  1. Comparison of Packed Beds and Qiagen Columns for Recovering Trace Amounts of B. anthracis DNA from Liquid Suspensions

    SciTech Connect (OSTI)

    Sorensen, K; Arroyo, E; Erler, A; Christian, A T; Camp, D; Wheeler, E K

    2006-06-23

The goal of this work was to optimize and evaluate LLNL's in-bed amplification technology to improve the level of detection for suspensions containing trace amounts of anthracis DNA. The binding/cleaning performance of the packed bed is compared to the conventional commercial approach; Qiagen column cleanup and elution, followed by detection through an ex-situ amplification process. Five liquid suspensions were spiked with B.anthracis DNA in concentration series. These suspensions were: (1) water, (2) water with EDTA, (3) dirty water from carpet extraction, (4) dirty carpet extraction with phosphate buffered saline (PBS) plus 0.1% Tween 20 plus 0.1% gelatin, and (5) a subway aerosol collected in water. Each suspension matrix was spiked with DNA and injected (in replicate) into either Qiagen Microcolumns (using the kit processing instructions) or LLNL's packed bed (using the LLNL in-bed purification and amplification protocol). The process output was assayed by quantitative polymerase chain reaction (QPCR). Table ES-1 shows the level of DNA (pg per 100 uL of input suspension) that resulted in successful amplification for all reactions (X=Y), and the level for which at least one of the reactions was successful (X>0). For each suspension and DNA concentration, there were Y QPCR assays of which X showed successful amplification. LLNL's packed bed technology outperformed Qiagen Microcolumns for all five suspensions, typically by one order of magnitude in both the limit of assured detection (all reactions positive), and the lower limit of detection (some reactions positive).

    2014-06-01

Purpose: In the commissioning and QA of surface isotope-based applicators, source-indexer distance (SID) has a great influence in the flatness, symmetry and output. To these purposes, methods described in the literature are the use of a special insert at the entrance of dwell chamber or radiochromic films. Here we present the experience with a micro-chamber array to perform the commissioning and QA of Valencia applicators. Methods: Valencia applicators have been used, the classic and the new extra-shielded version. A micro-chamber array has been employed, 1000 SRS (PTW), with 977 liquid filled, 2.3×2.3×0.5 mm{sup 3} sized ion chambers covering 11×11 cm{sup 2}, which spacing is 2.5 mm in the central 5.5×5.5 cm{sup 2}, dedicated mainly in principle, in conjunction with Octavius 4D (PTW), to IMRT, VMAT, SBRT verifications. Verisoft software that allows for 3D and planar analysis has been used to evaluate the results. Applicators were located on the surface of the array. To verify the SID, measurements corresponding to the reference value, SID ± 1 mm and SID ± 2 mm were acquired (integration time was fixed in order to discard the influence of the source entrance/exit). Once SID was determined, standard protocol treatments corresponding to 3 Gy and 7 Gy were acquired in order to establish typical patient dose distribution. Results: The method is fast and sensitive. The SID obtained was 1321 mm which is the nominal value included in the applicator manual. For example at 1319 mm an asymmetry of ±8% with respect to the central value was measured, along with a central deviation of −4% referred to 1321 mm. Conclusion: A practical method for the commissioning and QA of Valencia applicators has been described. It has been shown that it is an efficient and accurate tool for these purposes as well as for the verification of the absolute output constancy.

  19. Evaluation of models for estimating solar irradiation on vertical surfaces at Valencia, Spain

    SciTech Connect (OSTI)

    Utrillas, M.P.; Martinez-Lozano, J.A.; Casanovas, A.J. )

    1991-01-01

Hourly irradiation data recorded on vertical surfaces at north, east, south, and west orientations during the winter period going from December 1989 to March 1990 in Valencia, Spain, have been compared with estimated solar irradiation from several tilted-surface models. The isotropic-, Temps' and Coulson's-, Klucher's-, Hay's-, Shartveit's and Olseth's, Gueymard's-, and Perez' (simplified) models have been considered for this comparison. Root-mean-square-difference (RMSD), mean-bias-difference (MBD) and mean-absolute-difference (MAD) estimators have been used to measure the departure of models from experimental data. Modeled values are evaluated with the original coefficients proposed by the authors. Results of comparison show that the south orientation is less prone to modeling errors. Perez' 25{degree} circumsolar simplified model being the less affected by errors in north, eat and south orientations; for west orientation Klucher's model gives a lesser error. These results are consistent with those reported in previous studies.

