG and H{sub 2} infrastructure, technology for their use in transportation and stationary applications, and some of the major challenges and opportunities to more widespread use of these fuels. Two break-out sessions of three groups each addressed focus questions on: (1) infrastructure development needs; (2) deployment synergies; (3) natural gas and fuel cell vehicles (NGVs, FCVs), specialty vehicles, and heavy-duty trucks; (4) CHP (combined heat and power), CHHP (combined hydrogen, heat, and power), and synergistic approaches; and (5) alternative uses of natural gas.

    Binding of interleukin-2 (IL-2) to its specific receptor induces activation of two members of Jak family protein tyrosine kinases, Jak1 and Jak3. An IL-2 receptor (IL-2R)-reconstituted NIH 3T3 fibroblast cell line proliferates in response to IL-2 only when hematopoietic lineage-specific Jak3 is ectopically expressed. However, the mechanism of Jak3-dependent proliferation in the fibroblast cell line is not known. Here, I showed that Jak3 expression is dispensable for IL-2-induced activation of Jak1 and Stat proteins and expression of nuclear proto-oncogenes in the IL-2R-reconstituted fibroblast cell line. Jak3 expression markedly enhanced these IL-2-induced signaling events. In contrast, Jak3 expression was essential for induction of cyclin genes involved in the G1-S transition. These data suggest a critical role of Jak3 in IL-2 signaling in the fibroblast cell line and may provide further insight into the cell type-specific mechanism of cytokine signaling.

    The initial federal funding for the Colorado State University Smart Grid Integration Laboratory is through a Congressionally Directed Project (CDP), DE-OE0000070 Smart Grid Integration Laboratory. The original program requested in three one-year increments for staff acquisition, curriculum development, and instrumentation all which will benefit the Laboratory. This report focuses on the initial phase of staff acquisition which was directed and administered by DOE NETL/ West Virginia under Project Officer Tom George. Using this CDP funding, we have developed the leadership and intellectual capacity for the SGIC. This was accomplished by investing (hiring) a core team of Smart Grid Systems engineering faculty focused on education, research, and innovation of a secure and smart grid infrastructure. The Smart Grid Integration Laboratory will be housed with the separately funded Integrid Laboratory as part of CSU's overall Smart Grid Integration Center (SGIC). The period of performance of this grant was 10/1/2009 to 9/30/2011 which included one no cost extension due to time delays in faculty hiring. The Smart Grid Integration Laboratory's focus is to build foundations to help graduate and undergraduates acquire systems engineering knowledge; conduct innovative research; and team externally with grid smart organizations. Using the results of the separately funded Smart Grid Workforce Education Workshop (May 2009) sponsored by the City of Fort Collins, Northern Colorado Clean Energy Cluster, Colorado State University Continuing Education, Spirae, and Siemens has been used to guide the hiring of faculty, program curriculum and education plan. This project develops faculty leaders with the intellectual capacity to inspire its students to become leaders that substantially contribute to the development and maintenance of Smart Grid infrastructure through topics such as: (1) Distributed energy systems modeling and control; (2) Energy and power conversion; (3) Simulation of electrical power distribution system that integrates significant quantities of renewable and distributed energy resources; (4) System dynamic modeling that considers end-user behavior, economics, security and regulatory frameworks; (5) Best practices for energy management IT control solutions for effective distributed energy integration (including security with the underlying physical power systems); (6) Experimental verification of effects of various arrangements of renewable generation, distributed generation and user load types along with conventional generation and transmission. Understanding the core technologies for enabling them to be used in an integrated fashion within a distribution network remains is a benefit to the future energy paradigm and future and present energy engineers.

The Laboratory creates advanced technologies for emergency response operations, radiological incident response, and other remote sensing activities. RSL emergency response teams are on call 24-hours a day, and maintain the capability to deploy domestically and internationally in response to threats involving the loss, theft, or release of nuclear or radioactive material. Such incidents might include Nuclear Power Plant accidents, terrorist incidents involving nuclear or radiological materials, NASA launches, and transportation accidents involving nuclear materials. Working with the US Department of Homeland Security, RSL personnel equip, maintain, and conduct training on the mobile detection deployment unit, to provide nuclear radiological security at major national events such as the super bowl, the Indianapolis 500, New Year's Eve celebrations, presidential inaugurations, international meetings and conferences, just about any event where large numbers of people will gather.

    The Hydrogen Systems Laboratory in a unique partnership between Purdue University's main campus in West Lafayette and the Calumet campus was established and its capabilities were enhanced towards technology demonstrators. The laboratory engaged in basic research in hydrogen production and storage and initiated engineering systems research with performance goals established as per the USDOE Hydrogen, Fuel Cells, and Infrastructure Technologies Program. In the chemical storage and recycling part of the project, we worked towards maximum recycling yield via novel chemical selection and novel recycling pathways. With the basic potential of a large hydrogen yield from AB, we used it as an example chemical but have also discovered its limitations. Further, we discovered alternate storage chemicals that appear to have advantages over AB. We improved the slurry hydrolysis approach by using advanced slurry/solution mixing techniques. We demonstrated vehicle scale aqueous and non-aqueous slurry reactors to address various engineering issues in on-board chemical hydrogen storage systems. We measured the thermal properties of raw and spent AB. Further, we conducted experiments to determine reaction mechanisms and kinetics of hydrothermolysis in hydride-rich solutions and slurries. We also developed a continuous flow reactor and a laboratory scale fuel cell power generation system. The biological hydrogen production work summarized as Task 4.0 below, included investigating optimal hydrogen production cultures for different substrates, reducing the water content in the substrate, and integrating results from vacuum tube solar collector based pre and post processing tests into an enhanced energy system model. An automated testing device was used to finalize optimal hydrogen production conditions using statistical procedures. A 3 L commercial fermentor (New Brunswick, BioFlo 115) was used to finalize testing of larger samples and to consider issues related to scale up. Efforts continued to explore existing catalytic methods involving nano catalysts for capture of CO2 from the fermentation process.