    SciTech Connect (OSTI)

    Jain, Rajeev; Mahadevan, Vijay

    2015-09-30

This report gives documentation for using MeshKit’s Reactor Geometry (and mesh) Generator (RGG) GUI and also briefly documents other algorithms and tools available in MeshKit. RGG is a program designed to aid in modeling and meshing of complex/large hexagonal and rectilinear reactor cores. RGG uses Argonne’s SIGMA interfaces, Qt and VTK to produce an intuitive user interface. By integrating a 3D view of the reactor with the meshing tools and combining them into one user interface, RGG streamlines the task of preparing a simulation mesh and enables real-time feedback that reduces accidental scripting mistakes that could waste hours of meshing. RGG interfaces with MeshKit tools to consolidate the meshing process, meaning that going from model to mesh is as easy as a button click. This report is designed to explain RGG v 2.0 interface and provide users with the knowledge and skills to pilot RGG successfully. Brief documentation of MeshKit source code, tools and other algorithms available are also presented for developers to extend and add new algorithms to MeshKit. RGG tools work in serial and parallel and have been used to model complex reactor core models consisting of conical pins, load pads, several thousands of axially varying material properties of instrumentation pins and other interstices meshes.

  10. Kit for providing a technetium medical radioimaging agent

    DOE Patents [OSTI]

    Wildung, Raymond E.; Garland, Thomas R.; Li, Shu-Mei W.

    2000-01-01

The present invention is directed toward a kit for microbial reduction of a technetium compound to form other compounds of value in medical imaging. The technetium compound is combined in a mixture with non-growing microbial cells which contain a technetium-reducing enzyme system, a stabilizing agent and an electron donor in a saline solution under anaerobic conditions. The mixture is substantially free of an inorganic technetium reducing agent and its reduction products. The resulting product is Tc of lower oxidation states, the form of which can be partially controlled by the stabilizing agent. It has been discovered that the microorganisms Shewanella alga, strain Bry and Shewanella putrifacians, strain CN-32 contain the necessary enzyme systems for technetium reduction and can form both mono nuclear and polynuclear reduced Tc species depending on the stabilizing agent.

Highlights: • SCF receptor c-Kit is functionally expressed in primary and transformed osteoblasts. • SCF protects primary and transformed osteoblasts from H{sub 2}O{sub 2}. • SCF activation of c-Kit in osteoblasts, required for its cyto-protective effects. • c-Kit mediates SCF-induced Akt activation in cultured osteoblasts. • Akt activation is required for SCF-regulated cyto-protective effects in osteoblasts. - Abstract: Osteoblasts regulate bone formation and remodeling, and are main target cells of oxidative stress in the progression of osteonecrosis. The stem cell factor (SCF)-c-Kit pathway plays important roles in the proliferation, differentiation and survival in a range of cell types, but little is known about its functions in osteoblasts. In this study, we found that c-Kit is functionally expressed in both osteoblastic-like MC3T3-E1 cells and primary murine osteoblasts. Its ligand SCF exerted significant cyto-protective effects against hydrogen peroxide (H{sub 2}O{sub 2}). SCF activated its receptor c-Kit in osteoblasts, which was required for its cyto-protective effects against H{sub 2}O{sub 2}. Pharmacological inhibition (by Imatinib and Dasatinib) or shRNA-mediated knockdown of c-Kit thus inhibited SCF-mediated osteoblast protection. Further investigations showed that protection by SCF against H{sub 2}O{sub 2} was mediated via activation of c-Kit-dependent Akt pathway. Inhibition of Akt activation, through pharmacological or genetic means, suppressed SCF-mediated anti-H{sub 2}O{sub 2} activity in osteoblasts. In summary, we have identified a new SCF-c-Kit-Akt physiologic pathway that protects osteoblasts from H{sub 2}O{sub 2}-induced damages, and might minimize the risk of osteonecrosis caused by oxidative stress.

  14. Evaluation of aftermarket LPG conversion kits in light-duty vehicle applications. Final report

    SciTech Connect (OSTI)

    Bass, E.A.

    1993-06-01

SwRI was contracted by NREL to evaluate three LPG conversion kits on a Chevrolet Lumina. The objective of the project was to measure the Federal Test Procedure (FTP) emissions and fuel economy of these kits, and compare their performance to gasoline-fueled operation and to each other. Varying LPG fuel blends allowed a preliminary look at the potential for fuel system disturbance. The project required kit installation and adjustment according to manufacturer`s instructions. A limited amount of trouble diagnosis was also performed on the fuel systems. A simultaneous contract from the Texas Railroad Commission, in cooperation with NREL, provided funds for additional testing with market fuels (HD5 propane and industry average gasoline) and hydrocarbon (HC) emissions speciation to determine the ozone-forming potential of LPG HC emissions. This report documents the procurement, installation, and testing of these LPG conversion kits.

  19. An evaluation of chemical screening test kits for lead in paint

    SciTech Connect (OSTI)

    Oglesby, L.S.

    1996-04-01

The Residential Lead-Based Paint Hazard Reduction Act (Title X) requires abatement and management of lead-based paint. The purpose of this study was to evaluate three chemical screening test kits using materials and methods from one study and subjecting the results to the statistical analysis of another. The three kits were used to predict the presence of lead in paint at ten weight concentrations from 0.04 to 3.97%. Paint was applied to four wood boards yielding a sample size of 40. Four boards were painted with lead-free paint and used as blanks. All of the boards were tested with the three test kits by an untrained individual having no knowledge of the actual lead content. Sensitivity, specificity, and false positive and negative rates were calculated for the test kit results. The manufactures` detection limits, the observed sensitivity ranged from 1.00 to 0.80, specificity ranged from 1.00 to 0.42, false positive ranged from 0 to 58%, and false negatives ranged from 0 to 20%. At the 0.5% Federal threshold level, the observed sensitivity ranged from 1.00 to 0.94, specificity ranged from 1.00 to 0.5, false positives ranged from 0 to 11.1%, and false negatives ranged from 0 to 20%. The observed false positive and false negative rates for all three kits were found to be significantly lower than those reported in a previous study. These results indicate that the kits perform very well at the Federal threshold, with two of the kits having false negative rates below 12.5% and false positive rates of 3.13%. These results indicate that these two kits would probably be acceptable screening tests for lead in paint.

  20. Studies on KIT-6 Supported Cobalt Catalyst for Fischer–Tropsch Synthesis

    2010-01-01

KIT-6 molecular sieve was used as a support to prepare cobalt catalyst for Fischer-Tropsch synthesis (FTS) using an incipient wetness impregnation method to produce cobalt loadings of 15 and 25 wt%. The catalysts were characterized by BET surface area, X-ray diffraction, scanning transmission election microscopy (STEM), extended X-ray absorption fine spectroscopy and X-ray absorption near edge spectroscopy. The catalytic properties for FTS were evaluated using a 1L CSTR reactor. XRD, pore size distribution, and STEM analysis indicate that the KIT-6 mesostructure remains stable during and after cobalt impregnation and tends to form smaller cobalt particles, probably located inside the mesopores. The mesoporous KIT-6 exhibited a slightly higher cobalt dispersion compared to amorphous SiO{sub 2} supported catalyst. With the higher Co loading (25 wt%) on KIT-6, partial structural collapse was observed after the FTS reaction. Compared to an amorphous SiO{sub 2} supported cobalt catalyst, KIT-6 supported cobalt catalyst displayed higher methane selectivity at a similar Co loading, likely due to diffusion effects.

    2012-04-01

Long-term reconstituting hematopoietic stem cells first arise from the aorta of the aorta-gonad-mesonephros (AGM) region in a mouse embryo. We have previously reported that in cultures of the dispersed AGM region, CD45{sup low}c-Kit{sup +} cells possess the ability to reconstitute multilineage hematopoietic cells, but investigations are needed to show that this is not a cultured artifact and to clarify when and how this population is present. Based on the expression profile of CD45 and c-Kit in freshly dissociated AGM cells from embryonic day 9.5 (E9.5) to E12.5 and aorta cells in the AGM from E13.5 to E15.5, we defined six cell populations (CD45{sup -}c-Kit{sup -}, CD45{sup -}c-Kit{sup low}, CD45{sup -}c-Kit{sup high}, CD45{sup low}c-Kit{sup high}, CD45{sup high}c-Kit{sup high}, and CD45{sup high}c-Kit{sup very} {sup low}). Among these six populations, CD45{sup low}c-Kit{sup high} cells were most able to form hematopoietic cell colonies, but their ability decreased after E11.5 and was undetectable at E13.5 and later. The CD45{sup low}c-Kit{sup high} cells showed multipotency in vitro. We demonstrated further enrichment of hematopoietic activity in the Hoechst dye-effluxing side population among the CD45{sup low}c-Kit{sup high} cells. Here, we determined that CD45{sup low}c-Kit{sup high} cells arise from the lateral plate mesoderm using embryonic stem cell-derived differentiation system. In conclusion, CD45{sup low}c-Kit{sup high} cells are the major hematopoietic cells of mouse AGM.

The U.S. Environmental Protection Agency (EPA) grants Certificates of Conformity for alternative fuel conversion systems and also offers other forms of premarket registration of conversion kits for use in vehicles more than two model years old. Use of alternative fuels such as ethanol, natural gas, and propane are encouraged by the Energy Policy Act of 1992. Several original equipment manufacturers (OEMs) produce emissions-certified vehicles capable of using alternative fuels, and several alternative fuel conversion system manufacturers produce EPA-approved conversion systems for a variety of alternative fuels and vehicle types. To date, only one manufacturer (Flex Fuel U.S.) has received EPA certifications for ethanol fuel (E85) conversion kits. This report details an independent evaluation of a vehicle with a legal installation of a Flex Fuel U.S. conversion kit. A 2006 Dodge Charger was baseline tested with ethanol-free certification gasoline (E0) and E20 (gasoline with 20 vol % ethanol), converted to flex-fuel operation via installation of a Flex Box Smart Kit from Flex Fuel U.S., and retested with E0, E20, E50, and E81. Test cycles included the Federal Test Procedure (FTP or city cycle), the highway fuel economy test (HFET), and the US06 test (aggressive driving test). Averaged test results show that the vehicle was emissions compliant on E0 in the OEM condition (before conversion) and compliant on all test fuels after conversion. Average nitrogen oxide (NOx) emissions exceeded the Tier 2/Bin 5 intermediate life NO{sub X} standard with E20 fuel in the OEM condition due to two of three test results exceeding this standard [note that E20 is not a legal fuel for non-flexible-fuel vehicles (non-FFVs)]. In addition, one E0 test result before conversion and one E20 test result after conversion exceeded the NOX standard, although the average result in these two cases was below the standard. Emissions of ethanol and acetaldehyde increased with increasing ethanol

We present a web toolkit STructure mapper and Online Coarse-graining Kit for setting up coarse-grained molecular simulations. The kit consists of two tools: structure mapping and Boltzmann inversion tools. The aim of the first tool is to define a molecular mapping from high, e.g. all-atom, to low, i.e. coarse-grained, resolution. Using a graphical user interface it generates input files, which are compatible with standard coarse-graining packages, e.g. VOTCA and DL_CGMAP. Our second tool generates effective potentials for coarse-grained simulations preserving the structural properties, e.g. radial distribution functions, of the underlying higher resolution model. The required distribution functions can be provided by any simulation package. Simulations are performed on a local machine and only the distributions are uploaded to the server. The applicability of the toolkit is validated by mapping atomistic pentane and polyalanine molecules to a coarse-grained representation. Effective potentials are derived for systems of TIP3P (transferable intermolecular potential 3 point) water molecules and salt solution. The presented coarse-graining web toolkit is available at http://stock.cmm.ki.si.

Managing technical references for projects that have long histories is hampered by the large collection of documents, each of which might contain discrete pieces of information relevant to the site conceptual model. A database application has been designed to improve the efficiency of retrieving technical information for a project. Although many databases are currently used for accessing analytical and geo-referenced data, applications designed specifically to manage technical reference material for projects are scarce. Retrieving site data from the array of available references becomes an increasingly inefficient use of labor. The electronic-Knowledge Information Tool (e-KIT) is designed as a project-level resource to access and communicate technical information. The e-KIT is a living tool that grows as new information becomes available, and its value to the project increases as the volume of site information increases. Having all references assembled in one location with complete reference citations and links to elements of the site conceptual model offers a way to enhance communication with outside groups. The published and unpublished references are incorporated into the e-KIT, while the compendium of references serves as a complete bibliography for the project. (authors)

Military-authorized activities included military training exercises, facilities maintenance, new construction, controlled burning, livestock grazing, and public-access hunting. Positive effects of the military included habitat preservation, preactivity surveys, and natural resources management practices designed to conserve kit foxes and their habitat. Perceived negative effects such as entrapment in dens, shootings during military exercises, and accidental poisoning were not observed. Foxes were observed in areas being used simultaneously by military units. Authorized activities were known to have caused the deaths of three of 52 radiocollared foxes recovered dead: one became entangled in concertina wire, one was believed shot by a hunter, and one was struck by a vehicle. Entanglement in communication wire may have contributed to the death of another radiocollared fox that was killed by a predator. Approximately 10% of kit fox dens encountered showed evidence of vehicle traffic, but denning sites did not appear to be a limiting factor for kit foxes.

  8. SU-E-T-350: Verification of Gating Performance of a New Elekta Gating Solution: Response Kit and Catalyst System

    SciTech Connect (OSTI)

    2014-06-01

Purpose: In this work, we have tested the performance of new respiratory gating solutions for Elekta linacs. These solutions include the Response gating and the C-RAD Catalyst surface mapping system.Verification measurements have been performed for a series of clinical cases. We also examined the beam on latency of the system and its impact on delivery efficiency. Methods: To verify the benefits of tighter gating windows, a Quasar Respiratory Motion Platform was used. Its vertical-motion plate acted as a respiration surrogate and was tracked by the Catalyst system to generate gating signals. A MatriXX ion-chamber array was mounted on its longitudinal-moving platform. Clinical plans are delivered to a stationary and moving Matrix array at 100%, 50% and 30% gating windows and gamma scores were calculated comparing moving delivery results to the stationary result. It is important to note that as one moves to tighter gating windows, the delivery efficiency will be impacted by the linac's beam-on latency. Using a specialized software package, we generated beam-on signals of lengths of 1000ms, 600ms, 450ms, 400ms, 350ms and 300ms. As the gating windows get tighter, one can expect to reach a point where the dose rate will fall to nearly zero, indicating that the gating window is close to beam-on latency. A clinically useful gating window needs to be significantly longer than the latency for the linac. Results: As expected, the use of tighter gating windows improved delivery accuracy. However, a lower limit of the gating window, largely defined by linac beam-on latency, exists at around 300ms. Conclusion: The Response gating kit, combined with the C-RAD Catalyst, provides an effective solution for respiratorygated treatment delivery. Careful patient selection, gating window design, even visual/audio coaching may be necessary to ensure both delivery quality and efficiency. This research project is funded by Elekta.

  13. Household energy use in urban Venezuela: Implications from surveys in Maracaibo, Valencia, Merida, and Barcelona-Puerto La Cruz

    SciTech Connect (OSTI)

    Figueroa, M.J.; Sathaye, J.

    1993-08-01

This report identifies the most important results of a comparative analysis of household commercial energy use in Venezuelan urban cities. The use of modern fuels is widespread among all cities. Cooking consumes the largest share of urban household energy use. The survey documents no use of biomass and a negligible use of kerosene for cooking. LPG, natural gas, and kerosene are the main fuels available. LPG is the fuel choice of low-income households in all cities except Maracaibo, where 40% of all households use natural gas. Electricity consumption in Venezuela`s urban households is remarkably high compared with the levels used in households in comparable Latin American countries and in households of industrialized nations which confront harsher climatic conditions and, therefore, use electricity for water and space heating. The penetration of appliances in Venezuela`s urban households is very high. The appliances available on the market are inefficient, and there are inefficient patterns of energy use among the population. Climate conditions and the urban built form all play important roles in determining the high level of energy consumption in Venezuelan urban households. It is important to acknowledge the opportunities for introducing energy efficiency and conservation in Venezuela`s residential sector, particularly given current economic and financial constraints, which may hamper the future provision of energy services.

The Institute of Gas Technology (IGT) was contracted by the National Renewable Energy Laboratory (NREL) to evaluate three compressed natural gas (CNG) conversion systems using a 1993 Chevrolet Lumina baseline vehicle. A fourth conversion system was added to the test matrix through funding support from Brooklyn Union. The objective of this project was to measure the Federal Test Procedure (FTP) emissions and fuel economy of the different conversion systems, and to compare the performance to gasoline-fueled operation and each other. Different natural gas compositions were selected to represent the 10th percentile, mean, and 90th percentile compositions distributed in the Continental United States. Testing with these different compositions demonstrated the systems` ability to accommodate the spectrum of gas found in the United States. Each compressed natural gas conversion system was installed and adjusted according to the manufacturer`s instructions. In addition to the FTP testing, an evaluation of the comparative installation times and derivability tests (based on AGA and CRC guidelines) were conducted on each system.

As contained in the Regulations for Implementing the Procedural Provisions of the National Environmental Policy Act, 40 CFR 1500--1508, the Council on Environmental Quality (CEQ) directs federal agencies to adopt their own procedures for implementing the Act. The US Department of Energy (DOE) and the US Department of Agriculture Forest Service (USFS) are two examples of federal agencies with dissimilar but functionally equivalent CX processes. The DOE and USFS were selected as subjects for this study because of their distinctly different missions and as a results of the author`s familiarity with the policies of both agencies. The objectives of this study are to: (1) describe the CX policies and processes of the two agencies, (2) identify the similarities and differences between the two processes, and (3) suggest ways for improving these processes. In performing this evaluation, the authors will identify the components of each agency`s CX process that clearly contributes qualitative information for the purpose of making environmental planning decisions. Drawing from the best elements of each process, the authors will provide some general recommendations that should enable the agencies to fulfill their various obligations to the CX process while concurrently performing early, thorough, and expeditious environmental reviews under NEPA.

    SciTech Connect (OSTI)

    1997-07-01

In FY96, Enterprise Advisory Services, Inc. (EASI) continued to support efforts to protect endangered species and cultural resources at the Naval Petroleum Reserves in California (NPRC). These efforts are conducted to ensure NPRC compliance with regulations regarding the protection of listed species and cultural resources on federal properties. Population monitoring activities were conducted for San Joaquin kit foxes, giant kangaroo rats, blunt-nosed leopard lizards, and Hoover`s wooly-star. Kit fox abundance and distribution was assessed by live-trapping over a 329-km{sup 2} area. Kit fox reproduction and mortality were assessed by radiocollaring and monitoring 22 adults and two pups. Reproductive success and litter size were determined through live-trapping and den observations. Rates and sources of kit fox mortality were assessed by recovering dead radiocollared kit foxes and conducting necropsies to determine cause of death. Abundance of coyotes and bobcats, which compete with kit foxes, was determined by conducting scent station surveys. Kit fox diet was assessed through analysis of fecal samples collected from live-trapped foxes. Abundance of potential prey for kit foxes was determined by conducting transect surveys for lagornorphs and live-trapping small mammals.

This paper presents an enterprise plan for introducing solar power in a rural market, while providing economic development, and hence the ability of the user to pay for the power source. This plan is based on a product called GEEP - a solar sewing machine conversion kit. This kit can be retrofit onto pedal sewing machines and marketed to village tailors in India, as part of a marketing program which includes increased demand for tailored products which will allow the tailors to be able to finance the conversion kits.
